Telocytes are stromal cells defined by extremely long, moniliform prolongations termed telopodes and have been described in most mammalian organs since their formal designation in 2010. During the past two decades, the field has expanded from ultrastructural organ-mapping to hypotheses concerning stem-cell niche regulation, extracellular-vesicle-mediated communication, fibrosis, inflammation, and cancer-associated stromal remodelling. This expansion, however, has also generated methodological heterogeneity, with frequent reliance on non-specific markers such as CD34, PDGFRα, vimentin, and c-ki

The pancreas is a crucial digestive and endocrine organ, yet the histology of the animal pancreas remains incompletely elucidated, particularly in amphibians. This study aimed to elucidate the histology of the American bullfrog (Lithobates catesbeianus) pancreas using histological, immunohistochemical, transmission electron microscopic methods. Histologically, the pancreas displayed typical exocrine acini and scattered endocrine islets. Two types of endocrine cells were distinguished based on their secretory granules: Type I, found diffusely among acini and containing rounded granules (0.28 ±

This paper reports the presence of telocytes in the choroid, a pigmented vascular tissue layer of the eye, as examined by scanning and transmission electron microscopy. These cells are reported to occur in the stroma of heart, reproductive tissues, digestive tract, skin, lungs and excretory organs. In the choroid of the teleost Clarius gariepinus, telocytes are widespread over the entire area of the choroidal stroma. Their processes, called telopodes, are rather thin (<0.2 µm in diameter) and exceptionally longer (>50 µm), which branch into 3-5 short processes. These processes are intimately a

Acute salpingitis (AS) is an inflammatory disease which causes severe damage to a subset of classically described cells lining in oviduct wall and contributes to interstitial fibrosis and fertility problems. Telocytes (TCs), a newly discovered peculiar type of stromal cells, have been identified in many organs, including oviduct, with proposed multiple potential bio-functions. However, with recent increasing reports regarding TCs alterations in disease-affected tissues, there is still lack of evidence about TCs involvement in AS-affected oviduct tissues and potential pathophysiological roles.

Lately, spatial three-dimensional (3D) identity of cells and their interrelations with the environment that surrounds it represent a challenging trend with the purpose to achieve a holistic view over the functions. Combining data from different imaging of cells in the third dimension can offer insight into behavior modalities making a world of difference. This chapter outlines a breakthrough in telocyte research by volume electron microscopy with the aid of focused ion beam scanning electron microscopy (FIB-SEM). Reconstructing 3D (three-dimensional) appearance of telocytes from a set of two-d

Telocytes (TCs), a novel cell type, are briefly defined as interstitial cells with telopodes (Tps). However, a specific immunocytochemical marker has not yet been found; therefore, electron microscopy is currently the only accurate method for identifying TCs. TCs are considered to have a mesenchymal origin. Recently proteomic analysis, microarray-based gene expression analysis, and the micro-RNA signature clearly showed that TCs are different from fibroblasts, mesenchymal stem cells, and endothelial cells. The dynamics of Tps were also revealed, and some electrophysiological properties of TCs

Telocytes (TCs), a new type of interstitial cells, were identified in many different organs and tissues of mammalians and humans. In this study, we show the presence, in human oesophagus, of cells having the typical features of TCs in lamina propria of the mucosa, as well as in muscular layers. We used transmission electron microscopy (TEM), immunohistochemistry (IHC) and primary cell culture. Human oesophageal TCs present a small cell body with 2-3 very long Telopodes (Tps). Tps consist of an alternation of thin segments (podomers) and thick segments (podoms) and have a labyrinthine spatial a

Telocytes (TC) are a class of interstitial cells present in heart. Their characteristic feature is the presence of extremely long and thin prolongations (called telopodes). Therefore, we were interested to see whether or not TCs form networks in normal cardiac tissues, as previously suggested. Autopsy samples of cardiac tissues were obtained from 13 young human cadavers, without identifiable cardiac pathology and with a negative personal history of cardiovascular disease. Immunohistochemistry on formalin-fixed paraffin-embedded tissues was performed using monoclonal antibodies for CD117/c-kit.

Telocytes, a novel mesenchymal cell population, are characterized by their distinctive long and slender projections known as telopodes and have garnered significant interest since their formal introduction to the literature in 2010. These cells have been identified in various tissues, including the gastrointestinal (GI) tract, where they are suggested to play important roles in maintaining structural integrity, immune modulation, and barrier function. Inflammatory bowel diseases (IBD), which include Crohn's disease (CD) and ulcerative colitis (UC), are characterized by chronic inflammation and

CD34, vimentin, and vascular endothelial growth factor immunohistochemical analysis and electron microscopic tools were employed to record the initial appearance of telocytes (TCs) and stage-by-stage variations in TC localizations in the developing rabbit lung. TCs could not be identified in the primitive embryonic lung until day 18 of gestation. In the pseudoglandular lung, CD34+ TCs had been recorded under the cartilage of the main bronchus, in the wall of large-sized pulmonary vessels and large epithelial tubes. In the canalicular phase, TCs could be demonstrated in the smooth muscle layer

The telocyte (TC) is a new interstitial cell type described in a wide variety of organs and loose connective tissues around small vessels, but its presence in large arteries remains unexplored. TCs have small cell bodies and remarkably thin, long, moniliform processes called telopods (Tps). Using transmission electron microscopy and immunofluorescence, we identified TCs in normal human thoracic aortas and in those with aneurysm or acute dissection (TAAD). In normal aortas the TCs were distributed throughout the connective tissue of the adventitial layer, in its innermost portion and at the zon

Arsenic is a toxicant commonly found in drinking water. Even though its main route of exposure is oral, little is known of the impact of in vivo arsenic exposure on small intestine. In vitro studies have shown that arsenic decreases differentiation of stem and progenitor cells in several different tissues. Thus, small intestinal organoids were used to assess if arsenic exposure would also impair intestinal stem cell differentiation. Unexpectedly, no changes in markers of differentiated epithelial cells were seen. However, exposing mice to 100 ppb arsenic in drinking water for 5 weeks impaired

Telocytes are stromal cells characterized by small cell bodies and long cytoplasmic extensions termed telopodes. Although identified in the rat testicular interstitium, no standardized protocol for their isolation and in vitro enrichment has been established. This study aimed to develop an enriched primary culture model for rat testicular telocytes and characterize their morphology in vitro. Testicular cells from adult male Wistar rats were isolated by enzymatic digestion, subjected to short-term culture with differential adhesion, and enriched by fluorescence-activated cell sorting based on a

We studied telocytes/CD34+ stromal cells in the normal and pathological peripheral nervous system (PNS), for which we reviewed the literature and contributed our observations under light and electron microscopy in this field. We consider the following aspects: (A) general characteristics of telocytes and the terminology used for these cells (e.g., endoneurial stromal cells) in PNS; (B) the presence, characteristics and arrangement of telocytes in the normal PNS, including (i) nerve epi-perineurium and endoneurium (e.g., telopodes extending into the endoneurial space); (ii) sensory nerve ending

The aganglionic bowel in short-segment Hirschsprung's disease is characterized both by the absence of enteric ganglia and the presence of extrinsic thickened nerve bundles (TNBs). The relationship between the TNBs and the loss of enteric ganglia is unknown. Previous studies have described decreasing numbers of ganglia with increasing density of TNBs within the transition zone (TZ) between ganglionic and aganglionic gut, and there is some evidence of spatial contact between them in this region. To determine the cellular interactions involved, we have analysed the expression of perineurial marke

Insomnia is one of the main manifestations of subhealth, a condition that has become increasingly prevalent and seriously affects the physical and mental well-being of many individuals. This study aims to investigate the effects of γ-aminobutyric acid (GABA)-herbal medicine composite (GABA-HMC) on sleep in both healthy and insomnia-plagued rats. According to traditional Chinese medicine theory, the Du Meridian, situated along the dorsal midline fascia of the body, plays a crucial role in regulating neuropsychiatric disorders, such as insomnia. Recent reports morphologically confirmed that telo

Telocytes (TCs) are recently described interstitial cells, present in almost all human organs. Among many other functions, TCs regulate gastrointestinal motility together with the interstitial cells of Cajal (ICCs). TCs and ICCs have close localization in the human myenteric plexus; however, the exact spatial relationship cannot be clearly examined by previously applied double immunofluorescence/confocal microscopy. Data on TCs and submucosal ganglia and their relationship to intestinal nerves are scarce. The aim of the study was to analyse the spatial relationship among these components in th

Telocytes (TCs) are interstitial cells characterized by long, thin cytoplasmic extensions called telopodes. Although their presence has been established in various organs, data on their existence and organization within the human ovary remain limited. This study aimed to identify and describe telocytes in the ovarian stroma using immunohistochemical, immunofluorescence, and ultrastructural methods. Ovarian tissues from women aged 24-65 years who underwent total hysterectomy and bilateral salpingo-oophorectomy for nonmalignant uterine diseases were analyzed. TCs were identified by CD34 co-expre

Many studies have been carried out to investigate the histological structure of the trachea in many species of birds. However, the cellular organization of the trachea in the mallard duck is still unclear. This study was performed on 12 sexually mature male Mallard duck to demonstrate the cellular organization of the trachea using light and electron microscopy. The tracheal epithelium is considered the first line of defense against airborne pathogens. The mallard trachea was lined by a pseudostratified ciliated columnar epithelium that contained many morphologically distinct cell types: ciliat
