Tongue-on-a-Chip: Parallel Recording of Sweet and Bitter Receptor Responses to Sequential Injections of Pure and Mixed Sweeteners.

A microfluidic tongue-on-a-chip platform has been evaluated relative to the known sensory properties of various sweeteners. Analogous metrics of typical sensory features reported by human panels such as sweet taste thresholds, onset, and lingering, as well as bitter off-flavor and blocking interactions were deduced from the taste receptor activation curves and then compared. To this end, a flow cell containing a receptor cell array bearing the sweet and six bitter taste receptors was transiently exposed to pure and mixed sweetener samples. The sample concentration gradient across time was sepa

Beyond flavor perception: The role of the taste receptor family in immunity.

Taste receptor family 1 (T1R) and taste receptor family 2 (T2R) are G protein-coupled receptors (GPCRs) responsible for sweet, umami, and bitter taste perception. Their widespread ectopic expression of T1Rs and T2Rs in extraoral tissues suggest important physiological functions beyond taste. However, their roles in immune regulation and mucosal immunity have not been comprehensively summarized. We aimed to review the current evidence on the roles of T1Rs and T2Rs in immune cell function, respiratory and intestinal mucosal immunity, and disease susceptibility. Relevant studies investigating the

Combining molecular simulations and bitter receptor modelling reveals the bitter-taste mechanism of walnut protease hydrolysates.

Walnut protein hydrolysates (WPHs) have attracted considerable attention owing to their functional properties. However, the enzymatic hydrolysis process frequently leads to the formation of bitter peptides, which impart an undesirable taste to the hydrolysates, consequently limiting their applications. To elucidate the mechanisms underlying bitterness perception, this study used a combined computational and experimental approach to systematically explore bitter peptides derived from walnut proteins and their interaction mechanisms with bitter-taste receptors (type 2 taste receptors, T2Rs). Pathophysiology and possible treatments for olfactory-gustatory disorders in patients affected by COVID-19.

SARS-CoV-2 infects host cells mainly through the interaction between the virus's Spike protein and the viral receptors namely Angiotensin-Converting Enzyme 2 (ACE2) and transmembrane serine protease 2 (TMPRSS2). Both are highly expressed in the gastrointestinal tract, in the nasal and bronchial epithelium, as well as in the type II alveolar epithelial cells. The aim of this review is to report the evidences from the scientific literature on the pathophysiology and the available treatments for olfactory-gustatory disorders in patients with COVID-19. The mechanisms involved in these disorders ar

[Healing process of taste receptor disturbance].

Patients with taste receptor disturbance were treated with zinc dosage, and their healing process was traced through the filter-paper-disc method and electrogustometry. The number of cases studied was 119,45 with idiopathic taste disturbance, 38 with drug-induced taste disturbance and 36 with zinc-deficient taste disturbance. 1) Three patterns of taste recovery were observed on the tongue surface. Out of a total of 119 cases, 54 cases (45.4%) were found to be improving on the anterior as well as posterior parts of the tongue simultaneously (simultaneous improvement type). Fifty-three cases (44

Fenofibrate and Impaired Taste Perception in Type 2 Diabetes.

BACKGROUND Although reduced sweet taste perception has been found in studies of clofibrate in healthy volunteers, this phenomenon has not been reported for the chemically related and more widely used drug fenofibrate. CASE REPORT A 65-year-old woman with insulin-treated type 2 diabetes was initiated on fenofibrate for worsening diabetic retinopathy. She subsequently developed a marked loss of sweet taste perception. After 3 months of fenofibrate, her glycemic control had improved and her insulin requirements had decreased, probably as a result of anorexia. Her renal function had also worsened.

Role of the bitter taste receptor T2R38 in upper respiratory infection and chronic rhinosinusitis.

PURPOSE OF REVIEW: Taste receptor family 2 (T2R) bitter taste receptors were originally identified and named on the basis of their role in type 2 taste cells of the tongue, in which they serve to detect the presence of potentially harmful ingested chemicals. In 2009, researchers demonstrated that airway epithelial cells also express T2R receptors, but their role in airway physiology and human disease has only recently begun to be identified. RECENT FINDINGS: Recent research has demonstrated that at least one airway T2R receptor, taste receptor family 2 isoform 38 protein (T2R38) is activated

Functional Alteration and Differential Expression of the Bitter Taste Receptor T2R38 in Human Paranasal Sinus in Patients with Chronic Rhinosinusitis.

The bitter taste receptors (T2Rs) expressed in human sinonasal mucosae are known to elicit innate immune responses involving the release of nitric oxide (NO). We investigated the expression and distribution of two T2Rs, T2R14 and T2R38, in patients with chronic rhinosinusitis (CRS) and correlated the results with fractional exhaled NO (FeNO) levels and genotype of the T2R38 gene (TAS2R38). Using the Japanese Epidemiological Survey of Refractory Eosinophilic Chronic Rhinosinusitis (JESREC) phenotypic criteria, we identified CRS patients as either eosinophilic (ECRS, n = 36) or non-eosinophilic

Depletion of bitter taste transduction leads to massive spermatid loss in transgenic mice.

Bitter taste perception is an important sensory input warning against the ingestion of toxic and noxious substances. Bitter receptors, a family of ~30 highly divergent G-protein-coupled receptors, are exclusively expressed in taste receptor cells that contain the G-protein α-subunit gustducin, bind to α-gustducin in vitro, and respond to bitter tastes in functional expression assays. We generated a taste receptor type 2 member 5 (T2R5)-Cre/green fluorescent protein reporter transgenic mouse to investigate the tissue distribution of T2R5. Our results showed that Cre gene expression in these mic

Bitter Taste Receptors in Bacterial Infections and Innate Immunity.

BACKGROUND: Bitter taste receptors (TAS2Rs), originally identified for their role in gustation, are now recognized for their functions in extraoral tissues, particularly in innate immune responses. TAS2Rs detect bacterial quorum sensing molecules (QSMs) and other metabolites, enabling the host to sense and respond to pathogenic threats across mucosal surfaces. OBJECTIVE: This review synthesizes current knowledge of TAS2Rs in the context of bacterial infection, emphasizing their mechanisms of immune modulation, genetic polymorphisms, tissue-specific expression, and therapeutic potential. Mechanisms and novel therapeutic roles of bitter taste receptors in diseases.

Bitter taste receptors (TAS2R) are expressed in the oral cavity, intestine, airways, and vascular smooth muscle, where they regulate physiological processes, including immune responses. However, the activation of TAS2R triggers signaling pathways that influence inflammation, metabolism, and cell proliferation, suggesting their potential as therapeutic targets for diseases such as Alzheimer's disease, Parkinson's disease, asthma, and cancer. Bitter compounds capable of activating TAS2R have shown potential in modulating these pathways, presenting novel opportunities for drug development. This r

Bitter Taste Receptors TAS2R8 and TAS2R10 Reduce Proton Secretion and Differentially Modulate Cadmium Uptake in Immortalized Human Gastric Cells.

Beyond sensing bitter-tasting compounds, bitter taste receptors (TAS2Rs) have been demonstrated to play a functional role in proton secretion as a key mechanism of gastric acid secretion (GAS) and the cellular uptake of the zinc metal ion. Given its chemical similarity and comparable effects in GAS, we focused this work on cadmium and hypothesized that gastric TAS2Rs are involved in (i) cadmium-induced inhibition of proton secretion and (ii) in its cellular uptake. To test this hypothesis, immortalized human parietal HGT-1 cells were exposed to 62.5-1000 µM CdCl2 for 30 min to elucidate TAS2R-

Polycystic kidney disease 2-like 1 channel contributes to the bitter aftertaste perception of quinine.

Bitterness is an important physiological function in the defense responses to avoid toxic foods. The taste receptor 2 family is well known to mediate bitter taste perception in Type II taste cells. Here, we report that the polycystic kidney disease 2-like 1 (PKD2L1) channel is a novel sensor for the bitter aftertaste in Type III taste cells. The PKD2L1 channel showed rebound activation after the washout of quinine, a bitter tastant, in electrophysiological whole-cell recordings of the PKD2L1-expressing HEK293T cells and Ca2+-imaging analysis of Type III taste cells isolated from wild-type PKD2

Intracellular and dual-site inhibition of a bitter taste GPCR.

Bitter taste receptors (TAS2Rs) are G protein-coupled receptors expressed in both gustatory and extraoral tissues and activated by a broad range of compounds. TAS2R14 is among the most promiscuous members of this family, responding to many structurally diverse ligands. Cryo-electron microscopy structures of TAS2R14 have revealed agonists binding in an intracellular pocket, raising the question of the main sites of interaction for known TAS2R14 antagonists. To address this, we examined the effects of mutations at residues located in the extracellular and intracellular regions on receptor inhibi

How sensory experiences shape our eating behaviour: evidence for individual taste differences affecting weight management.

The modern surge in obesity can be seen as evidence that food choice remains an important area of study. Sensory science has also provided a necessary contribution to this area. This review will describe the latest scientific understanding on flavour perception and how metabolism, weight management, exercise and food design might affect this perception. Taste research has shifted from sensory testing paradigms to a receptor level scientific understanding of gustation with the discovery of the heterodimeric taste receptor type 1 proteins (T1R) for sweet and umami taste, and the family of taste

Cucurbitacin B Induces Hypoglycemic Effect in Diabetic Mice by Regulation of AMP-Activated Protein Kinase Alpha and Glucagon-Like Peptide-1 via Bitter Taste Receptor Signaling.

Taste receptors exist in several organs from tongue to colon and have diverse functions dependent on specific cell type. In enteroendocrine L-cells, stimulation of taste receptor signaling induces incretin hormones. Among incretin hormones, glucagon-like peptide-1 (GLP-1) induces insulinotropic action by activating GLP-1 receptor of pancreatic β-cells. However, GLP-1 mimetic medicines have reported clinical side effects, such as autoimmune hepatitis, acute kidney injury, pancreatitis, and pancreatic cancer. Here, we hypothesized that if natural components in ethnomedicines can activate agonist

The bitter end: T2R bitter receptor agonists elevate nuclear calcium and induce apoptosis in non-ciliated airway epithelial cells.

Bitter taste receptors (T2Rs) localize to airway motile cilia and initiate innate immune responses in retaliation to bacterial quorum sensing molecules. Activation of cilia T2Rs leads to calcium-driven NO production that increases cilia beating and directly kills bacteria. Several diseases, including chronic rhinosinusitis, COPD, and cystic fibrosis, are characterized by loss of motile cilia and/or squamous metaplasia. To understand T2R function within the altered landscape of airway disease, we studied T2Rs in non-ciliated airway cell lines and primary cells. Several T2Rs localize to the nucl

Menstrual cycle phase and sex influence muscle glycogen utilization and glucose turnover during moderate-intensity endurance exercise.

Based on the molecular findings that many bitter taste receptors (T2Rs) are expressed within the same receptor cells, it has been proposed that bitter taste is encoded by the activation of discrete neural elements. Here we examined how a variety of bitter stimuli are represented by neural activity in central gustatory neurons. Taste responses (spikes/s) evoked by bathing the tongue and palate with intensity-matched concentrations (in M) of 2 sugars (0.32 sucrose and 0.5 D-fructose), ethanol (40%), 4 salts (0.01 NaCl, 0.008 NaNO(3), 0.01 MgCl(2), and 0.05 KCl), 2 acids (0.003 HCl and 0.005 citr

Effects of chronic unpredictable mild stress-induced depression on bitter taste receptor expression in mice.

OBJECTIVE: With the rapid increase in the pace of life, people are facing increasing pressures of all kinds, and depression has gradually become a serious psychological disorder in human society, strongly affecting normal social and physiological activities. Depression can disrupt an individual's taste perception and potentially result in taste disorders by affecting and altering taste receptors. This disruption can consequently impact their food preferences and overall eating experiences. DESIGN: In this study, we used the chronic unpredictable mild stress (CUMS) method to establish a depres

Postoral receptors and alimentary taste: implications for energy intake and appetite.

Eating behavior is shaped by genetic, psychological, and physiological factors, with nutrient sensing playing a central role in modulating intake. The tongue, as the primary gustatory organ, initiates this process by influencing hedonic preferences, food choices, and feeding behavior. Recent sensory research has highlighted the potential role of an emerging class of taste modalities known as alimentary tastes. This concept refers to the gustatory detection of compounds that produce weak or subtle taste perceptions but elicit strong postoral effects. While most studies have focused on umami and
