Theileria parva, a protozoan parasite, is the causative agent of East Coast fever (ECF), an economically important disease of cattle in sub-Saharan Africa. The Muguga cocktail vaccine which comprises 3 T. parva strains, namely Muguga, Kiambu 5 and Serengeti transformed, is used for immunization of cattle to control ECF. However, the relative contributions of these T. parva strains to vaccine efficacy are not fully understood. This study compared the in vitro infectivity of the strains at varying concentrations of 2.75, 84.5, and 169 infected acini/ml using peripheral blood mononuclear cells is…

A comparison of ten methods for staining tick salivary glands for detection of Theileria parva infection from ticks fed on rabbits for various periods was undertaken. Staining with azure without hydrochloric acid hydrolysis was found to be the most reliable method for detection of the presporozoite stages (sporoblasts) of T. parva in the salivary gland acini of unfed Rhipicephalus appendiculatus and could be applied to field ticks. All the stains proved suitable for the detection and quantitation of sporozoites in ticks fed for 4 days on rabbits. The capacity of the stains to allow detection o…

Ticks and tickborne diseases (TBDs) are serious constraints to cattle production in Tanzania and other tropical and subtropical countries. Among the TBDs, East Coast fever (ECF) is the most important as it causes significant economic losses to the cattle industry in Tanzania. However, control of ECF in Tanzania has continued to be a challenge due to inadequate epidemiological information. The main objective of this study was to determine the epidemiological situation of Theileria parva infections in cattle kept under pastoral and agro-pastoral farming systems in Mara, Singida, and Mbeya region…

The effects of holding temperature, pH and medium on the infectivity of Theileria parva sporozoites were investigated using an in vitro infectivity assay. The sporozoite infectivity lasted for 72 h at a holding temperature of 4 C but for only 24 h at 24 degrees C. Sporozoite infectivity was found to be sensitive to pH variations and sporozoites were most infective between pH 7 and pH 8. There was a significant loss in infectivity at pH 5 and infectivity was almost totally abolished at pH 9. Theileria parva sporozoites are usually held and manipulated in Eagle's minimum essential medium (MEM) w…

The role played by complement and fibrinolysis in the pathogenesis of pulmonary oedema and the disseminated, subendothelial, petechial haemorrhages in cattle infected with Theileria parva was investigated. There was a marked reduction in total haemolytic complement in animals that died of this disease whereas the survivors showed only transient changes. The reduction in C3 levels in animals with severe disease, however, was equivalent to that in the survivors. Neither circulating soluble immune complexes nor free antibody against macroschizonts were detected in sera from cattle with severe inf…

The tick-transmitted apicomplexan Theileria parva causes East Coast fever, a bovine disease of great economic and veterinary importance in Africa. Papain-like cysteine proteases play important roles in protozoan parasite host cell entry and egress, nutrition and host immune evasion. This study reports the identification and characterisation of a T. parva strain Muguga cathepsin L-like (C1A subfamily) cysteine protease (ThpCP). Molecular modelling confirmed the papain-like fold of ThpCP, hydrophobic character of the S2 substrate binding pocket and non-covalent interaction between the pro- and c…

The apicomplexan parasite Theileria parva is the causative agent of East Coast fever, usually a fatal disease for cattle, which is prevalent in large areas of eastern, central, and southern Africa. Protective immunity against T. parva is mediated by CD8+ T cells, with CD4+ T-cells thought to be important in facilitating the full maturation and development of the CD8+ T-cell response. T. parva has a large proteome, with >4000 protein-coding genes, making T-cell antigen identification using conventional screening approaches laborious and expensive. To date, only a limited number of T-cell antige…

Babesia bovis BOV57, which is a homolog of the Theileria parva vaccine candidate antigen P67, is expressed in both the tick and blood stages of the life cycle of this parasite. However, the vaccine potential of BOV57 remained to be investigated. In the present study, we generated recombinant BOV57 (rBOV57) and prepared polyclonal antibodies against rBOV57 in mice and rabbits. Indirect immunofluorescence assays conducted with the mouse anti-rBOV57 antibody demonstrated that BOV57 localized at the apical end of paired merozoites in infected bovine red blood cells, whereas the antigen was found i…

The paper reviews the infectivity, cross-immunization experiments, and cattle vaccination of Zimbabwean cattle-derived Theileria parva (Boleni) sporozoite stabilates produced at the Central Veterinary Laboratory (CVL) in Harare between 1980 and 2003. The Boleni stock was first isolated in July 1978 during a theileriosis outbreak and was shown to be virulent in susceptible cattle. Thereafter, the reactions observed in susceptible cattle produced by different tick stabilates derived from the original have been mostly severe (76%) or moderate (24%). The parasite concentrations in the Boleni vacci…

Theileria parva causes widespread morbidity and mortality in cattle in endemic regions. An outbreak of theileriosis occurred on a farm near Ladysmith in KwaZulu-Natal, South Africa, which is not a declared Corridor disease-infected area. A survey of Red Brangus cattle from all age groups and areas of the farm was performed. Transmission of the parasite from infected animals on the farm to susceptible animals by tick transmission and tick-stabilate injection, was attempted. The survey indicated high numbers of animals with antibody titres to T. parva but only 6 infected animals, based on real-t…

Adult male and female Rhipicephalus appendiculatus ticks infected with Theileria parva (Muguga 3087) were fed on rabbits and the development of infection was monitored daily using light microscopy and an in vitro titration technique able to quantify the infectivity of sporozoite suspensions. The salivary glands stained with methyl green pyronine showed presence of infection in some unfed ticks. The intensity of staining was shown to increase with the number of days the ticks had fed. The in vitro technique, on the other hand, could detect infection only in ticks which had fed for 3-5 days. Fee…

Sporogony of the sporozoan Theileria parva in the salivary gland of the tick vector of East Coast fever was studied in electron micrographs. The findings differ in several respects from previous interpretations based upon light microscopy. Cytokinesis of the primary sporoblast to form secondary and tertiary sporoblasts is not substantiated. Instead it is suggested that the parasite develops as a ramifying, multinucleate syncytium rapidly increasing in size and complexity until it gives rise to myriad sporozoites in a terminal episode of cytoplasmic fission. The proliferating nuclei initially o…

Environmental changes and human activities such as deforestation and expansion of agricultural land are increasing tick-borne diseases including Anaplasmosis, Babesiosis, Ehrlichiosis, and Theileriosis. These diseases, which affect animals, can be transmitted to humans through tick bites. Kiambu County's warm, wet climate provides an environment conducive to tick breeding and development. This study investigated the circulation and genetic diversity of Theileria parva and Anaplasma spp. in tick samples collected from Kiambu County, Kenya. Ticks were collected from cattle, goats, and sheep usin…

Two Theileria parva sporozoite stabilates stored at -196 degrees C, then at -70 degrees C for six weeks (stabilate 1) and more than six months (stabilate 2) were inoculated into four eight-month old male calves, 1 and 2 (stabilate 1), and 3 and 4 (stabilate 2). Calves 1 and 2 developed pyrexia, enlargement of lymph nodes, and the former died of East Coast Fever. Calves 3 and 4 showed slight enlargement of lymph nodes without fever. Lymph node smears from all calves (from day 10 to 20 post-inoculation) showed lymphoblasts, phagocytic macrophages, and schizonts. Piroplasms were detected in eryth…

INTRODUCTION: East Coast fever (ECF), caused by Theileria parva, remains endemic in Malawi. ECF vaccines and chemical treatments are regarded as expensive by small-holder farmers. The aims of this study were to clarify the epidemiology of ECF and to evaluate the efficacy of ethnoveterinary medicine, Synadenium cupulare on T. parva infection.
METHODS: A cross-sectional study was conducted in three districts of northern Malawi (Chitipa, Karonga, and Mzimba). A structured questionnaire on feeding style, treatment of ECF, and tick control practices was administered to 79 farmers, and blood sample…

East Coast fever (ECF), caused by Theileria parva causes devastating loss to livestock keepers in sub-Saharan Africa. A longitudinal study was carried out in Narok County to estimate the incidence and risk factors associated with Theileria parva infection among calves. Calves were recruited within 5 weeks of birth and visited every six weeks to collect farm and calf level factors. A total of 1849 serum samples were collected and tested using an indirect enzyme-linked immunosorbent assay (ELISA). A multilevel multivariable logistic regression was used to determine associations between the serop…

The range of the protozoan parasite Theileria parva, which causes East Coast fever in cattle, has been expanding to countries where it has not previously been detected, as a result of cross-border domestic cattle movement. Countries where T. parva has not previously been observed until recently include Cameroon and South Sudan. This raises the issue of the conservation of the p104 antigen gene, on which the nested PCR assay that is widely used for T. parva surveillance in the blood of infected cattle is based. We sampled 40 isolates from six countries widely distributed across the geographical…

Intracellular parasites show host cell specificity, and precise information on the range of host cells is a prerequisite for the identification of host molecules that account for the specificity and are involved in entry processes. The sporozoite stage of the tick-borne protozoan parasite Theileria parva binds to and enters bovine lymphocytes, but precise information on the susceptibility of other cell types present at the tick attachment site is unavailable. We quantitatively examined the susceptibility of cell types known to be present at the tick attachment site by a previously established …
