CD86 immunohistochemical staining for the detection of Talaromyces marneffei in lesions.

Talaromycosis is an invasive fungal disease caused by the pathogenic thermodimorphic fungus Talaromyces marneffei (TM), which is often overlooked in tropical and subtropical regions of Asia. In view of the diversity of clinical manifestations in patients with TM infection, early diagnosis remains challenging. We assessed the sensitivity and specificity of a novel immunohistochemical staining by performing CD86 immunohistochemical staining on 56 tissue sections from patients with talaromycosis who had fungal culture or metagenomic next-generation sequencing confirmed to exist in clinical specim

Fatal Talaromyces marneffei Infection in a Patient with Autoimmune Hepatitis.

Talaromyces marneffei, previously known as Penicillium marneffei, is the most important pathogenic thermally dimorphic fungus causing systemic mycosis in Southeast Asia. Traditionally, T. marneffei infection in human was mainly associated with acquired immunodeficiency syndrome caused by HIV infection. In recent years, there has been an increasing number of T. marneffei infections reported in non-HIV-infected patients with other immunocompromised conditions, including autoantibodies against interferon-gamma, systemic lupus erythematosis, solid organ transplantation, Job's syndrome, hematologic

Allergic bronchopulmonary mycosis associated with Talaromyces boninensis: A case report.

Allergic bronchopulmonary aspergillosis is a chronic inflammatory pulmonary disorder characterized by type I and III hypersensitivity reactions to Aspergillus fumigatus in the airways of patients with asthma or cystic fibrosis. However, other filamentous fungi can cause similar pathophysiological changes, a condition referred to as allergic bronchopulmonary mycosis (ABPM). Data on ABPM involving non-Aspergillus fungi remain limited. To the best of our knowledge, we present the first case of ABPM associated with Talaromyces boninensis. The patient met the diagnostic criteria proposed by the Jap

Anti-inflammatory polyketides from the endophytic fungus Talaromyces aculeatus A884.

Chemical investigation on the EtOAc extract of endophytic fungus Talaromyces aculeatus A884 led to the isolation of one undescribed γ-butyrolactone aculeatone A (1), one rubralactone aculeatone B (2), and one polyester aculeatone C (5) together with 12 known compounds. Their structures were unambiguously elucidated by extensive spectral analyses. The absolute configurations of aculeatones A-B were assigned by the comparison of their experimental and calculated ECD data. In addition, all of the isolates (1-15) were evaluated for their anti-inflammatory activities by measuring the inhibition of

PANoptosis-mediated mechanisms underlying AST elevation in Talaromyces marneffei infection.

BACKGROUND: Talaromyces marneffei (T. marneffei), a life-threatening opportunistic fungal pathogen, is endemic to Southeast Asia. Although elevated aspartate aminotransferase (AST) levels are commonly observed in infected individuals, the origin and mechanism of this phenomenon remain unclear. This study aimed to determine whether AST is a specific clinical indicator of T. marneffei infection and to investigate the underlying mechanisms associated with tissue damage and cell death. METHODS: We retrospectively analyzed clinical and laboratory data of HIV/AIDS patients with or without T. marnef

Emergence of Next-Generation Sequencing for Laboratory Diagnosis of Talaromyces marneffei.

The first case of Talaromyces marneffei infection diagnosed by next-generation sequencing (NGS) was reported in 2018. By 31st December 2024, the number of T. marneffei infections picked up by NGS has increased to a total of at least 241. Most cases were reported from China, reflecting both the regional importance of this infection and its widespread use of NGS for laboratory diagnosis of infectious diseases. Among the cases with clinical details reported, 136 were HIV-negative, which was probably the main reason why T. marneffei was not suspected and the diagnosis was subsequently made with th

Pulmonary infection caused by Talaromyces purpurogenus in a patient with multiple myeloma.

A 66-year-old female patient with multiple myeloma (MM) was admitted to the emergency service on 29.09.2014 with an inability to walk, and urinary and faecal incontinence. She had previously undergone autologous bone marrow transplantation (ABMT) twice. The patient was hospitalized at the Department of Haematology. Further investigations showed findings suggestive of a spinal mass at the T5-T6-T7 level, and a mass lesion in the iliac fossa. The mass lesion was resected and needle biopsy was performed during a colonoscopy. Examination of the specimens revealed plasmacytoma. The patient also had

An Updated Taxonomy of Talaromyces (Trichocomaceae, Eurotiales): New Series and Species.

Species of Talaromyces affect human societies in many different ways. Infrageneric classifications of the genus at series level had been established in only two sections, Subinflati and Trachyspermi. In this study, phylogenies of Talaromyces were reconstructed section by section based on separate or concatenated multi-locus datasets: beta-tubulin (BenA), calmodulin (CaM) and RNA polymerase II second-largest subunit (RPB2). Fifty series belonging to nine sections were accordingly recognized, i.e., one in sections Brunneospori and Tenues, two in sections Bacillispori and Helici, three in sect. S

Genomic analysis provides insights into the transmission and pathogenicity of Talaromyces marneffei.

Talaromyces marneffei (T. marneffei) is a medically important opportunistic dimorphic fungus that infects both humans and bamboo rats. However, the mechanisms of transmission and pathogenicity of T. marneffei are poorly understood. In our study, we combined Illumina and PacBio sequencing technologies to sequence and assemble a complete genome of T. marneffei. To elucidate the transmission route and source, we sequenced three additional T. marneffei isolates using Illumina sequencing technology. Variations among isolates were used to develop a multilocus sequence typing (MLST) system comprising
