Heat stress poses a great threat to plant survival. Although great progress has been made in identifying multiple important regulatory factors in response to heat stress, the post-translational regulatory mechanisms in the process are largely unknown. SUMOylation is an important post-translational modification of proteins and plays key roles in the heat stress response. Here, we identified HAT1 as a novel substrate of SUMO E3 ligase SAPAND MIZ1DOMAIN-CONTAINING LIGASE1 (SIZ1). Further analysis revealed that high temperature induces the accumulation of the SIZ1 protein, which promotes its inter

Heat stress is a major abiotic constraint that severely limits maize (Zea mays L.) productivity under changing climate conditions. This study explored a novel integrative strategy to enhance thermotolerance in maize through the combined application of methyl jasmonate-loaded chitosan nanoparticles (MJNPs) and eucalyptus-derived biochar (EBB). Methyl jasmonate was nano-encapsulated using the ionic gelation method and characterized by SEM, TEM, and FTIR analyses, which confirmed uniform spherical nanoparticles and effective surface functionalization. A greenhouse experiment was conducted under c

The Peste des Petits Ruminants (PPR) live attenuated vaccines, the PPR virus (PPRV) Nigeria 75/1 strain (lineage II) and PPRV India Sungry 96 strain (lineage IV), currently used for control and eradication programme are very efficient vaccines as they provide the host, sheep and goats, a lifelong immunity after a single minimum recommended dose of 102.5 TCID50/mL. Unfortunately, both live attenuated vaccines are thermolabile and their use requires maintaining the cold chain from the manufactory premises to the field as most PPR-infected regions are facing of hot climate, with poor infrastructu

Increasing temperatures associated with climate change have the potential for far-reaching impacts on human health and disease vectors, including fungal pathogens. Pathogenic fungi inhabit a wide range of environments across the world, and their ranges have been slowly expanding in recent decades due, in part, to climate change. Despite these links between increasing temperature and higher prevalence of fungal disease, the direct effects of rising environmental temperatures on the evolution of pathogenic fungi remain unclear. In this study, we investigated how increasing temperatures drive ada

Light and temperature are major drivers in the ecology and biogeography of symbiotic dinoflagellates living in corals and other cnidarians. We examined variations in physiology among 11 strains comprising five species of clade A Symbiodinium We grew cultures at 26°C (control) and 32°C (high temperature) over a duration of 18 days while measuring growth and photochemical efficiency (Fv /Fm ). Responses to thermal stress ranged from susceptible to tolerant across species and strains. Most strains exhibited a decrease in cell densities and Fv /Fm when grown at 32°C. Tolerance to high temperature

UNLABELLED: The genetic basis of phenotypic differences between species is fundamental to our view of evolution. In contrast to variation within species, phenotypic differences between species can result from a long-term sequential process whereby prior steps and multiple changes at a single locus can play important roles. However, investigating the genetic basis of phenotypes that have evolved over long time periods is limited by reproductive barriers between species. In this study, we use hybrid genetic analysis to examine reproductively isolated Saccharomyces species that have diverged in t

Heat stress is emerging as a dominant constraint on global crop productivity by destabilizing membranes, disrupting photosynthesis, impairing reproductive development, and accelerating oxidative damage. In recent years, nanoparticles (NPs) have been widely proposed as new regulators of plants thermotolerance, but available literature is rather fragmented, often descriptive and frequently inconsistent across experimental systems. This review provides a critical synthesis of the nanoparticle-mediated heat-stress resistance in most important crops, such as rice, wheat, maize, tomato, and soybean

HSFA2 is a key regulator of acquired thermotolerance after mild-heat priming, but whether volatile-triggered priming depends on this pathway remains unclear. Here, we examined thermotolerance induced by the reactive green leaf volatile trans-2-hexenal (T2H) in Arabidopsis thaliana Col-0 and an hsfa2 T-DNA insertion mutant. Seedlings were primed with mild heat or a 30-min T2H pulse, recovered for 2 days, and challenged with lethal heat. T2H pretreatment improved survival in both genotypes. Survival analysis and image-based quantification of projected green area showed that T2H-supported recover

During its complex life cycle, the malaria parasite survives dramatic environmental stresses, including large temperature shifts. Protein prenylation is required during asexual replication of Plasmodium falciparum, and the canonical heat shock protein 40 protein (HSP40; PF3D7_1437900) is posttranslationally modified with a 15-carbon farnesyl isoprenyl group. In other organisms, farnesylation of Hsp40 orthologs controls their localization and function in resisting environmental stress. In this work, we find that plastidial isopentenyl pyrophosphate (IPP) synthesis and protein farnesylation are

Plasmodium falciparum, the parasite responsible for nearly all cases of severe malaria, must survive challenging environments to persist in its human host. Symptomatic malaria is characterized by periodic fevers corresponding to the 48-hour asexual reproduction of P. falciparum in red blood cells. As a result, P. falciparum has evolved a diverse collection of heat shock proteins to mitigate the stresses induced by temperature shifts. Among the assortment of heat shock proteins in P. falciparum, there is only one predicted canonical cytosolic J domain protein, HSP40 (PF3D7_1437900). Here, we ge

The MarR-like transcriptional regulator and two ABC transporters encoded by the rcrRPQ operon in the dental caries pathogen Streptococcus mutans have important regulatory roles related to oxidative stress tolerance, genetic competence and (p)ppGpp metabolism. A unique feature of the rcrRPQ operon, when compared to other bacteria, is the presence of two peptides, designated Pep1 and Pep2, encoded in alternative reading frames at the 3' end of rcrQ. Here, we show that the rcrRPQ operon, including Pep1 and 2, is essential for S. mutans to survive and maintain viability at elevated temperatures. N

Sixty-nine vaginal swabs and 138 rectal swabs collected from 195 equids were analysed for the presence of thermotolerant bacteria, that is, bacteria surviving at 60+/-0.1 degrees C for one hour. Thermotolerant Escherichia coli, Enterobacter species, Klebsiella pneumoniae, Proteus species and Pseudomonas species were isolated from 41, 16, nine, three and three of the 138 rectal swabs, respectively; seven of the E coli and two of the Enterobacter species isolates survived pasteurisation at 63.8+/-0.1 degrees C for 30 minutes. All except three E coli, two Enterobacter species and one Proteus spec

SRF6 is a heat-inducible, non-RD receptor-like kinase that contributes to thermotolerance in Arabidopsis by modulating heat-, ER stress-, ABA-, and ROS-responsive pathways. Receptor-like kinases (RLKs) are central regulators of plant growth, development, and stress perception, yet their roles in heat stress (HS) responses remain largely unclear. Here, we identify STRUBBELIG-RECEPTOR-FAMILY 6 (SRF6) as a heat-inducible, plasma membrane-localized, non-arginine-aspartate (non-RD) RLK that contributes to thermotolerance in Arabidopsis thaliana. HS significantly induces SRF6 expression, and this in

Chloroplast stress granules (cpSGs) are emerging as dynamic suborganellar condensates that play a crucial role in mediating stress response in plants. In this study, we demonstrate that the chlorophyll biosynthesis enzyme protochlorophyllide oxidoreductase C (PORC) localizes to cpSGs in response to acute and prolonged heat stress in Arabidopsis (Arabidopsis thaliana). While PORC promoter activity was developmentally regulated and remained unresponsive to heat, PORC protein re-localized from a diffuse chloroplast distribution into punctate structures under elevated temperatures. This condensati

More frequent short-term episodes of extreme high temperatures caused by global warming seriously threaten plant growth and development around the world. Although heat stress can occur throughout the plant life cycle, exposure during the seedling stage severely impacts subsequent plant growth and productivity. In this study, we found that the Arabidopsis metacaspase mutant atmc1 exhibited a heat-sensitive phenotype at the seedling stage, and restoration of AtMC1 expression in the atmc1 substantially rescued the heat-sensitive phenotype, indicating an important role of AtMC1 in thermotolerance.

BACKGROUND: Liriomyza trifolii is an economically significant, invasive pest of horticultural and vegetable crops. The larvae form tunnels in foliage and hasten senescence and death. Outbreaks of L. trifolii often erupt in hot weather and are driven by thermotolerance; furthermore, the poor effectiveness of pesticides has made outbreaks more severe. But it is still unclear whether the development of insecticide tolerance will contribute to thermotolerance in L. trifolii.
RESULTS: To explore potential synergistic relationships between insecticide exposure and thermotolerance in L. trifolii, we

Wild-type cells of Ustilago maydis which have been briefly heated at 42 °C subsequently acquire resistance to the lethal effects of a higher temperature (48 °C). This induced thermotolerance develops over a 2 h period after treatment at 42 °C and is dependent on protein synthesis. It is distinct from the cellular response to DNA-damaging agents, since mutants deficient in this and other repair processes show the same induced thermotolerance as wild-type cells. However, UV-light treatment does induce some resistance to subsequent heat treatment. A single 48 °C heat treatment increases recombina
