Effects of neonatal conductive hearing loss on brain stem auditory nuclei.

Both postnatal auditory deprivation and experimentally produced conductive hearing losses in mice result in incomplete maturation of most brain stem auditory neurons. The affected groups are: octopus cell, globular cell, small spherical cell, and large spherical cell groups in ventral cochlear nuclei; and the lateral superior olive and medial nucleus of the trapezoid body of the superior olivary complex. When 45 days of auditory deprivation are followed by 45 days of normal acoustic stimulation, there is incomplete maturation of neurons in: multipolar cell, globular cell, small spherical cell,

Acromioclavicular joint instability on cross-body adduction view: the biomechanical effect of acromioclavicular and coracoclavicular ligaments sectioning.

BACKGROUND: The acromioclavicular (AC) and coracoclavicular (CC) ligaments are important stabilizers of the AC joint. We hypothesized that AC and trapezoid ligament injuries induce AC joint instability and that the clavicle can override the acromion on cross-body adduction view even in the absence of conoid ligament injury. Accordingly, we investigated how sectioning the AC and CC ligaments contribute to AC joint instability in the cross-body adduction position. METHODS: Six fresh-frozen cadaveric shoulders were used in this study, comprising five male and one female specimen, with a mean age

Perfidious synaptic transmission in the guinea-pig auditory brainstem.

The presence of 'giant' synapses in the auditory brainstem is thought to be a specialization designed to encode temporal information to support perception of pitch, frequency, and sound-source localisation. These 'giant' synapses have been found in the ventral cochlear nucleus, the medial nucleus of the trapezoid body and the ventral nucleus of the lateral lemniscus. An interpretation of these synapses as simple relays has, however, been challenged by the observation in the gerbil that the action potential frequently fails in the ventral cochlear nucleus. Given the prominence of these synapses

Comparison of the hyperglycaemic effect of adrenaline and morphine introduced into the liquor space.

1. In unanaesthetized cats a comparison is made of the hyperglycaemic effects of adrenaline and morphine, when injected or infused through chronically implanted cannulae, into different regions of the cerebral ventricles or of the subarachnoid space, in order to determine their sites of action. 2. On injection into the cerebral ventricles both adrenaline and morphine have to reach the subarachnoid space beneath the ventral surface of the brain stem before they can exert their hyperglycaemic effect. The adrenaline has to reach the region rostral to the pons, i.e. the fossa interpeduncularis, an

Neck movement but not neck position modulates skin sympathetic nerve activity supplying the lower limbs of humans.

We previously showed that dynamic, but not static, neck displacement modulates muscle sympathetic nerve activity (MSNA) to lower limbs of humans. However, it is not known whether dynamic neck displacement modulates skin sympathetic nerve activity (SSNA). Tungsten microelectrodes inserted into the common peroneal nerve were used to record SSNA in 5 female and 4 male subjects lying supine on a table that fixed their head in space but allowed trapezoidal ramp (8.1 ± 1.2°/s) and hold (17.5° for 53 s) or sinusoidal (35° peak to peak at 0.33-0.46 Hz) horizontal displacement of the body about the hea

Olivocochlear projections contribute to superior intensity coding in cochlear nucleus small cells.

Understanding communication signals, especially in noisy environments, is crucial to social interactions. Yet, as we age, acoustic signals can be disrupted by cochlear damage and the subsequent auditory nerve fibre degeneration. The most vulnerable medium- and high-threshold-auditory nerve fibres innervate various cell types in the cochlear nucleus, among which the small cells are unique in receiving this input exclusively. Furthermore, small cells project to medial olivocochlear (MOC) neurons, which in turn send branched collaterals back into the small cell cap. Here, we use single-unit recor

Kv3.1 and Kv3.3 subunits differentially contribute to Kv3 channels and action potential repolarization in principal neurons of the auditory brainstem.

KEY POINTS: Kv3.1 and Kv3.3 subunits are highly expressed in the auditory brainstem, with little or no mRNA for Kv3.2 or Kv3.4. Changes in Kv3 currents and action potential (AP) firing were analysed from wild-type, Kv3.1 and Kv3.3 knockout (KO) mice. Both Kv3.1 and Kv3.3 immunostaining was present and western blots confirmed loss of subunit protein in the respective KO. Medial nucleus of the trapezoid body (MNTB) AP repolarization utilized Kv3.1 and/or Kv3.3; while in the lateral superior olive (LSO) Kv3.3 was essential. Voltage-gated calcium currents were unchanged between the genotypes. But

[Vestibular efferent neurons of the guinea pig forming projections into the saccule].

The distribution of vestibular efferent neurons projecting to the saccule and efferent neurons transmitting axons to the acoustic nerve (cochlear efferent neurons) has been studied in guinea pig by comparison using the method of retrograde axonal transport of horseradish peroxidase. The saccular efferent neurons are revealed bilaterally in the subendymal granular fundus of the fourth ventricle of the brain and more laterally of the facial nerve genu, ipsilaterally in the small-celled reticular nucleus as well as in nuclei of the supraolivary complex: nucleus of lateral oliva and lateral nucleu

Structure-function relation of the developing calyx of Held synapse in vivo.

KEY POINTS: During development the giant, auditory calyx of Held forms a one-to-one connection with a principal neuron of the medial nucleus of the trapezoid body. While anatomical studies described that most of the target cells are temporarily contacted by multiple calyces, multi-calyceal innervation was only sporadically observed in in vivo recordings, suggesting a structure-function discrepancy. We correlated synaptic strength of inputs, identified in in vivo recordings, with post hoc labelling of the recorded neuron and synaptic terminals containing vesicular glutamate transporters (VGluT)

Preliminary In Vivo Evaluation of Coracoclavicular Ligament Mechanics During Shoulder Elevation After Acromioclavicular Joint Reconstruction.

BACKGROUND: Persistent instability and altered scapular mechanics after acromioclavicular joint (ACJ) reconstruction may stem from inadequate replication of native coracoclavicular (CC) ligament function. However, in vivo data on postoperative CC ligament behavior during dynamic shoulder motion remain lacking. PURPOSE: To assess in vivo functional distance and timing characteristics of the conoid and trapezoid ligaments during active shoulder elevation after ACJ reconstruction, using dynamic stereo x-ray (DSX) imaging and patient-specific 3-dimensional (3D) models. Synaptic plasticity of inhibitory synapses onto medial olivocochlear efferent neurons.

The descending auditory system modulates the ascending system at every level. The final descending, or efferent, stage comprises lateral olivocochlear and medial olivocochlear (MOC) neurons. MOC somata in the ventral brainstem project axons to the cochlea to synapse onto outer hair cells (OHC), inhibiting OHC-mediated cochlear amplification. MOC suppression of OHC function is implicated in cochlear gain control with changing sound intensity, detection of salient stimuli, attention and protection against acoustic trauma. Thus, sound excites MOC neurons to provide negative feedback of the cochle

Anatomy of superior olivary complex and lateral lemniscus in Etruscan shrew.

Based on the auditory periphery and the small head size, Etruscan shrews (Suncus etruscus) approximate ancestral mammalian conditions. The auditory brainstem in this insectivore has not been investigated. Using labelling techniques, we assessed the structures of their superior olivary complex (SOC) and the nuclei of the lateral lemniscus (NLL). There, we identified the position of the major nuclei, their input pattern, transmitter content, expression of calcium binding proteins (CaBPs) and two voltage-gated ion channels. The most prominent SOC structures were the medial nucleus of the trapezoi

Designing Active Assistance for Gait With Hip Abductors Weakness Using Predictive Simulation.

Individuals with weak hip abductors commonly exhibit compensatory gait patterns and excessive lateral center of mass excursions, which may reduce walking efficiency and increase the risk of falling. In this study, we investigated the influence of hip abductor weakness and of ideal hip abduction assistive torque on gait patterns through an established predictive simulation framework and a full-body musculoskeletal model. Using this model and simulation framework, hip abductor muscle strength was reduced in several increments, and gait patterns were predicted and qualitatively compared to report

Kainic acid lesions of the superior olivary complex: a horseradish peroxidase study of surviving brain-stem projections.

Lesions of the superior olivary complex of the adult ferret were made by pressure injection of kainic acid (5 nM/microliters in Locke's solution) through a glass micropipette inserted into the lower brain-stem. Small injections of kainic acid (1.5 microliters) produced a localized loss of nerve cells in the superior olivary complex in the vicinity of the pipette tip without apparent damage to fibers of passage in the trapezoid body. The extent of neural damage was determined by the absence of cell bodies in Nissl-stained sections. Integrity of fibers in the trapezoid body and other decussating

Pressor response elicited by nose-up vestibular stimulation in cats.

The purpose of the present study was to determine whether selective activation of vestibular receptors produces changes in blood pressure. Blood pressure was recorded during trapezoidal head rotations in cats with extensive denervations to eliminate nonlabyrinthine inputs that could be produced by the movements. Large (50 degrees) nose-up trapezoidal head tilts produced an increase in blood pressure of approximately 18 mmHg; ear-down tilt produced little change in blood pressure. The changes in blood pressure began approximately 1.4 s after the plateau of the stimulus. The responses to nose-up

Mycophenolic acid therapeutic drug monitoring using area under the curve in pediatric heart transplant recipients.

INTRODUCTION: Pharmacokinetics of mycophenolic acid (MPA) display substantial interpatient variability, with up to 10-fold difference of exposure in individual patients under a fixed-dose regimen. MPA trough level (C0) monitoring is common in clinical practice but has not proven sufficiently informative in predicting MPA exposure or patient outcomes, especially in children. No limited sampling strategies (LSSs) have been generated from pediatric heart transplant (HTx) recipients to estimate MPA AUC. METHODS: Single-center, observational analysis of 135 de novo pediatric HTx recipients ≤21 yea

Myelin mosaicism and brain plasticity in heterozygous females of a canine X-linked trait.

The shaking (sh) pup, an animal model of Pelizaeus-Merzbacher disease, is characterized by severe central nervous system dysmyelination in affected males, and myelin mosaicism in some female heterozygotes as a result of X-linked inactivation. Heterozygous females develop a tremor of varying severity that usually disappears at 4 to 6 weeks, whereas male hemizygotes have severe, generalized tremor that persists throughout life. We have used these two myelin-deficient models to study the potential for recovery with time as reflected by brainstem auditory evoked responses (BAERs). At set time poin

Functional Role of γ-Crystallin N in the Auditory Hindbrain.

γ-crystallins are major components of the vertebrate lens but show expression in other tissues as well. Their extralenticular functions remain so far unclear. Here, we explored such roles in the rodent superior olivary complex in which previous analysis demonstrated developmentally regulated expression of Crygd, Cryge and Crygn. Immunohistochemistry with novel antibodies against Crygd/e and Crygn indicate that expression of Crygd/e was moderate and varied between the perinatal superior olivary complex of mice, rats, and gerbils. Crygn-immunoreactivity was more robust and consistently highest i

Protection from noise-induced hearing loss by Kv2.2 potassium currents in the central medial olivocochlear system.

The central auditory brainstem provides an efferent projection known as the medial olivocochlear (MOC) system, which regulates the cochlear amplifier and mediates protection on exposure to loud sound. It arises from neurons of the ventral nucleus of the trapezoid body (VNTB), so control of neuronal excitability in this pathway has profound effects on hearing. The VNTB and the medial nucleus of the trapezoid body are the only sites of expression for the Kv2.2 voltage-gated potassium channel in the auditory brainstem, consistent with a specialized function of these channels. In the absence of un

Impacts of perinatal nicotine exposure on nicotinic acetylcholine receptor expression and glutamatergic synaptic transmission in the mouse auditory brainstem.

In utero nicotine exposure from maternal smoking is linked to increased risk of auditory processing deficits. This study investigated the impact of developmental nicotine exposure during the critical period on nicotinic acetylcholine receptor (nAChR) functional expression, glutamatergic synaptic transmission and auditory processing in the mouse auditory brainstem. We assessed nAChR function at a central synapse and the consequences of perinatal nicotine exposure (PNE) on synaptic currents and auditory brainstem responses (ABRs) in mice. Our results indicate developmentally regulated changes in
