Histamine suppresses non-NMDA excitatory synaptic currents in rat supraoptic nucleus neurons.

Whole cell patch-clamp recordings were obtained from supraoptic neurons to investigate the effects of histamine on excitatory postsynaptic currents evoked by electrical stimulation of areas around the posterior supraoptic nucleus. When cells were voltage-clamped at -70 mV, evoked excitatory postsynaptic currents had amplitudes of 88.4 +/- 9.6 pA and durations of 41.1 +/- 3.0 ms (mean +/- SE; n = 43). With twin stimulus pulses (20 Hz) used, paired-pulse facilitation ratios were 1.93 +/- 0.12. Bath application of 6-cyano-7-nitroquinoxalene-2,3-dione (CNQX) abolished synaptic currents. Histamine

Microdissection of the supraoptic nucleus and posterior pituitary gland from fresh unfixed tissue.

The rat supraoptic nucleus (SON) contains magnocellular neurons that project long axons that terminate in the posterior pituitary gland. To perform molecular characterization of these regions, such as transcriptome and methylome profiling, it is necessary to obtain large quantities of high-quality RNA and DNA. Prior methods to isolate molecular material from these small regions required fixing or freezing and laser microdissection of whole tissue, which can compromise recovery and integrity. We have established a straight-forward method of dissecting out the SON and posterior pituitary gland f

Gene expression in the rat supraoptic nucleus induced by chronic hyperosmolality versus hyposmolality.

Magnocellular neurons of the hypothalamo-neurohypophysial system play a fundamental role in the maintenance of body homeostasis by secreting vasopressin and oxytocin in response to systemic osmotic perturbations. During chronic hyperosmolality, vasopressin and oxytocin mRNA levels increase twofold, whereas, during chronic hyposmolality, these mRNA levels decrease to 10-20% of that of normoosmolar control animals. To determine what other genes respond to these osmotic perturbations, we have analyzed gene expression during chronic hyper- versus hyponatremia. Thirty-seven cDNA clones were isolate

Isosmotic hypovolemia preserves inverse neurovascular coupling in the supraoptic nucleus during heart failure.

Vasopressin (AVP) neurons in the hypothalamic supraoptic nucleus (SON) are activated by systemic challenges that threaten fluid balance. We previously showed that a systemic salt challenge triggers inverse neurovascular coupling (iNVC) in the SON, in which activity-dependent dendritic AVP release induces parenchymal arteriole (PA) vasoconstriction and local hypoxia. In heart failure (HF), however, the polarity of this salt-evoked response is reversed: microglia-derived adenosine acting on A2A receptors overrides an enhanced AVP-mediated vasoconstriction, producing net vasodilation. Still, whet

A microinjection study of the control of antidiuretic hormone release by the supraoptic nucleus of the hypothalamus in the cat.

1. The release of antidiuretic hormone (ADH) has been studied in the chloralose anaesthetized cat after microinjection of various agents directly into the brain, in particular the supraoptic nucleus of the hypothalamus (SON). The concentration of ADH in jugular venous blood was determined using the waterloaded, alcohol anaesthetized rat assay. The position of the microinjection cannula was located post mortem in stained brain sections.2. Nicotine, noradrenaline (NA) and hypertonic saline caused release of ADH, whereas microinjections of isotonic saline did not affect the blood level of the hor

The afferent pathway for carotid body chemoreceptor input to the hypothalamic supraoptic nucleus in the rat.

The pathway for chemoreceptor input to hypothalamic supraoptic nuclei has been examined in anaesthetised lactating and non-lactating rats. In lactating rats, the increase in intramammary pressure following bilateral carotid occlusion, which is probably mainly due to vasopressin, was abolished by lesions in the septum, but not by lesions in more caudal regions of the hypothalamus. In non-lactating rats, electrophysiological experiments demonstrated that the input from carotid body chemoreceptors to phasically-discharging supraoptic neurones is ipsilateral only. The effects of chemoreceptor stim

Acquiring Social Safety Engages Oxytocin Neurons in the Supraoptic Nucleus: Role of Magel2 Deficiency.

INTRODUCTION: Exposure to social trauma may alter engagement with both fear-related and unrelated social stimuli long after. Intriguingly, how simultaneous discrimination of social fear and safety is affected in neurodevelopmental conditions remains underexplored. The role of the neuropeptide oxytocin is established in social behaviors and yet unexplored during such a challenge post-social trauma. METHODS: Using Magel2 knockout mice, an animal model of Prader-Willi syndrome (PWS) and Schaaf-Yang syndrome (SYS), we tested memory of social fear and safety after a modified social fear conditioni

Muscarinic receptor modulation of slow afterhyperpolarization and phasic firing in rat supraoptic nucleus neurons.

A slow posttrain afterhyperpolarization (sAHP) was studied in rat magnocellular neurosecretory cells (MNCs) in vitro. The sAHP was isolated from other afterpotentials by blocking the depolarizing afterpotential (DAP) with Cs(+) and the medium afterhyperpolarization (mAHP) with apamin. The sAHP amplitude increased logarithmically with activity ( approximately 3 mV per e-fold increase in number of impulses) and, when firing stopped, decayed exponentially with a time constant of 2 sec. The sAHP was associated with increased membrane conductance, and its amplitude varied linearly with voltage, rev

Structural reorganization in the supraoptic nucleus of withdrawn rats following long-term alcohol consumption.

We have recently shown in the supraoptic nucleus (SON) of the rat that prolonged ethanol consumption induces cell degeneration and enlargement of the surviving neurons and of their subcellular organelles. We analyzed the SON of withdrawn rats to evaluate whether it displays any evidence of morphological reorganization following abstinence from ethanol, inasmuch as in this condition the ethanol-induced changes in the plasma levels of neurohormones and plasma osmolality are no longer detectable. A group of 18-month-old withdrawn rats was compared with age-matched, pair-fed control and ethanol-tr

Hypovolemia Evokes Conserved Inverse Neurovascular Coupling in the Supraoptic Nucleus Independent of Heart Failure.

Vasopressin (AVP) neurons in the hypothalamic supraoptic nucleus (SON) are activated by systemic challenges that threaten fluid balance. Glial regulation of extrasynaptic NMDA receptor-mediated excitation of supraoptic nucleus neurones during dehydration.

Magnocellular neurosecretory cells (MNCs) of the supraoptic nucleus (SON) project to the posterior pituitary gland where they release the hormones, vasopressin and oxytocin into the circulation to maintain plasma osmolality. Hormone release is proportionate to SON MNC action potential (spike) firing rate. When activated by ambient extracellular glutamate, extrasynaptic NMDA receptors (eNMDARs) mediate a tonic (persistent) depolarisation to increase the probability of action potential firing. In the present study, in vivo single-unit electrophysiological recordings were made from urethane-anaes

Activity of oxytocinergic neurons in the supraoptic nucleus under stimulation of alpha2-adrenoceptors in Brattleboro rats.

Vasopressin (AVP) deficient homozygous Brattleboro rats exhibit severe osmotic challenges due to waterless chronic hypernatremia and hyperosmolality. We investigated the effect of xylazine, an alpha2-adrenoceptor agonist, on the activity of oxytocinergic (OXY) neurons in the supraoptic nucleus (SON) of homozygous (di/di), heterozygous (di/+), and control (+/+) rats. Ninety minutes after saline (0.1 mL/100 g b.w., i.p.) or xylazine injection (10 mg/kg, i.p.) rats were anaesthetized with pentobarbital (50 mg/kg i.p.) and sacrificed by transcardial perfusion with fixative. Activity of OXY neurons

The vasopressin and oxytocin neurons in the human supraoptic and paraventricular nucleus; changes with aging and in senile dementia.

The neuropeptides vasopressin (AVP) and oxytocin (OXT) are supposed to be involved not only in peripheral functions (e.g. diuresis, labour and lactation) but also in central processes that are frequently disturbed during aging and senile dementia (e.g. fluid and electrolyte homeostasis and cognitive functions). A concomitant decrease in activity of the hypothalamo-neurohypophyseal system (HNS) with aging has been postulated in the literature, but has not yet been established. In order to investigate possible age-related changes in the human HNS, immunocytochemically identified AVP and OXT neur

The influence of age on the hypothalamo-neurohypophyseal system of the mouse: a quantitative ultrastructural analysis of the supraoptic nucleus.

A quantitative study of various morphological parameters in endocrine cells of the neuroendocrine region of the laboratory mouse was carried out. The supraoptic nucleus of the hypothalamo-neurohypophyseal system synthesises the hormones vasopressin and, to a lesser extent, oxytocin, and transports them to the posterior pituitary. Female C57BL/Icrfat mice at 8 and 26 months of age, free of macroscopic pathology, were sampled when in a physiologically defined resting state. No significant differences in the volume fractions of most cell and subcellular components could be detected at the two age

Dystrophins in Structural Changes of the Supraoptic Nucleus During Seasonal Adaptation in Wild Jaculus orientalis.

Dystrophin (Dp), a 427 kDa membrane-associated cytoskeletal protein, encoded by the Duchenne muscular dystrophy (DMD) gene, is known for its deficiency in DMD. Emerging evidence suggests Dp expression in the hypothalamus of rats and mice, where it plays a role in functional plasticity. In this study, we focused on Jaculus orientalis, a desert-adapted rodent that serves as an excellent model for investigating seasonal adaptation challenges. However, due to challenges in capturing and properly maintaining these animals in the laboratory, only a limited number of studies have been carried out so

Excitatory Effects of Astrocytic Hydrogen Sulfide on the Electrical Activity of Oxytocin Neurons in the Supraoptic Nucleus.

INTRODUCTION: In the regulation of oxytocin (OT) neuronal activity, hydrogen sulfide (H2S), a gaseous neurotransmitter, likely exerts an excitatory role. This role is associated with increased expression of astrocytic cystathionine-β-synthase (CBS), the key enzyme for H2S synthesis. However, it remains unclear whether H2S is mainly produced in astrocytes and contributes to the autoregulation of OT neurons. METHODS: In hypothalamic slices of male rats, OT and H2S-associated drug effects were observed on the firing activity and spontaneous excitatory postsynaptic currents (sEPSCs) of putative O

Sustained outward rectification of oxytocinergic neurones in the rat supraoptic nucleus: ionic dependence and pharmacology.

1. Intracellular recordings were obtained in vitro from oxytocin and vasopressin neurones from dioestrous and lactating female rats. Oxytocin neurones were characterized under current clamp by the expression of a depolarization-activated, sustained outward rectification (SOR) and a rebound depolarization (RD). 2. An increment in extracellular K+ shifted the expression of the SOR and RD towards a more depolarized membrane potential, indicating that the mechanisms underlying these events are dependent on extracellular potassium. 3. The SOR and RD were blocked by external tetraethylammonium (10 m

The amount of mutant vasopressin precursor in the supraoptic and paraventricular nucleus of Brattleboro rats increases with age.

In the homozygous diabetes insipidus (di/di) Brattleboro rat an aberrant vasopressin (VP) precursor is synthesized. An antiserum raised against a 14 amino acid sequence of this mutant precursor (CP-14) stains neurons in the supraoptic (SON) and paraventricular nucleus (PVN). In the present study it was shown that during life (i.e. from 16 days up to 83 weeks postnatally) the number of CP-14 SON cells and their content gradually increases. Only a few CP-14 SON cells are present in young pre-pubertal di/di rats. The staining becomes increasingly more intense after puberty. In contrast, the numbe

Opioid receptor subtypes in the supraoptic nucleus and posterior pituitary gland of morphine-tolerant rats.

Morphine, given acutely, inhibits oxytocin secretion in adult female rats, but chronic intracerebroventricular infusion for five to six days induces tolerance and dependence in the mechanisms regulating oxytocin secretion. One explanation for tolerance could be that there is a loss of opioid receptors. To test this hypothesis cryostat sections of selected brain regions and the pituitary, from six control and six intracerebroventricular morphine-infused rats, were processed for quantitative in vitro receptor autoradiography. [3H]Etorphine or [3H](-)-bremazocine were used as ligands, and DAGO, D
