Identification of Antibodies to Chondrocyte and Synoviocyte Antigens in Equine Osteoarthritis.

Innate immune responses within the joint are now known to play a key role in osteoarthritis (OA) pathogenesis. However, comparatively little is known regarding the role of adaptive immune responses in OA, and whether they may be important for initiating and sustaining progressive low-level joint inflammation. Therefore, we evaluated spontaneous osteoarthritis in horses to investigate whether antibodies recognizing live joint cells (chondrocytes, synoviocytes) were present in blood or synovial fluid, and to identify possible cellular target antigens. We found that horses with advanced OA had an

TLRs Play Crucial Roles in Regulating RA Synoviocyte.

Rheumatoid arthritis (RA) is an autoimmune inflammatory disease comparing the inflammation of synovium. Macrophage-like synoviocytes and fibroblast-like synoviocytes (synoviocytes) are crucial ingredients of synovium. Therein, a lot of research has focused on synoviocytes. Researches demonstrated that TLR1, TLR2, TLR3, TLR4, TLR5, TLR6 TLR7 and TLR9 are expressed in synoviocyte. Additionally, the expression of TLR2, TLR3, TLR4 and TLR5 is increased in RA synoviocyte. In this paper, we review the exact role of TLR2, TLR3, TLR4 and TLR5 participate in regulating the production of inflammatory fa

Arthritogenicity of annexin VII revealed by phosphoproteomics of rheumatoid synoviocytes.

OBJECTIVE: To identify novel proteins involved in the pathogenesis of rheumatoid arthritis (RA) and to characterise the identified proteins based on pathogenic and therapeutic aspects. METHODS: The authors applied differential phosphoproteomic analysis to articular synoviocytes between RA and osteoarthritis (OA) to identify proteins differently phosphorylated between RA and OA. Focusing on annexin VII (Anx7), one of the highly phosphorylated proteins in RA, the authors prepared Anx7-transgenic C57BL/6 (Anx7-Tg-B6) mice to evaluate their susceptibility to collagen-induced arthritis (CIA). Effects of calmodulin inhibitors on rabbit synoviocyte phospholipase A2.

The effect of calmodulin inhibitors on synoviocyte phospholipase A2 activity was evaluated. Cells were incubated with [3H]arachidonic acid after 24 hours to label phospholipids. [3H]prostaglandin E2 synthesis was stimulated by Salmonella minnesota lipopolysaccharide (100 micrograms/ml). Trifluoperazine, 35 microM, reduced lipopolysaccharide-stimulated [3H]prostaglandin E2 synthesis by 50%. In sonicated suspensions of cells, calcium-dependent phospholipase A2 activity was inhibited by trifluoperazine 3-100 microM and by compound 48/80 (3 micrograms/ml). These agents inhibit calmodulin-dependent

Fibroblast-like synoviocytes in rheumatoid arthritis: Surface markers and phenotypes.

Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease that mainly affects synovial joints. During the course of RA, the synovium transforms into hyperplastic invasive tissue, leading to cartilage and bone destruction. Fibroblast-like synoviocytes (FLS) in the synovial lining develop aggressive phenotypes and produce pathogenic mediators that lead to the occurrence and progression of disease, playing a major role in RA pathophysiology. Therefore, research on FLS has become the main focus within the RA field. With technical advances and the development of multi-omics comprehensive a

Serum amyloid A protein stimulates CCL20 production in rheumatoid synoviocytes.

OBJECTIVE: Although serum amyloid A (SAA) has been used as a marker of inflammation, its role in leucocyte recruitment and angiogenesis has not been well established in RA. CCL20 is a chemokine involved in the migration of CCR6-expressing Th17 cells. To study the contribution of SAA to the recruitment of Th17 cells, we investigated the effects of SAA on CCL20 production by RA synoviotytes. METHODS: Synoviocytes isolated from RA patients were stimulated with recombinant SAA and cellular supernatants were analysed by CCL20-specific ELISA. CCL-20 mRNA expression was analysed by RT-PCR. RESULTS:

Fibrotic pathways and fibroblast-like synoviocyte phenotypes in osteoarthritis.

Osteoarthritis (OA) is the most common form of arthritis, characterized by osteophyte formation, cartilage degradation, and structural and cellular alterations of the synovial membrane. Activated fibroblast-like synoviocytes (FLS) of the synovial membrane have been identified as key drivers, secreting humoral mediators that maintain inflammatory processes, proteases that cause cartilage and bone destruction, and factors that drive fibrotic processes. In normal tissue repair, fibrotic processes are terminated after the damage has been repaired. In fibrosis, tissue remodeling and wound healing a

Phagocytosis of monosodium urate crystals by human synoviocytes induces inflammation.

Gout is distinguished by an inflammatory process that is mediated by phagocytosis of monosodium urate (MSU) crystals in synoviocytes by regulation of unknown mechanisms. Here we suggest that the synovial cells play a crucial role in gouty arthritis by activating inflammation by MSU uptake and increasing the secretion of pro-inflammatory cytokines IL-1β, IL-6, IL-8, TNF-α, MCP-1, and the growth factors NGF and HGF. We discuss some co-existing features in synoviocytes, including anomalous morphologies of the cells, and microvesicle formation, dysregulation in VEGF gene expression. We provide evi

Montelukast inhibits inflammatory response in rheumatoid arthritis fibroblast-like synoviocytes.

Montelukast, a potent selective antagonist of cysteinyl leukotriene (cysLT) receptors, has displayed its important anti-oxidative and anti-inflammatory effects in various tissues and organs. Rheumatoid arthritis (RA) is an immune disease defined by hyperplastic synovitis and joint destruction. Fibroblast-like synoviocytes in RA (RA-FLSs) are the main cell component of the hyperplastic synovium. Whether montelukast can protect against the inflammatory milieu of RA remains unclear. Here, it is shown that cysLT1R is present in FLSs and unregulated in RA-FLSs. Montelukast has an inhibitory effect

Orexin A may suppress inflammatory response in fibroblast-like synoviocytes.

Orexins are peptides found in the hypothalamus and other peripheral tissues including adipose tissue, the endocrine cells of the gut, adrenal gland testis, and the pancreas. They play important roles in neuro-protection by inhibiting oxidative stress and inflammatory response via their type 1 and 2 receptors (OX1R and OX2R). The expressions of OX1R and OX2R were detected and the physiological function of orexin A in fibroblast-like synoviocytes (FLSs) was investigated in this study. Only OX1R was found on FLSs and the expression of OX1R was decreased in fibroblast-like synoviocytes in rheumato

Niclosamide induces apoptosis in human rheumatoid arthritis fibroblast-like synoviocytes.

To explore the effects of niclosamide on the viability and apoptosis of rheumatoid arthritis of fibroblast-like synoviocytes (rheumatoid arthritis (RA)fibroblast-like synoviocytes (FLS)), FLS obtained from RA patients were treated with niclosamide. Niclosamide significantly inhibited the viability of RA FLS in a concentration-dependent manner. Niclosamide treated FLS showed a significant increase in the percentage of apoptosis and higher intracellular ROS levels. N-acetyl-l-cysteine (NAC) pretreatment significantly attenuated niclosamide-induced apoptosis. The apoptotic response was due to the

Synoviocytes synthesize, bind, and respond to basic fibroblast growth factor.

Rheumatoid arthritis (RA) is a systemic disease characterized by the destructive proliferation of synovial tissue. It has been suggested that this proliferative lesion resembles a malignancy. Although polypeptide growth factors have been implicated in malignant cell growth, their role in the pathogenesis of proliferative but non-neoplastic diseases such as RA has not been extensively studied. We tested the hypothesis that the synoviocyte itself may be a source of growth factor activity. We demonstrated that culture supernatants from synoviocytes obtained from patients with RA, osteoarthritis,

METTL3-mediated fibroblast-like synoviocytes senescence promotes temporomandibular joint osteoarthritis progression.

Temporomandibular joint osteoarthritis (TMJOA) is a disease that affects the TMJ and is associated with degenerative changes in the articular cartilage. Fibroblast-like synoviocytes (FLSs) have been found to contribute to osteoarthritis. Here, we aim to investigate the role of METTL3-mediated FLS senescence in the TMJOA process. TMJOA model rats were successfully generated, displaying typical structural and inflammatory alterations, and primary FLSs were isolated from monosodium iodoacetate (MIA)-induced TMJOA rats; these FLSs were accompanied by increased senescence, attenuated mitophagy, and

Antiviral potential of Tualang honey in Chikungunya virus-infected human synoviocytes.

Chikungunya is a mosquito-borne viral disease caused by chikungunya virus (CHIKV), characterised by fever, polyarthralgia, myalgia, rash, and headache, with chronic arthralgia persisting for months or years. Although vaccine is recently available, it is not widely used, and treatment focuses on symptom relief. Many natural products or their active compounds have been investigated for their anti-CHIKV activities on Vero cells which is irrelevant in CHIKV pathogenesis in human. Tualang Honey has also been shown to exert anti-CHIKV in Vero cells. This study advances the evaluation of Tualang hone

Epigenetic Regulation of Nutrient Transporters in Rheumatoid Arthritis Fibroblast-like Synoviocytes.

OBJECTIVE: Since previous studies indicate that metabolism is altered in rheumatoid arthritis (RA) fibroblast-like synoviocytes (FLS), we undertook this study to determine if changes in the genome-wide chromatin and DNA states in genes associated with nutrient transporters could help to identify activated metabolic pathways in RA FLS. METHODS: Data from a previous comprehensive epigenomic study in FLS were analyzed to identify differences in genome-wide states and gene transcription between RA and osteoarthritis. We utilized the single nearest genes to regions of interest for pathway analyses

Alogliptin inhibits IL-1β-induced inflammatory response in fibroblast-like synoviocytes.

Excessive production of pro-inflammatory cytokines such as interleukin (IL)-1β plays an important role in the chronic inflammation in fibroblast-like synoviocytes (FLS) in rheumatoid arthritis (RA). Alogliptin, an important selective dipeptidyl peptidase-4 (DPP-4) inhibitor licensed for the treatment of type 2 diabetes, has displayed a wide range of pharmacological capacities. In the present study, we aimed to investigate whether alogliptin possessed a protective effect against IL-1β-induced insult in FLS. Our results indicate that alogliptin treatment ameliorated IL-1β-induced production of r

Adrenal Steroids Modulate Fibroblast-Like Synoviocytes Response During B. abortus Infection.

Brucella abortus stimulates an inflammatory immune response that stimulates the endocrine system, inducing the secretion of dehydroepiandrosterone (DHEA) and cortisol. In humans, the active disease is generally present as osteoarticular brucellosis. In previous studies we showed that B. abortus infection of synoviocytes creates a proinflammatory microenvironment. We proposed to determine the role of cortisol and DHEA on synoviocytes and infiltrating monocytes during B. abortus infection.
