Novel insights into microbial DMSP/DMS cycling: from surface to deep ocean.

The microbial cycling of dimethylsulfoniopropionate (DMSP) and dimethyl sulfide (DMS) constitutes a crucial component of the global sulfur cycle, influencing climate regulation through the release of climate-active gases. Since DMSP production has historically been attributed to planktonic algae, research on its cycling has consequently centered on the surface ocean. However, the continuous discovery of bacterial DMSP synthesis genes (dsyB, mmtN, and dsyGD) has confirmed that the DMSP/DMS cycling process may also exist in the aphotic deep-sea environment. Further supporting this, the recently

High zinc exposure leads to reduced dimethylsulfoniopropionate (DMSP) levels in both the host and endosymbionts of the reef-building coral Acropora aspera.

Dimethylsulfoniopropionate (DMSP) is a biogenic compound that could be involved in metal detoxification in both the host and endosymbionts of symbiotic corals. Acropora aspera, a common reef-building coral of the Great Barrier Reef, was exposed to zinc doses from 10 to 1000μg/L over 96h, with zinc being a low-toxic trace metal commonly used in the shipping industry. Over time, significantly lower DMSP concentrations relative to the control were found in both the host and symbionts in the highest zinc treatment where zinc uptake by both partners of the symbiosis was the highest. This clearly in

Chemoselective addition of isocyanides to N-tert-butanesulfinimines.

The reaction of isocyanides with N-tert-butanesulfinimines shows remarkable chemoselectivity. β-Sulfinylamino isocyanides are formed exclusively with aromatic sulfinimines, while α-sulfeneimino acetamides result when using aliphatic derivatives. A mechanism is suggested for the latter transformation, together with an explanation for the observed selectivity. Finally, a scope study is presented for this remarkably chemoselective reaction.

Rhodobacteraceae methanethiol oxidases catalyze methanethiol degradation to produce sulfane sulfur other than hydrogen sulfide.

UNLABELLED: Methanethiol (MT) is a sulfur-containing compound produced during dimethylsulfoniopropionate (DMSP) degradation by marine bacteria. The C-S bond of MT can be cleaved by methanethiol oxidases (MTOs) to release a sulfur atom. However, the cleaving process remains unclear, and the species of sulfur product is uncertain. It has long been assumed that MTOs produce hydrogen sulfide (H2S) from MT. Herein, we studied the MTOs in the Rhodobacteraceae family-whose members are important DMSP degraders ubiquitous in marine environments. We identified 57 MTOs from 1,904 Rhodobacteraceae genomes

Transport of S-adenosylmethionine in Saccharomyces cerevisiae.

The properties of a specific system for the transport of S-adenosylmethionine in yeast are described. The process was pH-, temperature-, and energy-dependent, and showed saturation kinetics. The K(m) for the system was 3.3 x 10(-6)m. Of the S-adenosylmethionine moieties tested, only S-adenosylhomocysteine competitively inhibited the uptake of the adenosylsulfonium compound. Adenine, adenosine, methionine, homocysteine, and the sulfonium compound S-methylmethionine were without effect. The analogue S-adenosylethionine showed competitive inhibition. Under conditions of inhibition of protein synt

Light/copper catalysis in 1,2-alkylarylation of allylic alcohols with sulfonium salts involving radical 1,2-aryl migrations.

A visible-light-induced copper catalysis radical relay reaction of a,a-disubstituted allylic alcohols with sulfonium salts is described. The established protocol provides facile access to distal keto thioether derivatives via radical C-S bond cleavage and a 1,2-aryl migration relay strategy. A synthetic application of the product was achieved through a simple reduction or oxidation for the preparation of value-added compounds, including alcohols, sulfoxide, and sulfone-substituted thioether derivatives. Additionally, our methodology provided the direct assembly of keto-containing thioether mol

A Catalyst-Free Synthesis Platform for Functionalized Sulfur-Containing Molecules via Cyclic Sulfonium Salts.

Organosulfur chemistry has garnered significant attention due to the critical role of sulfur-containing compounds in bioactive natural products, pharmaceuticals, and functional materials. A unique class of sulfonium salts has been successfully synthesized, enabling a range of transformation reactions, such as elimination, nucleophilic substitution, and Kornblum oxidation, all without requiring an external catalyst. This development not only facilitates the integration of diazo chemistry and organosulfur chemistry but also establishes a simple and efficient platform for the diverse synthesis of

Metabolic partitioning between photosynthesis and osmotrophy in Collodaria photosymbiotic holobionts.

Collodaria (Radiolaria) are widespread marine planktonic protists that host photosynthetic microalgal symbionts. Within this photosymbiosis, they can contribute substantially to primary production, particularly in oligotrophic ocean regions. Despite their ecological importance, the mechanisms underlying the formation and maintenance of photosymbiosis are poorly understood. To uncover the metabolic dependencies within this photosymbiotic system, we develop a stable isotope labeling strategy that allows us to investigate the assimilation of inorganic and organic carbon in the Collodaria holobion

First clinical evidence that trimethylsulfonium can serve as a biomarker for the production of the signaling molecule hydrogen sulfide.

BACKGROUND: Hydrogen sulfide (H2S) is established as the third gaseous signaling molecule and is known to be overproduced in down syndrome (DS) due to the extra copy of the CBS gene on chromosome 21, which has been suggested to contribute to the clinical manifestation of this condition. We recently discovered trimethylsulfonium (TMS) in human urine and highlighted its potential as a selective methylation metabolite of endogenously produced H2S, but the clinical utility of this novel metabolite has not been previously investigated. We hypothesize that the elevation of H2S production in DS would

Sulfonium, an Underestimated Moiety for Structural Modification, Alters the Antibacterial Profile of Vancomycin Against Multidrug-Resistant Bacteria.

In the antibiotics arsenal, vancomycin is a last resort for the treatment of intractable infections. However, this situation is under threat because of the increasing appearance of vancomycin-resistant bacteria (VRB). Herein, we report a series of novel vancomycin derivatives carrying a sulfonium moiety. The sulfonium-vancomycin derivatives exhibited enhanced antibacterial activity against VRB both in vitro and in vivo. These derivatives also exhibited activity against some Gram-negative bacteria. The sulfonium modification enhanced the interaction of vancomycin with the bacterial cell membran

Photo/Copper-Catalyzed Sulfinamidation of Arenes and Alkenes.

Aryl sulfinamides are valuable intermediates and functional motifs in organic synthesis, with broad utility in pharmaceutical and agrochemical development. However, their broader application has been limited by underdeveloped synthetic methods, often requiring conventionally prefunctionalized aryl/alkenyl halides or organometallic reagents, protected sulfinylamine reagents, and harsh reaction conditions. Herein, we report a photo/copper-catalyzed sulfinamidation of arenes and alkenes, employing aryl and alkenyl sulfonium salts as radical linchpins. This protocol proceeds under mild conditions,

Copper-Catalyzed Formal [4+2] Cycloaddition of Enoldiazoimides with Sulfur Ylides.

Enoldiazoimides, a new subclass of enoldiazo compounds, generate enol-substituted carbonyl ylides whose reactions with sulfur ylides enable an unprecedented formal [4+2] cycloaddition. The resulting multifunctionalized indolizidinones, which incorporate sulfur, are formed in good yields under mild reaction conditions. The uniqueness of this transformation stems from the role of the silyl-protected enol, since the corresponding acetyldiazoimide failed to provide any cross-products in metal-catalyzed reactions with sulfur ylides. This copper-catalyzed cycloaddition is initiated with the generati

Transition-metal mediated carbon-sulfur bond activation and transformations: an update.

Carbon-sulfur bond cross-coupling has become more and more attractive as an alternative protocol to establish carbon-carbon and carbon-heteroatom bonds. Diverse transformations through transition-metal-catalyzed C-S bond activation and cleavage have recently been developed. This review summarizes the advances in transition-metal-catalyzed cross-coupling via carbon-sulfur bond activation and cleavage since late 2012 as an update of the critical review on the same topic published in early 2013 (Chem. Soc. Rev., 2013, 42, 599-621), which is presented by the categories of organosulfur compounds, t

Suplatast tosilate for treating cutaneous mastocytosis.

Pediatric cutaneous mastocytosis is a rare disease caused by mast cell hyperplasia. We report the case of an infant diagnosed as cutaneous mastocytosis and seasonal allergies. The wheals, flushing, and pruritus of the mastocytosis were unresponsive to combination therapy with an antihistamine, a mast cell stabilizer (sodium cromoglycate), and a leukotriene antagonist. Addition of suplatast tosilate as a treatment for the seasonal allergy also dramatically improved his cutaneous symptoms and signs. Further trials of suplatast tosilate in selected cases of cutaneous mastocytosis are warranted.

Design, synthesis of novel methyl-glycosyl furanose based sulfonium type α-glucosidase inhibitors with potent antihyperglycemic activity.

Developing effective α-glucosidase inhibitors is critical for managing hyperglycemia in individuals with high-carbohydrate diets; however, current drugs exhibit limitations in both efficacy and tolerability. In the present study, 25 derivatives of neoponkoranol, an active sulfonium type α-glucosidase inhibitor identified from Genus Salacia were synthesized. These compounds containing a methyl-glycosyl furanose scaffold modified with various hydrophobic groups at the 5'-O-position, were characterized using HRMS and NMR spectroscopy. Biological evaluations demonstrated a selectively enhanced mal

Decomposition kinetics and postmortem production of hydrogen sulfide and its metabolites.

BACKGROUND: Hydrogen sulfide (H2S), an endogenous gas, can also be generated from organics putrefaction. It is difficult for suspected cases of H2S poisoning to determine whether H2S in specimens is ingested by antemortem poisoning or generated from organics putrefaction. The aim of this study was to find the biomarkers of acute H2S poisoning via comparing the concentrations of H2S and its metabolites over time in specimens. METHODS: The H2S-spiked blood and blank blood group were established. The decomposition kinetics and the postmortem production of H2S were studied due to organics putrefa

Dimethylsulphoniopropionate as a water quality indicator of coral bleaching: Implications for monitoring studies on the Great Barrier Reef.

Short term stress experiments with dissolved inorganic phosphorus (DIP) and tripolyphosphate (TPP) have been carried out on the staghorn coral Acropora intermedia, collected from Heron Island in the southern Great Barrier Reef, at low and elevated seawater temperatures. Zooxanthellae, chlorophyll a, intracellular and tissue dimethylsulfoniopropionate (DMSP), and extracellular DMSP production were measured to assess the level of stress on A. intermedia at different winter and summer seasons from 2001 to 2003. Whilst no significant changes were measured in these stress indicators in 2001 and 200

The Met243 sulfonium ion linkage is responsible for the anomalous magnetic circular dichroism and optical spectral properties of myeloperoxidase.

The heme group of myeloperoxidase shows anomalous optical properties, and the enzyme possesses the unique ability to catalyze the oxidation of chloride. However, the nature of the covalently bound heme macrocycle has been difficult to identify. In this work, the electronic and magnetic properties of the heme groups in oxidized and reduced forms of wild-type and Met243Thr mutant myeloperoxidase and wild-type lactoperoxidase have been investigated using variable-temperature (1.6-273 K) magnetic circular dichroism (MCD) spectroscopy along with parallel optical absorption and electron paramagnetic

Carbene transfer from thianthrenium ylides for cyclopropanation.

Carbenes are reactive divalent carbon intermediates that serve as one-carbon synthons to access cyclopropanes. The synthetic utility of these three-membered carbocycles has led chemists to accept the dangers associated with the explosive starting materials1 or intermediates2 required for both conventional and modern cyclopropanation reactions. Sulfonium salts, although safer, have not been competitive for cyclopropanation because they typically do not function as efficient carbene donors3. In all cyclopropanation reactions reported, diversity can be obtained through either the olefin or carben

Dogger bank itch. 4. An eczema-causing sulfoxonium ion from the marine animal, Alcyonidium gelatinosum [Bryozoa].

Repeated exposure to the marine bryozoan, Alcyonidium gelatinosum, frequently provokes an eczematous allergic contact dermatitis known as the "Dogger Bank Itch". The dermatitis, representing a severe occupational disease, is especially widely distributed among trawlermen working in the Dogger Bank area in the North Sea. The allergy is shown to belong to the type of cell-mediated hypersensitivity. The hapten has been identified as the (2-hydroxyethyl)dimethylsulfoxonium ion. The isolation, structure determination and synthesis are discussed.
