Photosynthesis in Synechocystis sp. PCC 6803 is not optimally regulated under very high CO2.

One strategy for CO2 mitigation is using photosynthetic microorganisms to sequester CO2 under high concentrations, such as in flue gases. While elevated CO2 levels generally promote growth, excessively high levels inhibit growth through uncertain mechanisms. This study investigated the physiology of the cyanobacterium Synechocystis sp. PCC 6803 under very high CO2 concentrations and yet stable pH around 7.5. The growth rate of the wild type (WT) at 200 µmol photons m-2 s-1 and a gas phase containing 30% CO2 was 2.7-fold lower compared to 4% CO2. Using a CRISPR interference mutant library, we i

Evaluation of cell-disruption effects of pulsed-electric-field treatment of Synechocystis PCC 6803.

In order to use Synechocystis PCC 6803 as feedstock of nonpetroleum-based diesel fuel, pulsed electric field (PEF) technology was used for cell disruption prior to extraction of intracellular lipids. Severe cell disruption was evident after PEF treatment, especially with treatment intensity (TI) > 35 kWh/m(3). Temperature increase during the treatment brought about most of the destruction of autofluorescence compounds, as well as a fraction of inactivation and the destruction of the plasma and thylakoid membranes. However, the forces associated with the pulsing electric field caused significan

Synechocystis mutants defective in manganese uptake regulatory system, ManSR, are hypersensitive to strong light.

High affinity transport of manganese ions (Mn2+) in cyanobacteria is carried by an ABC-type transporter, encoded by the mntCAB operon, which is derepressed by the deficiency of Mn2+. Transcription of this operon is negatively regulated by the two-component system consisting of a sensory histidine kinase ManS and DNA-binding response regulator ManR. In this study, we examined two Synechocystis mutants, defective in ManS and ManR. These mutants were unable to grow on high concentrations of manganese. Furthermore, they were sensitive to high light intensity and unable to recover after short-term

Integration of carbon and nitrogen metabolism with energy production is crucial to light acclimation in the cyanobacterium Synechocystis.

Light drives the production of chemical energy and reducing equivalents in photosynthetic organisms required for the assimilation of essential nutrients. This process also generates strong oxidants and reductants that can be damaging to the cellular processes, especially during absorption of excess excitation energy. Cyanobacteria, like other oxygenic photosynthetic organisms, respond to increases in the excitation energy, such as during exposure of cells to high light (HL) by the reduction of antenna size and photosystem content. However, the mechanism of how Synechocystis sp. PCC 6803, a cya

The impact of inorganic tin on the planktonic cyanobacterium Synechocystis aquatilis: the effect of pH and humic acid.

The influence of inorganic tin compounds on the unicellular cyanobacterium Synechocystis aquatilis was studied, and its dependence on changing pH of the surrounding medium and the presence of humic acid. Both Sn(II) and Sn(IV), used as chlorides (at the concentrations 1-10 mg litre(-1)), inhibit the growth and chlorophyll a content of the cyanobacterium cultures, but only under alkaline conditions. Generally, the observed tin toxicity increased with increase of metal concentration, time of exposure and pH value of the medium (in the range 7-9.8). Sn(II) seems to be more toxic than Sn(IV). At t

A Simple and Easy Method for RNA Extraction from the Cyanobacterium Synechocystis sp. PCC 6803.

Cyanobacteria have been widely used as model organisms in photobiochemical research and have recently been exploited as hosts in numerous pilot studies to produce valuable biochemicals via genetic and metabolic modifications. Analyzing cellular RNA is a suitable method for studying genetic changes in cells. Several methods have previously been reported for cyanobacterial RNA extraction. However, the majority of these methods rely heavily on phenol and chloroform, which are hazardous. Additionally, these methods are time-consuming and difficult to perform. Using Synechocystis sp. PCC 6803 as a

13C-metabolic flux analysis of respiratory chain disrupted strain ΔndhF1 of Synechocystis sp. PCC 6803.

Cyanobacteria are advantageous hosts for industrial applications toward achieving sustainable society due to their unique and superior properties such as atmospheric CO2 fixation via photosynthesis. However, cyanobacterial productivities tend to be weak compared to heterotrophic microbes. To enhance them, it is necessary to understand the fundamental metabolic mechanisms unique to cyanobacteria. In cyanobacteria, NADPH and ATP regenerated by linear and cyclic electron transfers using light energy are consumed by CO2 fixation in a central metabolic pathway. The previous study demonstrated that

Polyphosphate accumulation dynamics in a population of Synechocystis sp. PCC 6803 cells under phosphate overplus.

In this study, a simple and rapid DAPI-based protocol was developed and optimized to visualize polyphosphates (polyPs) in the cyanobacterium Synechocystis sp. PCC 6803. The optimum dye concentration and incubation time were determined, and formaldehyde fixation was shown to significantly improve polyP detection in Synechocystis cells. Using the developed protocol, for the first time, it was shown that 80% of Synechocystis cells under phosphate overplus were able to accumulate phosphorus as polyP 3 min after the addition of K2HPO4. After 1 h, the number of cells with polyP began to decrease, an

Extracellular laccase activity in engineered Synechocystis sp. PCC 6803 for mediator-free textile dye decolorization.

Industrial dye discharge into aquatic ecosystems poses persistent environmental and health risks, requiring sustainable and scalable remediation strategies. Laccases are versatile multicopper oxidases capable of degrading a wide spectrum of intractable dyes; however, their industrial application is often constrained by production costs and downstream processing requirements. Harnessing phototrophic microorganisms as self-sustaining enzyme production platforms offers a promising alternative. In this study, we engineered the model cyanobacterium Synechocystis sp. Improving productivity of citramalate from CO2 by Synechocystis sp. PCC 6803 through design of experiment.

BACKGROUND: Cyanobacteria have long been suggested as an industrial chassis for the conversion of carbon dioxide to products as part of a circular bioeconomy. The slow growth, carbon fixation rates, and limits of carbon partitioning between biomass and product in cyanobacteria must be overcome to fully realise this industrial potential. Typically, flux towards heterologous pathways is limited by the availability of core metabolites. Citramalate is produced in a single enzymatic step through the condensation of the central metabolites pyruvate and acetyl-CoA; improvements in citramalate product

Overexpression, isolation, and spectroscopic characterization of the bidirectional [NiFe] hydrogenase from Synechocystis sp. PCC 6803.

The bidirectional [NiFe] hydrogenase of the cyanobacterium Synechocystis sp. PCC 6803 was purified to apparent homogeneity by a single affinity chromatography step using a Synechocystis mutant with a Strep-tag II fused to the C terminus of HoxF. To increase the yield of purified enzyme and to test its overexpression capacity in Synechocystis the psbAII promoter was inserted upstream of the hoxE gene. In addition, the accessory genes (hypF, C, D, E, A, and B) from Nostoc sp. PCC 7120 were expressed under control of the psbAII promoter. The respective strains show higher hydrogenase activities c

Probing hydrogen peroxide oxidation kinetics of wild-type Synechocystis catalase-peroxidase (KatG) and selected variants.

Catalase-peroxidases (KatGs) are unique bifunctional heme peroxidases that exhibit peroxidase and substantial catalase activities. Nevertheless, the reaction pathway of hydrogen peroxide dismutation, including the electronic structure of the redox intermediate that actually oxidizes H(2)O(2), is not clearly defined. Several mutant proteins with diminished overall catalase but wild-type-like peroxidase activity have been described in the last years. However, understanding of decrease in overall catalatic activity needs discrimination between reduction and oxidation reactions of hydrogen peroxid

In vitro reactivation of in vivo ammonium-inactivated glutamine synthetase from Synechocystis sp. PCC 6803.

Glutamine synthetase from Synechocystis sp. strain PCC 6803 is inactivated by ammonium addition to cells growing with nitrate as the nitrogen source. The enzyme can be reactivated in vitro by different methods such as alkaline phosphatase treatment, but not phosphodiesterase, by raising the pH of the crude extract to values higher than 8, by increasing the ionic strength of the cell-free extract, or by preincubation with organic solvents, such as 2-propanol and ethanol. These results suggest that the loss of glutamine synthetase activity promoted by ammonium involves the non-covalent binding o

The small protein SbtC is a functional component of the CO2 concentrating mechanism in Synechocystis sp. PCC 6803.

Oxygenic phototrophs fix CO2 via the enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO), which shows relatively low CO2 affinity and specificity. To circumvent low and fluctuating CO2 concentrations in aquatic systems, cyanobacteria and algae have evolved sophisticated inorganic carbon (Ci) concentrating mechanisms (CCMs). Bicarbonate transporters such as SbtA play a crucial role in the cyanobacterial CCM and hence display multiple layers of tight regulation. Control of sbtA gene expression and corresponding transporter activity involves the PII-like protein SbtB, whose gene is f

Production of phycobiliproteins, bioplastics and lipids by the cyanobacteria Synechocystis sp. treating secondary effluent in a biorefinery approach.

Cyanobacteria have been identified as promising organisms to reuse nutrients from waste effluents and produce valuable compounds such as lipids, polyhydroxyalkanoates (PHAs), and pigments. However, almost all studies on cyanobacterial biorefineries have been performed under lab scale and short cultivation periods. The present study evaluates the cultivation of the cyanobacterium Synechocystis sp. in a pilot scale 30 L semi-continuous photobioreactor fed with secondary effluent for a period of 120 days to produce phycobiliproteins, polyhydroxybutyrate (PHB) and lipids. To this end, the harveste

Order-of-magnitude enhancement in photocurrent generation of Synechocystis sp. PCC 6803 by outer membrane deprivation.

Biophotovoltaics (BPV) generates electricity from reducing equivalent(s) produced by photosynthetic organisms by exploiting a phenomenon called extracellular electron transfer (EET), where reducing equivalent(s) is transferred to external electron acceptors. Although cyanobacteria have been extensively studied for BPV because of their high photosynthetic activity and ease of handling, their low EET activity poses a limitation. Here, we show an order-of-magnitude enhancement in photocurrent generation of the cyanobacterium Synechocystis sp. PCC 6803 by deprivation of the outer membrane, where e

Characterization of a phototolerant mutant of Synechocystis sp. PCC 6803 created by random mutagenesis of psbAII gene.

Photosensitivity and photosynthetic characteristics have been analyzed in wild type (KC) and its psbAII mutant (I6) of Synechocystis having three point amino acid substitutions, i.e., N322I, I326F and F328S, which are localized in the C-terminal extension of D1 protein of the photosystem II reaction center. Wild type and mutant cells show almost an identical growth pattern under normal/low light (30 mumol m-2s-1, 30 degrees C) liquid culture (BG-11) condition. However, upon shifting the cultures to high light (500 mumol m-2s-1, 30 degrees C), these two types of cells exhibit entirely different

Deletion of the psbG1 gene of the cyanobacterium Synechocystis sp. PCC6803 leads to the activation of the cryptic psbG2 gene.

The genes psbG1 and psbG2 in the cyanobacterium Synechocystis sp. PCC6803 are homologous. The psbG1 gene is located on the chromosome and is part of the ndhC--psbG1--ORF157 operon, while psbG2 is located on a plasmid and is not flanked by equivalent ndhC or ORF157 genes. Mutants in which psbG1 is deleted grow well under autotrophic conditions, while their growth is impeded in mixotrophic medium. These results argue against a functional role for psbG1 in photosynthesis, i.e. photosystem II, and are more compatible with a function in respiration. The psbG2 gene is not transcribed in wild-type ce

Deciphering the directed iron-stress regulatory network of Synechocystis sp. PCC 6803 reveals novel master regulators.

Iron deficiency elicits extensive transcriptional reprogramming in cyanobacteria; however, the directed regulatory hierarchy that coordinates this response remains insufficiently resolved. In this study, we integrated 34 iron-stress transcriptomic samples into a customized deep graph transformer (DGT) framework to infer a directed, context-specific gene regulatory network (GRN) for Synechocystis sp. PCC 6803 under iron limitation. This analysis identified 82 differentially expressed genes, with isiB and the antisense RNA IsrR exhibiting the strongest transcriptional shifts. The resulting GRN d

Isolation of Synechocystis Mutants Overproducing Mannitol Directly from CO2 via Laboratory Evolution under Increasing Salt Concentration.

Mannitol is a naturally occurring C(6) polyol with a wide range of applications in the food and pharmaceutical industry. In a previous study, mannitol production was achieved via the direct conversion of CO2 in Synechocystis sp. PCC6803. However, a major barrier to future applications of this strain was its low production rate. In this study, three mutants were isolated after 164 generations of adaptive laboratory evolution under salt stress. These mannitol overproducing mutants were able to produce 27.71 mg L-1 OD730-1 mannitol under 350 mM salt stress when the OD730 reached 2, roughly 24 tim
