Chimerism patterns of long-term stable mixed chimeras posthematopoietic stem cell transplantation in patients with nonmalignant diseases: follow-up of long-term stable mixed chimerism patients.

Long-term stable mixed chimerism (MC) is a rare phenomenon after hematopoietic stem cell transplantation (HSCT) characterized by 5% to 95% residual recipient hematopoietic cells. The underlying mechanisms of MC are largely unknown. In this study we compared full donor chimerism with long-lasting stable MC for a median of 9.5 years (range, 5 to 16.5) post-HSCT in patients with nonmalignant diseases. Several factors significantly associated with the likelihood of stable MC development were identified by univariate analysis, eg, younger donor age, sibling donor, and conditioning regimen. Despite

Regulation of the immune response in experimental models of autoimmune disorders: I. Immunocompetence and transplantation tolerance in female (NZB X NZW)F1 hybrid mice immunosuppressed with total lymphoid irradiation and in reconstituted bone marrow chimeras.

Female (NZB X NZW)F1 (B/WF1) mice treated with fractionated total lymphoid irradiation (TLI) and TLI conditioned BALB/c leads to B/WF1 bone marrow (BM) chimeras were studied to determine the effects of TLI and reconstitution with normal BM on in vivo and in vitro immune reactivity. TLI treated B/WF1 females showed limited increased retention of semiallogeneic skin grafts (2 1/2 times as long as untreated) and allogeneic skin grafts (twice as long as untreated). In vitro proliferative responses to allogeneic lymphocytes were severely depressed at 5 days after TLI with partial recovery as early

Chimerism through the activation of invariant natural killer T cells prolongs graft survival after transplantation of induced pluripotent stem cell-derived allogeneic cardiomyocytes.

The loss of functional cells through immunological rejection after transplantation reduces the efficacy of regenerative therapies for cardiac failure that use allogeneic induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs). Recently, mixed-chimera mice with donor-specific immunotolerance have been established using the RGI-2001 (liposomal formulation of α-galactosyl ceramide) ligand, which activates invariant natural killer T (iNKT) cells. The present study aimed to investigate whether mixed chimerism, established using RGI-2001, prolongs graft survival in allogeneic iPSC-CM transpl

Adaptive differentiation of murine lymphocytes. I. Both T and B lymphocytes differentiating in F1 transplanted to parental chimeras manifest preferential cooperative activity for partner lymphocytes derived from the same parental type corresponding to the chimeric host.

The concept of adaptive (selective) differentiation preducts that early differentiation of lymphocytes is conditioned by the environment in which such differentiation takes place. These processes appear to involve selection of lymphocytes according to their self-recognition between interacting lymphocytes is, at least in part, controlled by major histocompatibility complex-linked genes, then adaptive differentiation is also controlled by these genes. In these studies, we have tested the capacities of helper T lymphocytes and hapten-specific B lymphocytes primed in the environments of various c

Successful Spermatogonial Stem Cells Transplantation within Pleuronectiformes: First Breakthrough at inter-family Level in Marine Fish.

As a promising biotechnology, fish germ cell transplantation shows potentials in conservation germplasm resource, propagation of elite species, and generation of transgenic individuals. In this study, we successfully transplanted the Japanese flounder (P. olivaceus), summer flounder (P. dentatus), and turbot (S. maximus) spermatogonia into triploid Japanese flounder larvae, and achieved high transplantation efficiency of 100%, 75-95% and 33-50% by fluorescence tracking and molecular analysis, respectively. Eventually, donor-derived spermatozoa produced offspring by artificial insemination. We

A Phase 1 Study of 4 Live, Recombinant Human Cytomegalovirus Towne/Toledo Chimera Vaccines in Cytomegalovirus-Seronegative Men.

BACKGROUND: Human cytomegalovirus (HCMV) infection causes disease in newborns and transplant recipients. A HCMV vaccine (Towne) protects transplant recipients. METHODS: The genomes of Towne and the nonattenuated Toledo strain were recombined, yielding 4 Towne/Toledo chimera vaccines. Each of 36 HCMV-seronegative men received 1 subcutaneous dose of 10, 100, or 1000 plaque-forming units (PFU) in cohorts of 3. Safety and immunogenicity were evaluated over 12 weeks after immunization and for 52 weeks for those who seroconverted. RESULTS: There were no serious local or systemic reactions. No subj

In utero bone marrow transplantation of fetal baboons with mismatched adult marrow: initial observations.

Recent advances in prenatal diagnoses of sickle cell anemia and thalassemia permit early identification of affected fetuses. However, the only intervention possible to date is abortion of the affected fetuses. Transplantation of normal marrow into fetuses in utero could correct these life-threatening disorders, but to accomplish this techniques must be developed for fetal transplantation in man. Therefore, we have transplanted fetal baboons with mismatched adult baboon bone marrow from donors that differed at the glucose phosphate isomerase locus. Twenty-two fetuses between 60 and 160 days of

Chimerism in the immunohematology laboratory in the molecular biology era.

Dual or multiple cell populations, induced by chimeras, have been the subject of many studies. This long-standing fascination with chimeras has revealed a good deal of knowledge about human inheritance. Although historically most chimeras were caused by natural events, certain current medical intervention therapies are increasing the number of situations that can lead to a mixed cell population, that is, the chimeric condition, in humans. Medical therapies such as transfusion, stem cell transplantation, kidney transplantation, and artificial insemination induce temporary and sometimes permanen

Sex determination in germ line chimeras of Drosophila melanogaster.

Of 55 flies developing from blastoderms which had received male or female pole cell transplants, 15 (7 females and 8 males) wer shown by progeny testing to be germ line chimeras. Since donor and host pole cells were genetically marked with contrasting X- or Y-linked alleles, the progeny testing scheme enabled the genotypic sex of the donor component undergoing gametogenesis to be identified as either the same as ('homosexual' chimeras) or opposite ('heterosexual' chimeras) that of the host. All seven of the female chimeras were identified as 'homosexual' chimeras carrying only chromosomally fe

Comparison of wasting syndrome in [MRL lpr/lpr-->MRL +/+] chimera and graft versus host disease in [B10.D2-->BALB/c] chimera and an attempt to transfer the wasting syndrome in [MRL lpr/lpr-->MRL +/+] to MRL +/+ mice.

We compared the findings in the wasting syndrome seen in [MRL lpr/lpr--> MRL +/+] chimeras with those of chronic graft versus host disease (GVHD) in [B10.D2-->BALB/c] chimeras. BALB/c mice were lethally irradiated and administered B10.D2 spleen and bone marrow cells. These mice are identical to MHC and Mls but differ as to genetic background. As a result of chronic GVHD, these [B10.D2-->BALB/c] chimeras showed hair loss, weight loss and atrophy of lymph nodes and spleen beginning 5 weeks after the transplantation. MRL lpr/lpr mice carry the lpr gene and spontaneously develop generalized lymph

Exploring the Ethics of Creating Chimeric 'Monkey-Human' Embryos: Could Xenotransplantation, the Treatment of Anencephalic Infants, or Synthetic Human Embryos Provide Insights?

Induced human extended pluripotent stem cells (hEPSC) have been injected into monkey blastocysts to create chimeric embryos. Chimera research is a scientific technique which seeks to develop human organs for transplantation, but it has raised concerns regarding human cells contributing to the brain formation of nonhuman animals. This article considered whether brain development should guide ethical considerations on chimera research by investigating the morality of terminating live anencephalic infants to procure their organs. It found that identifying the presence or absence of a human biolog

Will Human-Animal Chimeras Cause Moral Confusion? Exploring Public Attitudes.

Recent medical research involving human-monkey chimeras, human brain organoids in rats, and the transplantation of a gene-edited pig heart and gene-edited pig kidneys in living human beings have intensified the debate about whether we should create human-animal chimeras for biomedical purposes and, if so, how we should treat them. Influential views in the debate frequently appeal to assumptions regarding how people will react to such chimeras. It has, for example, been argued that the most important objection against creating such chimeras is that this will result in inexorable moral confusion

Innate and adaptive immune responses are tolerized in chimeras prepared with nonmyeloablative conditioning.

BACKGROUND: Mixed chimerism is an effective approach for tolerance induction in transplantation. Strategies to achieve mixed chimerism with relatively low toxicity have significantly expanded the clinical use of chimerism. METHODS: Allogeneic bone marrow transplants were performed between B6 (H2(b)) and BALB/c (H2(d)) mice. Recipient B6 mice were nonmyeloablatively conditioned with anti-αβ-T-cell receptor, anti-CD154, or rapamycin alone or in different combinations. A total of 15 × 10(6) BALB/c bone marrow cells were transplanted after varying doses of cGy of total body irradiation. RESULTS:

T lymphocytes of allophenic mice do not reject spleen colony-forming units of the parental genotypes.

Self-tolerance in BALB/c<-->C57BL/6 and BALB/c<-->B10.SM (SM) allophenic mice is examined in this study. Chimerism was determined by coat mosaicism, using electrophoresis of allozyme variants at the glucose phosphate isomerase-1 (GPI-1) in peripheral blood erythrocytes and a cytotoxity test for lymph node (LN) lymphocytes and blood nucleated cells. An age-dependent increase in the percent content of BALB/c erythrocytes was observed in both types of chimeras. The immunological status of chimeras was assessed by T cell interactions with spleen colony-forming cells (SFU-S) able to form colonies i

Humoral and cell-mediated immune responses in fully allogeneic bone marrow chimera in mice.

AKR mice were protected from lethal irradiation and established as long-lived chimeras by transplanting allogeneic C57BL/6 (B6) bone marrow that had been treated in vitro with anti-Thy-1 antiserum without complement. In these chimeras, which were designated [B6 {arrow} AKR], virtually all the thymus and spleen cells were shown to be derived from the B6 donor; several immune functions studied in these chimeras were as follows: (a) The chimeric mice were tolerant of histocompatibility antigens of both donor and recipient strain and nearly fully reactive to antigens of third party, as revealed by
