Double (fluorescent and spin) sensors for detection of reactive oxygen species in the thylakoid membrane.

A series of dansylated sterically hindered amines designed to trapping reactive oxygen species, were synthesized. Compounds were tested in isolated thylakoid membranes subjected to photoinhibition by excess photosynthetically active radiation (400-700 nm). DanePy showed good selectivity for singlet oxygen and the formation of nitroxide was detected by appearance of ESR signal and quenching fluorescence.

Evaluation of color photostability and structure of thylakoids in isolation and after undergoing food processing treatments.

Improving the photostability of natural chlorophyll remains a substantial challenge for its application in the food industry. Thylakoids in chloroplasts are pouch-like compartments contain over 90 % of total chlorophyll. In this study, the color photostability of thylakoids were evaluated. Accelerated photodegradation experiments were conducted at 5 °C under an intense illumination model (15,000 lm). Thylakoids exhibited a green retention rate of 79.2 % after five days of illumination, comparable to sodium copper chlorophyllin (78.1 %), a commonly used synthetic chlorophyll derivative. The lig

Thylakoid-based photosynthetic hydrogel for dual-phase ROS regulation in infected wounds.

Reactive oxygen species (ROS) play a dual-edged role in infected wounds: they are vital for initial pathogen clearance but exacerbate local oxidative stress and delay healing once the infection is contained. Traditional photodynamic therapy (PDT) fails to meet these dynamic needs because its initial bactericidal ROS generation is severely restricted by wound hypoxia, while it simultaneously lacks the ability to scavenge excess ROS in the later stages to relieve oxidative stress. In this work, we developed a self-oxygenating photodynamic hydrogel (denoted GCT) by co-encapsulating spinach-derive

Two inhibition targets by [Cr(2gb)(3)](3+) and [Co(2gb)(3)](3+) on redox enzymes of spinach thylakoids.

[Cr(2gb)(3)]Cl(ZnCl(4)), [Cr(2gb)(3)]Cl(3), and [Co(2gb)(3)]Cl(3) were synthesized and characterized. Their chemical structures and the oxidation states of their metal centers remained unchanged in solution. The effects of these compounds, CrCl(3) and [Co(NH(3))(6)]Cl(3), on photosynthesis were investigated. The coordination compounds inhibit ATP synthesis and electron flow (basal, phosphorylating, and uncoupled), behaving as Hill reaction inhibitors. The target for [Cr(2gb)(3)]Cl(ZnCl(4)) is located at the Q(B) level. In contrast, the interaction sites of [Cr(2gb)(3)]Cl(3) and [Co(2gb)(3)]Cl(

Plant sample integrity is important for efficient photosystem II repair and thylakoid remodelling in Arabidopsis.

The thylakoid is a dynamic membrane that houses the protein complexes involved in the light-dependent reactions of photosynthesis. Although light provides the energy that drives these reactions, it also damages the photosynthetic apparatus, particularly photosystem II. This damage can be repaired via the photosystem II repair cycle, a process thought to be accompanied by structural rearrangements of the thylakoid membrane. When the light-induced damage exceeds the capacity for repair, photoinhibition occurs, resulting in a decline in photosynthetic capacity. While photoinhibition has been exte

Genes and transcripts for the polypeptides of the cytochrome b6/f complex from spinach thylakoid membranes.

Cytochrome b6/f complex was prepared from washed thylakoid membranes by a procedure involving detergent treatment and centrifugation in sucrose gradients. The complex is composed of at least four polypeptide species, cytochrome f which occurs in two variant forms (mol. wt. 34/33 kd), cytochrome b6 (23 kd), the high-potential Rieske iron-sulfur protein (19 kd) and a fourth subunit (17 kd) of unknown function. Transcripts for the cytochromes f, b6 and subunit 4 were found in plastid RNA, those for the Rieske iron-sulfur protein in cytosolic poly(A) RNA. Transcripts for cytochrome b6 and subunit

The light-harvesting state of LHCII is remarkably robust to changes in thylakoid lipid composition and pH.

LHCII is the main antenna complex of plants and green algae. Under low light it maximizes light absorption for photosynthesis, whereas under excess light it becomes the principal site of nonphotochemical quenching (NPQ), a photoprotective process triggered by the acidification of the thylakoid lumen that dissipates excess excitation energy as heat. LHCII is thus believed to be able to switch between different conformations, associated with its light-harvesting and photoprotective roles. The thylakoid lipid environment has been proposed as an important determinant to modulate LHCs' structure an

Membrane lipid-driven thylakoid biogenesis coordinating chlorophyll synthesis and expression of photosynthetic proteins.

Chloroplasts in seed plants differentiate from proplastids or, occasionally, from other types of plastids. The development of the thylakoid membrane (TM) is a key process in chloroplast biogenesis, enabling plants to perform photosynthesis. The TM is a lipid bilayer membrane system densely packed with photosynthetic protein-cofactor complexes, and its formation requires the coordinated synthesis of membrane lipids, photosynthetic proteins, and cofactors particularly chlorophyll. During chloroplast biogenesis, membrane lipids are synthesized in the envelope membranes and transferred to the TM t

Photodamaged PSII does not accumulate in the non-appressed thylakoid membranes in the absence of PSII repair.

Photosystem II (PSII) is highly sensitive to light-induced damage. Photoinhibition, the light-dependent inactivation of PSII, is associated with an increase of excitation-energy quenching. Recovery from photoinhibition involves migration of PSII complexes from the appressed to the non-appressed region of the thylakoid membrane, where D1 (the PSII core protein most sensitive to photodamage) is degraded and repair occurs. However, it remains unclear whether damaged PSII core complexes accumulate in the stroma lamellae when repair is blocked. Here, we combined confocal Fluorescence Lifetime Imagi

Evaluating the efficiency of a photoelectrochemical electrode constructed with photosystem II-enriched thylakoid membrane fragments.

The photoelectrochemical electrode has been intensively studied in recent years as a means of generating electricity from light through the use of intact thylakoid membranes or highly purified photosystem II. PSII-enriched thylakoid membrane fragments (PSII-BBY), also have the potential to construct the photoelectrochemical anode. In this study, we examined the feasibility of utilizing PSII-BBY preparations to construct a relatively inexpensive photoelectrochemical anode with a comparable current density and a reasonable stability. Characteristics and functionality of appetite-reducing thylakoid powders produced by three different drying processes.

BACKGROUND: Thylakoids, a chloroplast membrane extracted from green leaves, are a promising functional ingredient with appetite-reducing properties via their lipase-inhibiting effect. Thylakoids in powder form have been evaluated in animal and human models, but no comprehensive study has been conducted on powder characteristics. The aim was to investigate the effects of different isolation methods and drying techniques (drum-drying, spray-drying, freeze-drying) on thylakoids' physicochemical and functional properties. RESULTS: Freeze-drying yielded thylakoid powders with the highest lipase-in

Specific role of phosphatidylglycerol and functional overlaps with other thylakoid lipids in Arabidopsis chloroplast biogenesis.

With phosphate deficiency, the role of phosphatidylglycerol is compensated by increased glycolipid content in thylakoid membrane biogenesis but not photosynthetic electron transport in Arabidopsis chloroplasts. In plants and cyanobacteria, anionic phosphatidylglycerol (PG) is the only major phospholipid in thylakoid membranes, where neutral galactolipids monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG) are predominant. In addition to provide a lipid bilayer matrix, PG plays a specific role in photosynthetic electron transport. Non-phosphorous sulfoquinovosyldiacylglyce

Carbonic anhydrase activity in Arabidopsis thaliana thylakoid membrane and fragments enriched with PSI or PSII.

The procedure of isolating the thylakoids and the thylakoid membrane fragments enriched with either photosystem I or photosystem II (PSI- and PSII-membranes) from Arabidopsis thaliana leaves was developed. It differed from the one used with pea and spinach in durations of detergent treatment and centrifugation, and in concentrations of detergent and Mg(2+) in the media. Both the thylakoid and the fragments preserved carbonic anhydrase (CA) activities. Using nondenaturing electrophoresis followed by detection of CA activity in the gel stained with bromo thymol blue, one low molecular mass carri

NIR-Driven Nanomotors Integrating With Platelet-Thylakoid Hybrid Membranes for Synchronized Thrombolysis and Vascular Remodeling.

The pathological interplay of oxidative stress, inflammation, and thrombosis driven by endothelial injury creates a self-perpetuating cycle that undermines conventional thrombolytic therapies. Herein, near-infrared (NIR)-responsive nanomotors are constructed by integrating thylakoid and platelet membranes on strontium-doped mesoporous polydopamine nanoparticles (PSr@PT NPs), which enables synchronized thrombus dissolution and vascular microenvironment restoration. Following intravenous administration, PSr@PT nanomotors exhibited preferential accumulation within thrombi and were internalized by

Phycoerythrins of the oxyphotobacterium Prochlorococcus marinus are associated to the thylakoid membrane and are encoded by a single large gene cluster.

An intrinsic divinyl-chlorophyll a/b antenna and a particular form of phycobiliprotein, phycoerythrin (PE) III, coexist in the marine oxyphotobacterium Prochlorococcus marinus CCMP 1375. The genomic region including the cpeB/A operon of P. marinus was analysed. It encompasses 10,153 nucleotides that encode three structural phycobiliproteins and at least three (possibly five) different polypeptides analogous to cyanobacterial or red algal proteins involved either in the linkage of subunits or the synthesis and attachment of chromophoric groups. This gene cluster is part of the chromosome and is

Thylakoids suppress appetite by increasing cholecystokinin resulting in lower food intake and body weight in high-fat fed mice.

Thylakoids are membranes isolated from plant chloroplasts which have previously been shown to inhibit pancreatic lipase/colipase catalysed hydrolysis of fat in vitro and induce short-term satiety in vivo. The purpose of the present study was to examine if dietary supplementation of thylakoids could affect food intake and body weight during long-term feeding in mice. Female apolipoprotein E-deficient mice were fed a high-fat diet containing 41% of fat by energy with and without thylakoids for 100 days. Mice fed the thylakoid-enriched diet had suppressed food intake, body weight gain and body fa

Transmembrane distribution of phospholipids and their involvement in electron transport, as revealed by phospholipids A2 treatment of spinach thylakoids.

Thylakoid membranes were treated with either pancreatic or snake venom phospholipase A2, and the residual phospholipid content of these membranes was determined and compared to the rates of Photosystem II and/or Photosystem I electron transports. The hydrolysis curves of both phosphatidylglycerol and phosphatidylcholine displayed a first, rapid phase which was almost temperature-insensitive, followed by a second, slower phase which depended strongly on the temperature. When pancreatic phospholipase A2 had access either to the outer face or to both faces of the thylakoid membrane, either only p

Exploring thylakoid emergence: evolution of membrane biogenesis and photosystem II assembly in early-diverging cyanobacteria.

Thylakoid membranes (TM) in cyanobacteria and chloroplasts host the light-dependent reactions of oxygenic photosynthesis. Gloeobacterales, the earliest-diverging cyanobacterial lineage, lack TM and perform photosynthesis in the cytoplasmic membrane (CM), representing an ancestral state relative to other cyanobacteria (Phycobacteria). This study investigates the evolutionary origin of TM. Phylogenomic analyses were performed across a phylogenetically diverse set of cyanobacteria, including extensive representation of basal lineages (Gloeobacterales, Thermostichales, Gloeomargaritales, and Pseud

CurT dosage quantitatively affects thylakoid structure and PSII performance in Synechocystis sp. PCC 6803.

The photosynthetic thylakoid membrane systems of many oxygenic photosynthesizers form elaborate three-dimensional structures. Curvature Thylakoid (CurT/CURT1) proteins are central regulators of thylakoid architecture in cyanobacteria and plants, driving both grana stacking and the formation of thylakoid convergence zones (TCZs) by promoting membrane curvature while also contributing to green-algal chloroplast and cyanobacterial cell division. While the functional role of grana stacks in terrestrial photosynthesis is well established, the physiological significance of cyanobacterial TCZs remain

The Tomato SlVIPP1 Gene Is Required for Plant Survival Through the Proper Development of Chloroplast Thylakoid Membrane.

Since membranes play essential roles in all living beings, all cells have developed mechanisms for efficient and fast repair of membrane damage. In Escherichia coli, the Phage shock stress A (PspA) protein is involved in the maintenance of the integrity of its inner membrane in response to the damage produced by exposure to stress conditions. A role in thylakoid membrane maintenance and reorganization has been proposed for Vesicle Inducing Protein in Plastid 1 (VIPP1), the putative PspA ortholog in Arabidopsis thaliana. While some membranes of plant cells have been extensively studied, the bio
