Mapping of sex hormones in the testicular vein of healthy men.

CONTEXT: The testis produces hormones controlling sex differentiation, development, and reproduction. Their quantification in physiological conditions can aid our understanding of the endocrine function of the testis. OBJECTIVE: To measure the concentrations of steroid hormones and FSH, LH, inhibin B, anti-Müllerian hormone (AMH), and insulin-like factor 3 (INSL3) in the testicular and femoral veins, femoral artery, and seminal fluid in healthy men. DESIGN: Sample collection was made by angiographic catheterization under fluoroscopy from the testicular and femoral veins. Femoral artery sampl

Testicular cancer and hormonally active agents.

Testicular cancer (TC) is a rare form of cancer, accounting for 1% of all new cancer cases in Canadian males. TC is the most common malignancy among young men, aged 25-34 yr old. Over previous decades, the incidence of TC has increased in many Western countries. Countries with a sufficiently long period of cancer registration, such as Denmark, document this trend back to the first half of the 20th century. The etiology of TC remains poorly understood. Most of the established risk factors are likely related to in utero events, including some factors that are purported to be surrogate measures f

Hormone Regulation in Testicular Development and Function.

The testes serve as the primary source of androgens and the site of spermatogenesis, with their development and function governed by hormonal actions via endocrine and paracrine pathways. Male fertility hinges on the availability of testosterone, a cornerstone of spermatogenesis, while follicle-stimulating hormone (FSH) signaling is indispensable for the proliferation, differentiation, and proper functioning of Sertoli and germ cells. This review covers the research on how androgens, FSH, and other hormones support processes crucial for male fertility in the testis and reproductive tract. Thes

Hormonal and genetic control of testicular descent.

Cryptorchidism has the potential to affect the health of the human male. Although it is often considered a mild malformation, it represents the best-characterized risk factor for reduced fertility and testicular cancer. The aetiology of cryptorchidism remains, for the most part, unknown and cryptorchidism itself might be considered a complex disease. This reflects the intricate mechanisms regulating testicular development and descent from intra-abdominal location into the bottom of the scrotum, involving different anatomical and hormonal factors. Major actors of testicular descent are the Leyd

Reproductive Hormones During Minipuberty Suggest Subtle Testicular Impairments in Boys With Hypospadias.

CONTEXT: The ontogeny of hypospadias and its implications remain incompletely understood. OBJECTIVE: To examine whether fetal outcomes and reproductive hormone concentrations in boys with hypospadias differ from reference standards during minipuberty. DESIGN: Prospective cohort study (May 2021-January 2023). SETTING: Tertiary hypospadias center. PATIENTS: Infants presenting with hypospadias (n = 139), of whom 113 were enrolled following parental consent (median postnatal age: 0.28 years). INTERVENTIONS: Examination of hypospadias grade, external masculinization score (EMS), placental and

Intracerebroventricular salusin-β infusion to rats increases hypothalamus-pituitary-testicular axis hormones.

Salusin-β (Sal-β), which originates from preprosalusin, is a multifunctional hormone with a peptide structure. Sal-β exists in the hypothalamus and can stimulate the pituitary gland. The present study was conducted to determine the effects of Sal-β on hormones that play roles in the male reproductive system. Forty male Wistar Albino rats were used in the study. No infusions were performed on the control group, and infusions were applied to the infusion groups (artificial cerebrospinal fluid to the sham group, 2 and 20 nM Sal-β to the experimental group) through intracerebroventricular infusion

Baldness and other correlates of sex hormones in relation to testicular cancer.

There is evidence that sex hormones and intrauterine factors are involved in the etiology of testicular cancer. We evaluated the importance of perinatal and adult life correlates of sex hormones as risk factors for testicular cancer in a case control study of 97 incident, histologically confirmed cases, residents of the Greater Athens area and environs, who were diagnosed in the 3 specialized cancer hospitals and the major General Hospital in Athens during the 2 year period 1993-94. Cases were age-matched to 2 healthy controls from the same study base. Both cases and controls as well as their

Correlation of oxidation-reduction potential with hormones, semen parameters and testicular volume.

Seminal oxidative stress (OS) is a major cause of male factor infertility and can be measured as oxidation-reduction potential (ORP). Studies showed significant negative relationships of ORP with sperm count, motility or DNA integrity. Since these parameters are also positively or negatively associated with reproductive hormones follicle-stimulating hormone (FSH), luteinising hormone (LH), testosterone, testicular volume and the occurrence of varicocele, it is important to understand the mechanistic relationship between ORP and hormonal and/or testicular parameters. Therefore, we studied the r

Serum hormones in patients with nonobstructive azoospermia after microdissection testicular sperm extraction.

PURPOSE: Microdissection testicular sperm extraction combined with intracytoplasmic sperm injection is currently used to treat infertility in cases of nonobstructive azoospermia. Even in patients with nonmosaic Klinefelter's syndrome, who usually present with small testes and hypogonadism, the procedure has been done successfully. We assessed serum hormones after microdissection testicular sperm extraction and compared postoperative testicular damage between 46XY males with nonobstructive azoospermia and those with Klinefelter's syndrome. MATERIALS AND METHODS: We retrospectively reviewed the

PURPOSE: To obtain information about preorchiectomy gonadal function in patients with testicular germ cell cancer to improve the clinical management of fertility and other andrologic aspects in these men. PATIENTS AND METHODS: In group 1, a group of 83 consecutive patients with testicular germ cell cancer (TGCC) investigated before orchiectomy, semen analysis was carried out in 63 patients and hormonal investigations, including measurement of follicle-stimulating hormone, luteinizing hormone (LH), testosterone, estradiol, sex hormone-binding globulin (SHBG), inhibin B, and human chorionic gon

In vitroreconstitution of the hormone-responsive testicular organoids from murine primary testicular cells.

Increasing rates of male infertility require more experimental models to understand the mechanisms underlying male infertility. In vitroorganoids hold unprecedented promise for this purpose; however, the development of organoids with tissue architecture similar to that of the testisin vivoremains a challenge. Here, we generated testicular organoids derived from testicular cells by combining a hanging drop culture and a rotation culture system. Our results indicated that testicular cells could self-assemble into spheroid organoids with tubule-like structures in hanging drop culture. The organoid

Hormonal control of testicular descent and the cause of cryptorchidism.

This paper briefly reviews the literature on testicular descent and presents new observations from the authors' laboratory which suggest new ways of looking at old problems. There is now good evidence that testicular descent occurs in two morphologically and hormonally distinct phases. Relative 'transabdominal migration' of the testis compared with the ovary occurs at 10-15 weeks of gestation in the human and 'inguinoscrotal' migration occurs at 26-35 weeks of gestation. We have proposed previously that the first phase is controlled by Müllerian inhibiting substance although this remains contr

Anti-Müllerian hormone, testicular descent and cryptorchidism.

Anti-Müllerian hormone (AMH) is a Sertoli cell-secreted glycoprotein involved in male fetal sex differentiation: it provokes the regression of Müllerian ducts, which otherwise give rise to the Fallopian tubes, the uterus and the upper part of the vagina. In the first trimester of fetal life, AMH is expressed independently of gonadotropins, whereas from the second trimester onwards AMH testicular production is stimulated by FSH and oestrogens; at puberty, AMH expression is inhibited by androgens. AMH has also been suggested to participate in testicular descent during fetal life, but its role re
