Is the test for irreversibility of tetanus toxoids still relevant?

Tetanus vaccines for human and veterinary use are based on toxoids resulting from a formaldehyde-mediated inactivation of tetanus neurotoxin (TeNT). Due to the high toxicity of TeNT, safety tests are mandatory for each batch of these toxoids. One of the tests addresses the irreversibility of inactivation: The toxoid is stored at 37 °C for 6 weeks and then subjected to in vivo toxicity testing. However, we found that TeNT solutions rapidly lose their activity at 37 °C. Accordingly, any active TeNT molecules arising in the toxoid due to reversion events may no longer be detectable after the 37 °

The serological assessment of a tetanus toxoid field trial.

To determine the effectiveness of a method for controlling tetanus neonatorum, a double-blind controlled trial involving 1 618 women was conducted between 1961 and 1966. Women in the study were given 1-3 injections (1 ml) of aluminium-phosphate-adsorbed tetanus toxoid or a placebo (influenza vaccine). At the conclusion of the trial, 5-ml samples of blood were obtained from 299 women. Sera were titrated for anti-tetanus antibodies by two methods.A comparison of the clinical and laboratory results showed a close relationship. It is suggested that the level of protection may be lower than is at p

Effects of detoxification process on toxicity and foreign protein of tetanus toxoid and diphtheria toxoid.

Formaldehyde detoxification is a process for converting tetanus toxin (TT) and diphtheria toxin (DT) into tetanus toxoid (TTd) and diphtheria toxoid (DTd), respectively. The mechanism of this detoxification process has been investigated by several previous studies based on lab-scale toxoids. To obtain greater insights of the effects induced by formaldehyde, industrial TTd and DTd batches obtained from different detoxification processes were studied in this work. Using liquid chromatography-mass spectrometry (LC-MS), 15 and 20 repeatable formaldehyde-induced modification sites of TTd and DTd we

Induction of protection against tetanus toxin in mice by tetanus toxoid-liposome conjugate.

Tetanus toxoid (Ttd) was coupled to liposomes via glutaraldehyde. Intraperitoneal injection in BALB/c mice with Ttd-liposomes induced a substantial amount of anti-Ttd IgG antibody production and an extremely low level of anti-Ttd IgE antibody production. Mice immunized with Ttd-liposomes were successfully protected against a subsequent challenge with a lethal dose of tetanus toxin (Ttx). On the other hand, aluminum hydroxide-adsorbed Ttd (Ttd-alum) and plain Ttd solution induced the production of both IgG and IgE antibodies against Ttd. Moreover, secondary immunization with Ttd-liposomes in mi

Tetanus toxoid as an antigen for delayed cutaneous hypersensitivity.

We studied the usefulness of tetanus toxoid (TT) as a skin test antigen in assessing cellular immune function. Hospitalized patients were skin tested with four antigens, and the response rates between these antigens were compared. Candida and mumps antigens resulted in significantly more positive responses than did TT or PPD. The response rate to the TT significantly declined in older patients, suggesting these persons may not be adequately immunized against tetanus. We found TT is not as useful as Candida or mumps antigens in the evaluation of cellular immune function in a hospitalized popula

[Prevalence of hyperimmunization against tetanus and systematic adverse reactions to tetanus toxoid in children].

BACKGROUND AND AIMS: during the last 30 years we have observed only two children (aged 6 and 3 years) with systemic reactions after tetanus vaccine, given unnecessarily 5 and 10 months after appropriate primary immunization. The adverse reactions after tetanic toxoid appear to be directly related to excessive titre of protecting antibodies. The aim of this paper is to investigate the concentration of tetanus antibodies, which may help to define the risk of adverse reactions. METHODS: tests were carried out in 190 children (86 male, 104 female) aged 7 (n=95) and 14 (n=95) years. Antibodies to

Strategies for stabilising tetanus toxoid towards the development of a single-dose tetanus vaccine.

The stability of tetanus toxoid (TT) has been investigated for the purpose of enhancing its immunogenicity when encapsulated in TT-polylactide-co-glycolide (PLGA) microspheres. In this paper we describe our simulations of various potential inactivating events during microsphere processing and release of the antigen. These include: i) processing aqueous TT solutions in the presence of organic solvents, ii) exposing lyophilized TT to moisture, and iii) incubating vaccine with the degrading PLGA. At 37 degrees C, TT began to aggregate in solution a few hours after the addition of either methylene

The combined use of analytical tools for exploring tetanus toxin and tetanus toxoid structures.

Aldehyde detoxification is a process used to convert toxin into toxoid for vaccine applications. In the case of tetanus toxin (TT), formaldehyde is used to obtain the tetanus toxoid (TTd), which is used either for the tetanus vaccine or as carrier protein in conjugate vaccines. Several studies have already been conducted to better understand the exact mechanism of this detoxification. Those studies led to the identification of a number of formaldehyde-induced modifications on lab scale TTd samples. To obtain greater insights of the changes induced by formaldehyde, we used three industrial TTd

Seroprevalence of tetanus antibody in Turkish population and effectiveness of single-dose tetanus toxoid.

The study presented here was performed to determine the rates of immunity to tetanus in a cross section of the Turkish population and to assess the impact of a single-dose tetanus toxoid in previously unvaccinated individuals. Among 18-year-old participants who received their last tetanus booster 4 years prior to the study and 22-year-old participants who received the booster 8 years previously, certain durable protective antibody (>1 IU/ml) was detected in 100%. However, only 3.3% of 20-year-old participants who did not know whether they had been vaccinated as children or young adults had a p

Comparison of CRM197, diphtheria toxoid and tetanus toxoid as protein carriers for meningococcal glycoconjugate vaccines.

Glycoconjugate vaccines are among the most effective and safest vaccines ever developed. Diphtheria toxoid (DT), tetanus toxoid (TT) and CRM197 have been mostly used as protein carriers in licensed vaccines. We evaluated the immunogenicity of serogroup A, C, W-135 and Y meningococcal oligosaccharides conjugated to CRM197, DT and TT in naïve mice. The three carriers were equally efficient in inducing an immune response against the carbohydrate moiety in immunologically naïve mice. The effect of previous exposure to different dosages of the carrier protein on the anti-carbohydrate response was s

Suspected Diphtheria Toxoid and Tetanus Toxoid (dTdap) Booster Vaccine-Induced Postural Orthostatic Tachycardia Syndrome (POTS).

We describe a case of a 40-year-old South Asian woman who presented with symptoms suggestive of postural orthostatic tachycardia syndrome (POTS) following a diphtheria toxoid and tetanus toxoid (dTdap) booster vaccination administered one week prior. The patient's POTS responded favorably to treatment with low-dose fludrocortisone and ivabradine. Clinicians should maintain a high index of suspicion for POTS as a possible vaccine adverse event (VAE) post-dTdap booster inoculation and be aware of appropriate management strategies.

[Primary response to tetanus toxoid stimulation in early infancy].

The authors attempt to evaluate the degree of immune response to the first dose of anti-tetanus vaccination in young infants and to study the correlation between maternal and infantile antitetanus antibody titers. The sample studied comprised 5 males and 5 females aged between 61 and 75 days old, without acute and/or chronic pathologies and uncircumsized. Three samples of peripheral venous blood were collected for each infant: 1 at time 0, and the other two during the 15 days after the administration of the first dose of tetanus toxoid. The results support the hypothesis that vaccinal stimulat

Detection of tetanus toxoid with fluorescent tetanus human IgG-AuNC-based immunochromatography test strip.

Assays for detecting tetanus toxoid are of great significance to be applied in the research of the safety testing of tetanus vaccine. Currently, guinea pigs or mice are usually used to evaluate the toxicity in these assays. Herein, a facile and quick biomineralization process was carried out to generate tetanus human immunoglobulin G (Tet-IgG)-functionalized Au nanoclusters (Tet-IgG-AuNCs). The obtained Tet-IgG-AuNCs exhibited strong red emission with a photoluminescence quantum yield of 13%. Based on surface plasmon resonance measurements, the apparent dissociation constant of the Tet-IgG-AuN
