Self-reactive B cells are not eliminated or inactivated by autoantigen expressed on thyroid epithelial cells.

Graves' Disease results from the production of autoantibodies against receptors for thyroid stimulating hormone (TSH) on thyroid epithelial cells, and represents the prototype for numerous autoimmune diseases caused by autoantibodies that bind to organ-specific cell membrane antigens. To study how humoral tolerance is normally maintained to organ-specific membrane antigens, transgenic mice were generated selectively expressing membrane-bound hen egg lysozyme (mHEL) on the thyroid epithelium. In contrast to the deletion of autoreactive B cells triggered by systemic mHEL (Hartley, S.B., J. Crosb

[A study on the mechanism of iodine-induced thyroid epithelial cell injury in the induction of autoimmune thyroiditis].

OBJECTIVE: To investigate the effect of iodine excess on thyroid follicle epithelial ultrastructure and the relationship between thyroid injury and autoimmune thyroiditis. METHODS: NOD. H-2(h4) mice and Kunming mice were randomly divided into four groups receiving plain water, 5 fold, 10 fold, and 100 fold excessive iodine water. 4, 8 and 24 weeks after receiving iodine water, the mice were killed. After fixation with osmic acid and dual staining with uranyl chloride and citrate lead, thyroid gland ultrastructure was examined with electron microscopy. RESULTS: Iodine treated NOD. H-2(h4) mic

Thyroid/cervical teratomas in children: immunohistochemical studies for specific thyroid epithelial cell markers.

Pathological and immunohistochemical findings for specific thyroid epithelial cell markers in thyroid/cervical teratomas from four children are reported. All tumors showed components from the three germinal cell layers. Neoplastic thyroglobulin-immunoreactive thyroid follicles occurred in three intrathyroid teratomas, but the findings were negative in a cervical teratoma in which evidence of its thyroid origin could not be ascertained. As previously suggested on histological grounds, thyroid tissue, other than normal trapped thyroid follicles, is not an infrequent component of cervical teratom

Serotonin activates cell survival and apoptotic death responses in cultured epithelial thyroid cells.

Anatomic and physiological interactions between central serotonergic system and thyroid gland are well established. However, the effects of locally available serotonin on the thyroid functions are poorly known. Here, we first demonstrate the expression of serotonin transporter SERT and 5-HT2A receptor subtype in rat thyroid epithelial cell line FRT both at mRNA and protein levels. In order to investigate the molecular mechanisms of serotonin action, FRT cells were exposed to increasing concentrations of the amine. Low concentrations of serotonin (up to 5 μM) enhanced FRT cell growth, and ERK1/

Thyrotropin preparations are mitogenic for thyroid epithelial cells in follicles in suspension culture.

We have been investigating the extent to which separated thyroid follicles in suspension culture, free of endothelium and fibroblasts, have the properties of follicles in vivo. To test whether thyrotropin (TSH) can cause thyroid epithelial cells to undergo mitosis, preparations of follicles suspended in Coon's modified F-12 medium with 0.5% calf serum were incubated with 10 milliunits of impure or pure TSH per ml. Three results were obtained: (i) TSH preparations stimulated the incorporation of [3H]thymidine into cell nuclei; (ii) mitotic figures were induced and they had the same characterist

One- and two-step transformations of rat thyroid epithelial cells by retroviral oncogenes.

A system of epithelial cells is described in which it is possible to study the number and the nature of genes capable of conferring the malignant phenotype. Two fully differentiated, hormone-responsive cell lines from rat thyroid glands are presented which are susceptible to one-step or two-step transformation upon infection with several murine acute retroviruses. After infection, both cell lines became independent from their thyrotropic hormone requirement for growth. However, complete transformation was achieved with one of the cell lines (FRTL-5 Cl 2), whereas the other cell line (PC Cl 3)

Thyroid hormone regulation of intestinal epithelial stem cell biology.

The gastrointestinal tract is a well-characterized target of thyroid hormones and thyroid hormone nuclear receptors TRs, as extensively described in the literature. The paradigm is its important remodelling in amphibians during thyroid hormone-dependent metamorphosis. Interestingly, several studies have described the conservation of this hormonal signal during intestinal development in mammals. Additional data suggested that it may also play a role in intestinal homeostasis, stem cell physiology and progenitor commitment as well as in tumour development. It is worth underlining that in the mam

Thyroid-specific T cells in the normal Wistar rat. II. T cell clones interact with cloned wistar rat thyroid cells and provide direct evidence for autoantigen presentation by thyroid epithelial cells.

Strains of rat differ in their susceptibility to experimental autoimmune thyroiditis (EAT). We recently observed that the normal Wistar rat has lymph node (LN) T cells which recognize the newly available cloned Wistar thyroid cell line (WRT) and/or rat thyroglobulin (rTg). We have now cloned thyroid-specific T cells and characterized their interaction with the WRT target cell. Twenty-three T cell clones were tested for their reactivity to syngeneic thymocytes, WRT cells alone, or WRT cells with thymocytes. All the clones were of the CD4+CD8- phenotype. Seven of 23 T cell clones proliferated in

Two distinct E3 ligases, SCFFBXL19 and HECW1, degrade thyroid transcription factor 1 in normal thyroid epithelial and follicular thyroid carcinoma cells, respectively.

Thyroid transcription factor 1 (TTF1) regulates the tissue-specific expression of genes. However, the molecular regulation of TTF1 in thyroid normal and carcinoma cells has not been revealed. Here we identify 2 distinct ubiquitin E3 ligases that are responsible for TTF1 degradation in normal thyroid cells and carcinoma cells, respectively. Phorbol myristate acetate induced TTF1 protein degradation in the ubiquitin-proteasome system in both HTori3 thyroid follicular epithelial cells and follicular thyroid carcinoma 133 (FTC133) cells. Lysine 151 residue was identified as a ubiquitin acceptor si

[Rare thyroid tumors: spindle-cell epithelial tumor and thyroid carcinoma with thymus-like differentiation].

The paper describes 2 cases of spindle-cell epithelial thyroid tumors (SETTLE) and carcinoma showing thymus-like differentiation (CASTLE). They are rare types of thyroid tumors and must be distinguished from medullary carcinoma, anaplastic carcinoma, synovial sarcoma and malignant teratoma. The presented cases show both common and unusual features as compared with the previously described tumors. CASTLE probably arises from ectopic thymus tissue. The further examination is needed to highlight the histogenesis of SETTLE, and to collect additional data on its aggressiveness and clinical outcome.

Ultrastructure of blebbing and phagocytosis of blebs by hyperplastic thyroid epithelial cells in vivo.

In addition to pseudopods, somewhat pleomorphic blebs were consistently found protruding from the apical surfaces of hyperplastic rat thyroid epithelial cells into the follicular lumens in vivo. Many blebs were knobby, roughly hemispherical protrusions, with a diameter of 2-3 mum. Such blebs had a densely packed microfilamentous core and contained numerous apparent ribosomes. They were morphologically similar to blebs that have been observed in a variety of cultured cells. Other blebs were larger, more elongate, and less knobby, but had a similar ultrastructural organization. Blebs of all size

Syngeneic sensitization of mouse lymphocytes on monolayers of thyroid epithelial cells. III. Induction of thyroiditis by thyroid-sensitized T lymphoblasts.

During in vitro sensitization of CBA spleen lymphocytes on syngeneic monolayers of either thyroid epithelial cells (TEC) or fibroblasts, stimulation occurs, as assessed by thymidine incorporation and lymphoblast generation. When lymphoblasts generated on syngeneic TEC are injected either into thyroid lobes of intact CBA recipients or i.v., thyroiditis appears at least 21 days after injection. Thyroiditis is assessed by both optical and electron microscopic blind studies and by the presence of antibodies in the sera directed against mouse thyroglobulin. In contrast, control animals which receiv

Embryologic origin of various epithelial cell types in the thyroid gland of the rat.

Ventral pharyngeal outpocketings and ultimobranchial outpocketings from the 14-day-old fetus of the Fischer rat were isolated before they fused to form thyroid glands. The outpocketings were implanted into different kidney capsules of adult male Fischer rats, and were allowed to grow for several months. Transplants were then excised and examined by electron microscopy. The ultimobranchial outpocketing gave rise to two types of follicles. One contained ciliated cells, cells with an abundant agranular reticulum, U cells with basal hemidesmosomes, and two types of cells with secretory vesicles. T

Cell polarity and water transport in thyroid epithelial cells in separated follicles in suspension culture.

Separated thyroid follicles maintained in suspension culture can be used to study the properties of thyroid epithelium in the virtual absence of other cell types and to study the effect of extracellular materials on the follicles. They can be prepared by enzymatic separation of thyroid into single cells followed by reaggregation of the epithelial cells and also by collagenase treatment of thyroids to release follicles and sheets of epithelia that can be separated from other materials by differential filtration. The follicles can exist with normal orientation or inverted (inside out). The folli

Cell-cell interactions in the process of differentiation of thyroid epithelial cells into follicles: a study by microinjection and fluorescence microscopy on in vitro reconstituted thyroid follicles.

Thyroid cells, cultured in the presence of thyroid stimulating hormone, reorganized within 36-48 hr into follicular structures, the in vitro reconstituted thyroid follicles or RTF. By microinjection of fluorescent probes either into the neoformed intrafollicular lumen (IL) or into cells forming the follicles, we have studied the development and some functional properties of cell-cell contacts involved in a) the formation of the thyroid follicular lumen and b) the communication between thyrocytes within the follicle. The probes were compounds of either low (Lucifer Yellow: LY) or high molecular

Molecular Pathology of Thyroid Follicular Epithelial Cell-Derived Neoplasia: An Update.

CONTEXT.—: Thyroid gland neoplasia comprises the most common group of tumors arising within organs of the endocrine system. Pathogenic molecular changes can now be identified in up to 90% of thyroid carcinomas. These alterations not only shape our understanding of tumor biology and classification, but also increasingly guide therapeutic approaches to these diseases. OBJECTIVE.—: To revisit a frequently cited review article from 2011 that outlined fundamental differences between tumor types-primarily papillary thyroid carcinoma and follicular neoplasia-in terms of their underlying genetic prof

RET receptor expression in thyroid follicular epithelial cell-derived tumors.

The RET proto-oncogene encodes a receptor tyrosine kinase for transforming growth factor-beta-related neurotrophic factors, which include GDNF and neurturin. The expression of RET proto-oncogene was detected in several tissues, such as spleen, thymus, lymph nodes, salivary gland, and spinal cord, and in several neural crest-derived cell lines. RET expression in the thyroid gland was reported to be restricted to neural crest-derived C cells. The presence of RET mRNA or protein has not yet been reported in thyroid follicular cells. We previously demonstrated the expression of oncogenic rearrange

Culture of hormone-dependent functional epithelial cells from rat thyroids.

Primary cultures of rat thyroid cells were made in medium supplemented with 0.1--0.5% calf serum and containing six hormones or growth factors: insulin, thyrotropin, transferrin, hydrocortisone, somatostatin, and glycyl-L-histidyl-L-lysine acetate. The FRTL strain was purified by successive colonial isolations and was found to maintain highly differentiated features (secretion into the culture medium of physiological amounts of thyroglobulin and concentration of iodide by 100-fold). The FRTL strain has been observed for more than 3 years in continuous culture. It has maintained the same bioche

Thyroid hormone protects human lung epithelial cells from cold preservation and warm reperfusion-induced injury.

BACKGROUND: Cellular stress associated with static-cold storage (SCS) and warm reperfusion of donor lungs can contribute to ischemia-reperfusion (IR) injury during transplantation. Adding cytoprotective agents to the preservation solution may be conducive to reducing graft deterioration and improving post-transplant outcomes. METHODS: SCS and warm reperfusion were simulated in human lung epithelial cells (BEAS-2B) by exposing cells to low potassium dextran glucose solution at 4 °C for different periods and then switching back to serum-containing culture medium at 37 °C. Detection of in vivo production of tumour necrosis factor-alpha by human thyroid epithelial cells.

We have established previously that human thyroid epithelial cells (TEC) from patients with autoimmune thyroiditis are able to synthesize cytokines, such as interleukin-1 (IL-1) and interleukin-6 (IL-6). This paper examines TEC in sections from autoimmune thyroiditis for the in vivo production of tumour necrosis factor-alpha (TNF-alpha) and interferon-gamma (IFN-gamma) using the combined techniques of in situ hybridization and immunohistochemistry. Thyroid tissue from patients with Graves' disease, Hashimoto's disease and non-toxic goitre was examined and both mRNA and the protein of TNF-alpha
