Evaluation of color photostability and structure of thylakoids in isolation and after undergoing food processing treatments.

Improving the photostability of natural chlorophyll remains a substantial challenge for its application in the food industry. Thylakoids in chloroplasts are pouch-like compartments contain over 90 % of total chlorophyll. In this study, the color photostability of thylakoids were evaluated. Accelerated photodegradation experiments were conducted at 5 °C under an intense illumination model (15,000 lm). Thylakoids exhibited a green retention rate of 79.2 % after five days of illumination, comparable to sodium copper chlorophyllin (78.1 %), a commonly used synthetic chlorophyll derivative. The lig

The Tomato SlVIPP1 Gene Is Required for Plant Survival Through the Proper Development of Chloroplast Thylakoid Membrane.

Since membranes play essential roles in all living beings, all cells have developed mechanisms for efficient and fast repair of membrane damage. In Escherichia coli, the Phage shock stress A (PspA) protein is involved in the maintenance of the integrity of its inner membrane in response to the damage produced by exposure to stress conditions. A role in thylakoid membrane maintenance and reorganization has been proposed for Vesicle Inducing Protein in Plastid 1 (VIPP1), the putative PspA ortholog in Arabidopsis thaliana. While some membranes of plant cells have been extensively studied, the bio

Evidence for an association of the early light-inducible protein (ELIP) of pea with photosystem II.

The precursor to the nuclear-coded 17 kDa early light-inducible protein (ELIP) of pea has been transported into isolated intact chloroplasts. The location of the mature protein in the thylakoid membranes was investigated after using cleavable crosslinkers such as DSP and SAND in conjunction with immunofractionation methods and by application of mild detergent fractionation. We show that ELIP is integrated into the membranes via the unstacked stroma thylakoids. After isolation of protein complexes by solubilization of membranes with Triton X-100 and sucrose density-gradient centrifugation the c

Plant sample integrity is important for efficient photosystem II repair and thylakoid remodelling in Arabidopsis.

The thylakoid is a dynamic membrane that houses the protein complexes involved in the light-dependent reactions of photosynthesis. Although light provides the energy that drives these reactions, it also damages the photosynthetic apparatus, particularly photosystem II. This damage can be repaired via the photosystem II repair cycle, a process thought to be accompanied by structural rearrangements of the thylakoid membrane. When the light-induced damage exceeds the capacity for repair, photoinhibition occurs, resulting in a decline in photosynthetic capacity. While photoinhibition has been exte

Transcriptomic and proteomic analyses reveal new insight into chlorophyll synthesis and chloroplast structure of maize leaves under zinc deficiency stress.

Zinc (Zn) deficiency reduces chlorophyll content and damages chloroplast structure in leaves. However, little is known about the responses of chlorophyll synthesis and chloroplast structure of plants to Zn deficiency stress at the transcriptomic and proteomic level. In the present study, maize seedlings of cultivar ZD958 were grown in nutrient solution with 0 and 1.0 μmol·L-1 Zn treatments. Zn deficiency stress reduced photosynthetic rate at 10 and 15 days after treatment (DAT) and the chlorophyll content (SPAD value) at 15 DAT. Eight differentially expressed genes and 6 differential abundance

Heterogeneous origin of carbonic anhydrase activity of thylakoid membranes.

Carbonic anhydrase activities of pea thylakoids as well as thylakoid fragments enriched either in Photosystem 1 (PS1-membranes) or Photosystem 2 (PS2-membranes) were studied. The activity of PS1-membranes if calculated on chlorophyll basis was much higher than the activity of PS2-membranes. Acetazolamide, a non-permeable inhibitor of carbonic anhydrases, increased carbonic anhydrase activity of PS2-membranes at concentrations lower than 10(-6) M and suppressed this activity only at higher concentrations. A lipophilic inhibitor of carbonic anhydrases, ethoxyzolamide, effectively suppressed the

Systems biology approach in Chlamydomonas reveals connections between copper nutrition and multiple metabolic steps.

In this work, we query the Chlamydomonas reinhardtii copper regulon at a whole-genome level. Our RNA-Seq data simulation and analysis pipeline validated a 2-fold cutoff and 10 RPKM (reads per kilobase of mappable length per million mapped reads) (~1 mRNA per cell) to reveal 63 CRR1 targets plus another 86 copper-responsive genes. Proteomic and immunoblot analyses captured 25% of the corresponding proteins, whose abundance was also dependent on copper nutrition, validating transcriptional regulation as a major control mechanism for copper signaling in Chlamydomonas. The impact of copper deficie

Photodamaged PSII does not accumulate in the non-appressed thylakoid membranes in the absence of PSII repair.

Photosystem II (PSII) is highly sensitive to light-induced damage. Photoinhibition, the light-dependent inactivation of PSII, is associated with an increase of excitation-energy quenching. Recovery from photoinhibition involves migration of PSII complexes from the appressed to the non-appressed region of the thylakoid membrane, where D1 (the PSII core protein most sensitive to photodamage) is degraded and repair occurs. However, it remains unclear whether damaged PSII core complexes accumulate in the stroma lamellae when repair is blocked. Here, we combined confocal Fluorescence Lifetime Imagi

Thylakoid membrane transporter SecE1 is required for chloroplast development and photosynthetic function in Arabidopsis.

Chloroplast biogenesis depends on protein transport, pigment biosynthesis, and thylakoid membrane assembly, which are coordinated through anterograde (nucleus-to-plastid) and retrograde (plastid-to-nucleus) signaling. The chloroplast Sec translocon mediates the import of nuclear-encoded precursor proteins across the thylakoid membrane, yet the regulatory role of its core subunit SecE1 in chloroplast development and photosynthetic performance remains incompletely understood. Here, we characterized a chlorotic Arabidopsis mutant with partial loss of SecE1 function. Compared with the wild type an

The pathogenic effector PHYL1JWB interacts with ZjCURT1A to mediate jujube witches' broom-induced photosynthesis impairment of jujube trees.

Photosynthesis impairment and the accompanying leaf yellowing represent a prominent and physiologically critical symptom of phytoplasma infection. However, the underlying molecular mechanisms remain poorly understood. In this study, we identified the Jujube Witches' Broom pathogenic effector PHYL1JWB, which was previously reported to induce phyllody in jujube, as a key inducer of photosynthesis impairment. Overexpression of PHYL1JWB in sour jujube and Arabidopsis thaliana disrupted photosynthesis by causing severe disorganization of chloroplast ultrastructure. PHYL1JWB directly interacts with

Evaluating the efficiency of a photoelectrochemical electrode constructed with photosystem II-enriched thylakoid membrane fragments.

The photoelectrochemical electrode has been intensively studied in recent years as a means of generating electricity from light through the use of intact thylakoid membranes or highly purified photosystem II. PSII-enriched thylakoid membrane fragments (PSII-BBY), also have the potential to construct the photoelectrochemical anode. In this study, we examined the feasibility of utilizing PSII-BBY preparations to construct a relatively inexpensive photoelectrochemical anode with a comparable current density and a reasonable stability. Expression profiling and bioinformatic analyses of a novel stress-regulated multispanning transmembrane protein family from cereals and Arabidopsis.

Cold acclimation is a multigenic trait that allows hardy plants to develop efficient tolerance mechanisms needed for winter survival. To determine the genetic nature of these mechanisms, several cold-responsive genes of unknown function were identified from cold-acclimated wheat (Triticum aestivum). To identify the putative functions and structural features of these new genes, integrated genomic approaches of data mining, expression profiling, and bioinformatic predictions were used. The analyses revealed that one of these genes is a member of a small family that encodes two distinct groups of

Structural basis of HSP90C, a highly active chloroplastic HSP90 chaperone from Arabidopsis thaliana.

Chloroplasts are the main energy-producing organelles in plants, responsible for photosynthesis, CO2 fixation, and O2 production. These processes rely on the import of numerous nucleus-encoded proteins into the chloroplast and, eventually, the thylakoids. While translocation systems across chloroplast and thylakoid membranes are well characterized, the stromal route between these membranes remains poorly understood. The chloroplastic HSP90 (HSP90C) is likely to play a key role in this process, yet its structure and molecular mechanisms are unknown. Here, we combine structural and biophysical a

Calcium binding to the subunit c of E. coli ATP-synthase and possible functional implications in energy coupling.

The 8-kDa subunit c of the E. coli F0 ATP-synthase proton channel was tested for Ca++ binding activity using a 45Ca++ ligand blot assay after transferring the protein from SDS-PAGE gels onto polyvinyl difluoride membranes. The purified subunit c binds 45Ca++ strongly with Ca++ binding properties very similar to those of the 8-kDa CF0 subunit III of choloroplast thylakoid membranes. The N-terminal f-Met carbonyl group seems necessary for Ca++ binding capacity, shown by loss of Ca++ binding following removal of the formyl group by mild acid treatment. The dicyclohexylcarbodiimide-reactive Asp-61

Protein identification of purified particles isolated from spinach thylakoids by deoxycholate electrophoresis.

Three thylakoid complexes were isolated by deoxycholate preparative electrophoresis. The protein composition of each fraction was analyzed by SDS analytical electrophoresis. No protein of the PS 1 enriched fraction (fraction 1) was found in the PS 2 enriched fraction (fraction 2) and inversely. The antenna complex (fraction 3) did not have any contamination by proteins of fraction 1 or fraction 2. Fraction 1 was mainly composed of the CP1, the reaction center complex of the PS1, and by low molecular weight proteins, previously found in other PS 1 preparations. Tentative assignments of these pr

Cell membrane-encased thylakoid as white light triggered PDT therapy for facile and targeted choroidal melanoma treatment.

Choroidal melanoma, the most prevalent subtype of uveal melanoma with a bleak prognosis, necessitates safer and more efficient treatment methods. Tumor cell membrane-modified nanodelivery systems have exhibited significant enhancement in photodynamic therapy (PDT). Currently, photosensitizers utilized in PDT encompass porphyrin-related organic molecules and other types, requiring complex and intricate systems construction. This study focuses on harnessing the naturally occurring photodynamic active photosynthetic unit-thylakoid which serve as a major source of singlet oxygen. Initially, thylak

Requirement of phosphatidylglycerol for maintenance of photosynthetic machinery.

Phosphatidylglycerol (PG) is a ubiquitous component of thylakoid membranes. Experiments with the pgsA mutant of the cyanobacterium Synechocystis sp. PCC6803 defective in biosynthesis of PG have demonstrated an indispensable role of PG in photosynthesis. In the present study, we have investigated the light susceptibility of the pgsA mutant with regard to the maintenance of the photosynthetic machinery. Growth of the mutant cells without PG increased the light susceptibility of the cells and resulted in severe photoinhibition of photosynthesis upon a high-light treatment, whereas the growth in t

Development and use of domain-specific antibodies in a characterization of the large subunits of soybean photosystem 1.

The molecular architecture of the soybean photosystem 1 reaction center complex was examined using a combination of surface labeling and immunological methodology on isolated thylakoid membranes. Synthetic peptides (12 to 14 amino acids in length) were prepared which correspond to the N-terminal regions of the 83 and 82.4 kDa subunits of photosystem 1 (the PsaA and PsaB proteins, respectively). Similarly, a synthetic peptide was prepared corresponding to the C-terminal region of the PsaB subunit. These peptides were conjugated to a carrier protein, and were used for the production of polyclona

The properties of the positively charged loop region in PSI-G are essential for its "spontaneous" insertion into thylakoids and rapid assembly into the photosystem I complex.

The PSI-G subunit of photosystem I (PSI) is an 11-kDa membrane protein that plays an important role in electron transport between plastocyanin and PSI and is involved in the stability of the PSI complex. Within the complex, the PSI-G subunit is bound to PSI-B and is in contact with Lhca1. PSI-G has two transmembrane spans connected by a positively charged stromal loop. The loop is inaccessible to proteases, indicating a tightly bound location within the PSI complex. Here, we have studied the insertion mechanism and assembly of PSI-G. We show that the protein inserts into thylakoids by a direct

CP26 is not involved in qE- or qZ-type non-photochemical quenching in Arabidopsis.

CP26 is a monomeric minor light-harvesting complex of Photosystem II (LHCII) protein located at the interface between LHCII trimers and the PSII core in thylakoid membranes. Previous studies have proposed that CP26 plays a role in non-photochemical quenching (NPQ) in addition to light harvesting. Here, we utilized biophysical and pharmacological approaches to investigate this role using single- and higher-order Arabidopsis (Arabidopsis thaliana) cp26 mutants, examining its relationship to known NPQ regulators (Photosystem II subunit S, PsbS, violaxanthin de-epoxidase, and the pH gradient acros
