Phoneutria nigriventer toxins block tityustoxin-induced calcium influx in synaptosomes.

Neurotoxins can help the understanding of mechanisms involved in neurotransmission. We here report that two neurotoxin isoforms, Tx3-3 and Tx3-4 obtained from the venom of the spider Phoneutria nigriventer inhibited the 45Ca2+ influx in rat cortical synaptosomes induced by the scorpion venom tityustoxin. The IC50 for Tx3-3 and Tx3-4 were 0.32 and 7.9 nM, respectively. The neurotoxins Tx3-3 and Tx3-4 are very effective in inhibiting 45Ca2+ influx and they should be useful in studies involving Ca(2+)-dependent processes.

Chlorpromazine stimulatory effect on iron uptake by rat brain synaptosomes.

Clinical long-term neuroleptic administration induces extrapyramidal motor side-effects, of which tardive dyskinesia is the most important. Experimentally, dopamine D2 supersensitivity is observed after phenothiazine and butyrophenone treatment. Neuroleptic-induced tardive dyskinesia and D2 modulation have been linked to impaired iron homeostasis in the central nervous system. Increased nonheme iron levels found in the basal ganglia of patients with extrapyramidal symptomology support the connection between iron and neuronal dopaminergic modulation. We now report the effect of chlorpromazine o

[Internalization and degradation of latrotoxin bound to rat brain synaptosomes].

The accessibility to trypsin of 125I-labeled latrotoxin bound to rat brain synaptosomes was investigated. It was shown that latrotoxin bound to synaptosomes in the cold can be practically completely removed by trypsin treatment. The resistance of latrotoxin to proteolysis increases during its incubation with synaptosomes (37 degrees C). Concanavalin A (10(-6) M) decreases toxin binding by 30%, but fully prevents internalization (incorporation). Moreover, latrotoxin is not incorporated into synaptosomal membrane fragments irrespective of duration and temperature of incubation. Latrotoxin incorp

[3H]adenosine uptake and release from synaptosomes. Alterations by barbiturates.

The effects of barbiturates on adenosine movements across the synaptic plasma membrane have been investigated using rodent whole brain synaptosomes. The hypothesis tested was that some of the depressant actions of these drugs may be mediated through interference with an endogenous adenosine system. Adenosine uptake was studied using synaptosomes prepared from Swiss-Webster mice. After preincubation at 37 degrees, [3H]adenosine was added to the synaptosomes in the presence or absence of pentobarbital, methohexital, phenobarbital, or 5-(2-cyclohexylideneethyl)-5-ethyl barbituric acid (CHEB) at v

Scorpion neurotoxin. Mode of action on neuromuscular junctions and synaptosomes.

Electrophysiological analysis of the effects of scorpion toxin I, one of the neurotoxins from the venom of the scorpion Androctonus australis Hector, upon crayfish neuromuscular junctions has shown that the toxin strongly associates with the nerve terminal to stimulate release of neurotransmitters. The biochemical approach has shown that the binding of scorpion toxin I to rat brain synaptosomes is accompanied by a decrease in their capacity to accumulate gamma-aminobutyric acid. The main effect of the toxin is to stimulate neurotransmitter release. The apparent dissociation constant of the tox

Local anesthetics inhibit glutamate release from rat cerebral cortex synaptosomes.

Local anesthetics have been widely used for regional anesthesia and the treatment of cardiac arrhythmias. Recent studies have also demonstrated that low-dose systemic local anesthetic infusion has neuroprotective properties. Considering the fact that excessive glutamate release can cause neuronal excitotoxicity, we investigated whether local anesthetics might influence glutamate release from rat cerebral cortex nerve terminals (synaptosomes). Results showed that two commonly used local anesthetics, lidocaine and bupivacaine, exhibited a dose-dependent inhibition of 4-AP-evoked release of gluta

Tityustoxin-induced release of ATP from rat brain cortical synaptosomes.

Tityustoxin, a scorpion toxin that alters the Na+ channel activity, induces release of ATP from rat brain cortical synaptosomes. The effect of tityustoxin is dependent on its concentration and incubation time. Continuously or cumulative release of ATP evoked by tityustoxin was calcium-dependent and interestingly only partially inhibited by tetrodotoxin. We suggest that tityustoxin mainly releases ATP from the vesicular pool but other pools may also be involved.

Increased lipid fluidity in synaptosomes from brains of hyperosmolal rats.

Taurine, a product of sulfur amino acid metabolism, is important in cerebral osmoregulation. To understand the adaptive changes in transport which accompany different hyperosmolal states, we determined lipid composition and fluorescence anisotropy of synaptosomal liposomes from rats with chronic hypernatremic dehydration (CHD), streptozocin-induced (STZ) diabetes, and insulin treated diabetes. Induction of CHD increased serum osmolality, and enhanced in vitro synaptosomal taurine uptake (P < 0.01, n = 3, vs. control). Fluorescence anisotropy studies showed that the fluidity of lipids from CHD

Studies of receptor-mediated inhibition of 45Ca accumulation into synaptosomes.

1. The effects of alpha 2-adrenoceptor and kappa-opiate receptor activation on 45Ca accumulation into rat cortical synaptosomes were examined. 2. Clonidine (1 microM) and U50488H (1 microM) significantly reduced 45Ca accumulation under both resting (5 mM K+) and depolarizing (15-30 mM K+) conditions. 3. The inhibitory effects of the agonists on 45Ca accumulation into synaptosomes were enhanced in the presence of the Na(+)-Ca2+ exchange inhibitor sodium orthovanadate (vanadate, 2 mM), and were not present in mitochondrial preparations. 4. When the agonists were used together, their inhibitory e

Kainate-induced uptake of calcium by synaptosomes from rat brain.

Kainic acid induces a rapid increase in 45Ca2+ uptake by crude synaptosomal fractions isolated from rat brain. This enhanced Ca2+ permeability occurs with a half-time of approx. 1 s, similar to the fast phase of depolarization-induced calcium uptake. The depolarization-induced uptake of calcium is inhibited 85% by 3 mM CoCl2, 80% by 100 microM quinacrine and 50% by 15 microM trifluoperazine while these agents had little effect on the kainate-induced uptake. It is proposed that kainate induces receptor-mediated opening of a class of calcium channels with properties different from those of the v

Characterization of the zinc-induced Shank3 interactome of mouse synaptosome.

Variants of the SHANK3 gene, which encodes a core scaffold protein of the postsynaptic density of excitatory synapses, have been causally associated with numerous brain disorders. Shank3 proteins directly bind zinc ions through their C-terminal sterile α motif domain, which enhances the multimerization and synaptic localization of Shank3, to regulate excitatory synaptic strength. However, no studies have explored whether zinc affects the protein interactions of Shank3, which might contribute to the synaptic changes observed after zinc application. To examine this, we first purified Shank3 prot

On the kaliotoxin and dendrotoxin binding sites on rat brain synaptosomes.

The toxic polypeptides alpha-, beta-, gamma- and delta-dendrotoxin (DTX), known to be potent blockers of voltage-dependent potassium channels of the Kv1 family, were purified from the venom of the green mamba Dendroaspis angusticeps. Their binding behaviour to synaptosomal membranes of rat brain was analysed and compared with that of kaliotoxin (KTX), in a competition assay using [(125)I] KTX. alpha-DTX and delta-DTX were found to compete with radioiodinated-KTX (IC(50) of 8 pM and 0.2 nM respectively), whereas gamma-DTX did not. Several minor components that competed with radioiodinated-KTX b

The effect of tityustoxin in the turnover of inositol phosphates in crude synaptosomes.

The formation of inositol phosphates in response to stimulation by tityustoxin was studied in crude synaptosomes (P2). Phosphoinositides were labelled by incubation in the presence of [3H]inositol. Tityustoxin stimulated the turnover of inositol phosphate (PI). Tityustoxin specifically stimulated the turnover of glycerophosphoinositol (GP1), inositol 1-monophosphate (IP1), inositol 1,4-biphosphate (IP2) and inositol 1,4,5-triphosphate (IP3). The tityustoxin-stimulated turnover of water-soluble inositol phosphates was inhibited by tetrodotoxin, atropine and phorbol myristate acetate. It is sugg

[Binding of latrotoxin to synaptosomes and synaptic membranes of the rat brain].

The binding of [125I] alpha-latrotoxin to synaptosomes from the rat brain is studied. It is shown that the constant rate of toxin association with the synaptosome receptor at 37 degrees C is equal to 8.2 +/- 1.3 x 10(7) M-1.s-1, while that of synaptosomal membrane -7.6 +/- 2.7 x 10(6) M-1 s-1. Depolarization of the synaptosome membrane induced by 55 mM KCl decreases the binding rate of toxin to the receptor, the rate constant being equal to 3.9 +/- 1.5 x 10(7) m-1 s-1. Perturbational actions of barbiturate analogues on erythrocyte and synaptosomal membranes.

The membrane perturbational actions of pentobarbital, methohexital, and phenobarbital were analyzed in terms of their effects on the incorporation of group-specific chemical probes into protein and phospholipid structural components and on the activity of various functionally distinct ATPases in erythrocyte and brain synaptosomal membranes. When compared at concentrations producing similar degrees of antihemolysis in red cells, the three analogues differed most markedly in the nature and degree of the structural and functional alterations induced in membrane proteins, with the most highly lipo

The effect of scorpion venom tityustoxin on the uptake of calcium in synaptosomes.

The effect of tityustoxin on calcium uptake in synaptosomes incubated in hypotonic medium low sodium and physiological medium high sodium was studied. In the hypotonic medium, calcium uptake was ATP and temperature dependent and TsTX inhibits this uptake. The inhibition of calcium uptake was not due to an osmotic effect since it was also observed in conditions of physiological cell osmolarity. In the physiological medium high sodium, tityustoxin stimulated calcium uptake in synaptosomes. The effect was inhibited by tetrodotoxin and cocaine. The stimulating effect of tityustoxin was specific fo

Studies on valinomycin inhibition of synaptosome-fraction protein synthesis.

The ionophore valinomycin inhibited adult and neonatal synaptosome fraction protein synthesis with half-maximal inhibition at approximately 10nM. Valinomycin had no effect on [3H]leucine uptake into synaptosomes at high or low external [K+]. Synaptosome-fraction protein synthesis was dependent on [K+]e reaching a maximum at 25mM-K+. Valinomycin inhibition of protein synthesis was not reversed at high [K+]e. Valinomycin failed to influence the intrasynaptosomal [K+] even at zero [K+]e. A significant increase in State-4 respiration of synaptosomal fractions was found at 5nM-valinomycin with a de

Action of barbiturates on activity of acetylcholinesterase from synaptosomal membranes.

The acetylcholinesterase from synaptosomal membranes is inhibited by anesthetics: Nembutal, brietal, and thiopental. Nembutal and brietal decrease the Km for acetylthiocholine, without changes in Vmas. A noncompetitive type of inhibition is produced by thiopental. This anesthetic decreases Arrhenius plot discontinuity by about 4 degrees C and increases activation energies. Nembutal and brietal do not change Arrhenius plot discontinuities, but they increase activation energies. These results suggest that barbiturates change lipid-protein interactions in synaptosomal membranes.
