Induction of a torpor-like hypothermic and hypometabolic state in rodents by ultrasound.

Torpor is an energy-conserving state in which animals dramatically decrease their metabolic rate and body temperature to survive harsh environmental conditions. Here, we report the noninvasive, precise and safe induction of a torpor-like hypothermic and hypometabolic state in rodents by remote transcranial ultrasound stimulation at the hypothalamus preoptic area (POA). We achieve a long-lasting (>24 h) torpor-like state in mice via closed-loop feedback control of ultrasound stimulation with automated detection of body temperature. Ultrasound-induced hypothermia and hypometabolism (UIH) is trig

Estrous cycle modulates fasting-induced torpor propensity via hypothalamic estrogen signalling.

Torpor is a state of transient hypometabolism and hypothermia that is engaged by many species in adverse conditions such as food scarcity. Chemo- or optoactivation of neurons in the hypothalamic preoptic area (POA) drives torpor-like hypometabolism and hypothermia. Estrogens, principally estradiol, modulate thermogenesis and energy balance by central actions, and POA neurons express the canonical estrogen receptor ERα. We explored the role of POA estrogen signalling in fasting-induced torpor in the mouse. We found that torpor depth and duration vary across the estrus cycle in mice, whereby the

Prolonged activation of EP3 receptor-expressing preoptic neurons underlies torpor responses.

Many species use a temporary drop in body temperature and metabolic rate (torpor) as a strategy to survive food scarcity. A similar profound hypothermia is observed with activation of preoptic neurons that express the neuropeptides Pituitary Adenylate-Cyclase-Activating Polypeptide (PACAP)1, Brain Derived Neurotrophic Factor (BDNF)2, or Pyroglutamylated RFamide Peptide (QRFP)3, the vesicular glutamate transporter, Vglut24,5 or the leptin receptor6 (LepR), estrogen 1 receptor (Esr1)7 or prostaglandin E receptor 3 (EP3R) in mice8. However, most of these genetic markers are found on multiple popu

Human torpor: translating insights from nature into manned deep space expedition.

During a long-duration manned spaceflight mission, such as flying to Mars and beyond, all crew members will spend a long period in an independent spacecraft with closed-loop bioregenerative life-support systems. Saving resources and reducing medical risks, particularly in mental heath, are key technology gaps hampering human expedition into deep space. In the 1960s, several scientists proposed that an induced state of suppressed metabolism in humans, which mimics 'hibernation', could be an ideal solution to cope with many issues during spaceflight. In recent years, with the introduction of spe

Long-term pharmacological torpor of rats with feedback-controlled drug administration.

The maintenance of pharmacological torpor and hypothermia (body temperature 28 °C - 33 °C) in rats for a week is presented. For this purpose, our laboratory has developed a device (BioFeedback-2) for the feed-back controlled multiple injections of small doses of a pharmacological composition that we created earlier. On the 7th day, the rat spontaneously come out of the pharmacological torpor, the body temperature returned to normal, and on the 8th day, the animal could consume food and water. The proposed approach for maintaining multi-day pharmacological torpor can be applied in medicine, as

Torpor-responsive microRNAs in the heart of the Monito del monte, Dromiciops gliroides.

The marsupial Monito del monte (Dromiciops gliroides) utilizes both daily and seasonal bouts of torpor to preserve energy and prolong survival during periods of cold and unpredictable food availability. Torpor involves changes in cellular metabolism, including specific changes to gene expression that is coordinated in part, by the posttranscriptional gene silencing activity of microRNAs (miRNA). Previously, differential miRNA expression has been identified in D. gliroides liver and skeletal muscle; however, miRNAs in the heart of Monito del monte remained unstudied. In this study, the expressi

[Neural Circuits Involved in Torpor and Hibernation and Prospects for Artificial Hibernation Technology].

Mammals and birds are categorized as thermostatic animals and consume significant quantities of energy to maintain their body temperature. Body temperature in mammals is usually maintained within a narrow range of 36-37℃ despite fluctuations in the external temperature. However, during periods of food scarcity, which leads to a diminished source of energy, such as in winter, some species adopt a strategy to overcome the energy shortage by lowering their body temperature, which significantly reduces their basal metabolism (energy demand). This phenomenon is referred to as hibernation or torpor.

Thermoregulatory role of ghrelin in the induction of torpor under a restricted feeding condition.

Ghrelin, a circulating orexigenic hormone secreted from the stomach, stimulates appetite and food intake by activating the hypothalamic arcuate nucleus. Administration of exogenous ghrelin exerts anabolic effects, causing weight gain, increased adiposity, and decreased metabolism. Body temperature (BT), which is determined by the balance of heat production and heat loss, must be strictly regulated to maintain proper cellular function and metabolism. However, the role of ghrelin in thermoregulation remains unclear. In this study, we found that ghrelin was essential for decreasing BT when mice a

The effects of dietary cholesterol on metabolism and daily torpor patterns in Siberian hamsters.

The concentrations of fatty acids in the diet influence torpor in numerous species of mammals. Much less is known, however, about the potential effects of other types of dietary lipids. One study demonstrated that increasing dietary cholesterol levels during fall feeding increased torpor bout length and also decreased minimum body temperatures during hibernation by chipmunks. Another hibernation study with ground squirrels revealed that the cholesterol contents of both the cerebral cortex and the microsomes were significantly greater during arousal episodes than during torpor bouts, suggesting

CO2 balance of a heterothermic rodent: comparison of sleep, torpor, and awake states.

CO2 homeostasis of different thermal states have been compared in a heterothermic ground squirrel, Spermophilus tereticaudus. Gas exchange (MO2, MCO2), lung ventilation (VE), and body temperature (Tb) were simultaneously measured during sleep, shallow torpor (Tb 25-29 degrees C), deep torpor (Tb 11-15 degrees C), awake heterothermia (Tb 30-42.5 degrees C), and transitions between these states. CO2 retention (falling MCO2/MO2 and VE/MCO2) accompanied entrance into sleep and torpor. CO2 retention lowered MO2 in sleeping and torpid squirrels beyond that caused by reduced Tb. In torpor at steady s

Mitochondrial proteomic adaptations to daily torpor in the Djungarian hamster (Phodopus sungorus).

Hibernation is an adaptive strategy that conserves energy in response to environmental challenges. While mitochondrial proteomic adaptations are well-documented in deep hibernators, the proteomic changes underlying daily torpor remain less clear. We investigated mitochondrial proteomic adaptations in the liver of a daily hibernator, the Djungarian hamster (Phodopus sungorus), across different hibernation phases. Hamsters were maintained under long-day (summer) or short-day photoperiods (winter), to induce torpor. Livers from summer, torpor, and interbout euthermia phases were analyzed by liqui

Preoptic activation induces a torpor-like hypothermic and hypometabolic state that is cerebroprotective.

Therapeutic hypothermia for stroke has been limited by shivering, increased metabolic demand, and poor patient tolerance. Engaging endogenous thermoregulatory circuits to lower body temperature may overcome these limitations and modulate metabolism, offering an integrated approach to cerebroprotection. Here, we show that chemogenetic activation of neurons in the preoptic area (POA) elicits a torpor-like state in mice, characterized by sustained hypothermia and hypometabolism. In an animal stroke model, this endogenous hypothermic state significantly reduced infarct volume and improved motor ou

Adenosine and P1 receptors: Key targets in the regulation of sleep, torpor, and hibernation.

Sleep, torpor, and hibernation are three distinct hypometabolic states. However, they have some similar physiological features, such as decreased core body temperature and slowing heart rate. In addition, the accumulation of adenosine seems to be a common feature before entry into these three states, suggesting that adenosine and its receptors, also known as P1 receptors, may mediate the initiation and maintenance of these states. This review, therefore, summarizes the current research on the roles and possible neurobiological mechanisms of adenosine and P1 receptors in sleep, torpor, and hibe

Reversible Tau Phosphorylation Induced by Synthetic Torpor in the Spinal Cord of the Rat.

Tau is a key protein in neurons, where it affects the dynamics of the microtubule system. The hyperphosphorylation of Tau (PP-Tau) commonly leads to the formation of neurofibrillary tangles, as it occurs in tauopathies, a group of neurodegenerative diseases, including Alzheimer's. Hypothermia-related accumulation of PP-Tau has been described in hibernators and during synthetic torpor (ST), a torpor-like condition that has been induced in rats, a non-hibernating species. Remarkably, in ST PP-Tau is reversible and Tau de-phosphorylates within a few hours following the torpor bout, apparently not

The relationship between fasting-induced torpor, sleep, and wakefulness in laboratory mice.

STUDY OBJECTIVES: Torpor is a regulated and reversible state of metabolic suppression used by many mammalian species to conserve energy. Whereas the relationship between torpor and sleep has been well-studied in seasonal hibernators, less is known about the effects of fasting-induced torpor on states of vigilance and brain activity in laboratory mice. METHODS: Continuous monitoring of electroencephalogram (EEG), electromyogram (EMG), and surface body temperature was undertaken in adult, male C57BL/6 mice over consecutive days of scheduled restricted feeding. RESULTS: All animals showed bouts

Frequent nocturnal torpor in a free-ranging Australian honeyeater, the noisy miner.

Torpor in birds is considered to be far less common than in mammals. This is particularly true for passerine birds for which knowledge of torpor expression is scarce, although almost all are small, have high energy expenditure and could profit energetically from using torpor. To assess whether the extent and diversity of avian and especially passerine torpor expression and heterothermy may be currently underestimated because of limited long-term data on free-ranging birds, core body temperature fluctuations were quantified over ~ 4.3 months in a medium-sized honeyeater, the noisy miner (Manori

A pharmacological composition for induction of a reversible torpor-like state and hypothermia in rats.

AIMS: To initiate a state of artificial torpor we suggested a pharmacological multi-targeting strategy for simulation of the physiological pattern of natural hibernation including a significant reduction in heart rate, respiratory rate, body temperature and oxygen consumption as well as a decline in brain activity known as torpor. MATERIALS AND METHODS: We have developed a composition which initiates a pharmacologically induced torpor-like state (PITS-composition), made up of eight therapeutic agents, inert gas xenon and lipid emulsion served as a drug vehicle. KEY FINDINGS: After a single i

Huddling and food availability shape seasonal torpor and energetics of juvenile garden dormice.

Environmental fluctuations challenge juvenile hibernators to efficiently balance growth, fattening, and energy conservation prior to their first winter. This study investigates how social thermoregulation (huddling) and food availability affect the use of torpor, growth, fat accumulation, and hibernation in late-born juvenile garden dormice. Juveniles exhibited flexible energy-saving strategies, with huddling influencing torpor patterns and food intake during pre-hibernation. Despite these behavioral differences, individuals reached similar body mass and size at the onset and end of hibernatio

Persistent testicular glycogenolysis and active lipid metabolism during torpor in hibernating Siberian chipmunks.

Numerous hibernating species within the family Sciuridae display distinct hibernation bout patterns and initiate reproductive activities shortly after arousal from torpor. However, the influence of different hibernation phases on testicular function has not yet been fully elucidated. To address this knowledge gap, we investigated alterations in testicular morphology and glycogen as well as lipid metabolism in Siberian chipmunks (Tamias sibiricus) across key physiological stages associated with the hibernation cycle. We found that serum lipid levels were high during torpor, while glucose peaked

Hippocampal neuroimmune response in mice undergoing serial daily torpor induced by calorie restriction.

Hibernating animals demonstrate a remarkable ability to withstand extreme physiological brain changes without triggering adverse neuroinflammatory responses. While hibernators may offer valuable insights into the neuroprotective mechanisms inherent to hibernation, studies using such species are constrained by the limited availability of molecular tools. Laboratory mice may serve as an alternative, entering states of hypometabolism and hypothermia similar to the torpor observed in hibernation when faced with energy shortage. Notably, prolonged calorie restriction (CR) induces serial daily torpo
