The study of the effects of low-level exposure to ionizing radiation using a bio-indicator system.

The object of the present study was to apply the high sensitivity of a botanical mutagenicity test (Tradescantia stamen-hair mutation bioassay), in "in situ" bioassays to determine the responses induced by the exposure to low levels of ionizing radiation. Mutagenesis was evaluated during 28 days, in an environment that presented gamma radiation exposure rates of 1.6 (control), 25.0 and 75.0 microR min(-1). From the results we observed that there was no linear response throughout the exposure time when compared mutagenic events and gamma radiation exposure rates were compared. The results obtai

Detection of genotoxic effects of heavy metal contaminated soils with plant bioassays.

Aim of the present study was the development of a bioassay which enables the detection of genotoxic effects of heavy metal contaminated soils. In the first part of the present study, the data base on metal effects in plant bioassays was extended. Four metal salts, namely Cr(VI)O3, Cr(III)Cl3, Ni(II)Cl2 and Sb(III)Cl3 were tested comparatively in MN tests with pollen tetrad cells of Tradescantia clone #4430 and in meristematic root tip cells of Vicia faba. With Cr6+ and Ni2+, clear-cut dose-effects were observed in a range between 0.75 and 10.0 mM, whereas this was not the case with Cr3+ (range

Varied chromosome distribution behaviours during meiosis in triploid Chinese chives contribute to the formation of viable pollen.

Triploids play an important role in the polyploidization process and are considered a bridge between diploids and polyploids. To inform plant polyploidization research and polyploid breeding, it is important to explore chromosome behaviour during triploid pollen development, pollen fertility problems in triploids and the potential value of utilizing triploids. In this study, acetocarmine, carbol fuchsin and fluorescence staining methods were used to observe microsporogenesis and microspore development in fertile triploid Chinese chives. The results revealed that some of the pollen mother cells

Nitric oxide induces stomatal closure and enhances the adaptive plant responses against drought stress.

Nitric oxide (NO) is a very active molecule involved in many and diverse biological pathways where it has proved to be protective against damages provoked by oxidative stress conditions. In this work, we studied the effect of two NO donors, sodium nitroprusside (SNP) and S-nitroso-N-acetylpenicillamine SNP-treated on the response of wheat (Triticum aestivum) to water stress conditions. After 2 and 3 h of drought, detached wheat leaves pretreated with 150 microM SNP retained up to 15% more water than those pretreated with water or NO(2)(-)/NO(3)(-). The effect of SNP treatment on water retentio

Higher-order structure of interphase chromosomes and radiation-induced chromosomal exchange aberrations.

PURPOSE: To discuss the effects of chromosome structure on the inter- and intrachromosomal exchanges in the framework of the model, which argues that the higher-order chromosome structure contains 10-30 Mbp loop clusters (micelles). RESULTS AND CONCLUSIONS: Intermicelle contacts determine exchange-type chromosome aberrations. Ratios of inter- to intrachromosomal exchanges calculated in the model are similar to the experimental data (literature) for human lymphocytes and flat fibroblasts. The frequency of interchanges is affected by nuclear shape; this might explain the greater number of inter

Evaluation of herbicides action on plant bioindicators by genetic biomarkers: a review.

The use of pesticides has increased worldwide, owing to the demand for products of good quality and to satisfy a growing population. Herbicides represent almost half of the total amount of pesticides used. Although important to the reduction of costs and an increase of productivity, their indiscriminate use, as well as that of the other pesticides, is a global environmental problem, since they affect the living organisms. To evaluate the damage caused by herbicides to the environment, different organisms have been used as bioindicators, especially higher plants, due to several advantages. This

Cryofixation rapidly preserves cytoskeletal arrays of leaf epidermal cells revealing microtubule co-alignments between neighbouring cells and adjacent actin and microtubule bundles in the cortex.

Accurate preservation of microtubule and actin microfilament arrays is crucial for investigating their roles in plant cell development. Aldehyde fixatives such as paraformaldehyde or glutaraldehyde preserve cortical microtubule arrays but, unless actin microfilaments are stabilized with drugs such as m-maleimidobenzoyl N-hydroxysuccinimide ester (MBS), ethylene glycol bis[sulfosuccinimidylsuccinate] (sulfo-EGS) or phalloidin, their arrays are often poorly preserved. Cryofixation, used primarily for electron microscopy, preserves actin microfilaments well but is used rarely to fix plant cells f

Silent Signals: Correlating Plant Bioelectric Activity with Human Emotional States via Wearable Sensing.

We present a bio-hybrid sensing system that uses a living plant (Tradescantia pallida) as an ambient biosensor for human stress states as a single-participant proof-of-concept study. An AD8232 biosignal amplifier captures plant bioelectric activity, while a Happimeter smartwatch simultaneously measures the wearer's mood via machine learning on wrist-worn sensor data. Over 129 paired observations across eleven days in a naturalistic desk-work setting, a within-day fixed-effects analysis reveals robust stress-plant coupling: seven correlations survive Benjamini-Hochberg false discovery rate corr

Synergistic and antagonistic effects on genotoxicity of chemicals commonly found in hazardous waste sites.

Synergistic and antagonistic effects on genotoxicity of mixtures of four chemicals; i.e., lead tetraacetate (LTA), arsenic trioxide (ATO), dieldrin (DED), and tetrachloroethylene (TCE), were evaluated by the Tradescantia-micronucleus (Trad-MCN) assay. The chemicals were mixed in ratios of 1:1, 1:2 and 2:1 for mixtures of two chemicals and 1:1:1 each for three chemicals. The concentration of stock solution of these chemicals was around the minimum effective dose (MED) or below the MED for these chemicals as reported by Sandhu et al. (1989). Treatments were applied to plant cuttings by hydroponi

A simple non-toxic ethylene carbonate fluorescence in situ hybridization (EC-FISH) for simultaneous detection of repetitive DNA sequences and fluorescent bands in plants.

The major drawbacks of standard plant fluorescence in situ hybridization (FISH) designed for double-stranded DNA probes include requirement for experimentally determined heat denaturation of chromosomes at high temperatures and at least overnight hybridization. Consequently, processing with chromosomal preparations may easily result in heat-induced deterioration of chromosomal structural details, is time-consuming, and involves the use of toxic formamide and formaldehyde. Here, I have described a simple and appealing non-toxic procedure with ethylene carbonate (EC)-a formamide-substituting sol

Apoptotic/cell cycle arrest potential of dusty miller methanol extract against Paca-2 pancreatic cancer cells via upregulating Bax/Bcl2: A HPLC-ESI-MS/MS, GNPS-based molecular networking and network pharmacology studies.

Cancer continues to be a major worldwide health problem, necessitating the continuous discovery of novel therapeutics. The current study investigated the anti-cancer potential of Jacobaea maritima (JM) and Tradescantia pallida (TP) extracts via MTT assay. Initial screening revealed JM's superior cytotoxicity across multiple human cancer cell lines (lung, pancreatic, colon, breast, osteosarcoma), with maintaining moderate safety on normal cells, while TP showed limited activity. Further analysis highlighted JM's potent effect on the pancreatic cells (Paca2) (IC50 = 29.1 μg/mL), where it induced

Immediate hypersensitivity to leaf extracts of Callisia fragrans (inch plant) in a dog.

Contact with members of the plant family Commelinaceae, which includes wandering jew (Commelina spp. formerly called Tradescantia spp.) and inch plant (Callisia fragrans), can cause cell-mediated contact dermatitis in dogs. However, reports of canine IgE-mediated hypersensitivity to these plants have not been published. The purpose of this study was to discover whether IgE antibodies specific for extractable components of C. fragrans could be identified in serum from a dog that had anaphylactic shock after exposure to the plant and after skin patch testing with the sap from a leaf of C. fragra

A new efficient immunoprotocol to detect chromosomal/nuclear proteins along with repetitive DNA in squash preparations of formalin-fixed, long-stored root tips.

BACKGROUND: Protein detection on large somatic chromosomes typically includes paraformaldehyde fixation and squashing of enzymatically softened root tips in a buffer. It often suffers from chromosome clumping, poor chromosome morphology, non-specific fluorescence, insufficient immunoreactivity, which collectively reduce the credibility of immunolabeling, hindering its effective combination with fluorescence in situ hybridization (FISH). Material harvesting and pre-detection steps must be completed within a short time, usually one day, which complicates research. The aim of this study was to de

New spatial records of vascular plants in the Azores Archipelago: the PRIBES project and the Azorean Biodiversity Portal (ABP) initiatives - I. São Jorge Island (Azores).

BACKGROUND: The Azores Archipelago is known for its important natural heritage, yet its ecosystems face a "green tsunami" in the form of numerous exotic and invasive species. This influx has wrought serious biodiversity loss and degradation of ecosystem services, representing one of the greatest threats to conservation across the islands. Originating from accelerated global trade and travel, these invasions impact human activities, public health and economic sectors alike. The PRIBES project intends to contribute to "The Regional Strategy for the Management of Terrestrial and Freshwater Exotic
