Mechanical stress induces biotic and abiotic stress responses via a novel cis-element.

Plants are continuously exposed to a myriad of abiotic and biotic stresses. However, the molecular mechanisms by which these stress signals are perceived and transduced are poorly understood. To begin to identify primary stress signal transduction components, we have focused on genes that respond rapidly (within 5 min) to stress signals. Because it has been hypothesized that detection of physical stress is a mechanism common to mounting a response against a broad range of environmental stresses, we have utilized mechanical wounding as the stress stimulus and performed whole genome microarray a

Clinical evaluation of manual stress testing, stress ultrasound and 3D stress MRI in chronic mechanical ankle instability.

BACKGROUND: Chronic ankle instability (CAI) arises from the two etiological factors of functional (FAI) and mechanical ankle instability (MAI). To distinguish the contributions of the two etiologies, it is necessary to quantitively assess functional and mechanical deficits. Validated and reproducible assessment of mechanical instability remains a challenge in current research and practice. Physical examination, stress sonography and a novel 3D stress MRI have been used, while stress radiography has been called into question and arthrometry is limited to research purposes. The interaction of th

Collagen glycation-mediated mechanical stress aggravates ischemia-reperfusion injury.

Ischemia-reperfusion (IR) injury is a grave concern because of its substantial risk of organ failure in organ transplantation. Understanding its mechanism is essential for exploring potential management strategies to alleviate IR injury. In this study, we investigate the development of IR injury from a biomechanical perspective. Our results reveal a significant increase in tissue stiffness in IR- affected areas, driven by extracellular matrix crosslinking through collagen glycation. A stiffened matrix induces mechanical stretch, leading to excessive intracellular force transmission, which trig

Osmotic tablet coatings: Drying stress, mechanical properties and microstructure.

The coatings in osmotic tablets play a critical role in controlling the release of active pharmaceutical ingredient. Coatings are formed by spraying dilute polymer solution onto the tablet surface. During drying, the films develop shrinkage stress, which can cause cracking. The coatings are also subject to large tensile stress generated by osmotic pressure during the dissolution process, which may rupture the coating. Despite the role of tensile stress in causing fracture in osmotic tablet coatings, a rigorous quantification of the drying stress, mechanical properties and microstructure is mis

Mechanical Stress as a Pathomechanism of Plantar Acral Melanoma.

Sun exposure is widely recognized as a major etiologic factor in cutaneous melanoma. However, melanomas also occur in skin areas unexposed to the sun, such as the palmoplantar surface. Although the risk factors and mutational processes underlying acral melanoma remain poorly understood, these tumors frequently occur at sites of increased mechanical stress on the hand and foot, suggesting that mechanical stress may contribute to the development of plantar melanoma. Furthermore, mechanical forces regulate skin homeostasis, especially during skin development, regeneration, and wound healing. In p

Tendinopathy: The Interplay between Mechanical Stress, Inflammation, and Vascularity.

Tendinopathy is a long-lasting, debilitating disease that not only affects patients' individual lives, but also imposes a significant socioeconomic burden. Due to an insufficient understanding of the underlying pathomechanisms, treatment options remain limited. Although tendinopathy is recognized as a multifactorial disorder, known risk factors and predisposing conditions have largely been studied in isolation. In this review, it is aimed to examine the various pathomechanisms identified to date and synthesize them into a unified concept. Particular emphasis is placed on key drivers believed t

Molecular crowding suppresses mechanical stress-driven DNA strand separation.

Molecular crowding influences DNA mechanics and DNA-protein interactions and is ubiquitous in living cells. Quantifying the effects of molecular crowding on DNA supercoiling is essential to relating in vitro experiments to in vivo DNA supercoiling. We use single-molecule magnetic tweezers to study DNA supercoiling in the presence of dehydrating or crowding cosolutes. To study DNA supercoiling, we apply a stretching force of 0.8 pN to the DNA and then rotate one end of the DNA to induce supercoiling. In a 200 mM NaCl buffer without cosolutes, negatively supercoiled DNA absorbs some of the torti

FAK inhibition protects condylar cartilage under excessive mechanical stress.

OBJECTIVES: Excessive mechanical stress is assumed to be a major cause of temporomandibular joint (TMJ) osteoarthritis (OA). +Focal adhesion kinase (FAK) is a cytoplasmic non-receptor tyrosine kinase involved in a variety of signaling pathways. Little has been reported on the function of FAK in TMJ-OA. In the present study, we investigated the effect of FAK inhibition on TMJ cartilage under excessive mechanical loading stress. MATERIALS AND METHODS: Articular cartilage explants were harvested from the TMJ of rats and subjected to mechanical loading in the presence of an FAK inhibitor in organ

Heat stress protects against mechanical ventilation-induced diaphragmatic atrophy.

Mechanical ventilation (MV) is a life-saving intervention in patients who are incapable of maintaining adequate pulmonary gas exchange due to respiratory failure or other disorders. However, prolonged MV is associated with the development of respiratory muscle weakness. We hypothesized that a single exposure to whole body heat stress would increase diaphragm expression of heat shock protein 72 (HSP72) and that this treatment would protect against MV-induced diaphragmatic atrophy. Adult male Wistar rats (n = 38) were randomly assigned to one of four groups: an acutely anesthetized control group

The role of mechanical wall stress and wall shear stress on coronary artery disease.

BACKGROUND AND OBJECTIVE: Although the association of wall shear stress (WSS) with coronary artery disease has been well studied, that of mechanical wall stress (MWS) is mainly overlooked. In this study, we performed in-silico artery-specific modeling to investigate the involvement of both MWS and WSS in coronary artery disease. METHODS: Fifteen coronary arteries from five adult familial hypercholesterolemic pigs were imaged by coronary computed tomography angiography, intravascular ultrasound, and optical coherence tomography at three time points (3, 9, and 12 months). Local WSS and MWS in 3

A mechanical acupuncture instrument mitigates the endoplasmic reticulum stress and oxidative stress of ovariectomized rats.

BACKGROUND: Acupuncture has become a common complementary and alternative treatment approach for anxiety and depression. However, there is little research on the detailed mechanism of acupuncture therapy relieving depression. Previously, 17β-estradiol (E2) was shown to prevent oxidative stress and endoplasmic reticulum (ER) stress in ovariectomized (OVX) rats. This study investigated whether stimulation of Sanyinjiao (SP6) using a mechanical acupuncture instrument can alleviate depression-like behavior caused by estrogen deficiency in OVX rats. Furthermore, we found that acupuncture reduced ER

Mechanical models and measurement methods of solid stress in tumors.

In addition to genetic mutations, biomechanical factors also affect the structures and functions of the tumors during tumor growth, including solid stress, interstitial fluid pressure, stiffness, and microarchitecture. Solid stress affects tumors by compressing cancer and stromal cells and deforming blood and lymphatic vessels which reduce supply of oxygen, nutrients and drug delivery, making resistant to treatment. Researchers simulate the stress by creating mechanical models both in vitro and in vivo. Cell models in vitro are divided into two dimensions (2D) and three dimensions (3D). 2D mod

Mechanical stress can regulate temporomandibular joint cavitation via signalling pathways.

The temporomandibular joint (TMJ), composed of temporal fossa, mandibular condyle and a fibrocartilage disc with upper and lower cavities, is the biggest synovial joint and biomechanical hinge of the craniomaxillofacial musculoskeletal system. The initial events that give rise to TMJ cavities across diverse species are not fully understood. Most studies focus on the pivotal role of molecules such as Indian hedgehog (Ihh) and hyaluronic acid (HA) in TMJ cavitation. Although biologists have observed that mechanical stress plays an irreplaceable role in the development of biological tissues and o

Mechanical stress-induced hemolysis of bovine blood is donor-dependent.

INTRODUCTION: In vitro hemolysis testing is an essential method for assessing the hemolytic potential of blood pumps, but has poor reproducibility. Further investigations are needed to determine the sources and extent of variability and to find a practical way to reduce the variation. METHODS: A small volume blood circulating loop driven by a Centrimag pump was established to provide relatively higher hemolysis readouts within a short run time and to be able to sequentially perform multiple repeated hemolysis tests in a working day. RESULTS: The repeatability with this system was demonstrate

Assessing mechanical vibration-altered wall shear stress in digital arteries.

The aim of this study is to implement and validate a method for assessing acute vibration-altered Wall Shear Stress (WSS) in the proper volar digital artery of the non-exposed left forefinger when subjecting the right hand to mechanical vibration. These changes of WSS may be involved in Vibration White Finger. Hence, an experimental device was set-up to link a vibration shaker and an ultra-high frequency ultrasound scanner. The Womersley-based WSS was computed by picking up the maximum velocity from pulse Wave Doppler measurements and extracting the artery diameter from B-mode images through a

Palmdelphin Regulates Nuclear Resilience to Mechanical Stress in the Endothelium.

BACKGROUND: PALMD (palmdelphin) belongs to the family of paralemmin proteins implicated in cytoskeletal regulation. Single nucleotide polymorphisms in the PALMD locus that result in reduced expression are strong risk factors for development of calcific aortic valve stenosis and predict severity of the disease. METHODS: Immunodetection and public database screening showed dominant expression of PALMD in endothelial cells (ECs) in brain and cardiovascular tissues including aortic valves. Mass spectrometry, coimmunoprecipitation, and immunofluorescent staining allowed identification of PALMD par

Inhibition of mechanical stress-induced hypertrophic scar inflammation by emodin.

At least 50% of hypertrophic scarring (HS) is characterized by inflammation, for which there is currently no effective treatment available. Emodin is a major component of the widely used Chinese herb, rhubarb, which has been used to treat inflammation in several types of disease. However, few studies have investigated the efficacy of emodin in the treatment of HS. In the present study, a mouse model with mechanical stress‑induced HS was used to investigate the effects of emodin (20, 40, 80, or 120 mg/ml) on HS, and to determine the potential underlying mechanisms. Treatment with emodin signifi
