Quantitative taste bud distribution in the hamster.

The distribution of taste buds was examined in hamsters to reveal the sources of convergent neural activity on taste neurons of the medulla and pons. A total of 723 taste buds was found in adult animals. Fungiform papillae contained 130 taste buds (18%). A single vallate papilla had 168 taste buds (23%). There were 230 taste buds (32%) located on bilateral foliate papillae. The palate contained 100 taste buds (14%) divided among the soft palate (88, 12%) and the incisal papilla (12, 2%). On the epiglottis and adjacent region of the esophagus were found 70 taste buds (10%). About 10 taste buds

Lineage tracing of Twist2-expressing cells in mouse taste buds.

OBJECTIVES: It is widely accepted that Taste bud cells originate from the epithelium; however, evidence indicates that these cells are also derived from the mesenchyme beneath the epithelium. In this study, the cell lineage expressing Twist2, a transcription factor that is specifically expressed in the mesenchyme within taste buds, were examined to gain a deeper understanding of its biological characteristics. METHODS: Immunohistochemical analyses and taste organoid cultures were performed using Twist2 lineage-tracing reporter mice (Twist2-Cre mice crossed with tdTomato mice) to identify Twis

Cyclophosphamide-Induced Inflammation of Taste Buds and Cytoprotection by Amifostine.

Taste buds in the oral cavity have a complex immune system regulating normal functions and inflammatory reactions. Cyclophosphamide (CYP), a chemotherapy drug, has wide-ranging disruptive effects on the taste system including loss of taste function, taste sensory cells, and capacity for taste cell renewal. In bladder epithelium, CYP also induces inflammation. To determine if CYP induces inflammation in taste buds, we used immunohistochemistry to examine tumor necrosis factor alpha (TNF-α) (a proinflammatory cytokine) expression over a 72-hour period. Expression of TNF-α increased in a subset o

Subepithelial nerve plexus (with ganglion cells) associated with taste buds.

OBJECTIVE: The purpose of this investigation was to confirm the presence of subepithelial nerve plexuses associated with taste buds in the human tongue and to study the histomorphologic features of each plexus. STUDY DESIGN: Through use of a light microscope, several hundred microslides representing biopsies of the human tongue were reviewed so that the histomorphologic character of subepithelial nerve plexuses could be detected and studied. Tissue sections were stained with hematoxylin-erythrosin, Bodian, and Masson's trichrome, as well as with immunohistochemical stains for neuron-specific

Taste bud distribution in the rat pharynx and larynx.

The present study evaluated the number and distribution of taste buds in the rat pharynx and larynx using light microscopic techniques. The average number of taste buds in this region was 141.9 (S.D. = 29.4, n = 10). Pharyngeal and laryngeal taste buds could be grouped into three subpopulations: those associated with laryngeal structures (LA), the nasopharynx (NP), or the palatopharyngeal eminence (PE). Laryngeal taste buds were most numerous (mean = 103.2, S.D. = 23.6). They were observed on the laryngeal surface of the epiglottis and extended caudally along the aryepiglottic folds, reaching

Taste bud homeostasis in health, disease, and aging.

The mammalian taste bud is an onion-shaped epithelial structure with 50-100 tightly packed cells, including taste receptor cells, supporting cells, and basal cells. Taste receptor cells detect nutrients and toxins in the oral cavity and transmit the sensory information to gustatory nerve endings in the buds. Supporting cells may play a role in the clearance of excess neurotransmitters after their release from taste receptor cells. Basal cells are precursor cells that differentiate into mature taste cells. Similar to other epithelial cells, taste cells turn over continuously, with an average li

Mouse Mandibular Retromolar Taste Buds Associated With a Mucus Salivary Gland.

We have characterized a recently rediscovered chemosensory structure at the rear of the mandibular mucosa in the mouse oral cavity originally reported in the 1980s. This consists of unorganized taste buds, not contained within troughs, associated with the ducts of an underlying minor salivary gland. Using whole-mount preparations of transgenic mice expressing green fluorescent protein under the promoter of taste-signaling-specific genes, we determined that the structure contains taste bud clusters and salivary gland orifices at the rear of each mandible, distal to the last molar and anterior t

Ontogeny and innervation of taste buds in mouse palatal gustatory epithelium.

We investigated the relationship between mouse taste bud development and innervation of the soft palate. We employed scanning electron microscopy and immunohistochemistry using antibodies against protein gene product 9.5 and peripherin to detect sensory nerves, and cytokeratin 8 and α-gustducin to stain palatal taste buds. At E14, nerve fibers were observed along the medial border of the palatal shelves that tracked toward the epithelium. At E15.5, primordial stages of taste buds in the basal lamina of the soft palate first appeared. At E16, the taste buds became large spherical masses of colu

[Research progress in functional regeneration methods and mechanisms of taste buds].

Gustation is one of the most important human senses. Taste dysfunctions, which may be due to aging, tongue cancer surgery, radiotherapy and chemotherapy, affect life quality. That is why the need for taste bud regeneration has received more attention. At present, research on development and renewal of taste cells provides a basis for taste bud regeneration; molecular mechanisms related to taste bud regeneration are being continuously uncoverd, aiding in the identification of more accurate targets for therapy. New methods such as nerve regeneration, tissue engineering, and cytokine therapy have

Type II/III cell composition and NCAM expression in taste buds.

Taste buds are localized in fungiform (FF), foliate (FL), and circumvallate (CV) papillae on the tongue, and taste buds also occur on the soft palate (SP). Mature elongate cells within taste buds are constantly renewed from stem cells and classified into three cell types, Types I, II, and III. These cell types are generally assumed to reside in respective taste buds in a particular ratio corresponding to taste regions. A variety of cell-type markers were used to analyze taste bud cells. NCAM is the first established marker for Type III cells and is still often used. However, NCAM was examined

Postnatal development of palatal and laryngeal taste buds in the hamster.

Mammalian taste buds are distributed within several distinct subpopulations, innervated by branches of three cranial nerves. These taste bud populations originate and mature at different times in various mammalian species and are thought to play differential roles in the control of taste-mediated behaviors. The hamster is a common animal for the electrophysiological study of the gustatory system, and it has been shown that taste buds innervated by the IXth nerve develop postnatally in this species. To delineate further the development of the gustatory system of hamsters, we quantified the numb

Cyclophosphamide induces the loss of taste bud innervation in mice.

Many common chemotherapeutics produce disruptions in the sense of taste which can lead to loss of appetite, nutritional imbalance, and reduced quality of life, especially if taste loss persists after treatment ends. Cyclophosphamide (CYP), an alkylating chemotherapeutic agent, affects taste sensitivity through its cytotoxic effects on mature taste receptor cells (TRCs) and on taste progenitor cell populations, retarding the capacity to replace TRCs. Mechanistic studies have focused primarily on taste cells, however, taste signaling requires communication between TRCs and the gustatory nerve fi

The Obese Taste Bud study: Objectives and study design.

AIMS: Taste modifies eating behaviour, impacting body weight and potentially obesity development. The Obese Taste Bud (OTB) Study is a prospective cohort study launched in 2020 at the University of Leipzig Obesity Centre in cooperation with the HI-MAG Institute. OTB will test the hypothesis that taste cell homeostasis and taste perception are linked to obesity. Here, we provide the study design, data collection process and baseline characteristics. MATERIALS AND METHODS: Participants presenting overweight, obesity or normal weight undergo taste and smell tests, anthropometric, and taste bud d

Establishment of a Three-Dimensional In Vitro Model of Taste Bud Cells for Fat Taste Investigation.

Fat taste is emerging as a sixth primary taste modality, with CD36 identified as a key receptor for oral detection of long-chain fatty acids. Tastants bind to receptors on taste bud cells (TBCs), triggering intracellular calcium mobilization, neurotransmitter release, activation of gustatory afferent fibers, and transmission of sensory information to the brain. Conventional two-dimensional (2D) culture systems fail to reproduce the complex three-dimensional (3D) architecture and cell-to-cell interactions essential in vivo. Here, we developed a 3D model of mouse TBCs to investigate the cellular

Insulin Is Transcribed and Translated in Mammalian Taste Bud Cells.

We and others have reported that taste cells in taste buds express many peptides in common with cells in the gut and islets of Langerhans in the pancreas. Islets and taste bud cells express the hormones glucagon and ghrelin, the same ATP-sensitive potassium channel responsible for depolarizing the insulin-secreting β cell during glucose-induced insulin secretion, as well as the propeptide-processing enzymes PC1/3 and PC2. Given the common expression of functionally specific proteins in taste buds and islets, it is surprising that no one has investigated whether insulin is synthesized in taste

Evidence that taste buds are not present on the human adult uvula.

We conducted a study to investigate whether taste buds are present on the human adult uvula. Our impetus was to determine whether surgical procedures that involve removal of the uvula can affect taste perception. Five human uvulae were removed via a modified carbon dioxide laser-assisted uvulopalatoplasty in an outpatient office setting. The uvulae were serially sectioned and stained with a solution specific for membrane-bound calcium-modulated adenosine triphosphatase, a high concentration of which is found in taste receptors. Examination of the stained sections under light microscopy failed

Maturation of taste buds on the soft palate of the postnatal rat.

Taste bud distribution on the soft palate and within three types of tongue papillae (fungiform, foliate, and circumvallate) were examined histologically in the rat at different postnatal ages. After paraffin embedding, serial sections (10 microm) were made and stained by HE, and digitized images of each section were examined. The existence of a taste pore was used to identify mature taste buds. At birth, 53% (68 of 127 observed) of the taste buds on the soft palate, but only 14% (14 of 110 observed) within fungiform papillae, contained a taste pore. One week after birth, the number of mature t
