Exploring the Function of OPTN From Multiple Dimensions.

Autophagy is an essential intracellular degradation system responsible for delivering cytoplasmic components to lysosomes. Within this intricate process, optineurin (OPTN), an autophagy receptor, has attracted extensive attention due to its multifaceted roles in the autophagy process. OPTN is regulated by various posttranslational modifications and actively participates in numerous signaling pathways and cellular processes. By exploring the regulatory mechanism of OPTN posttranslational modification, we can further understand the critical role of protein posttranslational modification in biolo

Optineurin Restricts ZIKV Replication Through Promoting IFN-Mediated JAK/STAT Signaling Pathway.

Optineurin (OPTN) is a multifunctional adapter protein involved in several inflammatory and anti-viral signaling pathways. However, its role in Zika virus (ZIKV) infection remains unclear. This study aims to investigate the effect of OPTN on ZIKV replication and to elucidate the underlying molecular mechanisms. Differential expressions of OPTN during ZIKV infection were analyzed in both A549 and U251-MG cells. OPTN was overexpressed using an EGFP-OPTN plasmid, with an empty EGFP-N1 vector as a control. ZIKV NS5 mRNA and NS1 protein levels were quantified in A549 cells following ZIKV infection

OPTN recruitment to a Golgi-proximal compartment regulates immune signalling and cytokine secretion.

Optineurin (OPTN) is a multifunctional protein involved in autophagy and secretion, as well as nuclear factor κB (NF-κB) and IRF3 signalling, and OPTN mutations are associated with several human diseases. Here, we show that, in response to viral RNA, OPTN translocates to foci in the perinuclear region, where it negatively regulates NF-κB and IRF3 signalling pathways and downstream pro-inflammatory cytokine secretion. These OPTN foci consist of a tight cluster of small membrane vesicles, which are positive for ATG9A. Disease mutations in OPTN linked to primary open-angle glaucoma (POAG) cause a

Absence of the OPTN mutation in a patient with ALS and familial primary open angle glaucoma.

The optineurin (OPTN) gene, known to be implicated in primary open-angle glaucoma (POAG), is the more recent genetic factor linked to ALS. We report the case of a 75year-old man who developed ALS and whose medical history was dominated by a familial POAG. The absence of OPTN gene mutation in a patient who suffered from two conditions linked to mutations of this gene does not support involvement of OPTN in ALS.

Autophagic signatures in peripheral blood mononuclear cells from Parkinson's disease patients.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor impairments and the accumulation of misfolded α-synuclein. Dysregulation of the autophagy-lysosomal pathway (ALP), responsible for degrading misfolded proteins, has been implicated in PD pathogenesis. However, current diagnostic approaches rely heavily on motor symptoms, which occur due to substantial neurodegeneration, limiting early detection and intervention. This study investigated the potential of ALP-associated proteins in peripheral blood mononuclear cells (PBMCs) as diagnostic biomarkers for ear

Oligomerization of optineurin and its oxidative stress- or E50K mutation-driven covalent cross-linking: possible relationship with glaucoma pathology.

The optineurin gene, OPTN, is one of the causative genes of primary open-angle glaucoma. Although oligomerization of optineurin in cultured cells was previously observed by gel filtration analysis and blue native gel electrophoresis (BNE), little is known about the characteristics of optineurin oligomers. Here, we aimed to analyze the oligomeric state of optineurin and factors affecting oligomerization, such as environmental stimuli or mutations in OPTN. Using BNE or immunoprecipitation followed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), we demonstrated that both

The genetics of autosomal recessive ALS: a review of the common forms and their phenotypes.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease marked by progressive degeneration of upper and lower motor neurons. Most forms of ALS associated with a suspected causal variant are inherited in an autosomal dominant manner. However, there is an important subset of autosomal recessive (AR) variants, often associated with early-onset or atypical clinical features. Advances in genetic sequencing have led to increased recognition of AR ALS. In this review, we focus on four key confirmed AR ALS-associated genes, which appear to be most common-ALS2, SPG11, OPTN, and the D90

Distinct autophagy impairment mechanisms of huntingtin aggregates with different polyQ lengths.

Huntington's disease (HD) is characterized by the aggregation of mutant huntingtin (mHTT) containing elongated polyglutamine (polyQ) tracts. mHTT aggregates that fail to be cleared by autophagy cause neurotoxicity. While the polyQ length in patients with HD ranges from 40 to over 90 repeats, how these varying polyQ lengths affect autophagy impairment remains unclear. Using polyQ aggregation sensors based on bimolecular fluorescence complementation (BiFC), we uncovered distinct autophagy impairment mechanisms: PolyQ103 aggregates evade recognition by autophagy receptor SQSTM1/p62, whereas polyQ

Model reactions of Cr (VI) with DNA mediated by thiol species.

Model reactions were devised to investigate the capacity of physiologically interesting thiol compounds to mediate reactions between CrO4(2-) (Cr (VI)) and DNA. The sulfhydryl containing reagents included cysteine, glutathione, apo-metallothionein (apoMT). Zinc finger 3 of transcription factor IIIA (Zn-F3) of Xenopus laevis was investigated as a potential redox active site of reaction of Cr (VI) and thiol compounds. The DNA samples were calf thymus DNA and two oligomers, one of them specific for binding Zn-F3. Results showed that in the presence of Cr (VI) apoMT readily participated in damagin

Clinical heterogeneity within the ALS-FTD spectrum in a family with a homozygous optineurin mutation.

OBJECTIVE: Mutations in the gene encoding for optineurin (OPTN) have been reported in the context of different neurodegenerative diseases including the amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) spectrum. Based on single case reports, neuropathological data in OPTN mutation carriers have revealed transactive response DNA-binding protein 43 kDa (TDP-43) pathology, in addition to accumulations of tau and alpha-synuclein. Herein, we present two siblings from a consanguineous family with a homozygous frameshift mutation in the OPTN gene and different clinical presentatio

Adult-onset primary glaucoma and molecular genetics: a review.

PURPOSE: To evaluate recent molecular genetic studies focused on localizing and identifying the genes involved in adult-onset primary glaucoma, characterizing the gene products, and investigating the molecular mechanisms implicated in the pathophysiology of the disease. METHODS: Several studies have aimed at understanding gene expression and protein processing and attempting to correlate the mutations identified in the involved genes, particularly the TIGR/MYOC gene, with the overall spectrum of the disease, ranging from juvenile glaucoma to typical late-onset primary open-angle glaucoma. Phase separation of OPTN initiates mitophagy to orchestrate craniofacial bone mineralization.

Recently, mitophagy-mediated bone mineralization of mesenchymal stem cells has emerged as another bone formation pattern, but whether mitophagy-mediated bone mineralization shapes craniofacial development remains unknown. Here, we demonstrate that loss of OPTN, a keystone macroautophagy/autophagy receptor, impairs mitophagy and acidic calcium phosphate (ACP) transport in orofacial bone mesenchymal stem cells (OMSCs), leading to craniofacial bone mineralization defects. We substantiate that OPTN undergoes LLPS both in vitro and in vivo, driven by S173 phosphorylation within its intrinsically di

Role of Optineurin in the Mitochondrial Dysfunction: Potential Implications in Neurodegenerative Diseases and Cancer.

Optineurin (Optn) is a 577 aa protein encoded by the Optn gene. Mutations of Optn are associated with normal tension glaucoma and amyotrophic lateral sclerosis, and its gene has also been linked to the development of Paget's disease of bone and Crohn's disease. Optn is involved in diverse cellular functions, including NF-κB regulation, membrane trafficking, exocytosis, vesicle transport, reorganization of actin and microtubules, cell cycle control, and autophagy. Besides its role in xenophagy and autophagy of aggregates, Optn has been identified as a primary autophagy receptor, among the five

A Dutch family with autosomal recessively inherited lower motor neuron predominant motor neuron disease due to optineurin mutations.

Approximately 10% of motor neuron disease (MND) patients report a familial predisposition for MND. Autosomal recessively inherited MND is less common and is most often caused by mutations in the superoxide dismutase 1 (SOD1) gene. In 2010, autosomal recessively inherited mutations in the optineurin (OPTN) gene were found in 1% of Japanese patients with sporadic amyotrophic lateral sclerosis (ALS). Autosomal dominantly inherited OPTN mutations have been described as a cause of primary open-angle glaucoma in the Netherlands and were also found in two Dutch sporadic MND patients. We report the fi

Optineurin in primary open angle glaucoma.

The authors' initial estimate indicated that mutations in Optineurin are responsible for a significant proportion of LPG/POAG families. Currently, there are up to 1.2 million persons with LPG and up to 2.47 million persons with POAG in the United States alone. Perhaps twice as many individuals are already affected with this condition without any identifiable clinical signs or symptoms. Investigators are eagerly awaiting confirmation of OPTN mutations in other glaucoma populations. Although additional mutations have already been identified in the sporadic cases of LPG, the significance of this

Optineurin mutation-associated language variant frontotemporal dementia.

A right-handed woman in her early 50s presented with progressive language difficulties with visual and auditory hallucinations. Upper motor neuron signs were present without lower motor neuron signs, insufficient to make a diagnosis of amyotrophic lateral sclerosis (ALS). MRI showed bilateral superior temporal atrophy (right>left) and F-18 fluorodeoxyglucose-positron emmission tomography (FDG-PET) scan showed right temporal hypometabolism. Her clinical diagnosis was atypical or mixed language variant frontotemporal dementia (FTD) given the presence of agrammatism and phonemic intrusions. There

Paeoniflorin Ameliorates Osteoarthritis via Ring Finger Protein 216/Optineurin-Mediated Autophagy Activation in Chondrocytes.

Osteoarthritis (OA) is a prevalent degenerative joint disorder. Paeoniflorin (PF), a primary bioactive compound from Paeonia lactiflora roots, demonstrates therapeutic potential for OA. This study investigated the effects and mechanisms of PF in OA. In vitro, an OA model was established using interleukin-1β (IL-1β)-treated chondrocytes. In vivo, an OA mouse model was generated via destabilization of the medial meniscus (DMM). Molecular docking, bioinformatics analysis, and experimental validation identified ring finger protein 216 (RNF216) as a PF target. RNF216 overexpression and optineurin (

Toward an integrative view of Optineurin functions.

BACKGROUND: The development of fundamental motor skills (FMS) and changes in body composition among children and adolescents with intellectual and developmental disabilities (IDD) are an emerging public health issue. Physical activity serves as a dual protective mechanism, enhancing motor skills and optimizing body composition. Yet no systematic reviews have been published examining the effects of structured exercise programs on both FMS progression and weight management in this population. METHODS: Web of Science, PubMed, Cochrane Library, and Medline databases were searched for peer-reviewe
