Controlling Pepper Mild Mottle Virus (PMMoV) Infection in Pepper Seedlings by Use of Chemically Synthetic Silver Nanoparticles.

We investigated the roles of different concentrations of chemical synthetic spherical silver nanoparticles (AgNPs) in protecting pepper seedlings of the Mecca region, which were naturally and artificially infected by the pepper mild mottle virus (PMMoV). The virus shows many infection symptoms, including pepper leaf deformation with filiform leaves and severe mosaic symptoms. Our study focused on the antiviral activity of different concentrations of spherical nanoparticles in controlling PMMoV infecting pepper seedlings. PMMoV identification was confirmed via DAS-ELISA using the following anti

Non-toxic concentrations of cadmium inhibit systemic movement of turnip vein clearing virus by a salicylic acid-independent mechanism.

Systemic movement of plant viruses is a central event in viral infection. To better understand this process, the heavy metal cadmium was used to inhibit systemic spread of turnip vein clearing virus (TVCV), a tobamovirus, in tobacco plants. Study of the mechanism by which cadmium exerts this inhibitory effect may provide insights into the essential steps of the TVCV systemic movement pathway. Our results demonstrated that cadmium treatment did not affect TVCV transport from the inoculated non-vascular tissue into the plant vasculature but blocked viral exit into uninoculated non-vascular tissu

Geographic Structuring of the Tomato Brown Rugose Fruit Virus Populations in Morocco.

Tomato brown rugose fruit virus (Tobamovirus fructirugosum, ToBRFV) is an emerging virus that affects tomatoes, capsicum, and chili. Since its first detection in Jordan in 2015, the virus has been reported in more than 40 countries across all the continents. In Morocco, the virus was reported for the first time in October 2021. However, its genetic diversity remains unexplored. In this work, we used a collection of 100 tomato fruits from local markets to investigate the virus's variability. Thirty-eight sequences were recovered and used to study evolutionary pressures acting on the N-terminus

Predicting the Potential Suitable Distribution of Tomato Brown Rugose Fruit Virus in China Using a BIOMOD2 Ensemble Model.

Tomato brown rugose fruit virus (ToBRFV) is an emerging tobamovirus that threatens tomato production and seedling trade. We used a BIOMOD2 ensemble to model present and future environmental suitability in China from 35 screened occurrence records, climatic predictors, and agricultural context data. Climate projections were based on BCC-CSM2-MR under SSP126, SSP245, SSP370, and SSP585 for the 2050s and 2090s. The ensemble achieved calibration TSS = 0.831 and AUC/ROC = 0.958. Spatial-block evaluation was lower and more variable, so the calibration metrics do not demonstrate equally strong geogra

An effective antiviral strategy based on silence of susceptibility genes through cucumber mosaic virus (CMV) attenuated vaccine vector.

Plant viruses represent a major threat to agriculture, affecting a wide range of crops with substantial economic losses. This study presented a novel strategy for managing plant viral diseases through the development an attenuated vaccine utilizing cucumber mosaic virus (CMV) for virus-induced gene silencing (VIGS) targeting susceptibility gene. TOBAMOVIRUS MULTIPLICATION 2A (TOM2A) gene was identified as a critical factor that enhances susceptibility to TMV infection in plants. Two vaccines were constructed based on CMV attenuated vaccine vector-CR2V. The first vaccine was constructed by inco

Novel Cyclized Derivatives of Ferulic Acid as Potential Antiviral Agents through Activation of Photosynthesis.

To further develop new antiviral agents, several novel cyclized derivatives of ferulic acid were designed and synthesized. Their antiviral activities were evaluated against the cucumber mosaic virus (CMV), pepper mild mottle virus (PMMoV), and tomato spotted wilt virus (TSWV). The results showed that some ferulic acid derivatives exhibited desirable antiviral activities. Particularly, compound 5e exhibited excellent protective activities against CMV, PMMoV, and TSWV, with EC50 values of 167.2, 102.5, and 145.8 μg mL-1, respectively, which were superior to those obtained for trans-ferulic acid

Development of a magnetic nanoparticle-based method for concentrating SARS-CoV-2 in wastewater.

Several virus concentration methods have been developed to increase the detection sensitivity of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in wastewater, as part of applying wastewater-based epidemiology. Polyethylene glycol (PEG) precipitation method, a method widely used for concentrating viruses in wastewater, has some limitations, such as long processing time. In this study, Pegcision, a PEG-based method using magnetic nanoparticles (MNPs), was applied to detect SARS-CoV-2 in wastewater, with several modifications to increase its sensitivity and throughput. Molecular Mechanisms Underlying Cucumber Resistance to Cucumber Green Mottle Mosaic Virus: From Host-Virus Interactions to Precision Breeding Strategies.

Cucumber (Cucumis sativus L.) is an economically important vegetable crop cultivated worldwide, but its sustainable production is increasingly threatened by Cucumber green mottle mosaic virus (CGMMV). CGMMV is a positive-sense single-stranded RNA virus belonging to the genus Tobamovirus and represents a major constraint on cucurbit production because of its efficient seed transmission, mechanical spread, environmental persistence, and broad host range. Infection can cause leaf mosaic and mottling, growth suppression, and severe fruit symptoms, including internal discoloration, water-soaked tis

miR403a and SA Are Involved in NbAGO2 Mediated Antiviral Defenses Against TMV Infection in Nicotiana benthamiana.

RNAi (RNA interference) is an important defense response against virus infection in plants. The core machinery of the RNAi pathway in plants include DCL (Dicer Like), AGO (Argonaute) and RdRp (RNA dependent RNA polymerase). Although involvement of these RNAi components in virus infection responses was demonstrated in Arabidopsis thaliana, their contribution to antiviral immunity in Nicotiana benthamiana, a model plant for plant-pathogen interaction studies, is not well understood. In this study, we investigated the role of N. benthamiana NbAGO2 gene against TMV (Tomato mosaic virus) infection.

A viral protein promotes host L-asparaginase activity and weakens the asparagine-induced resistance pathway for the benefit of virus infection.

Amino acids are not only essential for plant nutrition but also serve as critical immune signaling molecules, particularly during pathogen invasion. Pathogens can manipulate amino acid metabolic pathways to counteract host immune defenses, yet the underlying mechanisms remain poorly understood. Here, we demonstrate that the pepper mild mottle virus (PMMoV) 126 kDa protein interacts with the host L-asparaginase (LA), identified as a negative regulator of antiviral defense. LA converts asparagine (Asn) to aspartic acid (Asp). Exogenous application of Asn markedly enhanced resistance to PMMoV, wh

Viral Contaminants in a Philippine Wastewater Treatment Plant: Quantification and Treatment Reduction.

In the Philippines, most of the population relies on decentralized wastewater systems, particularly septic tanks, and the rest on centralized wastewater systems. However, the performance of wastewater treatment systems in the Philippines in reducing viral contaminants remains to be characterized; thus, in this study, the occurrence and reduction of viruses in wastewater before and after treatment were assessed at a wastewater treatment plant (WWTP) that utilizes biological treatment technologies in the Philippines. Influent (n = 18) and effluent (n = 18) samples collected from WWTP from April

Characterizing the chemical and microbial fingerprint of unsheltered homelessness in an urban watershed.

Unsheltered homelessness is rapidly becoming a critical issue in many cities worldwide. The worsening situation not only highlights the socioeconomic plight, but it also raises awareness of ancillary issues such as the potential implications for urban water quality. The objective of this study was to simultaneously leverage diverse source tracking tools to develop a chemical and microbial fingerprint describing the relative contribution of direct human inputs into Las Vegas' tributary washes. By evaluating a wide range of urban water matrices using general water quality parameters, fecal indic

Tobamoviruses CP Proteins Hijack Light-Induced Protein (NbLIP1) to Promote Viral Replication by Facilitating VRO Formation.

Tobamoviruses establish viral replication organelles (VROs) on the host endoplasmic reticulum (ER) for their replication, a process demanding substantial different types of lipids. However, how viruses efficiently transfer these lipids from other compartments like chloroplasts remains incompletely understood. Fibrillin (FBN) proteins are primarily localised to chloroplasts and are intimately linked with lipid metabolism and stress responses in chloroplasts. Here, we report that NbLIP1, a light-induced FBN1 family protein in Nicotiana benthamiana, directly interacts with the coat protein (CP) o

Infectious Clone Development of Zucchini Green Mottle Mosaic Virus Infecting Medicinal Plant Trichosanthes kirilowii and Establishment of a Serological Assay System.

Trichosanthes kirilowii has long been cultivated for application in traditional Chinese medicine. In this study, we identified two isolates of zucchini green mottle mosaic virus (ZGMMV; species Tobamovirus cucurbitae) from T. kirilowii plants. We determined the complete genome sequences of the ZGMMV isolates named ZGMMV-GL-1 and ZGMMV-GL-2. Each ZGMMV genome was 6,517 nucleotides in length, with only a single nucleotide variation detected between two sequences. Sequence analysis revealed that the ZGMMV isolates from this study shared 88.07 to 91.62% nucleotide identity with five other ZGMMV is

Correlation between microbial source tracking markers and pathogens at beaches and estuaries in southern California.

Fecal indicator bacteria (FIB) have historically been used to assess public health risks associated with contact water recreation, and microbial source tracking (MST) has been introduced to distinguish human-associated fecal pollution from natural sources of animal feces. While MST can identify human-associated fecal pollution, without interpretation it cannot differentiate between small amounts of fresh fecal contamination and large amounts of decayed pollution, which can present vastly different health risks. This study examined the ratio of HF183 and pepper mild mottle virus (PMMoV) as an i
