Previous microarray analyses have shown a key role for the two-component system PhoBR (SYNW0947, SYNW0948) in the regulation of P transport and metabolism in the marine cyanobacterium Synechococcus sp. WH8102. However, there is some evidence that another regulator, SYNW1019 (PtrA), probably under the control of PhoBR, is involved in the response to P depletion. PtrA is a member of the cAMP receptor protein transcriptional regulator family that shows homology to NtcA, the global nitrogen regulator in cyanobacteria. To define the role of this regulator, we constructed a mutant by insertional ina

Marine Synechococcus are frequently found in environments where iron (Fe) is a limiting nutrient. To understand their capacity to respond to Fe stress, we screened picoplankton genomes and the Global Ocean Survey metagenome for known Fe stress genes. Many open ocean strains of Synechococcus lack most known genes for Fe stress, while coastal and upwelling strains contain many, suggesting that maintaining multiple Fe limitation compensation strategies is not a selective advantage in the open ocean. All genomes contained iron deficiency-induced protein A (IdiA) and its complementary Fe(3+) transp

Calcium (Ca²⁺) functions as a universal signaling ion across biological systems, yet its role in cyanobacterial metabolic regulation remains underexplored. Here, we investigated the association between Ca²⁺ availability and physiological responses in Synechococcus elongatus PCC 7942 in relation to the conserved regulatory protein PII. Ca²⁺ supplementation significantly increased cell viability, pigment accumulation, photosynthetic efficiency, and ATP/ADP ratios in wild-type cells, while a ΔglnB mutant lacking PII failed to respond, demonstrating PII's central role. Notably, Ca²⁺ enhanced the s

The cyanobacterium Synechococcus sp. PCC 7002 produces a monomeric hemoglobin (GlbN) implicated in the detoxification of reactive nitrogen and oxygen species. GlbN contains a b heme, which can be modified under certain reducing conditions. The modified protein (GlbN-A) has one heme-histidine C-N linkage similar to the C-S linkage of cytochrome c. No clear functional role has been assigned to this modification. Here, optical absorbance and NMR spectroscopies were used to compare the reactivity of GlbN and GlbN-A toward nitric oxide (NO). Both forms of the protein are capable of NO dioxygenase a

The genomes of two closely related thermophilic cyanobacterial isolates, designated Synechococcus isolate OS-A and Synechococcus isolate OS-B', from the microbial mats of Octopus Spring (Yellowstone National Park) have been sequenced. An extensive suite of genes that are controlled by phosphate levels constitute the putative Pho regulon in these cyanobacteria. We examined physiological responses of an axenic OS-B' isolate as well as transcript abundances of Pho regulon genes as the cells acclimated to phosphorus-limiting conditions. Upon imposition of phosphorus deprivation, OS-B' stopped divi

Current ambient UV-B levels can significantly depress productivity in aquatic habitats, largely because UV-B inhibits several steps of photosynthesis, including the photooxidation of water catalyzed by photosystem II. We show that upon UV-B exposure the cyanobacterium Synechococcus sp. PCC 7942 rapidly changes the expression of a family of three psbA genes encoding photosystem II D1 proteins. In wild-type cells the psbAI gene is expressed constitutively, but strong accumulations of psbAII and psbAIII transcripts are induced within 15 min of moderate UV-B exposure (0.4 W/m2). This transcription

Constraint-based reconstruction and analysis (COBRA) is a powerful systems biology approach for computational bioengineering. Synechococcus elongatus PCC 11801 and PCC 11802 are fast-growing, stress-tolerant cyanobacteria that are promising platforms for photosynthetic biomanufacturing. Here, we present constraint-based models (CBMs) iLV1052 and iLV1087 of PCC 11801 and PCC 11802, respectively, to facilitate and streamline strain engineering efforts. Following draft reconstruction using a template model, the models underwent extensive manual curation to reduce redundancy, and verification usin

As diazotrophic cyanobacteria of tremendous biomass, Trichodesmium continuously provide a nitrogen source for carbon-fixing cyanobacteria and drive the generation of primary productivity in marine environments. However, ocean iron deficiencies limit growth and metabolism of Trichodesmium. Recent studies have shown the co-occurrence of Trichodesmium and siderophore-producing Synechococcus in iron-deficient oceans, but whether siderophores secreted by Synechococcus can be used by Trichodesmium to adapt to iron deficiency is not clear. We constructed a mutant Synechococcus strain unable to produc

The co-occurrence of nanoplastics (NPs) and antibiotics in marine environments poses complex threats to primary producers, yet the multi-level mechanisms of their combined toxicity remain poorly understood. In this study, we systematically investigated the physiological, biochemical, and transcriptomic responses of the marine cyanobacterium Synechococcus sp. WH8102 to single and combined exposure of polystyrene NPs and ofloxacin (OFL). Interestingly, Independent Action analysis based on population growth indicated an apparent antagonistic interaction, which may be associated with altered OFL a

Cyanobacteria, such as Synechococcus sp. PCC 7002 (Syn7002), are promising chassis strains for "green" biotechnological applications as they can be grown in seawater using oxygenic photosynthesis to fix carbon dioxide into biomass. Their other major nutritional requirements for efficient growth are sources of nitrogen (N) and phosphorus (P). As these organisms are more economically cultivated in outdoor open systems, there is a need to develop cost-effective approaches to prevent the growth of contaminating organisms, especially as the use of antibiotic selection markers is neither economicall

The Amt proteins constitute a ubiquitous family of transmembrane ammonia channels that permit the net uptake of ammonium by cells. In many organisms, there is more than one amt gene, and these genes are subjected to nitrogen control. The mature Amt protein is a homo- or heterooligomer of three Amt subunits. We previously characterized an amt1 gene in the unicellular cyanobacterium Synechococcus elongatus strain PCC 7942. In this work, we describe the presence in this organism of a second amt gene, amtB, which encodes a protein more similar to the bacterial AmtB proteins than to any other chara

We have determined the complete nucleotide sequence of a Synechococcus sp. PCC7942 gene encoding the homologue of mammalian uroporphyrinogen decarboxylase (UROD). The gene, designated hemE, encoded a polypeptide of 354 amino acids with a molecular weight of 39,283. The primary sequences of the polypeptide encoded by hemE and human and rat UROD had 32.5% identical amino acid residues. No invariant cysteine residues were found, despite the fact that UROD isolated from different sources has been shown to be inhibited by sulfhydryl reagents. The knowledge of the primary structure of this cyanobact

Cyanobacteria hold promise as cell factories for the photoautotrophic conversion of carbon dioxide to useful chemicals. For the eventual commercial viability of such processes, cyanobacteria need to be engineered for (i) efficient channeling of carbon flux toward the product of interest and (ii) improved product tolerance, the latter being the focus of this study. We chose the recently reported, fast-growing, high light and CO2 tolerant cyanobacterium Synechococcus elongatus PCC 11801 for adaptive laboratory evolution. In two parallel experiments that lasted over 8400 h of culturing and 100 se

Responsible for fixing 25% of carbon dioxide (CO2) globally, cyanobacteria use carboxysomes to house their CO2 fixing machinery. The formation and permeability of the proteinaceous shell of carboxysomes is an area of active study. While necessary in air (0.04% CO2), the shell is not required when cyanobacteria are in high CO2 levels representative of early Earth. To understand how the carboxysome shell responds to increased CO2 conditions, we used a Grx1-roGFP2 redox sensor and single cell timelapse fluorescence microscopy to track subcellular redox states of Synechococcus sp. PCC 7002. Compar

Microbial volatile organic compounds (VOCs) are cell metabolites that affect many physiological functions of prokaryotic and eukaryotic organisms. Earlier we have demonstrated the inhibitory effects of soil bacteria volatiles, including ketones, on cyanobacteria. Cyanobacteria are very sensitive to ketone action. To investigate the possible molecular mechanisms of the ketone 2-nonanone influence on cyanobacterium Synechococcus elongatus PCC 7942, we applied a genetic approach. After Tn5-692 transposon mutagenesis, several 2-nonanone resistant mutants have been selected. Four different mutant s

In the practical scale of cyanobacterial cultivation, the golden algae Poterioochromonas malhamensis is a well-known predator that causes devastating damage to the culture, referred to as pond crash. The establishment and maintenance of monoculture conditions are ideal for large-scale cultures. However, this is a difficult challenge because microbial contamination is unavoidable in practical-scale culture facilities. In the present study, we unexpectedly observed the pond crash phenomenon during the pilot-scale cultivation of Synechococcus elongatus PCC 7942 using a 100-L photobioreactor. This

Glutaredoxins regulate cellular redox homeostasis, protect against oxidative stress, and modulate protein function crucial for the fitness of photosynthetic organisms and diseases progression in human. Protein functionality is intricately linked to structural integrity, which is maintained by conserved amino acids residues. This study investigates the structural and functional effect of single-point mutations at evolutionarily highly conserved residues in the dithiol glutaredoxin (Grx3) protein of cyanobacterium Synechococcus elongatus PCC 7942. Conservation analysis identified key residues su

The single crossover occurring via homologous recombination is a common phenomenon existing among microbes like Escherichia coli, Bacillus subtilis, Vibrio natriegens, Gluconobacter oxydans and most cyanobacteria species, threatening the stability of engineered strains and challenging iterative genetic engineering. Among them, we take the fast-growing cyanobacterium Synechococcus elongatus UTEX 2973 (Syn2973) as a representative study due to its promising roles for CO2 fixation and bioconversion. We established three marker-free platforms to achieve stable genome recombination: (i) T4CROSS, wh

To promote the utilization of industrial flue gases by microalgae, this study performed adaptive laboratory evolution (ALE) with CO2 as the selective pressure, aiming to improve the carbon fixation efficiency of microalgae under high CO2 conditions. Synechococcus elongatus PCC7942 was sequentially cultured with CO2 concentrations ranging from 5 % to 20 %, resulting in a domesticated strain S20 with growth advantage and capable of acclimatizing to 20 % CO2. At a CO2 concentration of 20 %, the biomass of S20 reached 1.37 g L-1, and the lipid contents of S20 were significantly elevated, reaching

Cyanobacteria engineered for production of biofuels and biochemicals from carbon dioxide represent a promising area of research in relation to a sustainable economy. Previously, we have succeeded in producing isopropanol from cellular acetyl-CoA by means of Synechococcus elongatus PCC 7942 into which a synthetic metabolic pathway was introduced. The isopropanol production by this synthetic metabolic pathway requires acetate; therefore, the cells grown under photosynthetic conditions have to be transferred to a dark and anaerobic conditions to produce acetate. In this study, we achieved acetate
