Insight into oriental theileriosis: Indian perspective.

Theileria orientalis, an emerging haemoprotozoa, primarily targets large ruminants, especially cattle. Being a globally distributed haemoprotozoa, it causes oriental theileriosis, which is transmitted by the tick vector Haemaphysalis longicornis. In tropical countries, such as India, Rhipicephalus microplus (Asian Blue tick) is another potential vector for disease transmission. Unlike T. parva and T. annulata, schizonts of T. orientalis are non-lymphoproliferative in nature and the main pathogenic effect is contributed by the piroplasms destroying erythrocytes, resulting in anemia. The T. orie

Interactions between chemotherapy and immunity in bovine theileriosis.

In bovine theileriosis the use of chemotherapy to control an infection sufficiently long to permit the establishment of a solid protective immune response has been developed as a routine vaccination procedure. Infections with Theileria parva and T. annulata can be prevented by the administration of carefully controlled numbers of sporozoites simultaneously with a long acting tetracycline and this form of immunization has been widely used for the control of East Coast fever in Africa with considerable success. In this review, the nature of the chemotherapy, the immune response and the interacti

Chemotherapy of field cases of East Coast fever using halofuginone lactate.

The chemotherapeutic effect of halofuginone lactate (Terit, Hoechst) was tested against natural pathogenic Theileria parva infections (East Coast fever, ECF) in 24 cattle. Halofuginone lactate, administered per os, 1.2 mg per kg b.wt and repeated after 48 h manifested a potent schizonticidal effect, observed between 5 and 11 days post treatment. Disappearance of erythrocytic forms (EF) took long, ranging from 6 to 34 days post treatment. Differences on efficacy between halofuginone lactate tablets and solution were not observed. The use of furosemide (Dimazon, Hoechst) to promote diuresis in p

Epidemiology of East Coast fever: some effects of temperature on the development of Theileria parva in the tick vector, Rhipicephalus appendiculatus.

The moulting behaviour of nymphal Rhipicephalus appendiculatus from the laboratory colony and 3 field strains from Kenya was greatly influenced by constant temperatures between 18 and 37 degrees C but was not significantly different for the 4 tick strains. Six batches of R. appendiculatus (Muguga laboratory) nymphs which had engorged on cattle parasitaemic with 4 stocks of Theileria parva (Kiambu 4, Kiambu 5, Muguga and Mbita 4) isolated in Kenya were incubated at constant temperatures between 18 and 37 degrees C during their pre-moult and post-moult periods. The isolation of Theileria? Taurotragi in South Africa.

In 3 out of 4 attempts strains of a Theileria sp. of low virulence were isolated in the laboratory by feeding adult Rhipicephalus appendiculatus collected from the field on susceptible cattle. One of the strains, previously identified as Theileria? taurotragi (Tzaneen), was found to be serologically cross reactive with the other 2 strains. It was concluded that T.? taurotragi is prevalent in South Africa in those parts where the vector exists. Infection was characterized by a transient fever and small numbers of macroschizonts and piroplasms. Subinoculation of the infection with small volumes

In vitro infection of bovine erythrocytes with Theileria annulata merozoites as a key step in completing the T. annulata life cycle in vitro.

Theileria annulata is a protozoan parasite with a complex life cycle involving a bovine host and a tick vector. It is transmitted by Hyalomma ticks and is the causative agent of tropical theileriosis, a debilitating and often fatal disease in southern Europe, northern Africa and large parts of Asia. Understanding the biology of different life cycle stages is critical for the control of tropical theileriosis and requires the use of experimental animals which poses an ethical concern. We present for the first time the in vitro infection of red blood cells (RBCs) with T. annulata differentiated s

The role of sheep (Ovis aries) in maintaining Theileria orientalis Ikeda type infection.

Theileria orientalis is a tick‒borne intracellular parasite of red blood cells that causes severe and mild infections in various ruminants worldwide. To date there have been 11 types identified within this species, of which 4 types are presently found in New Zealand cattle. Since 2012, New Zealand has suffered a substantial epidemic of infectious bovine anaemia in both dairy and beef cattle associated with the Ikeda type. The speed at which the disease spread through the North Island suggested that other species could have been involved in transmission. The aim of a series of related experimen

A molecular and haematological study of Theileria equi in Balkan donkeys.

Equine piroplasmosis in donkeys has been recognised as a serious problem of major economic importance. The present molecular study is the first investigation of the presence of Theileria equi and Babesia caballi in Balkan donkeys and of the possible haematological alterations related to it. A total of 70 apparently healthy donkeys from Serbia were included in this study. The overall prevalence of T. equi infection in donkeys tested with multiplex PCR was 50%. There was no B. caballi-positive sample. Infections in donkeys included in this study seem to be associated with decreased red blood cel

Monitoring piroplasms infection in three cattle farms in Minorca (Balearic Islands, Spain) with previous history of clinical piroplamosis.

Bovine piroplasmosis are tick-borne protozoan diseases caused by parasites of the genera Theileria and Babesia. Three Friesian cattle farms (F1-F3) with previous history of clinical piroplamosis were selected in Minorca (Balearic Islands, Spain). Blood samples were collected from 8 to 11 animals every two months throughout a year and, a newly developed multiplex DNA bead-based suspension array based on the Luminex(®) xMAP technology was used to monitor for the presence of piroplasms. The assay incorporated probes for Babesia divergens, Babesia bovis, Babesia bigemina, Babesia major, Babesia oc

Genetic diversity of Theileria species identified from questing and parasitic ticks in selected areas in Qinghai, China.

Piroplasmosis caused by tick-borne piroplasm is a vital zoonotic disease that poses significant concerns for human and animal health. However, the information on the molecular prevalence of the piroplasm transmitted by ticks is limited in Qinghai, China. Here, we investigated the geographic distribution of 544 collected ticks and the genetic diversity of tick-borne piroplasm in four areas of Qinghai. The results of the tick species identification revealed that the obtained ticks were Haemaphysalis qinghaiensis, Dermacentor everestianus, D. silvarum and D. nuttalli. Nested PCR revealed that the

Epigenetic landscape of the H3K27me3 mark in macrophages transformed by Theileria annulata.

Theileria annulata is an obligate intracellular parasite that induces bovine leukocyte transformation leading to uncontrolled proliferation and heightened dissemination of infected leukocytes. Early passage T. annulata-transformed macrophages are virulent, but with long-term culture, they become attenuated for dissemination. To investigate this phenotype from an epigenetic perspective, we focused on the important repressive histone marks, tri-methylated lysine 27 of histone H3 (H3K27me3) catalyzed by the Polycomb Repressive Complex 2. ChIP-seq revealed that the genomic distribution of H3K27me3

A note on exophthalmia in Theileria annulata infection: Clinical presentation, pathobiology, and the central role of cytokine-mediated response.

Exophthalmia is an important but poorly documented complication of tropical theileriosis caused by Theileria annulata, especially in neonatal and young calves. It involves forward displacement of the eyeball, periorbital swelling, conjunctival congestion, excessive tearing, and enlarged regional lymph nodes. Systemic signs like fever, anaemia, depression, and weakness often occur. The condition arises through complex pathogenetic mechanisms linked to the schizont stage of the parasite, where infected leukocytes transform into rapidly dividing, invasive cells. These cells infiltrate retro-orbit

First report of Theileria orientalis Ikeda genotype associated with disease in cattle, Georgia, USA.

Theileria orientalis genotype Ikeda is a tickborne parasite that infects red and white blood cells and causes bovine infectious anemia. The primary tick vector for T. orientalis Ikeda is the longhorned tick (Haemaphysalis longicornis). This tick was first detected in the United States in 2017 and in the state of Georgia in 2021, however, to date, no clinical cases of theileriosis in cattle have been reported in the state of Georgia. A Highland calf was clinically diagnosed with anemia, marked lethargy, tachycardia, tachypnea, decreased rumen contraction rate, dry and flaking skin, and thin and

[Equine piroplasmosis: Case descriptions and overview of the epidemiological situation in Europe with focus on Germany].

Equine piroplasmosis (EP) is caused by Babesia (B.) caballi and Theileria (T.) equi and is transmitted by hard ticks. Predominantly, the Mediterranean region is known as being endemic for both pathogens in Europe. However, autochthonous infections in central European countries such as Germany can no longer be ruled out due to individual case reports in horses without any stays abroad as well as the geographical expansion of the habitats of different tick species. The case reports presented underline the risk of infection for horses travelling to endemic areas and in horses imported from such a

First molecular evidence of Theileria lestoquardi in small ruminants in northern Iran.

Ovine theileriosis as a critical agent in small ruminant production, can cause lethal infections. Different species of Theileria have been reported in various parts of the world, and each species causes different diseases in the host. This is the first molecular study to investigate the prevalence of ovine theileriosis and identify the dominant Theileria species in northern Iran. A number of 220 small ruminants, including sheep and goats, were randomly sampled from 22 flocks. Peripheral blood smears were stained by the Giemsa staining method. As well as for species identification, all samples
