Evidence for neuronal and structural changes in submucous ganglia of patients with functional dyspepsia.

OBJECTIVES: An intact and well-functioning enteric nervous system is necessary to efficiently organize gut function. Functional gastrointestinal disorders are pathological entities in which gut function is impaired without a clearly established pathophysiology. On the basis of the relative ease with which intestinal biopsies can be obtained, and taking advantage of a recently developed optical recording technique, we evaluated whether functional neuronal defects exist in enteric nerves of patients with functional dyspepsia (FD). METHODS: The submucous plexus isolated from duodenal biopsies ta

Nitric oxide synthase immunoreactivity in the enteric nervous system of the developing human digestive tract.

We have investigated indirectly the presence of nitric oxide in the enteric nervous system of the digestive tract of human fetuses and newborns by nitric oxide synthase (NOS) immunocytochemistry and nicotinamide adenine dinucleotide phosphate diaphorase (NADPHd) histochemistry. In the stomach, NOS immunoactivity was confined to the myenteric plexus and nerve fibres in the outer smooth musculature; few immunoreactive nerve cell bodies were found in ganglia of the outer submucous plexus. In the pyloric region, a few nitrergic perikarya were seen in the inner submucous plexus and some immunoreact

Neuronal nitric oxide synthase activity is increased during granulomatous inflammation in the colon and caecum of pigs infected with Schistosoma japonicum.

Neuronal nitric oxide is a non-adrenergic non-cholinergic neurotransmitter in the enteric nervous system and plays a role in a variety of enteropathies including Crohn's and Chagas' diseases, ulcerative colitis, diabetes, atrophy and hypertrophy. The content of neuronal nitric oxide synthase (nNOS) in the colon and the caecum from pigs infected with Schistosoma japonicum was studied using immunohistochemical and histochemical staining for nNOS and nicotinamide adenine dinucleotide phosphate diaphorase (NADPH-diaphorase), respectively. In the infected pigs, lightly, moderately and less severely

Histochemical study of the distribution of nicotinamide adenine dinucleotide phosphate diaphorase (NADPHd)-positive neurons in the chicken caecum.

The distribution of nicotinamide adenine dinucleotide phosphate diaphorase (NADPHd)-positive neurones was investigated in the chicken caecum. Double staining combined NADPHd histochemistry with immunohistochemistry for neural nitric oxide synthase (nNOS) indicated that NADPHd-positive neurones also showed immunoreactivity for nNOS. NADPHd-positive nerve cell bodies were observed in both the myenteric and the submucous plexuses. Nerve fibres showing enzyme activity were mainly distributed in the circular muscle layer, but only a few fibres in the mucosal layer. Fine nerve fibres showing NADPHd

Submucosal hypoganglionosis causing chronic idiopathic intestinal pseudo-obstruction.

A 39-year-old woman presented with recurrent symptoms suggestive of intestinal obstruction. She was put on total parenteral nutrition (TPN) and consequently developed sepsis and endocarditis. TPN was stopped and a venting enterostomy was performed. Biopsies of mucosa and submucosa were taken at surgery; immunohistochemistry for neuronal proteins, protein gene product 9.5 (PGP 9.5) and the glial S-100-protein was done. Many enlarged nerve fiber strands were found in the submucosa. Few small ganglia containing a small number of nerve cells could be observed, suggesting hypoganglionosis. This pat

Morphometric determination of the methodological criteria for the diagnosis of intestinal neuronal dysplasia (IND B).

Intestinal neuronal dysplasia of the submucous plexus (IND B) is an indicator of a developmental abnormality of vegetative gut innervation. It is the mildest form of an inborn error of intestinal innervation. The diagnosis of IND B does not result in a functional conclusion or clinical recommendation but is often accompanied by oligoneuronal hypoganglionosis of the myenteric plexus or an aganglionosis of the rectum. The aim of this study was to demonstrate by morphometric means a way in which the diagnosis of IND B could be made much more reliable. In 20 control subjects, 40 IND B cases and 10

Cholinergic innervation of the gastric wall of the cat.

The inbuilt intrinsic cholinergic nervous apparatus of the gastric wall of the cat was studied by using two thiocholine methods for mapping the acetylcholinesterase-positive nerves and nerve cells. A rich distribution of acetylcholinesterase-positive nerves was observed in all layers of the gastric wall, except the superficial half of the lamina propria (with the epithelium), which was completely devoid of acetylcholinesterase activity, and the submucosa, in which a scarce distribution of large nerve fascicles and nerve trunks was observed. Acetylcholinesterase-positive ganglia were observed b

Distribution of galanin-immunoreactive nerves in the canine gastrointestinal tract.

The distribution of nerves containing galanin immunoreactivity (GAL-IR) in the canine gastrointestinal tract was determined by immunohistochemistry. GAL-IR was observed in nerve cell bodies and nerve fibers in all layers of the lower esophagus, gastric antrum, pylorus, ileum, and colon, and in the sphincters of lower esophagus and pylorus. There were numerous GAL-IR-containing nerve cell bodies in the myenteric plexus of gastric antrum. GAL-IR nerve somata were also common in the myenteric plexus of lower esophagus, ileum, pylorus, and colon, and in the submucous plexus of stomach, ileum, and

Nitric oxide synthase-containing neurons in the pig large intestine: topography, morphology, and viscerofugal projections.

The distribution of neurons that are capable of synthesizing nitric oxide (NO) has been demonstrated in the porcine large intestine by means of NO synthase (NOS) immunocytochemistry and nicotinamide adenine dinucleotide phosphate diaphorase (NADPHd) histochemistry. An overall colocalization of NOS immunoreactivity and NADPHd staining was observed. Nitrergic neurons were abundant in the myenteric and outer submucous plexus of the caecum, colon, and rectum. Only a few nitrergic perikarya were seen in the inner submucous plexus of the colon and caecum, whereas a substantially larger number was ob

Improving morphological and functional properties of enteric neuronal networks in vitro using a novel upside-down culture approach.

The enteric nervous system (ENS) comprises millions of neurons and glia embedded in the wall of the gastrointestinal tract. It not only controls important functions of the gut but also interacts with the immune system, gut microbiota, and the gut-brain axis, thereby playing a key role in the health and disease of the whole organism. Any disturbance of this intricate system is mirrored in an alteration of electrical functionality, making electrophysiological methods important tools for investigating ENS-related disorders. Microelectrode arrays (MEAs) provide an appropriate noninvasive approach

Nitric oxide synthase in neurons of the gastrointestinal tract of an avian species, Coturnix coturnix.

The distribution of reduced nicotinamide adenine dinucleotide phosphate (NADPH) diaphorase staining and its colocalisation with nitric oxide synthase immunoreactivity was examined in neurons of the quail gastrointestinal tract. Immunoreactivity and enzyme activity were found in identical populations of neurons. Nitric oxide synthase activity was present in 30-40% of myenteric nerve cells in each region. Numerous reactive cells were also in the submucous plexus and many positive nerve fibres innervated the muscle. In the small intestine, some of the fibres innervating the circular muscle formed

Annotated translation of Georg Meissner's first description of the submucosal plexus.

BACKGROUND: The discovery and detailed descriptions of the enteric nervous system dates back only ˂200 years. The 19th century was a golden age of histological, morphological, and physiological breakthroughs propelled by technological advances in microscopy, electricity, and scientific methodology. As a matter of fact, German-speaking scientists were highly successful during this period as can still be appreciated by the sheer number of German eponyms in anatomy. Therefore, the main language in scientific literature of this field was German at the time, thus, limiting the accessibility to the

Intestinal Region-Specific and Layer-Dependent Induction of TNFα in Rats with Streptozotocin-Induced Diabetes and after Insulin Replacement.

Tumour necrosis factor alpha (TNFα) is essential in neuroinflammatory modulation. Therefore, the goal of this study is to reveal the effects of chronic hyperglycaemia and insulin treatment on TNFα expression in different gut segments and intestinal wall layers. TNFα expression was mapped by fluorescent immunohistochemistry and quantitative immunogold electron microscopy in myenteric ganglia of duodenum, ileum and colon. Tissue TNFα levels were measured by enzyme-linked immunosorbent assays in muscle/myenteric plexus-containing (MUSCLE-MP) and mucosa/submucosa/submucous plexus-containing (MUC-S

How big is the little brain in the gut? Neuronal numbers in the enteric nervous system of mice, Guinea pig, and human.

BACKGROUND: Despite numerous studies on the enteric nervous system (ENS), we lack fundamental knowledge on neuronal densities or total neuron numbers in different species. There are more anecdotal than actual figures on nerve counts. METHODS: We used standardized preparation techniques and immunohistochemistry with validated panneuronal markers (human or mouse anti-HuD/C) to determine neuronal densities in specimen from the entire gastrointestinal tract of mice, guinea pig, and humans. In parallel, we measured the dimensions of the gastrointestinal regions in mouse and guinea pig. For humans,

Changes in the Enteric Neurons Containing Selected Active Substances in the Porcine Descending Colon after the Administration of Bisphenol A (BPA).

Bisphenol A (BPA) is an endocrine disruptor widely distributed in the environment due to its common use in the plastics industry. It is known that it has a strong negative effect on human and animal organisms, but a lot of aspects of this impact are still unexplored. This includes the impact of BPA on the enteric nervous system (ENS) in the large intestine. Therefore, the aim of the study was to investigate the influence of various doses of BPA on the neurons located in the descending colon of the domestic pig, which due to similarities in the organization of intestinal innervation to the huma

Nitric oxide synthase in the enteric nervous system of the guinea-pig: a quantitative description.

The distribution and abundance of nitric oxide synthase (NOS)-containing neurons and their terminals in the gastrointestinal tract of the guinea-pig were examined in detail using NADPH diaphorase histochemistry and NOS immunohistochemistry. NOS-containing cell bodies were found in the myenteric plexus throughout the gastrointestinal tract and in the submucous plexus of the stomach, colon and rectum. NOS-containing neurons comprised between 12% (in the duodenum) and 54% (in the esophagus) of total myenteric neurons. In the ileum, NOS neurons represented 19% of total myenteric neurons. Most of t

Submucosal enteric neurons of the cavine distal colon are sensitive to hypoosmolar stimuli.

KEY POINTS: Neurons of the enteric submucous plexus are challenged by osmolar fluctuations during digestion and absorption of nutrients. Central neurons are very sensitive to changes in osmolality but knowledge on that issue related to enteric neurons is sparse. The present study focuses on investigation of osmosensitivity of submucosal neurons including potential molecular mediating mechanisms. Results show that submucosal neurons respond to hypoosmolar stimuli with increased activity which is partially mediated by the transient receptor potential vanilloid 4 channel. We provided important in

Intestinal neuronal dysplasia.

Intestinal neuronal dysplasia (IND) is a disorder of the enteric nervous system that was first described by Meier-Ruge in 1971. IND may be associated with Hirschsprung's disease or may occur as an isolated disorder. The incidence of isolated IND varies from 0.3% to 40% of suction rectal biopsies. The uncertainty regarding the incidence of IND has resulted from considerable confusion concerning essential diagnostic criteria. The diagnostic difficulties reflect the wide variability in the literature in terms of diagnostic criteria, biopsy procedures, staining techniques, and patient age. The rec
