Co-Culture of Lactobacillus bulgaricus With Streptococcus thermophilus and Bifidobacterium Impact the Metabolism and Flavor of Fermented Milk.

Incorporating Bifidobacterium into fermented milk alters the balance between Lactobacillus bulgaricus and Streptococcus thermophilus. We investigated the bacterial interaction and metabolism post-fermentation and during 21-day storage. Utilizing non-targeted metabolomics and electronic nose technology, we assessed impacts on product quality and flavor. Bifidobacterium significantly increased the viability of the other two species, with AI-2 levels rising in the mixed culture. Metabonomic analysis revealed distinct metabolic profiles, with Bifidobacterium-fermented milk showing enriched key met

Characterization of tetA-like gene encoding for a major facilitator superfamily efflux pump in Streptococcus thermophilus.

Efflux pumps are membrane proteins involved in the active extrusion of a wide range of structurally dissimilar substrates from cells. A multidrug efflux pump named TetA belonging to the major facilitator superfamily (MFS) of transporters was identified in the Streptococcus thermophilus DSM 20617(T) genome. The tetA-like gene was found in the genomes of a number of S. thermophilus strains sequenced to date and in Streptococcus macedonicus ACA-DC 198, suggesting a possible horizontal gene transfer event between these two Streptococcus species, which are both adapted to the milk environment. Identification of Streptococcus thermophilus Genes Specifically Expressed under Simulated Human Digestive Conditions Using R-IVET Technology.

Despite promising health effects, the probiotic status of Streptococcus thermophilus, a lactic acid bacterium widely used in dairy industry, requires further documentation of its physiological status during human gastrointestinal passage. This study aimed to apply recombinant-based in vivo technology (R-IVET) to identify genes triggered in a S. thermophilus LMD-9 reference strain under simulated digestive conditions. First, the R-IVET chromosomal cassette and plasmid genomic library were designed to positively select activated genes. Second, recombinant clones were introduced into complementar

Streptococcus thermophilus DM287 and DM294 as candidate oral probiotic strains with anti-biofilm activity against cariogenic pathogens.

Streptococcus mutans and Streptococcus sobrinus are key contributors to dental caries within a broader polymicrobial biofilm community, driving pathogenicity through biofilm and dental plaque formation rather than direct tissue invasion. Probiotic-based strategies that target colonization rather than viability represent a promising non-antibiotic approach to caries prevention. Streptococcus thermophilus is a food-grade organism with established GRAS status and a long history of safe human consumption, yet it remains largely unexplored as an oral care ingredient. In this study, two S. thermophi

Glycosylated Carbonated Apatite Particles for Macrophage-Targeted Delivery of CpG Oligodeoxynucleotides From Streptococcus thermophilus Enhance Immune Activation.

Immunostimulatory CpG oligodeoxynucleotides (CpG-ODNs) are potent activators of innate immunity with promising applications in human and veterinary medicine. We previously developed carbonate apatite-based oligonucleotide particles (ODNcaps) for oral delivery, which are efficiently taken up by macrophages in jejunal Peyer's patches. To enhance macrophage-specific targeting, we introduced glycan modifications to ODNcaps, exploiting carbohydrate-binding receptors on macrophages. In this study, glycosylated ODNcaps were synthesized through N-acetylglucosamine (GlcNAc) conjugation and evaluated fo

Physicochemical, microbiological, and sensory properties of low-lactose yogurt using Streptococcus thermophilus with high β-galactosidase activity.

BACKGROUND: Lactose maldigestion or intolerance affects a large number of individuals worldwide. If lactose is hydrolyzed by the β-galactosidase enzyme during the fermentation process, lactose-intolerant individuals can consume milk products without experiencing diarrhea, flatulence, or other symptoms. RESULTS AND CONCLUSION: We isolated and characterized Streptococcus thermophilus, which exhibits high β-galactosidase activity. This was then used as a starter culture with Lactobacillus delbrueckii subsp. bulgaricus in yogurt to determine the effects of different starter ratios and fermentatio

The impact of Streptococcus thermophilus IDCC 2201 on gut microbiota and its potential as a prophylactic agent for colorectal cancer.

Colorectal cancer (CRC) is one of the leading causes of cancer-related deaths. The gut microbiota is known to be closely associated with CRC development, interacting with each other and influencing host metabolism. Probiotic interventions have been applied to prevent CRC; however, the mechanisms underlying their effects on CRC remain unclear. In this study, we evaluated the effects of the probiotic strain Streptococcus thermophilus IDCC 2201 on its interactions with gut commensal bacteria and CRC cell viability. S. thermophilus inhibited the growth of most bacterial species comprising the huma

Comparison of volatile metabolic profiles in fermented milk of Streptococcus thermophilus during the postripening period at different incubation temperatures.

Streptococcus thermophilus has been extensively applied in fermented milk. This study used gas chromatography-ion mobility spectroscopy to determine and evaluate the volatile metabolites in raw milk, milk fermented at 37°C, and milk fermented at 42°C. Ten discriminatory volatile metabolites were identified at different incubation temperatures: acetone, 2-heptanone, 2-pentanone, 2-hexanone, butanal, hexanal, ethyl acetate, 3-methylbutanal, 3-methylbutanoic acid, and 2-methylpropanoic acid, indicating that fermentation temperature affected the spectrum of volatiles in milk fermented by different

Use of chemical mutagenesis for the isolation of food grade beta-galactosidase overproducing mutants of bifidobacteria, lactobacilli and Streptococcus thermophilus.

A classical chemical mutagenesis protocol was evaluated for increasing beta-galactosidase production by probiotic bacteria to improve their potential to treat symptoms of lactose malabsorption in humans. Two Bifidobacterium species (B. breve and B. longum) and one strain each of Lactobacillus delbrueckii ssp. bulgaricus and Streptococcus thermophilus were tested by a single exposure to two chemical mutagens, ethyl methanesulfonate (EMS) and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). To screen for beta-galactosidase (beta-gal) overproducing mutants, optimized EMS and MNNG mutant pots for each

Metabolomic and proteomic profiling reveals the formation mechanism of volatile flavor in egg whites during fermentation by Streptococcus thermophilus.

In this study, the flavor of egg whites was significantly improved by lactic acid fermentation, and the metabolic networks of metabolites, volatile compounds, and enzymes were established using gas chromatography-mass spectrometry, metabolomic, and proteomic. Results indicate that among ten types of common lactic acid bacteria, Streptococcus thermophilus endowed egg white with the most pleasant flavor through increasing aldehydes, ketones, alcohols, esters, terpenoids, and aromatic compounds. Amino acid catabolism was the predominant pathway for generating most aldehydes, alcohols, acids, and

CpG-ODN from Streptococcus thermophilus as a tool for inducing extrinsic apoptosis via caspase-8 activation in mouse splenocytes.

Synthetic oligodeoxynucleotides (ODNs) containing unmethylated CpG motifs activate Toll-like receptor 9 (TLR9), thereby triggering a diverse range of immunomodulatory responses. Although the immune-stimulating properties of CpG-ODNs have been extensively studied, their potential application as apoptosis-inducing tools remains underexplored, particularly for probiotic-derived sequences. In this study, we established a method for inducing extrinsic apoptosis in mouse splenocytes using MsST, a CpG-ODN identified from the lacZ gene of Streptococcus thermophilus ATCC19258. Interplay between different SHP/Rgg quorum sensing systems that control production of post-translationally modified peptides in Streptococcus thermophilus.

In Bacillota, the Rap-Rgg-NprR-PlcR-PrgX-AimR (RRNPPA) superfamily of regulators mediates quorum sensing via autoinducing peptides (AIPs). AIPs are secreted, processed and reimported before interacting with their cognate regulators, modulating the latter's activity. Within RRNPPA families, short hydrophobic peptides (SHPs) interact with Rgg transcriptional regulators. A widely used bacterium in the dairy industry, Streptococcus thermophilus, possesses an unusually high number of Rgg regulators (mean of 5 and max of 10 per genome). These regulators mainly control the expression of operons invol

Development of a simple IgE-independent anaphylactic model using buckwheat antigen and B-type CpG oligodeoxynucleotide from Streptococcus thermophilus.

We developed a severe anaphylactic model in mice using buckwheat antigen and B-type CpG-oligodeoxynucleotides (CpG-ODNs) from Streptococcus thermophilus genome. In typical systemic anaphylaxis models, animals are challenged with large quantity of antigens via an intravenous (i.v.) route. Here, we showed a simple anaphylactic shock after challenge via intraperitoneal (i.p.) route. The i.p. method is simpler than i.v. administration and has a lower risk for failure. To generate this anaphylactic model, 5-week-old female BALB/c mice were first i.p. sensitized with buckwheat antigen mixed with B-t

A novel GH66 family glycoside hydrolase pseudo-dextranase from Streptococcus thermophilus 17140 in catalyzing multi-step deglycosylation of ginsenoside Rb1.

Rare ginsenosides exhibit enhanced bioavailability and strong pharmacological activities such as anti-aging and immunomodulatory effects, making them valuable in health care food and medicine. We previously identified a Streptococcus thermophilus strain 17140 (St17140) capable of converting rare ginsenosides. In this study, we cloned and expressed a novel GH66 family glycoside hydrolase gene, pseudo-Dextranase from St17140 (PStDex). Despite lacking canonical catalytic residues for β-glucan hydrolysis, PStDex specifically cleaves the glycosidic bonds of ginsenoside Rb1 to produce rare ginsenosi
