Chicken thymocyte antigen which participates in cell proliferations of thymocytes and tumour-derived lymphoid cell lines.

A novel antigen on chicken thymocytes was defined by CETHB1 and CETH46 monoclonal antibodies (mAbs) that were prepared against chick embryonic thymocytes. CETHB1 and CETH46 mAbs recognized different epitopes on the same membrane antigen (molecular weight: 76.2 kDa). These mAbs reacted with >80% of thymocytes of 14-day-old embryos to 8-week-old chickens. Almost all splenocytes, peripheral blood lymphocytes and bursa cells were negative, and only 7.7% of bone marrow cells were positive for both antibodies. In two-colour analysis with mAbs reacting to T cell markers (CD4 or CD8), most CETHB1 posi

Signaling and Transcriptional Control of Thymocyte Differentiation.

This review summarizes key topics discussed in the session "Biology within the Thymus" at the 54th Annual Meeting of the Japanese Society for Immunology, which highlighted recent advances in our understanding of T-cell development and fate determination. In particular, the session focused on transcriptional regulation of T-lineage commitment and the mechanisms by which T-cell receptor (SP) signaling governs thymocyte differentiation. Early T-lineage commitment is established through the integration of thymic microenvironmental cues and intrinsic transcription factors, including Bcl11b, PU.1, a

Cholinergic regulation of thymocyte negative selection.

The immune and nervous systems use a common chemical language for communication, namely, the cholinergic signaling involving acetylcholine (ACh) and its receptors (AChRs). Whether and how this language also regulates the development of the immune system is poorly understood. Here, we show that mouse CD4+CD8+ double-positive thymocytes express high levels of α9 nicotinic AChR (nAChR) and that this receptor controls thymic negative selection. α9 nAChR-deficient mice show an altered T cell receptor (TCR) repertoire and reduced CD4+ and CD8+ T cells in a mixed bone marrow chimera setting. α9 nAChR

An integrative mechanistic model of thymocyte dynamics.

BACKGROUND: The thymus plays a central role in shaping human immune function. A mechanistic, quantitative description of immune cell dynamics and thymic output under homeostatic conditions and various patho-physiological scenarios are of particular interest in drug development applications, e.g., in the identification of potential therapeutic targets and selection of lead drug candidates against infectious diseases. METHODS: We here developed an integrative mathematical model of thymocyte dynamics in human. It incorporates mechanistic features of thymocyte homeostasis as well as spatial const

A model for preservation of thymocyte-depleted thymus.

DiGeorge syndrome is a disorder caused by a microdeletion on the long arm of chromosome 22. Approximately 1% of patients diagnosed with DiGeorge syndrome may have an absence of a functional thymus, which characterizes the complete form of the syndrome. These patients require urgent treatment to reconstitute T cell immunity. Thymus transplantation is a promising investigational procedure for reconstitution of thymic function in infants with congenital athymia. Here, we demonstrate a possible optimization of the preparation of thymus slices for transplantation through prior depletion of thymocyt

The fate of thymocytes labeled in vivo with CFSE.

The fate of thymic emigrants had so far been studied using a variety of markers, each of which had inherent limitations as to stability, toxicity, or selectivity. We describe a new technique which relies on the in vivo injection of CFSE, an esterified vital dye hitherto used at 80 times lower concentrations for in vitro cell labeling. We show that CFSE labels a representative sample of all thymocyte subsets and that these migrate at a rate of approximately 2-3 x 10(6) cells/day to peripheral lymphoid organs. We show that they enter lymph nodes at day 1 postinjection and stay for at least 21 da

P2x purinoceptor is not important in thymocyte apoptosis.

Since the P2X purinoceptor has been shown to be homologous with RP-2, a gene activated in thymocytes undergoing apoptosis after glucocorticoid treatment, we have investigated the possible role of P2X purinoceptors in thymocyte apoptosis. We report that P2X purinoceptor mRNA is expressed both in untreated rat thymocytes and thymocytes treated with the synthetic glucocorticoid, 6 alpha-methylprednisolone, in a dose that can induce apoptosis. Furthermore, the P2X receptor agonist, extracellular ATP, was unable to trigger apoptosis in rat thymocytes and had no effect on methylprednisolone-induced

Serological identification of thymocyte differentiation antigens.

We have examined subfractions of human thymocytes for the expression of novel differentiation antigens. Non-HLA alloantisera procured from multiparous women served as antibody probes. Thymocytes from five individuals were sequentially separated by discontinuous Percoll density gradient centrifugation and a peanut agglutinin (PNA) panning technique. Subfractions were selected and examined for their relative intensity of HLA class I and CD1 antigens as determined by cytofluorometric analysis. Two subfractions were characterized as follows: an immature population (Fr6 PNA-) expressed a high level

Different roles for glucocorticoids in thymocyte homeostasis?

Glucocorticoids (GCs) have important immunoregulatory effects on thymocytes and T cells. Ectopic production of GCs has been demonstrated in thymic epithelial cells (TECs) but the role of GCs in thymocyte homeostasis is controversial. Studies in several different mouse models, genetically modified for the GC receptor (GR) expression or function, have demonstrated conflicting results in terms of the effect of the hormone on thymocytes. Here, we summarize these data and suggest that GCs can mediate both positive and negative effects in the organ depending on the local hormonal concentration. Basa

Androgens indirectly accelerate thymocyte apoptosis.

Apoptotic processes, or the disturbance of the natural regulation of these processes, may be involved in the pathogenesis of autoimmune diseases (AID). Women are, in general, more susceptible than men to develop AID like rheumatoid arthritis. Androgens and glucocorticoids, in contrast to oestrogens, have favourable effects in AID models as well as in human AID. It is known that glucocorticoids (GC), used for treatment of AID, increase apoptosis in the thymus resulting in decreased numbers of CD4+ CD8+ thymocytes. It was asked whether androgens, in contrast to oestrogens, exert their favourable

Development of double-positive thymocytes at single-cell resolution.

BACKGROUND: T cells generated from thymopoiesis are essential for the immune system, and recent single-cell studies have contributed to our understanding of the development of thymocytes at the genetic and epigenetic levels. However, the development of double-positive (DP) T cells, which comprise the majority of thymocytes, has not been well investigated. METHODS: We applied single-cell sequencing to mouse thymocytes and analyzed the transcriptome data using Seurat. By applying unsupervised clustering, we defined thymocyte subtypes and validated DP cell subtypes by flow cytometry. We classifi

TRAF6 directs commitment to regulatory T cells in thymocytes.

Regulatory T cells (Tregs), a subset of CD4(+) helper T cells, are crucial for immunological self-tolerance. Defect in development or function of Tregs results in autoimmune disease in human and mice. Whereas it is known that Tregs mainly develop in the thymus, the molecular mechanism underlying development of Treg is not fully understood. TRAF6-deficient mice showed a severe defect in the Treg development in thymus. In vitro fetal thymic organ culture experiments indicated that the defect is ascribed to the absence of TRAF6 in thymic cells. Moreover, mixed fetal liver transfer experiments rev

Evaluation of GM1 ganglioside-mediated apoptosis in feline thymocytes.

Cats with inherited GM1 gangliosidosis (GM1 mutant cats) have premature thymic involution characterized by decreased total thymocytes primarily affecting the CD4+ CD8+ subpopulation. While GM1 mutant cats have increased cell surface GM1 gangliosides, as determined by cholera toxin B binding, on both thymocytes and peripheral lymph node cells only thymocytes show increased apoptosis. To determine if GM1 gangliosides can increase the occurrence of apoptosis in feline thymocytes directly, we added exogenous GM1 ganglioside (GM1) to feline thymocyte primary cultures and compared the results to apo

Ethanol increases apoptotic cell death of thymocytes in vitro.

Exposure of animals to ethanol causes thymic atrophy in adults and fetuses. Whether direct effects of ethanol contribute to thymic atrophy or whether indirect effects are entirely responsible is at present unknown. In the normal animal, large numbers of thymocytes undergo a physiological form of cell death referred to as "apoptosis." To determine if ethanol affects the process of apoptosis, studies were undertaken in which mouse thymocytes were cultured overnight in the presence or absence of ethanol. Apoptotic cell death was analyzed by flow cytometric quantitation of apoptotic nuclei, by flu

Tolerogenic ability of thymocytes in organ-cultured thymus lobes.

It is generally believed that macrophages and dendritic cells are the major cell populations that present tolerogenic self antigens to developing thymocytes. However, it is still controversial whether self antigens expressed on thymocytes themselves work as tolerogens in the thymus. To evaluate this possibility, Thy-1 bright cells were sorted out from fetal thymus cells on the 15th gestation day, and were colonized into 2'-deoxyguanosine-treated allogeneic thymus lobes. The repopulated thymus lobes were organ-cultured, and the allo-specific killer activity of thymocytes recovered from the lobe

Phencyclidine blocks voltage-dependent potassium currents in murine thymocytes.

We investigated the effect of phencyclidine (PCP) on the voltage-dependent K+ current in whole cell recordings from murine thymocytes. PCP caused a dramatic, reversible and dose-dependent decrease of the current. An analysis of the dose-response relationship suggests a single site of action for PCP with an IC50 of 4.7 microM. Dextrorphan and naloxone also inhibited the current, although their effects were of a lower magnitude than those of PCP. Neither 10 microM dextrorphan nor 100 microM naloxone antagonized the inhibitory action of 10 microM PCP. The analysis of the dose-response curve for P

[The creatine kinase system in rat thymus and thymocytes].

The content of creatine phosphate, creatine and creatine kinase activity in thymus is shown to be 17.6, 5 and 4 times respectively higher, than in thymocytes isolated from this organ, both the level of adenine nucleotides and adenylate energy charge being practically the same. The creatine phosphate content in thymocytes decreases with addition of papaverine and remains unchanged under the influence of adenosine and concanavalin A. The creatine kinase activity increases considerably during the concanavalin A-induced thymocyte blasttransformation reaction. Creatine inhibits blasttransformation

MicroRNA-653 Inhibits Thymocyte Proliferation and Induces Thymocyte Apoptosis in Mice with Autoimmune Myasthenia Gravis by Downregulating TRIM9.

OBJECTIVES: Myasthenia gravis (MG) is an organ-specific autoimmune neuromuscular disorder that occurs as a result of the impairment in neuromuscular junction and autoantibody attack on the postsynaptic receptors. Increasing evidence suggests that microRNAs (miRs) might be involved in the development of MG. Therefore, the present study aimed to investigate the regulatory function of miR-653 on MG and its relationship with tripartite motif 9 (TRIM9). METHODS: The thymic tissues obtained from MG patients with thymic hyperplasia were prepared for establishing an MG mouse model in BALB/c mice. Aft

Met-enkephalin-containing peptides encoded by proenkephalin A mRNA expressed in activated murine thymocytes inhibit thymocyte proliferation.

Murine thymocytes activated with the mitogen Con A express proenkephalin A mRNA (PEA mRNA) and met-enkephalin and/or met-enkephalin-containing peptides ("enkephalins"). This Con A-induced expression of PEA mRNA is modulated by the delta opioid receptor agonist, deltorphin I, in a biphasic, dose-dependent manner. That is, 10(-13) M to 10(-11) M deltorphin enhanced PEA mRNA expression 3- to 3.5-fold over the level induced by Con A alone, and 10(-9) M to 10(-7) M deltorphin inhibited it 40 to 70%. delta opioid receptor antagonists recognizing the delta-2 (naltrindole (NTI) and naltriben (NTB)), b
