Phytotoxicity effect of a highly toxic isolate of Alternaria alternata metabolites from Iran.

Alternaria species produce several mycotoxins, such as alternariol (AOH), alternariol monomethyl ether (AME), altenuene (ALT), altertoxin (ATX), tentoxin (TTX) and tenuazonic acid (TeA). This research aimed to isolate and identify mycotoxins from highly toxic Alternaria alternata (w19) and A. tennuisima isolates and their phytotoxicity effects. Fungal metabolites were extracted from 21-day cultures of Alternaria in a Czapek broth medium with the organic solvent chloroform/acetone and identified using the HPLC method. Alternaria metabolites were infiltrated in vivo into several plant leaves for

Bacillus sp. Z26 suppresses Alternaria alternata and its mycotoxins by targeting AaPex13-mediated peroxisome biogenesis.

Alternaria alternata is a destructive postharvest pathogen that causes fruit decay and produces multiple mycotoxins, including alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA), posing a serious threat to crop quality and food safety. Biocontrol stands out as a promising strategy to mitigate both fungal infection and mycotoxin production. This study investigated that Bacillus sp. Z26 effectively inhibits the mycelial growth and mycotoxin accumulation of A. alternata. Comparative transcriptomic analysis revealed that strain Z26 did not directly repress canonical m

Gastrointestinal localization of metronidazole by a lactobacilli-inspired tetramic acid motif improves treatment outcomes in the hamster model of Clostridium difficile infection.

OBJECTIVES: Metronidazole, a mainstay treatment for Clostridium difficile infection (CDI), is often ineffective for severe CDI. Whilst this is thought to arise from suboptimal levels of metronidazole in the colon due to rapid absorption, empirical validation is lacking. In contrast, reutericyclin, an antibacterial tetramic acid from Lactobacillus reuteri, concentrates in the gastrointestinal tract. In this study, we modified metronidazole with reutericyclin's tetramic acid motif to obtain non-absorbed compounds, enabling assessment of the impact of pharmacokinetics on treatment outcomes. METH

The membrane as a target for controlling hypervirulent Clostridium difficile infections.

OBJECTIVES: The stationary phase of Clostridium difficile, which is primarily responsible for diarrhoeal symptoms, is refractory to antibiotic killing. We investigated whether disrupting the functions of the clostridial membrane is an approach to control C. difficile infections by promptly removing growing and non-growing cells. METHODS: The bactericidal activities of various membrane-active agents were determined against C. difficile logarithmic-phase and stationary-phase cultures and compared with known antibiotics. Their effects on the synthesis of ATP, toxins A/B and sporulation were also

Occurrence of tropane alkaloids, pyrrolizidine alkaloids and mycotoxins in rooibos: a comprehensive safety assessment.

Rooibos is a naturally caffeine-free herbal tea, widely consumed in Europe and perceived as a safe drink. However, contamination by natural toxins can pose a potential food safety concern for regular consumers. This study investigated the presence of 32 pyrrolizidine alkaloids (PAs), 16 tropane alkaloids (TAs), and 56 mycotoxins in 203 rooibos samples purchased from 18 European countries, using UHPLC-MS/MS. PAs were detected in 98% of the samples and were dominated by Senecio-type alkaloids, particularly senecionine N-oxide, senecionine, and retrorsine N-oxide. The mean and median total PA con

Comprehensive profiling of potato responses to Alternaria alternata and its toxins (TeA and AOH): ultrastructural, biochemical and multivariate insights.

BACKGROUND: Leaf spot caused by Alternaria alternata poses a major threat to potato production, yet the distinct impacts of its key toxins remain unresolved. This pathogen releases a range of secondary metabolites that interfere with normal leaf function and accelerate disease progression. Among them, alternariol (AOH) and tenuazonic acid (TeA) stand out as potent phytotoxins, known for triggering oxidative damage and weakening host tissues during infection. In the present work, we employed a multidisciplinary approach to investigate the physiological, biochemical, histochemical, ultrastructur

Metabolites contributing to Rhizoctonia solani AG-1-IA maturation and sclerotial differentiation revealed by UPLC-QTOF-MS metabolomics.

Rhizoctonia solani is a causative agent of sheath blight, which results in huge economic losses every year. During its life cycle, the formation of sclerotia helps Rhizoctonia solani withstand a variety of unfavorable factors. Oxidative stress is a key factor that induces sclerotium formation. The differentiated and undifferentiated phenotypes of R. solani AG-1-IA were obtained by controlling aerial conditions. Metabolomics based on the mass spectrometry technique combined with multivariate and univariate analyses was used to investigate the metabolic variation in vegetative, differentiated an

Discovery of the Alternaria mycotoxins alterperylenol and altertoxin I as novel immunosuppressive and antiestrogenic compounds in vitro.

Alternaria mycotoxins may pose significant challenges to food safety and public health due to the wide spectrum of reported adverse effects. Despite this, critical information on the immunomodulatory and antiestrogenic properties of most of these contaminants is still lacking. The present study aimed to identify the mycotoxins responsible for the immunosuppressive and antiestrogenic effects of a complex extract of Alternaria mycotoxins (CE) obtained by growing an Alternaria alternata strain on rice. Through a toxicity-guided fractionation procedure involving the production of CE-fractions by s

In vitro characterization of hepatic toxicity of Alternaria toxins.

Alternaria mycotoxins are secondary fungal metabolites which can contaminate food and feed. They are produced by Alternaria species with alternariol (AOH), alternariol monomethyl ether (AME), tenuazonic acid (TeA), and tentoxin (TEN) as the main representatives for Alternaria mycotoxins in food. Once passing the intestinal barrier, Alternaria toxins can reach the liver to exert yet uncharacterized molecular effects. Therefore, hepatic in vitro systems were used to examine selected Alternaria mycotoxins for their induction of metabolism-dependent cytotoxicity, phosphorylation of the histone H2A

Glycerol Monolaurate, an Analogue to a Factor Secreted by Lactobacillus, Is Virucidal against Enveloped Viruses, Including HIV-1.

The vaginal microbiota influences sexual transmission of human immunodeficiency virus type 1 (HIV-1). Colonization of the vaginal tract is normally dominated by Lactobacillus species. Both Lactobacillus and Enterococcus faecalis may secrete reutericyclin, which inhibits the growth of a variety of pathogenic bacteria. Increasing evidence suggests a potential therapeutic role for an analogue of reutericyclin, glycerol monolaurate (GML), against microbial pathogens. Previous studies using a macaque vaginal simian immunodeficiency virus (SIV) transmission model demonstrated that GML reduces transm

Fungi and mycotoxins during conventional and improved postharvest processing and storage of local rice in Nigeria.

This study applied a polyphasic characterization approach and LC-MS/MS analysis to elucidate fungal and mycotoxin contamination dynamics during conventional (CPRP) and improved (IPRP) postharvest processing and a 90-day storage of rice in Nigeria. Three hundred and twenty-five fungal isolates belonging to 21 genera (10 moulds, one crust fungus, 10 yeasts) and 54 species were recovered from rice across both processing chains. The CPRP chain had greater diversity of fungal genera compared to the IPRP chain. Aspergillus predominated both processing chains while yeasts occurred only in the CPRP ch

The Slt2-Swi6 signaling pathway plays a pivotal role in regulating cell wall biosynthesis, secondary metabolite production, and the pathogenicity of pear fungal A. alternata.

The cell wall integrity (CWI) MAPK signal pathway is crucial for the assembly of fungal cell walls and their associated virulence, as it activates the expression of the downstream transcription factors Mbp1, Swi6, and RlmA. However, the transcription factors directly regulated by this pathway in Alternaria alternata have yet to be identified. In this study, we delineated the functional roles of two transcription factors, AaSwi6 and AaMbp1, through targeted gene deletion. Disruption of either gene resulted in impaired hyphal extension, reduced mycelial accumulation, decreased conidiation, alter

Multiple mycotoxin exposure during pregnancy and risks of adverse birth outcomes: a prospective cohort study in rural Ethiopia.

INTRODUCTION: Mycotoxin exposure during pregnancy has been associated with adverse birth outcomes in low- and middle-income countries. The evidence, however, is inconsistent and mainly limited to the assessment of a single mycotoxin. We assessed biomarkers of exposure to multiple mycotoxins during pregnancy and their associations with adverse birth outcomes in rural Ethiopia. METHODS: We analyzed data from 579 pregnant women between 8 and 24 weeks of completed gestation enrolled in a prospective cohort study. Serum mycotoxin concentrations were determined using liquid chromatography coupled w
