In-situ preparation of hollow CdCoS2 heterojunction with enhanced photocurrent response for highly photoelectrochemical sensing of organophosphorus pesticides.

In this study, a porous hollow CdCoS2(2) microsphere was synthesized based on the ZIF-67-S MOFs derived method of sulfurization reaction and calcination process. Under visible light irradiation, the resulting CdCoS2(2) composite showed a markedly enhanced photoelectrochemical (PEC) response. The photocurrent value of the CdCoS2(2) modified ITO electrode was 93-fold and 41-fold than that of CoS and CdS materials, respectively. Promoting the photo-absorption ability by internal multilight scattering/reflection was due to the porous and hollow nature of CdCoS2(2). Furthermore, obtained CdCoS2(2)

Heart autonomic innervation during the acute phase of experimental American trypanosomiasis in the dog.

Heart autonomic innervation was studied in dogs during the acute phase of the experimental infection with the Berenice-78 strain of Trypanosoma cruzi. A glyoxylic acid-induced fluorescence method for catecholamines and a thiocholine method for demonstrating acetylcholinesterase activity showed the sympathetic and the parasympathetic nerve fibers, respectively. At day 34 of infection, moderate-to-intense rarefaction of both cholinergic and noradrenergic nerve fibers occurred in the atria of all animals coincident with moderate to intense myocarditis. In the ventricles, sympathetic denervation w

Analytical applications of enzymatic growth of quantum dots.

We have developed an analytical assay to detect the enzymatic activity of acetylcholine esterase and alkaline phosphatase based on the generation of quantum dots by enzymatic products. Acetylcholine esterase converts acetylthiocholine into thiocholine. The latter enhances the rate of decomposition of sodium thiosulfate into H(2)S, which in the presence of cadmium sulfate yields CdS quantum dots showing a time dependent exponential growth, typical of autocatalytic processes. This assay was also applied to detect acetylcholine esterase inhibitors. Alkaline phosphatase hydrolyzes thiophosphate an

Encapsulating gold nanoclusters into metal-organic frameworks to boost luminescence for sensitive detection of copper ions and organophosphorus pesticides.

Gold nanoclusters (Au NCs) with luminescence property are emerging as promising candidates in fluorescent methods for monitoring contaminants, but low luminescence efficiency hampers their extensive applications. Herein, GSH-Au NCs@ZIF-8 was designed by encapsulating GSH-Au NCs with AIE effect into metal-organic frameworks, achieving high luminescence efficiency and good stability through the confinement effect of ZIF-8. Accordingly, a fluorescent sensing platform was constructed for the sensitive detection of copper ions (Cu2+) and organophosphorus pesticides (OPs). Firstly, the as-prepared G

Adrenergic and acetylcholinesterase-positive innervation of palatine tonsils in mammals.

Innervation of human, feline and rabbit palatine tonsils was investigated. Adrenergic nerve components were visualized by formaldehyde-induced fluorescence of catecholamines and 5-HT, or by glyoxylic acid fluorescence, whereas acetylcholinesterase (AChE)-positive nerve structures were demonstrated by the direct thiocholine method. The largest density of adrenergic and AChE-positive nerve profiles was found in the adventitia of arterial branches in the fibrous capsula and septa, mainly in the form of periarterial nerve plexuses of different density. Fine nerve fibres lined the wall of small art

[A kinetic study of the blood serum cholinesterase activity in the freshwater fish Abramis ballerus].

In reaction of hydrolysis of choline and thiocholine esters of carbonic acids at 25 degrees C, cholinesterase activity of the blood serum from the fish A. ballerus has been studied by modified Ellman's method and potentiometric titration method. The activity is maximal in pH region 7.5-9.0 and is not inhibited by high concentration of substrates. Michaelis constants and maximal rates for the enzyme reactions were determined. Butyrylcholine and butyrylthiocholine were hydrolyzed with the highest rates by the serum. Some of the organophosphorus inhibitors (diisopropylfluorphosphate and DDVF) inh

Acetylcholinesterase-assisted photoelectrochemical solution gated graphene field-effect transistor for organophosphates pesticide detection.

This study develops a photoelectrochemical solution-gated graphene field-effect transistor (PEC-SGGT) for ultrasensitive organophosphates (OPs) detection, merging optoelectronic modulation with enzymatic signal amplification. The sensor employs a hybrid system of cadmium sulfide quantum dots (CdS QDs) and acetylcholinesterase (AChE), using acetylthiocholine (ATCh) as the substrate. Under light, CdS QDs generate electron-hole pairs, while AChE hydrolyzes ATCh into thiocholine (TCh), which enhances charge separation and amplifies photocurrent. OPs-induced inhibition of AChE reduces TCh productio

AChE and CoFe NPs-initiated cascade amplification for sensitive detection of organophosphorus pesticides.

Traditional methods for pesticide residue detection often rely on expensive instruments, are time-consuming, and lack cost efficiency. This investigation developed a fluorescence-based assay that utilizes cascade amplification through cholinesterase hydrolysis and thiamine (TH) oxidation for the detection of organophosphorus pesticides (OPs). A Prussian blue analog (CoFe NPs) with high catalytic activity for TH oxidation was synthesized. Cholinesterase hydrolyzes acetylthiocholine to produce thiocholine, which subsequently inhibits the CoFe NPs-catalyzed oxidation of TH. In the presence of par

A dual-channel colorimetric/fluorescent sensor array for discriminating aminoglycoside antibiotics in food via MOF-based inhibition.

Discriminating structurally similar Aminoglycoside antibiotics (AG) subtypes is highly desired yet remains a challenge. In this study, we developed a colorimetric and fluorescent dual-channel sensor array based on the inhibition of acetylcholinesterase-like activity MOF-808-Al for the discrimination of four AGs. The sensor leverages the competitive binding of AGs to Lewis acid and metal-OH sites on MOF-808-Al, which inhibits its AChE-like activity and prevents the generation of thiocholine. This in turn modulates the oxidation of o-phenylenediamine by Cu2+, leading to concentration-dependent c

Contrast of the biological activity of negatively and positively charged microwave synthesized CdSe/ZnS quantum dots.

Quantum dots (QDs) are semiconductor nanocrystals that have found use in bioimaging, cell tracking, and drug delivery. This article compares the cytotoxicity and cellular interactions of positively and negatively charged CdSe/CdS/ZnS QDs prepared by a microwave method using a murine alveolar macrophage-like cell culture model. Keeping the core semiconductor the same, QD charge was varied by altering the surface capping molecule; negatively charged QDs were formed with mercaptopropionic acid (MPA-QDs) and positively charged QDs with thiocholine (THIO-QDs). The size and charge of these two QDs w

AI-driven Cu/Mn-CeO2 nanozyme-functionalized paper-based biosensor for quantitative monitoring of alternariol in food.

The lack of rapid, user-friendly methods for alternariol (AOH), a prevalent emerging mycotoxin, presents a considerable analytical challenge. This work introduces an integrated sensing strategy advancing from classical homogeneous liquid-phase detection to a portable AI-enhanced paper-based biosensor. The system employed a Cu/Mn-CeO2 nanozyme as signal generator and amplifier, and exploited the inhibitory effect of AOH on acetylcholinesterase (AChE) to suppress the thiocholine (TCh)-mediated reduction of oxidized 3,3',5,5'-tetramethylbenzidine (oxTMB), enabling quantitative colorimetric readou

Construction of a copper nanocluster/MnO2 nanosheet-based fluorescent platform for butyrylcholinesterase activity detection and anti-Alzheimer's drug screening.

An abnormal level of butyrylcholinesterase (BChE) activity is highly connected with hepatic damage and Alzheimer's disease. Herein, a facile and efficient method was proposed for BChE detection by incorporating polyethyleneimine-capped copper nanoclusters (PEI-CuNCs) with manganese dioxide (MnO2) nanosheets. The emission of PEI-CuNCs can be significantly quenched by MnO2 nanosheets via the inner filter effect. With the addition of BChE, the hydrolysis of butyrylthiocholine iodide produces thiocholine which can reduce MnO2 nanosheets to Mn2+, thus resulting in the fluorescence recovery of PEI-C

Acidic pH and thiol-driven homogeneous cathodic electrochemiluminescence strategy for determining the residue of organophosphorus pesticide in Chinese cabbage.

Electrochemiluminescent (ECL) sensors for organophosphorus pesticides (OPs) have received considerable attention, whereas complicated electrode's immobilization, response to single hydrolysate and anodic emission correlated with ECL assays restrict their potential utilization. Herein, we developed a homogeneous dual-response cathodic ECL system for highly sensitive and reliable analysis of OP using CdTe QDs as emitters. CdTe QDs, emitting red light, were fabricated through a hydrothermal reaction and generated anodic and cathodic ECL emission upon stimulation of tripropyl amine and K2S2O8, res

Acetylcholinesterase and butyrylcholinesterase measurement in the pre-natal detection of neural tube defects and other fetal malformations.

Acetylcholinesterase activity in amniotic fluid was measured at 30 degree C by a reaction rate method employing acetyl-beta-methyl thiocholine as substrate and ethopropazine as a selective inhibitor of butyrylcholinesterase. This assay proved more specific than previously reported methods. Activity was greater in five cases of anencephaly (4.8-9.7 U/l) and nine cases of spinal bifida (5.1-8.6 U/l) than in 50 pregnancies with normal outcome (mean activity 2.0 +/- 0.9 (S.D.) U/l). There was no overlap between results from normal and neural-tube-defect groups, and the results showed no significan

Neuronal localization of pseudocholinesterase in the rat cerebellum: sagittal bands of Purkinje cells in the nodulus and uvula.

The histochemical distribution of pseudocholinesterase was studied in the rat cerebellum using Koelle's copper-thiocholine method. Throughout the cerebellum, pseudocholinesterase is uniformly localized in the endothelial cells of blood vessels and in the cell bodies and processes of the Bergmann glia. In addition, we demonstrate that exclusively in the ventral uvula and in the nodulus (lobules IXc and X of Larsell) pseudocholinesterase is localized in a small subpopulation of Purkinje cells. The cell bodies and dendrites of these labeled Purkinje cells form at least 4 distinct parallel bands e

Study on the ultrastructure and acetylcholinesterase activity in von Recklinghausen's neurofibromatosis.

The cutaneous nodules obtained from seven patients with von Recklinghausen's neurofibromatosis were investigated by electron microscopy, and ultrastructural localization of acetylcholinesterase activity was demonstrated in the nerve fibers of this tumor for the first time using Karnovsky's thiocholine method. The enzymatic activity was mainly found in unmyelinated fibers, exactly associated with their axonal membranes, the interspace between the apposing axonal and Schwann cell membrane, and some different mesaxons, which indicated their cholinergic nature. Almost all myelinated fibers and som

A fluorometric assay for acetylcholinesterase activity and inhibitor detection based on DNA-templated copper/silver nanoclusters.

A novel label-free, rapid, cost-effective, and highly sensitive fluorometric sensor has been constructed for the detection of acetylcholinesterase (AChE) activity and its inhibitor based on the fluorescence quenching of DNA-templated copper/silver nanoclusters (DNA-Cu/AgNCs). In this assay, AChE catalyzes the hydrolysis of acetylthiocholine (ATCh) to form thiocholine which induces fluorescence quenching of DNA-Cu/AgNCs. The AChE activity could be detected as low as 0.05mU/mL and with a linear range from 0.05 to 2.0mU/mL. This assay offers a very convenient "mix and detect" approach for AChE ac

A sensitive fluorescence assay of organophosphorus pesticides using acetylcholinesterase and copper-catalyzed click chemistry.

Organophosphorus pesticides (OPs) are widely used in agricultural fields, but exhibit high toxicity to human beings. A sensitive fluorescence assay for organophosphorus pesticides was developed using the inhibition of acetylcholinesterase (AChE) activity and the copper-catalyzed click chemical reaction. In the click reaction, two hybridized DNA probes can be ligated with copper ions, inducing a fluorescence quenching during the strand displacement reaction. AChE can hydrolyze acetylthiocholine (ATCh) to form thiocholine (TCh) which contains a thiol group. TCh will react with copper ions, block
