Signaling Pathways Critical for Tooth Root Formation.

Tooth is made of an enamel-covered crown and a cementum-covered root. Studies on crown dentin formation have been a major focus in tooth development for several decades. Interestingly, the population prevalence for genetic short root anomaly (SRA) with no apparent defects in crown is close to 1.3%. Furthermore, people with SRA itself are predisposed to root resorption during orthodontic treatment. The discovery of the unique role of Nfic (nuclear factor I C; a transcriptional factor) in controlling root but not crown dentin formation points to a new concept: tooth crown and root have different

Could the pulp be preserved in tooth with root resorption caused by embedded tooth?

BACKGROUND/PURPOSE: At present, there are no recognized guidelines or consensus for the treatment strategy of the asymptomatic tooth with external root resorption caused by an embedded tooth (et-ERR). Most clinicians would like prophylactic or concomitant root canal therapy (RCT) along with the extraction of the embedded tooth. The purpose of this study was to report the prognosis of external root resorption (ERR) and investigate the possibility to preserve the vital pulp of ERR tooth. MATERIALS AND METHODS: The patients who had asymptomatic et-ERR teeth were included. After extraction of the

Application of cryopreservation to tooth germ transplantation for root development and tooth eruption.

We cryopreserved mouse tooth germs with widely open cervical margins of the enamel organ to overcome difficulties in cryoprotectant permeation and tested their efficacy by transplanting them into recipient mice. The upper right first molar germs of 8-day-old donor mice were extracted and categorized into the following four groups according to cryopreservation time: no cryopreservation, 1 week, 1 month, and 3 months. The donor tooth germs were transplanted into the upper right first molar germ sockets of the 8-day-old recipient mice. The upper left first molars of the recipient mice were used a

[Management of accidental tooth root displacement into the mandibular canal during tooth extraction].

The accidental displacement of tooth roots into the mandibular canal is a serious complication during tooth extractions in oral and maxillofacial surgery, often resulting in direct damage to the structural and functional integrity of the inferior alveolar neurovascular bundle (IANB). This article reviews the anatomical features of the mandibular canal, the IANB, and adjacent tooth roots; identifies high-risk factors and anatomically vulnerable sites for root displacement; and outlines the clinical manifestations and radiographic characteristics of intraoperative root intrusion into the mandibu

Multiple essential MT1-MMP functions in tooth root formation, dentinogenesis, and tooth eruption.

Membrane-type matrix metalloproteinase 1 (MT1-MMP) is a transmembrane zinc-endopeptidase that breaks down extracellular matrix components, including several collagens, during tissue development and physiological remodeling. MT1-MMP-deficient mice (MT1-MMP(-/-)) feature severe defects in connective tissues, such as impaired growth, osteopenia, fibrosis, and conspicuous loss of molar tooth eruption and root formation. In order to define the functions of MT1-MMP during root formation and tooth eruption, we analyzed the development of teeth and surrounding tissues in the absence of MT1-MMP. In sit

Mechanisms of Osteoclastogenesis in Orthodontic Tooth Movement and Orthodontically Induced Tooth Root Resorption.

Orthodontic tooth movement (OTM) is achieved by the simultaneous activation of bone resorption by osteoclasts and bone formation by osteoblasts. When orthodontic forces are applied, osteoclast-mediated bone resorption occurs in the alveolar bone on the compression side, creating space for tooth movement. Therefore, controlling osteoclastogenesis is the fundamental tenet of orthodontic treatment. Orthodontic forces are sensed by osteoblast lineage cells such as periodontal ligament (PDL) cells and osteocytes. Of several cytokines produced by these cells, the most important cytokine promoting os

[Temporal pattern of tooth root migration following coronectomy].

INTRODUCTION: Injury to the inferior alveolar nerve is a potential complication during the removal of mandibular third molars, with a substantially increased risk when a direct anatomical relationship exists between the tooth roots and the mandibular canal. In such cases, coronectomy offers a surgical alternative that reduces the risk of nerve injury; however, the clinical relevance of postoperative root migration remains incompletely understood. OBJECTIVE: This study analyzed the timing and extent of root migration after coronectomy, examined influencing factors, assessed radiological indica

Delayed tooth replantation: MTA as root canal filling.

MTA has been investigated as a root-end filling material. Its mechanism of action has some similarities to that of Ca(OH)2. The purpose of this study was to evaluate the repair process taking place in the delayed replantation of monkey teeth using calcium hydroxide and MTA as root canal filling materials. Five monkeys had their lateral incisors extracted and bench-dried for 60 minutes. After root canal preparation, the teeth were assigned to two groups according to root canal filling material: I, calcium hydroxide; and II, MTA. The same treatment sequence was followed for both groups: coronal

High-altitude illness induced by tooth root infection.

High-altitude illness may occur after recent pulmonary infection, but high-altitude illness after root canal therapy has not been described previously. A 44-year-old man is presented who skied to a 3333 m high peak in the Eastern Alps one day after he had undergone root canal therapy because of a tooth root infection. After 4 hours above 3000 m severe symptoms of high-altitude illness, including pulmonary oedema, developed. His condition improved after immediate descent. The next day he presented with local and general signs of infection which were successfully treated with gingival incisions

Cellular and molecular mechanisms of tooth root development.

The tooth root is an integral, functionally important part of our dentition. The formation of a functional root depends on epithelial-mesenchymal interactions and integration of the root with the jaw bone, blood supply and nerve innervations. The root development process therefore offers an attractive model for investigating organogenesis. Understanding how roots develop and how they can be bioengineered is also of great interest in the field of regenerative medicine. Here, we discuss recent advances in understanding the cellular and molecular mechanisms underlying tooth root formation. We rev

Root resorption in relation to orthodontic tooth movement.

Root resorption is an undesirable sequela of orthodontic tooth movement. The odontoclasts are responsible for root resorption and the process of hyalinization is known to preceed the orthodontic root resorption. It is found that there are several predisposing factors, therefore an evaluation of these factors should be done by careful examination of personal medical history, severity of malocclusion and dental treatment (if any due to previous history of trauma), anterior crossbite etc. The evaluation becomes an essential factor as it helps the orthodontists in detecting the occurance and sever

[Pathological tooth root resorption and its clinical significance].

The author examined 316 teeth, on which total pulpotomy had been performed, for frequency and extent of resorption. Marked resorptions, always associated with chronic apical periodontitis, were radiographically disclosed in 43 teeth. It is pointed out that the clinical radiographic technique permits to visualize only resorptions of greater extent.

Effects of antiresorptive medications on tooth root formation and tooth eruption in paediatric patients.

Tooth eruption is a pivotal milestone for children's growth and development. This process involves with the formation of the tooth root, the periodontal ligament (PDL) and the alveolar bone, as the tooth crown penetrates the bone and gingiva to enter the oral cavity. This review aims to outline current knowledge of the adverse dental effects of antiresorptive medications. Recently, paediatric indications for antiresorptive medications, such as bisphosphonates (BPs), have emerged, and these agents are increasingly used in children and adolescents to cure pathological bone resorption associated

Root fracture in immature tooth: report of a case.

Root fracture injuries affect 0.5-7% of permanent teeth. Although this type of injury is rarely seen in teeth with immature root formation, the prognosis is generally good depending on the site of the fracture. A case report of horizontal root fracture in maxillary central incisor of an 8-year and 3-month-old girl and its treatment was presented.

Systemic lead absorption in human tooth roots.

One-hundred and five impacted and erupted sound teeth were analysed for lead content by graphite furnace atomic absorption spectrophotometry. The results showed that lead accumulated before eruption, presumably soon after the onset of dentine formation. The accumulation in dentine was systemic, without any contribution from the oral environment. The amount of lead in dentine may denote the amount of absorption into the body and serve as a precise index to body burden as no absorption from the mouth can take place. In adults, impacted and erupted teeth can be used together in any research and t

Management of an avulsed tooth with severe root resorption.

Delayed replantation of an avulsed tooth with an open apex resulted in failure of revascularization, necrosis of the pulp and extensive external root resorption. Removal of the necrotic pulp and repeated intra-canal dressings of calcium hydroxide over a three year period arrested the inflammatory resorptive process. The process was replaced by replacement resorption leading to ankylosis and infra-occlusion of the affected tooth. As the tooth was asymptomatic, it was retained in the arch to act as a space maintainer. To improve aesthetics, the crown was recontoured using a light-cured resin.
