Factors related to gastric hypersecretion during pregnancy and lactation in rats.

We attempted to elucidate the factors involved in gastric hypersecretion of rats during pregnancy and lactation. Acid secretion in pylorus-ligated and vagally denervated fistula rats stimulated with histamine, tetragastrin, and methacholine increased from midterm pregnancy and persisted during lactation. Pepsin secretion remained unaltered during pregnancy but increased during lactation. Vagal denervation itself abolished this hypersecretion. In late pregnancy, a delayed appearance of maximal acid response to histamine was apparent, as compared to nonpregnant rats, and was abolished by aminogu

Antisecretory effect of DS-4574, a mast cell stabilizer with peptidoleukotriene antagonism, on gastric acid secretion in the pig.

The antisecretory effect of DS-4574, a mast cell stabilizer with peptidoleukotriene antagonism, on the hypersecretion of gastric acid stimulated by several secretagogues was examined in the pig. Goettingen miniature pigs with chronic gastric fistula were used. Intramuscular injection of carbachol (60 micrograms/kg), tetragastrin (50 micrograms/kg) or histamine (200 micrograms/kg)-induced gastric acid hypersecretion. Intraduodenal administration of DS-4574 (10 and 20 mg/kg) significantly inhibited both the hypersecretion induced by carbachol and that by tetragastrin. On the other hand, DS-4574

Somatostatin in gastric juice in normal subjects and patients with duodenal ulcer.

Somatostatin in gastric juice was determined in normal subjects and patients with duodenal ulcer. Gel exclusion chromatography of gastric juice revealed that the main immunoreactivity existed at the position of somatostatin-14. A large amount of somatostatin was present in gastric juice, and the quantity increased following tetragastrin stimulation. Furthermore, there was a good inverse correlation between somatostatin concentration and acidity of gastric juice; however, there was no difference between normal subjects and patients with duodenal ulcer in the amount of somatostatin released into

Clinical study on gastric secretion with special reference to pepsin secretion.

The stimulating effect of AOC-tetragastrin, caerulein, Histalog and secretin on human gastric acid and pepsin secretion was studied in gastric ulcer patients. The pattern of gastric acid and pepsin secretion after the administration of caerulein was closely resembled to that of gastrin. Slight increase of pepsin secretion after gastrin or caerulein could be based on "wash-out" action caused by the increase of acid secretion after the stimulants. Stimulating effect on gastric pepsin secretion of histalog and secretin would be independent of gastric acid secretion.

[Study on the kinetics of mucus secreting cells in the gastrointestinal tract --effects of various drugs and hormones on the cell kinetics of the generative zone in mouse gastric mucosa].

The effects of various antiulcer drugs and hormones on the cell kinetics of the mouse gastric mucosa were studied using an autoradiographic technique with 3H-thymidine. The drugs or hormones were administered orally or parenterally once or twice a day for 7 consecutive days, and 3H-thymidine was injected after the last administration of the drug. The autoradiograph was prepared and then the labeling index was counted. Cimetidine (100 mg/kg X 2/day, p.o.), geranylgeranylacetone (GGA, 100 mg/kg X 2/day, p.o.) and Cu-chlorophyllin-Na (300 mg/kg X 2/day, p.o.) did not show any effect on the labeli

Chemical deglycosylation can induce methylation, succinimide formation, and isomerization.

Interpretation of deglycosylation studies relies heavily on the absence of modifications to the polypeptide chain. We have found that by using a common chemical deglycosylation technique, one can effect at least three changes in a peptide's structure: methylation, isomerization, and ring formation. It was determined that the conditions of chemical deglycosylation introduce a +14 Da shift in the masses of our model peptides, RKDVY, RKEVY, and horseradish peroxidase. This shift is localized to acidic functional groups and is interpreted as methylation of the free carboxylates in our models. An a

Chronic alcohol abuse leads to gastric atrophy and decreased gastric secretory capacity: a histological and physiological study.

We performed morphological and physiological studies in 43 male patients with alcohol dependence (ALC) who had no other apparent lesions in the upper gastrointestinal tract except atrophic and erosive gastritis. A gastric secretory study in which tetragastrin was used as the stimulant revealed that acid and pepsin secretion was less in ALC patients than in hospital controls (p less than 0.001). Endoscopic biopsy specimens of gastric mucosa from ALC patients revealed that atrophy of the gastric mucosa advanced with age. A strong negative correlation was also found between the secretory capacity

Amino acid in gastric juice of peptic ulcer patients.

To clarify the protein metabolism in peptic ulcer disease, the amino acid content was determined in gastric juice of gastric ulcer patients (n = 30), duodenal ulcer patients (n = 16), gastroduodenal ulcer patients (n = 8), and hospital controls (n = 8). The amino acid output in the gastric ulcer group was greater than that of the hospital control both in basal and maximal secretion. In the duodenal ulcer group of patients who were high secretors of gastric juice, the amino acid concentration was low, so the amino acid output was also lower than that of the hospital control group. Tetragastrin

Effects of experimentally induced panic attacks on neuroimmunological markers.

Since little is known concerning regulation of immunological parameters in rapid changing psychiatric states like panic attacks, we measured cytokines at different time points in healthy subjects, which underwent experimental panic induction using the CCK-4 paradigm. Apart from a challenge related IL-6 increase, we could not observe any changes of neuroimmunological markers in relation to acute anxiety with regard to time and group. Herein we conducted for the first time a new approach to immunological research in panic disorder, suggesting immune changes are more related to long term disease

Acid secretory effect of extracted guinea-pig gastrin on isolated guinea-pig parietal cells.

It was confirmed that histamine, cholinergic drugs and gastrin separately produce acid secretory effects of isolated parietal cells. However, the acid secretory response of isolated parietal cells to gastrin stimulation is known to be weaker than that to histamine or cholinergic drugs, which is different from the acid secretory response found in vivo studies. Tetragastrin and pentagastrin were less potent than histamine and carbachol in stimulating acid secretion in our previous study performed using parietal cells isolated from guinea-pigs. In this study, the effect of gastrin extracted from

Papain reduces gastric acid secretion induced by histamine and other secretagogues in anesthetized rats.

We studied the effect of papain on rats' gastric acid secretion and found that: 1. Feeding of latex of unripe papaya fruit significantly reduced gastric acid secretion induced by methacholine; 2. Feeding of crystalline papain in doses of 3.2 mg/kg reduced gastric acid secretion induced by histamine, methacholine and tetragastrin; 3. The reduction of gastric acid secretion was observed as early as 2 hours after papain feeding, lasted up to 48 hours, and waned within 96 hours; 4. Intraperitoneal injection of papain had no effect on acid secretion. These results led us to believe tha the effect o

Cholecystokinin and Panic Disorder: Reflections on the History and Some Unsolved Questions.

The classic gut hormone cholecystokinin (CCK) and its CCK2-receptor are expressed in almost all regions of the brain. This widespread expression makes CCK by far the most abundant peptidergic transmitter system in the brain. This CNS-ubiquity has, however, complicated the delineation of the roles of CCK peptides in normal brain functions and neuropsychiatric diseases. Nevertheless, the common panic disorder disease is apparently associated with CCK in the brain. Thus, the C-terminal tetrapeptide fragment of CCK (CCK-4) induces, by intravenous administration in a dose-related manner, panic atta

Evidence for central neuropeptidergic control of rumination in sheep.

Effects of intracerebroventricular (ICV) vs. intravenous (IV) administration of tetragastrin, pentagastrin, CCK8 and gastrin 17 on rumination were investigated in conscious sheep. Administered at 26 pmoles/kg ICV both tetra and pentagastrin induced a premature short (15-27 min) period of rumination only 24 +/- 7 and 23 +/- 9 min after food distribution in place of 112 +/- 44 min in controls. Similar but less pronounced effects were observed for ICV administration of an equimolar dose of gastrin 17 whereas CCK8 did not promote an early period of rumination despite its anorectic effects. Plasma gamma-amino-butyric acid (GABA) levels in cholecystokinine-tetrapeptide (CCK-4) induced anxiety.

INTRODUCTION: Dysregulation of the GABA system is supposed to play an important role in the pathophysiology of panic disorder. Moreover, prior investigations revealed an association between GABA plasma levels and experimental induced panic with lactate. To evaluate a possible relationship between plasma GABA and CCK-4 induced panic plasma GABA levels in healthy volunteers were investigated. METHODS: 12 healthy subjects were challenged with 50 micro g CCK-4. Blood samples for determination of plasma GABA were drawn at baseline, 5 and 10 minutes after CCK-4 injection. RESULTS: Plasma GABA leve

Isolation from porcine antral mucosa of a hexapeptide corresponding to the C-terminal sequence of gastrin.

The present studies were undertaken to confirm reports of high concentrations of the C-terminal tetrapeptide of gastrin in hog antral mucosa. A method was developed whereby synthetic tetrapeptide added to boiling water extracts of hog antral mucosa could be purified to homogeneity by adsorption to Amberlite XAD2 resin, ion exchange chromatography on DEAE cellulose, and reverse phase HPLC. The product had the amino acid composition of gastrin tetrapeptide. When the same method was used on antral mucosa without prior addition of synthetic G4, several small peaks of material with C-terminal immun

Excitatory and inhibitory effects of gastrin peptides on gastric motility in sheep.

In conscious sheep, tetragastrin, pentagastrin, and synthetic human gastrin I, injected either subcutaneously or intravenously in doses of 156-5,200 pmol/kg body wt, inhibited the vagally dependent cyclical motility of the reticulum and rumen, whereas in vitro pentagastrin (10(-12) to 10(-6) M) had no demonstrable inhibitory or excitatory effects on intrinsically active or quiescent muscle of the reticulum, rumen, and omasal leaves. In vitro pentagastrin (10(-18) to 10(-4) M) stimulated quiescent and intrinsically active longitudinal and circular muscles of the body of the omasum and the body

[Studies on predilection area of gastric ulcer viewed from gastric motility (author's transl)].

Clinically, most gastric ulcers are located in the region of the incisura angularis, but they are least found along the greater curvature. To elucidate this difference, following experiments were carried out. A total of 22 mongrel dogs were used. A set of bipolar electrodes and/or a water-proof strain gage were chronically implanted at both the incisura and the corresponding site of the greater curvature. Electrical and contractile activities of these two sites were compared at the fasted state and under the stimulations by food, neostigmine and tetragastrin. Results are summarized as follows.

Inhibition of gastric acid secretion by saiboku-to, an oriental herbal medicine, in rats.

Intraduodenal saiboku-to (250-1000 mg/kg) dose-dependently reduced gastric acid secretion and histamine output, without altering acetylcholine output in pylorus-ligated rats. Saiboku-to also inhibited subcutaneous bethanechol (1 mg/kg) and tetragastrin (0.3 mg/kg) -induced increases in gastric acid secretion in vagotomized pylorus-ligated rats; however, it did not inhibit subcutaneous histamine (20 mg/kg) -induced increase in acid secretion. These results, taken together, suggest a possibility that saiboku-to may inhibit histamine release. Thus, the effect of saiboku-to on histamine release wa

[Control of ingestive behavior and rumination by neuropeptides].

In sheep as in other species, intracerebroventricular (ICV) administration of picomolar doses of several peptide hormones affects food intake and rumination, suggesting that these hormones play a role in the physiological control of feeding behavior. Their effects depend on their duration (short vs long-term) and/or their nature (orexigenic vs anorexigenic). Among the short-term satiety factors, CCK8, CRF and calcitonin, administered ICV, decrease food intake by reducing the rate (CCK8) or the duration (CRF, calcitonin) of ingestion; in contrast, the gastrin group of peptides (gastrin 17, pent
