The impact of natural menstrual cycle and oral contraceptive pill phase on substrate oxidation during rest and acute submaximal aerobic exercise.

Previous research has identified sex differences in substrate oxidation during submaximal aerobic exercise including a lower respiratory exchange ratio (RER) in females compared with males. These differences may be related to differences in sex hormones. Our purpose was to examine the impact of the natural menstrual cycle (NAT) and second- and third-generation oral contraceptive pill (OCP2 and OCP3) cycle phases on substrate oxidation during rest and submaximal aerobic exercise. Fifty female participants (18 NAT, 17 OCP2, and 15 OCP3) performed two experimental trials that coincided with the l

Impact of aerobic fitness status, menstrual cycle phase, and oral contraceptive use on exercise substrate oxidation and metabolic flexibility in females.

The influence of menstrual cycle phase and fitness status on metabolism during high-intensity interval exercise (HIIE) was assessed. Twenty-five females (24.4 (3.6) years) were categorized by normal menstrual cycle (n = 14) vs. oral contraceptive (OC) use (n = 11) and by aerobic fitness, high-fitness females (HFF; n = 13) vs. low-fitness females (LFF; n = 12). HIIE was four sets of four repetitions with a 3 min rest between intervals on a cycle ergometer at a power output halfway between the ventilatory threshold and V̇O2peak and performed during follicular (FOL: days 2-7 or inactive pills) an

The Insulin Receptor Adaptor IRS2 is an APC/C Substrate That Promotes Cell Cycle Protein Expression and a Robust Spindle Assembly Checkpoint.

Insulin receptor substrate 2 (IRS2) is an essential adaptor that mediates signaling downstream of the insulin receptor and other receptor tyrosine kinases. Transduction through IRS2-dependent pathways is important for coordinating metabolic homeostasis, and dysregulation of IRS2 causes systemic insulin signaling defects. Despite the importance of maintaining proper IRS2 abundance, little is known about what factors mediate its protein stability. We conducted an unbiased proteomic screen to uncover novel substrates of the Anaphase Promoting Complex/Cyclosome (APC/C), a ubiquitin ligase that con

Identification of active site residues involved in metal cofactor binding and stereospecific substrate recognition in Mammalian tyrosinase. Implications to the catalytic cycle.

Tyrosinase (Tyr) and tyrosinase-related proteins (Tyrps) 1 and 2 are the enzymes responsible for mammalian melanogenesis. They display high similarity but different substrate and reaction specificities. Loss-of-function mutations lead to several forms of albinism or other pigmentation disorders. They share two conserved metal binding sites (CuA and CuB) which, in Tyr, bind copper. To define some structural determinants for these differences, we mutated Tyr at selected residues on the basis of (i) conservation of the original residues in most tyrosinases, (ii) their nonconservative substitution

Pacing cycle length-dependent electrophysiologic changes in left atrium: Poor validity of using low-voltage area and slow conduction area under specific pacing cycle length as absolute substrates of atrial fibrillation.

BACKGROUND: Pacing cycle length (PCL)-dependent changes in left atrial (LA) electrophysiologic properties have not been fully elucidated. OBJECTIVE: We aimed to elucidate these changes using a high-resolution mapping system. METHODS: Forty-eight patients underwent atrial fibrillation ablation with RHYTHMIA HDx. Paired LA maps under a baseline PCL (600 ms) and rapid PCL (300 ms) were acquired after pulmonary vein isolation under right atrial appendage pacing. The PCL-dependent change in the low-voltage area (LVA; area with <0.5 mV bipolar voltage), LA activation time (interval from first LA a

Cycle length statistics during human atrial fibrillation reveal refractory properties of the underlying substrate: a combined in silico and clinical test of concept study.

AIMS: The treatment of atrial fibrillation beyond pulmonary vein isolation has remained an unsolved challenge. Targeting regions identified by different substrate mapping approaches for ablation resulted in ambiguous outcomes. With the effective refractory period being a fundamental prerequisite for the maintenance of fibrillatory conduction, this study aims at estimating the effective refractory period with clinically available measurements. METHODS AND RESULTS: A set of 240 simulations in a spherical model of the left atrium with varying model initialization, combination of cellular refract

MOF Derivatives Confined Within Self-Supporting Bamboo Substrates with Hierarchical Porous Architectures for Long-Term Cycling Stability in Zinc-Air Batteries.

The relatively poor cycle stability of zinc-air batteries (ZABs) hinders their widespread application, while self-supporting electrode materials have shown great potential in enhancing the cycling stability of ZABs. To construct a self-supporting electrode, bamboo was employed as a sustainable precursor, and a two-step pyrolysis strategy was implemented to integrate ZIF-67-derived catalysts onto a hierarchically porous carbon framework, yielding the composite material Co-N@CB. Benefiting from its structural and electronic advantages, Co-N@CB exhibits outstanding electrocatalytic performance. T

Differential expression of genes involved with apoptosis, cell cycle, connective tissue proteins, fuel substrate utilization, inflammation and mitochondrial biogenesis in copper-deficient rat hearts: implication of a role for Nfkappab1.

We hypothesized that the increase in mitochondrial proliferation in hearts from copper-deficient rats is due to an increase in expression of the transcriptional factor peroxisomal-like proliferating related coactivator 1alpha (Ppargc1a), which regulates transcriptional activity for many of the genes that encode for mitochondrial proteins. In addition to several transcriptional factors implicated in mitochondrial biogenesis, we also looked at a number of genes involved in cell cycle regulation and fuel substrate utilization. Long-Evans rats were placed on either a copper-adequate (n=4) or coppe

Substrate Specificity of Biofilms Proximate to Historic Shipwrecks.

The number of built structures on the seabed, such as shipwrecks, energy platforms, and pipelines, is increasing in coastal and offshore regions. These structures, typically composed of steel or wood, are substrates for microbial attachment and biofilm formation. The success of biofilm growth depends on substrate characteristics and local environmental conditions, though it is unclear which feature is dominant in shaping biofilm microbiomes. The goal of this study was to understand the substrate- and site-specific impacts of built structures on short-term biofilm composition and functional pot

Standing or Very Low-Intensity Cycling as Sedentary Breaks: Does Physical Activity Level Matter?

BACKGROUND: Active breaks and very low- to low-intensity exercises such as walking or cycling at an active desk have been shown to significantly counteract the negative effect of prolonged sedentary behaviors. The objective was to investigate the effect of physical activity level (PAL) on changes in energy expenditure (EE), heart rate, and substrate oxidation from sit-to-stand and sit-to-light cycling. METHODS: Fifty healthy young males and females (age: 23.9 [3.9] y, body mass index: 22.9 [2.3] kg/m2) were submitted to a fixed 1 hour session of different posture allocations: 15-minute sittin

On modeling and nanoanalysis of caries-affected dentin surfaces restored with Zn-containing amalgam and in vitro oral function.

The aim of this research was to assess the influence of mechanical loading on the ability of Zn-free versus Zn-containing amalgams to promote remineralization at the dentin interface. Sound and caries-affected dentin surfaces (CAD) were restored using Zn-free or Zn-containing dental amalgams. Midcoronal dentin surfaces were studied by (1) atomic force microscopy analysis (including plot and phase imaging, nanoindentation test [modulus of Young (Ei), nanoroughness measurements, and fibril diameter assessment], (2) Raman spectroscopy/cluster analysis, (3) x-ray diffraction, (4) field emission el

Artificial gravity during prolonged bed rest preserves resting metabolic rate but not muscle function: Evidence from the BRACE study.

Prolonged head-down tilt bed rest induces metabolic and neuromuscular deconditioning. This study examined whether supine cycling exercise, with/without artificial gravity (AG), preserves resting metabolic rate (RMR) during 60 days of bed rest, and whether RMR responses align with integrated multivariate physiological adaptations. Twenty-four healthy men were randomly assigned to control (C), cycling (EX) or cycling plus AG (EX-AG). RMR, substrate oxidation, body composition (dual-energy X-ray absorptiometry/magnetic resonance imaging), muscle function and maximal cycling power were assessed be

Structured Sanda training as an innovative exercise intervention for young men with obesity: metabolic load distribution and adaptive responses compared with moderate-intensity continuous cycling.

BACKGROUND: Innovative and engaging exercise interventions are needed to improve cardiometabolic health and exercise adherence in young adults with obesity. Sanda, a Chinese combat sport characterized by footwork, striking combinations, defensive movements, and attack-recovery transitions, may provide a practical intermittent exercise modality when delivered in a structured and low-contact format. However, its metabolic-load characteristics and adaptive effects in individuals with obesity remain insufficiently understood. OBJECTIVE: This study compared the effects of 12 weeks of structured lo

Grain-Optimized Copper Current Collectors for Highly Stable Anode-Free Sodium Batteries.

Improving cycling stability and capacity retention is critical for the development of high-energy-density and low-cost anode-free sodium batteries (AFSBs). However, unstable Na plating/stripping and uncontrolled solid electrolyte interphase (SEI) evolution still hinder their practical application. In this work, we investigate the impact of grain-boundary density in the Cu substrate on the performance of AFSBs. The results reveal that higher grain-boundary density Cu exhibits a stronger affinity for sodium and can lower the sodium nucleation energy barriers, facilitating the formation of highly

Bioresource amendment promotes early mineralization and nitrification recovery in ammonium-rich rare earth tailings.

Rare earth tailings soil retains high residual NH4+-N after in-situ leaching with ammonium sulfate, creating coupled risks of nitrogen loss and poor revegetation. The substrate and microbial pathways that reopen ammonia oxidation and mediate nitrification restart in this ammonium rich rare earth tailings soil remain unclear. To address this gap, a microcosm incubation over time compared an unamended soil, MgO, and manure, integrating nitrogen transformations, net mineralization, enzymes, qPCR, and shotgun metagenomics. Under OM, extractable NH4+-N declined by 88.3 % from its initial value, whi

A stem cell-based assay platform demonstrates alpha-synuclein dependent synaptic dysfunction in patient-derived cortical neurons.

Alpha-synuclein (αS)-rich Lewy bodies and neurites in the cerebral cortex correlate with the presence of dementia in Parkinson disease (PD) and Dementia with Lewy bodies (DLB), but whether αS influences synaptic vesicle dynamics in human cortical neurons is unknown. Using a new iPSC-based assay platform for measuring synaptic vesicle cycling, we found that in human cortical glutamatergic neurons, increased αS from either transgenic expression or triplication of the endogenous locus in patient-derived neurons reduced synaptic vesicle cycling under both stimulated and spontaneous conditions. Exogenous Carbohydrate Oxidation Is Higher When Consuming Glucose + Fructose Compared with Glucose Alone during Endurance Exercise Regardless of Energy Status.

BACKGROUND: When glucose (GLU) is consumed during endurance exercise in an energy deficit state (DEF), exogenous carbohydrate oxidation is lower than in energy balance (BAL). Whether consuming multiple transportable carbohydrates, such as glucose + fructose (GLU+FRU), better maintains exogenous carbohydrate oxidation during DEF is unclear. OBJECTIVES: This study assessed the effects of consuming GLU + FRU compared with GLU on substrate oxidation and physical performance during BAL and DEF. METHODS: Eleven participants (sex: n = 2 females, 9 males) completed this double-blind randomized cross

Brain O-GlcNAcylation in Neurodegenerative Diseases: Context-Dependent Mechanisms and Precision Therapeutic Translation.

O-linked β-N-acetylglucosamine modification (O-GlcNAcylation) is a dynamic, nutrient-sensitive post-translational modification that couples hexosamine biosynthesis pathway flux to protein function in neurons and glia. This reversible cycling, catalyzed by O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA), integrates glucose, glutamine, acetyl-CoA, and nucleotide metabolism with synaptic activity, mitochondrial adaptation, transcriptional regulation, proteostasis, and neuroimmune signaling. Dysregulated O-GlcNAc cycling has been implicated in major neurodegenerative diseases, including Alzheimer

The effect of β-alanine supplementation on high intensity cycling capacity in normoxia and hypoxia.

The availability of dietary beta-alanine (BA) is the limiting factor in carnosine synthesis within human muscle due to its low intramuscular concentration and substrate affinity. Carnosine can accept hydrogen ions (H+), making it an important intramuscular buffer against exercise-induced acidosis. Metabolite accumulation rate increases when exercising in hypoxic conditions, thus an increased carnosine concentration could attenuate H+ build-up when exercising in hypoxic conditions. This study examined the effects of BA supplementation on high intensity cycling capacity in normoxia and hypoxia.

Effects of Organic Substrate Concentrations on Biological Mn Oxidation and Reduction under Aerobic Conditions.

Biological Mn oxidation mediated by manganese-oxidizing bacteria (MnOB) plays a central role in Mn cycling in natural and engineered systems. Although most MnOB are heterotrophic and frequently associated with oligotrophic environments, the quantitative relationship between organic substrate concentrations and Mn oxidation activity remains unclear. We herein systematically evaluated the effects of organic substrate concentrations on MnOB activity using a down-flow hanging sponge (DHS) reactor (0-1,000 mg COD L-1), batch experiments with an enriched biomass, and the pure MnOB strain Pseudomonas
