[Thyroid hormones, obesity and brown adipose tissue thermogenesis].

Brown adipose tissue (BAT) is the main site for hormone-dependent (non-shivering) thermogenesis in response to cold in lower mammals. The hypothalamus controls the cold-induced BAT activation by stimulating the sympathetic nerves and the secretion of norepinephrine (NE) in BAT. Mediated by beta-3 noradrenergic receptor and in the presence of triiodothyronine (T3), NE promotes the synthesis of the uncoupling protein 1 (UCP1). UCP1 is a 32 kDa protein located in the inner membrane of BAT mitochondria, where it dissipates the proton gradient created by oxidations in the mitochondria. UCP1 functio

Brain energy allocation is consistent with adaptive thermogenesis.

Caloric restriction and weight loss reduce resting metabolic rate (RMR) beyond what is explained by the loss of fat-free mass, a phenomenon known as adaptive thermogenesis (ATRMR). Here, we frame ATRMR conceptually as the result of shifts in energy budgets within the body. Given the brain's central role as both a key regulator of energy intake and a major energy consumer within the body, we hypothesize that ATRMR represents one component of the weight-loss-associated reduction in energy expenditure and may reflect a redistribution of energy from the body to the brain, consistent with the hypot

Exosome trafficking is a key regulator of adipocyte thermogenesis.

Activation of beige adipocytes enhances energy expenditure and promotes metabolic health, presenting a promising approach for combating obesity and diabetes. As part of this process, thermogenesis, fueled by uncoupled mitochondrial respiration, plays a central role in converting calories into thermal energy, thereby preventing their storage as fat. Here, we identify exosome trafficking as an intrinsic regulator of thermogenic adipocyte function. Exosomes are small extracellular vesicles that mediate cell-cell and intracellular communication by transporting regulatory cargo, including microRNAs

Pharmacodynamic characterization of insulin on MDMA-induced thermogenesis.

Sympathomimetic drugs (MDMA; ecstasy) induce a potentially catastrophic hyperthermia that involves free fatty acid (FFA) activation of mitochondrial uncoupling proteins (UCP). Insulin is an important regulator of plasma FFA levels, although its role in thermogenesis is unclear. The aims of the present study were 1) to characterize the pharmacodynamic effects of MDMA on plasma insulin and glucose, 2) to examine the effects of insulin on MDMA-induced thermogenesis and 3) to examine MDMA-induced thermogenesis in an animal model of insulin resistance, the obese Zucker rat. Insulin levels peaked 15

Thyroid hormone control of thermogenesis and energy balance.

The mechanisms whereby thyroid hormone increases heat production have been analyzed with emphasis in more recent developments. Thyroid hormone increases obligatory thermogenesis as a result of the stimulation of numerous metabolic pathways involved in development, remodeling, and delivery of energy to the tissues. In addition, thyroid hormone may specifically stimulate some thermogenic mechanisms selected during evolution of homeotherms (e.g., Na/K-ATPase, Ca2+ cycling in muscle). Thyroid hormone also plays an essential role in facultative thermogenesis interacting with the sympathetic nervous

Effect of fetal thyroidectomy on newborn thermogenesis in lambs.

We investigated the effect of the transient neonatal hyperthyroid state on thermogenesis at birth by measuring rectal temperature, plasma free fatty acids, plasma catecholamines, and in vitro brown adipose tissue respiration in thyroidectomized (n = 6) and sham operated (n = 5) fetal sheep. Surgery was performed at an average of 133 days of gestation followed by cesarean delivery at 146 days. Fetuses were delivered into a constant room temperature of 25 degrees C. Serial measurements were made in utero before delivery and at timed intervals after birth. Serum 3,3',5 triiodothyronine and thyrox

Projection-defined hypothalamic outputs differentially regulate thermogenesis and lipolysis.

The ventromedial hypothalamus (VMH) is a key regulator of energy homeostasis, linking central neural activity to peripheral metabolic function. Within the VMH, neurons expressing steroidogenic factor-1 (SF-1; VMHSF1) are essential for regulating energy expenditure, yet the projection-defined pathways through which they differentially control distinct adipose depots remains unclear. Here, we use projection-specific optogenetic and chemogenetic manipulations in SF1-Cre mice to identify two anatomically and functionally distinct VMHSF1 output pathways with complementary metabolic roles. Bidirectional shifts in Pm20d1 expression impact thermogenesis and metabolism.

BACKGROUND: Peptidase M20 domain containing 1 (PM20D1) is a secreted N-fatty acyl amino synthase and hydrolase that controls tissue and blood levels of N-fatty acyl amino acids. In brown adipocytes, N-fatty acyl amino acids bind to mitochondria and act as uncouplers of mitochondria, independent of UCP1. Interventions aimed at increasing or inhibiting PM20D1 expression considerably impact energy balance and metabolism; however, little is known about naturally occurring variants of the PM20D1/Pm20d1 gene and their impact on phenotype. METHODS: In vivo, gene expression of Pm20d1 in BALB/c, C57BL

Maternal arginine supplementation enhances thermogenesis in the newborn lamb.

Body temperature maintenance is one of the most important physiological processes initiated after birth. Brown adipose tissue (BAT) is an essential mediator of thermogenesis in many species and is responsible for 50% of the heat generated in the newborn lamb. To determine if maternal arginine supplementation could enhance thermogenesis in the neonate, we randomly assigned 31 multiparous Suffolk ewes, gestating singletons or twins, to receive intravenous injections of either l-arginine (27 mg/kg body weight; n = 17) or sterile saline (n = 14) three times daily from day 75 to 125 of gestation (t

Optogenetic-induced sympathetic neuromodulation of brown adipose tissue thermogenesis.

The brown adipose tissue (BAT) is a thermogenic organ that plays a major role in energy balance, obesity, and diabetes due to the potent glucose and lipid clearance that fuels its thermogenesis, which is largely mediated via sympathetic nervous system activation. However, thus far there has been little experimental validation of the hypothesis that selective neuromodulation of the sympathetic nerves innervating the BAT is sufficient to elicit thermogenesis in mice. We generated mice expressing blue light-activated channelrhodopsin-2 (ChR2) in the sympathetic nerves innervating the BAT using tw

Uncoupling protein 2-mediated thermogenesis in vulnerable atherosclerotic plaques.

Timely detection of vulnerable atherosclerotic plaques is of the utmost importance to prevent acute clinical events, such as unstable angina, myocardial infarction and sudden death. Advanced plaques show a dense infiltration of macrophages, of which a subpopulation strongly expresses uncoupling protein 2 (UCP2). This protein uncouples adenosine triphosphate (ATP) production from mitochondrial respiration and thereby converts the loss of potential energy in heat production. In early atherosclerotic lesions, UCP2 seems to fulfil an atheroprotective effect by reducing reactive oxygen species (ROS

Reversible umbilical cord occlusion: effects on thermogenesis in utero.

The initiation of thermogenesis at birth is an important adaptation for survival. We examined the sequential effects of cooling, increased oxygenation, and repeated episodes of umbilical cord occlusion on nonshivering thermogenesis in six fetal sheep at 139 to 145 d of gestation. The fetal sheep were cooled by circulating cold water through a coil placed around the trunk for 4 h. The fetal core temperature fell 2.47 +/- 0.24 degrees C in the first 60 min of cooling with minimal changes in plasma FFA and glycerol levels. After fetal arterial O2 tension was increased above 6.65 kPa by ventilatio

The potential role of hypothalamic POMCTRPM2 in interscapular BAT thermogenesis.

The major function of primary order neurons in the arcuate nucleus of the hypothalamus is control of energy homeostasis. Among these neurons, proopiomelanocortin (POMC) neurons play a significant role in controlling anorexigenic feeding behavior and upregulating energy expenditure. In addition, transient receptor potential melastatin 2 (TRPM2) is a well-established temperature sensor, but no evidence of regulation of brown adipose tissue (BAT) thermogenesis via POMCTRPM2 neurons in the arcuate nucleus has been reported so far. Here, through single-cell reverse-transcription and immunohistochem

Thyroid hormones in the regulation of brown adipose tissue thermogenesis.

A normal thyroid status is crucial for body temperature homeostasis, as thyroid hormone regulates both heat loss and conservation as well as heat production in the thermogenic tissues. Brown adipose tissue (BAT) is the major site of non-shivering thermogenesis and an important target of thyroid hormone action. Thyroid hormone not only regulates the tissue's sensitivity to sympathetic stimulation by norepinephrine but also the expression of uncoupling protein 1, the key driver of BAT thermogenesis. Vice versa, sympathetic stimulation of BAT triggers the expression of deiodinase type II, an enzy

Is there a role for sarcolipin in avian facultative thermogenesis in extreme cold?

Endotherms defend their body temperature in the cold by employing shivering (ST) and/or non-shivering thermogenesis (NST). Although NST is well documented in mammals, its importance to avian heat generation is unclear. Recent work points to a prominent role for the sarco/endoplasmic reticulum Ca2+ ATPase (SERCA) in muscular NST. SERCA's involvement in both ST and NST, however, posits a tradeoff between these two heat-generating mechanisms. To explore this tradeoff, we assayed pectoralis gene expression of adult songbirds exposed to chronic temperature acclimations. Counter to mammal models, we

Exogenous induction of thermogenesis in Arum concinnatum by salicylic acid.

Arum concinnatum Schott is a highly thermogenic species, with the temperature of the appendix exceeding ~10.9°C above the ambient temperature during thermogenesis, whereas the rates of respiration of the male florets in intact inflorescences peak at 0.92μmol s-1 g-1, which is the highest rate so far measured among the plants. Here, we attempt the ex situ exogenous induction of thermogenesis in whole inflorescences and in separate appendices of the spadix, and explore the thermogenic patterns under controlled laboratory conditions of light and temperature. Mature but unopened inflorescences and

An adipose-specific control of thermogenesis in body weight regulation.

Much of our understanding about 'adaptive thermogenesis' as a control system in mammalian weight regulation derives from studies of experimental starvation and overfeeding, and these have served to characterize its functional role as an 'attenuator' of energy imbalance. By applying a system-analysis approach in evaluating data on the energetics of starvation and refeeding, evidence is presented here in support of the hypothesis that there are in fact two distinct control systems underlying adaptive thermogenesis. In one of them, the efferent limb is primarily under the control of the sympathet

Lack of nonshivering thermogenesis in infants anesthetized with fentanyl and propofol.

BACKGROUND: Sweating, vasoconstriction, and shivering have been observed during general anesthesia. Among these, vasoconstriction is especially important because-once triggered-it minimizes further hypothermia. Surprisingly, the core-temperature plateau associated with vasoconstriction appears to preserve core temperature better in infants and children than adults. This observation suggests that vasoconstriction in anesthetized infants may be accompanied by hypermetabolism. Consistent with this theory, unanesthetized infants rely on nonshivering thermogenesis to double heat production when vas

Neostigmine in the hippocampus increases thermogenesis and T4-to-T3 conversion.

The firing rate of the nerves innervating interscapular brown adipose tissue (IBAT), IBAT and colonic temperatures (TIBAT and TC), and O2 consumption were monitored in urethan-anesthetized male Sprague-Dawley rats. These variables were measured for 40 min before (baseline values) and 40 min after a neostigmine (5 x 10(-7) mol) or saline injection in the hippocampus. The blood level of 3,5,3'-triiodothyronine and L-thyroxine (T3 and T4) and the 5'-deiodinating activity of IBAT, liver, and kidneys were determined in other rats with neostigmine or saline injection. Direct microcalorimetry was used for measurements of heat production in cell suspensions of platelets and adipocytes, obtained from hypothyroid patients before and after 3 months on full L-thyroxine substitution. Platelet heat production was significantly lower than normal before treatment and increased in all 10 patients studied; the mean value increased from 51.3 +/- 1.6 fW/cell before to 57.1 +/- 1.8 fW/cell after therapy (P less than 0.001). Similarily, adipocyte heat production was initially significantly lower than normal and increased during treatment in all 6 patients investigated. The
