T-2 toxin is an inevitable mycotoxin in food products and feeds. It is a proven toxicant impairing the male reproductive system. However, previous studies have concentrated on the toxic effect of T-2 toxin on Leydig cells, with little attention on the Sertoli cell cytotoxicity. Therefore, this study aimed to establish the toxic mechanism of T-2 toxin on Sertoli cells. The Sertoli cell line (TM4 cell) was cultured and exposed to different concentrations of T-2 toxin with/without N-acetyl-L-cysteine (NAC) for 24 h. A CCK-8 assay then measured the cell viability. In addition, the expression of TM

T-2 toxin, a mycotoxin produced by Fusarium species, has been shown to cause diverse toxic effects in animals and is also a possible pathogenic factor of Kashin-Beck disease (KBD). The role of mitochondria in KBD is recognized in our recent research. The aim of this study was to evaluate the role of mitochondria in T-2 toxin-induced human chondrocytes apoptosis to understand the pathogenesis of KBD. T-2 toxin decreased chondrocytes viabilities in concentration- and time-dependent manners. Exposure to T-2 toxin can reduce activities of mitochondrial complexes III, IV and V, ΔΨm and the cellular

Purified T-2 toxin was fed to S.C.W.L. hens at levels of 0 (control), 0.5, 1.0, 2.0, 4.0 and 8.0 p.p.m. of an otherwise balanced diet. Feed consumption, egg production and shell thickness were significantly (P less than 0.05) decreased in hens fed 8 p.p.m. as compared with control hens. The fertility and progeny performance were not depressed by feeding T-2 toxin, but the hatchability of fertile eggs of hens fed 2 and 8 p.p.m. was significantly (P less than 0.05) lower than that of hens fed the control diet. The weights of liver, heart, gizzard and spleen were not influenced by T-2 toxin. Seru

Cutaneous absorption and decontamination of [3H]T-2 mycotoxin using various treatment modalities incorporating water, detergent, sprays, and scrubbing of application sites were examined in the rat model at 5, 30, 60, and 1440 min (24 h) postexposure. Rats were killed immediately after treatment and radiolabeled T-2 remaining in full-thickness skin samples were determined. Absorption and decontamination were followed over time, and decontaminating treatment modalities were evaluated for efficacy. Less than 1% of the applied dose was absorbed in 5 min, and 50% was absorbed in 24 h. At 5 min, 99.

Trichothecene mycotoxins commonly contaminate cereal grains and are often linked to human and animal food poisoning. The rapid onset of anorexia is a common hallmark of trichothecenes-induced toxicity. Although the neurotransmitters 5-hydroxytryptamine (5-HT) and substance P (SP) are known to regulate appetite, it remains unknown whether these two neurotransmitters are involved in type A trichothecenes-induced anorectic response. The goal of this study is to relate plasma 5-HT and SP to anorectic responses induced by type A trichothecenes T-2 toxin (T-2), HT-2 toxin (HT-2), diacetoxyscirpenol

T-2 toxin, a mycotoxin that frequently causes hidden contamination in food and animal feed, poses a substantial threat to both human and animal health. Staphylococcus saprophyticus (S. saprophyticus) is an opportunistic pathogen that widely infects humans and various animals. However, the specific conditions under which it becomes pathogenic, as well as the mechanisms underlying its pathogenicity remain unknown. In this study, it is found that a sub-cytotoxic dose of T-2 toxin in piglet and mouse models promotes the proliferation of intestinal S. saprophyticus and facilitates its translocation

As one of the most common worldwide contaminants in agricultural production, the T-2 mycotoxin is commonly found in moldy feed and its raw materials. It can slow the growth and suppress the immune function of farm animals, resulting in reduced economic benefits. As a feed additive, sodium butyrate can enhance immune function. However, the toxicological effects of the T-2 toxin on the spleen, thymus, and bursa of Fabricius and the protective mechanism of sodium butyrate against the T-2 toxin in quails are not known. In this study, 1-day-old Korean quails were fed either with T-2 toxin (0.9 mg/k

The objective of the present study was to review the potential protective effects of Se against T-2 toxin-induced adverse effects in cartilage and other tissues as well as to discuss the potential molecular mechanisms by which Se counteracts T-2 toxicity. Laboratory studies demonstrate that Se attenuates T-2 toxin-induced chondrocyte death by inhibition of the mitochondrial pathway of apoptosis. Protective effects of Se against T-2 toxin-induced oxidative stress in chondrocytes are mediated by improvement of antioxidant selenoprotein expression, which is altered upon mycotoxin exposure. In add

T-2 toxin, a natural secondary sesquiterpenoid metabolite produced by numerous strains of Fusarium fungi, is prevalent in both contaminated food and the environment. T-2 toxin is known to be highly toxic to the cardiovascular system, but the precise mechanisms that lead to T-2 toxin-induced cardiotoxicity are not yet fully understood. Recent findings indicate that ferroptosis is a pivotal factor in cardiovascular damage and exhibits a strong correlation with the detrimental impacts of T-2 toxin. The present study was designed to examine the involvement of ferroptosis in T-2 toxin-induced cardi

T-2 toxin, a trichothecene mycotoxin, is a common contaminant in food and animal feed, and is also present in processed cereal products. The most common route of T-2 toxin exposure in humans is through dietary ingestion. The cytotoxic effects of T-2 toxin include modifications to feeding behavior, nervous disorders, cardiovascular alterations, immunosuppression, and hemostatic derangements. However, to date, effects on the central nervous system (CNS) have rarely been reported. In the present study, female Wistar rat were given a single dose of T-2 toxin at 2 mg/kg b.w. and were sacrificed at

The role of T-2 toxin-induced gastrointestinal lesions in T-2 toxin-enhanced resistance to listeriosis in mice was evaluated. The T-2 toxin-induced lesions did not cause a starvation effect sufficient to enhance resistance to listeriosis. Administration of polymyxin E markedly reduced the gram-negative intestinal microflora and did not eliminate the toxin-induced resistance to listeriosis. The T-2 toxin did not cause an increased expression of Ia surface antigens on peritoneal macrophages. Thus, toxin-induced anorexia and starvation or absorption of gram-negative intestinal bacteria and endoto

The modulating effect of preinoculation and postinoculation treatment with a single oral 4.0 mg/kg dosage of T-2 toxin on cell-mediated resistance was studied in mice inoculated with Listeria monocytogenes. Toxin treatment caused significant decreases in thymus and spleen weights, bone marrow cellularity, and in the total number of circulating leukocytes, lymphocytes, and neutrophils. Enhancement or suppression of resistance to listeriosis was dependent on the time of toxin administration relative to the time of Listeria challenge. Preinoculation treatment on Day 2 or 4 prior to Listeria chall

Contamination of feed with mycotoxins has become a severe issue worldwide. Among the most prevalent trichothecene mycotoxins, T-2 toxin is of particular importance for livestock production, including poultry posing a significant threat to animal health and productivity. This review article aims to comprehensively analyze the pathological consequences, metabolism, and toxic effects of T-2 toxin in poultry. Trichothecene mycotoxins, primarily produced by Fusarium species, are notorious for their potent toxicity. T-2 toxin exhibits a broad spectrum of negative effects on poultry species, leading

l-arginine is beneficial for reproductive health; however, whether l-arginine may confer protection against T-2 toxin-induced reproductive impairment is not known. To address this, we used a mice model treated with T-2 toxin to investigate protective effects of l-arginine. Experimentally, we pre-treated mice with designed diet of l-arginine supplementation prior to the T-2 toxin-injected intraperitoneally exposure and then assessed semen quality, fertility and serum testosterone concentration. The results showed that l-arginine improved semen quality (e.g., live spermatozoa, abnormal spermatoz

T-2 toxin and other trichothecene mycotoxins experimentally impair normal immune function and may predispose humans and animals to infectious disease. In this study, the histopathologic effects of Salmonella typhimurium challenge concurrently with sublethal T-2 toxin exposure were examined in the Salmonella-resistant C3H/HeN mouse. Oral administration of T-2 toxin (1 mg/kg) every other day for 10 d had little effect on the tissues examined when compared to control animals. Mice challenged with S. typhimurium and then treated with T-2 toxin every other day for 10 d had markedly larger and more

Changes in hematopoietic and lymphoid tissues of young Swiss mice fed a balanced semipurified diet containing T-2 toxin (20 ppm) were examined after one, two, three, four or six weeks. During the first three weeks of exposure of T-2 toxin, lymphoid tissues, bone marrow and splenic red pulp became hypoplastic, resulting in anemia, lymphopenia and eosinopenia. Subsequently, during continued exposure to T-2 toxin, hematopoietic cells regenerated in bone marrow and splenic red pulp and became hyperplastic by six weeks. Granulopoiesis and thrombopoiesis resumed in advance of erythropoiesis. All lym

T-2 toxin, a potent mycotoxin produced by Fusarium species, is recognized for its neurotoxic, hepatotoxic, and reproductive toxic effects. However, its impact on chondrocyte differentiation and cartilage development is not well understood. This study combines transcriptomic and metabolomic analyses to investigate the impact of T-2 toxin on ATDC5 cells during differentiation at days 3, 7, 14, and 21, following 48 h of exposure. Additionally, real-time quantitative PCR (RT-qPCR) was used to evaluate the expression levels of key chondrocyte differentiation-related genes (e.g., Fn1, Sox9, Runx2, A

Growing evidence has revealed the deleterious influence of environmental and food contaminants on puberty onset and development in both animals and children, provoking an increasing health concern. T-2 toxin, a naturally-produced Type A trichothecene mycotoxin which is frequently found in cereal grains and products intended for human and animal consumption, has been shown to impair the reproduction and development in animals. Nevertheless, whether this trichothecene mycotoxin can disturb the onset of puberty in females remains unclear. To clarify this point, infantile female rats were given a
