Mirror alignment by particle swarm optimization for synchrotron radiation beamlines.

This article proposes a particle swarm optimization (PSO)-based automated mirror-alignment methodology that enables the utilization of high-flux beams for measurements in synchrotron radiation beamlines. The method is designed to improve beam flux by adjusting two mirrors within a target alignment time of 5 min, thereby satisfying critical operational demands. The proposed PSO-based method incorporates three adaptations: (i) particle initialization and dynamics (which are modified to locate a narrow flux peak efficiently), (ii) mirror-movement optimization (which reduces the total mirror-trave

Synchrotron x-ray diffraction study of liquid and glassy toluene.

We measured the X-ray diffraction data of liquid toluene at several temperatures between 300 and 180 K and those of glasses prepared by liquid quenching and vapor deposition (VD) at 3 K. The VD glasses annealed at 90 and 112 K were also measured at 3 K. Throughout the measurements, we used a custom-made cryostat for VD experiments and a high-energy X-ray diffractometer at BL04B2, SPring-8. The structure factor S(Q) of each state was successfully fitted using potential fitting with molecular dynamics simulation and reverse Monte Carlo methods. Interestingly, in both the liquid and glassy states

Synchrotron Phase-Contrast Imaging and Cochlear Otosclerosis: A Case Report.

INTRODUCTION: Otosclerosis is a bone disorder affecting the labyrinthine capsule that leads to conductive and occasionally sensorineural hearing loss. The etiology of otosclerosis remains unknown; factors such as infection, hormones, inflammation, genetics, and autoimmunity have been discussed. Treatment consists primarily of surgical stapes replacement and cochlear implantation. High-resolution computed tomography is routinely used to visualize bone pathology. In the present study, we used synchrotron radiation phase-contrast imaging (SR-PCI) to examine otosclerosis plaques in a temporal bone

Alanine response to low energy synchrotron x-ray radiation.

Objective. The radiation response of alanine is very well characterized in the MV photon energy range where it can be used to determine the dose delivered with an accuracy better than 1%, making it suitable as a secondary standard detector in cancer radiation therapy. This is not the case in the very low energy keV x-ray range where the alanine response is affected by large uncertainties and is strongly dependent on the x-ray beam energy. This motivated the study undertaken here.Approach. Alanine pellets with a nominal thickness of 0.5 mm and diameter of 5 mm were irradiated with monoenergetic

Aeration of the Human Prussak's Space: A 3D Synchrotron Imaging Study.

OBJECTIVES: Prussak's space (PS) is an intricate middle ear region which may play an essential role in the development of middle ear disease. The three-dimensional (3D) anatomy of the human PS and its drainage routes remain relatively unknown. Earlier studies have histologically analyzed PS, by micro-dissection and endoscopy. Here, we used synchrotron-radiation phase-contrast imaging (SR-PCI), 3D reconstructions, and modeling to study the framework of the human PS, including aeration pathways. It may lead to increased understanding of development of middle ear pathology. DESIGN: Nine human te

The lung in acute respiratory failure: insights from synchrotron radiation imaging.

Acute respiratory failure and the acute respiratory distress syndrome (ARDS) are characterized by profound spatial and temporal heterogeneity in lung aeration, mechanics, and inflammation. While mechanical ventilation is often vital to ensure gas exchange, it can also cause or worsen lung injury as a result of the superimposed mechanical stress due to the application of positive pressure on a heterogeneous parenchyma. Characterizing the lung microstructure and function at small length scales is essential for understanding the pathogenesis of ventilator-induced lung injury. This is a challengin

Three-dimensional structure analysis of mouse nails using synchrotron radiation.

Until now, studies on nail diseases have been performed through microscopic diagnosis and microscopic computed tomography (micro-CT). However, these kinds of conventional methods have some limitations. Firstly, the microscopic method is considered the gold standard for medical diagnosis. However, due to the use of fluorescent materials, the sample is damaged and it takes a long time to get results. Secondly, while micro-CT is a noninvasive method to get inner structure images of the sample with high resolution, the penetration and spatial resolution are insufficient for studying the microstruc

Methods for dynamic synchrotron X-ray respiratory imaging in live animals.

Small-animal physiology studies are typically complicated, but the level of complexity is greatly increased when performing live-animal X-ray imaging studies at synchrotron and compact light sources. This group has extensive experience in these types of studies at the SPring-8 and Australian synchrotrons, as well as the Munich Compact Light Source. These experimental settings produce unique challenges. Experiments are always performed in an isolated radiation enclosure not specifically designed for live-animal imaging. This requires equipment adapted to physiological monitoring and test-substa

Jawbone remodeling: a conceptual study based on Synchrotron High-resolution Tomography.

One of the most important aspects of bone remodeling is the constant turnover mainly driven by the mechanical loading stimulus. The remodeling process produces changes not only in the bone microarchitecture but also in the density distribution of the mineralized matrix - i.e. in calcium concentrations- and in the osteocyte lacunar network. Synchrotron radiation-based X-ray microtomography (microCT) has proven to be an efficient technique, capable to achieve the analysis of 3D bone architecture and of local mineralization at different hierarchical length scales, including the imaging of the lac

Dynamic Detection of Thrombolysis in Embolic Stroke Rats by Synchrotron Radiation Angiography.

A rodent model of embolic middle cerebral artery occlusion is used to mimic cerebral embolism in clinical patients. Thrombolytic therapy is the effective treatment for this ischemic injury. However, it is difficult to detect thrombolysis dynamically in living animals. Synchrotron radiation angiography may provide a novel approach to directly monitor the thrombolytic process and assess collateral circulation after embolic stroke. Thirty-six adult Sprague-Dawley rats underwent the embolic stroke model procedure and were then treated with tissue plasminogen activator. The angiographic images were

Broadband terahertz heterodyne spectrometer exploiting synchrotron radiation at megahertz resolution.

A new spectrometer allowing both high resolution and broadband coverage in the terahertz (THz) domain is proposed. This instrument exploits the heterodyne technique between broadband synchrotron radiation and a quantum-cascade-laser-based molecular THz laser that acts as the local oscillator. Proof of principle for exploitation for spectroscopy is provided by the recording of molecular absorptions of hydrogen sulfide (H2S) and methanol (CH3OH) around 1.073 THz. Ultimately, the spectrometer will enable to cover the 1-4 THz region in 5 GHz windows at Doppler resolution.

Ferruginous bodies resolved by synchrotron XRF in a dog with peritoneal malignant mesothelioma.

Mesothelioma is a malignant tumor mainly correlated to occupational asbestos exposure. Rare reports describe its occurrence also in animals, mainly linked to asbestos in the environment. Asbestos exposure is demonstrated by the appearance of characteristic histological hallmarks: asbestos containing ferruginous bodies that are iron-based structures forming around fibers and also other dust particles. Here we present a clinical case of a suspect of mesothelioma in the peritoneum of a dog with parallel histological observation of ferruginous bodies. To possibly correlate the dog tumor to environ

Subcellular mapping of living cells via synchrotron microFTIR and ZnS hemispheres.

FTIR imaging is a label-free, non-destructive method valuably exploited in the study of the biological process in living cells. However, the long wavelength/low spatial resolution and the strong absorbance of water are still key constrains in the application of IR microscopy ex vivo. In this work, a new retrofit approach based on the use of ZnS hemispheres is introduced to significantly improve the spatial resolution on live cell FTIR imaging. By means of two high refractive index domes sandwiching the sample, a lateral resolution close to 2.2 μm at 6 μm wavelength has been achieved, i.e. belo

Histological structure of human nail as studied by synchrotron X-ray microdiffraction.

Three layers (characterized by different orientations of the keratin molecules) from the outer to the inner side of human nail were observed by synchrotron X-ray microdiffraction. These layers are associated with the histological dorsal, intermediate and ventral plates. The hair-like type alpha-keratin filaments (81 A in diameter), are only present in the intermediate layer (accounting for approximately 2/3 of the nail width) and are perfectly oriented perpendicular to the growth axis, in the nail plane. Keratin filaments of stratum corneum (epidermis) type, found in the dorsal and ventral cel

A method for examining the chemical basis for bone disease: synchrotron infrared microspectroscopy.

Infrared microspectroscopy combines microscopy and spectroscopy for the purpose of chemical microanalysis. Light microscopy provides a way to generate and record magnified images and visibly resolve microstructural detail. Infrared spectroscopy provides a means for analyzing the chemical makeup of materials. Combining light microscopy and infrared spectroscopy permits the correlation of microstructure with chemical composition. Inherently, the long wavelengths of infrared radiation limit the spatial resolution of the technique. However, synchrotron infrared radiation significantly improves bot

[Virtual histopathology of the pancreas: 3D insights using synchrotron-based imaging].

BACKGROUND: Conventional histopathology faces methodological limitations when assessing complex three-dimensional tissue architectures. In particular, for heterogeneous tissues such as the pancreas or in complex tissue pathologies, restriction to two-dimensional sections hampers comprehensive recognition of morphological features. OBJECTIVE: This study aims to demonstrate the potential of synchrotron-based phase-contrast imaging (SRµCT) as a tool for high-resolution visualization of pancreatic tissue. Three representative case examples were analyzed to capture morphological parameters volumet
