Proteinase and gelatinolytic properties of a Triatoma infestans extract.

An extract of Triatoma infestans has previously been demonstrated to produce specific IgG and IgE both in animals and in atopic humans with rhinitis/asthma as well as hypersensitivity pneumonitis in guinea pigs aerosolized with T. infestans. We attempted to determine whether the antigen or antigens responsible belonged to the protease group, as occurs with other allergens such as house dust mites and cockroaches. To do this, T. infestans was studied by SDS-PAGE, Western blots and gelatinolysis with and without the use of specific protease inhibitors such as E-64, TLCK, TPCK, PMSF, leupeptin, o

Metabolomics of developmental changes in Triatoma sanguisuga gut microbiota.

Triatoma sanguisuga is one of the major vectors of Trypanosoma cruzi in the southeastern US, where it sustains a robust zoonotic parasite transmission cycle and occasional human infections. A better understanding of triatomine development may allow for alternative approaches to insecticide-based vector control. Indeed, the role of the gut microbiota and bacterial endosymbionts in triatomine development and in their vectorial capacity is emerging. We investigated here the differences in microbiota among nymph and adult T. sanguisuga, to shed light on the metabolomic interactions occurring durin

Hindgut microbiota in laboratory-reared and wild Triatoma infestans.

Triatomine vectors transmit Trypanosoma cruzi, the etiological agent of Chagas disease in humans. Transmission to humans typically occurs when contaminated triatomine feces come in contact with the bite site or mucosal membranes. In the Southern Cone of South America, where the highest burden of disease exists, Triatoma infestans is the principal vector for T. cruzi. Recent studies of other vector-borne illnesses have shown that arthropod microbiota influences the ability of infectious agents to colonize the insect vector and transmit to the human host. This has garnered attention as a potenti

[Toxic effect of beta-cipermethrin, deltamethrin and fenitrothion in colonies of Triatoma dimidiata (Latreille, 1811) and Triatoma maculata (Erichson, 1848) (Hemiptera, Reduviidae)].

INTRODUCTION: The susceptibility to insecticides of triatomine species must be evaluated because of their involvement in the transmission of the Chagas disease. In each region with Chagas endemicity, evaluation of insecticide response is necessary to predict the success of the control campaigns. OBJECTIVE: The baseline susceptibility was determined for the active principles deltamethrin, beta-cypermethrin and fenitrothion in nymphs of first and fifth instar of Triatoma dimidiata and nymphs of first instar of Triatoma maculata. Host-Feeding Sources and Infection With Trypanosoma cruzi of Triatoma infestans and Triatoma eratyrusiformis (Hemiptera: Reduviidae) From the Calchaqui Valleys in Northwestern Argentina.

We assessed the prevalence of infection with Trypanosoma cruzi, parasite genotypes (discrete typing units, DTUs), and the host-feeding sources of domestic and peridomestic Triatoma infestans Klug and Triatoma eratyrusiformis Del Ponte in eight rural communities of the subandean Calchaqui valleys in northwestern Argentina. We sought to analyze their epidemiological role in the context of routine vector surveillance and control actions. Infection with T. cruzi was determined by optic microscopy or polymerase chain reaction (PCR) amplification of the hypervariable region of kinetoplast DNA minici

Viability of Trypanosoma cruzi in Triatoma pallidipennis Post-mortem.

PURPOSE: Given that contact with the feces of dead triatomine bugs poses a risk of Trypanosoma cruzi infection, this study evaluated parasite viability and infectious potential in dead triatomines across various post-mortem times and temperatures. METHODS: Three groups of nine of 5th-instar Triatoma pallidipennis nymphs (n = 27) were euthanized via insecticide 30 days post-infection (Ninoa strain). Nymphs were maintained at room temperature (10 -23 °C), refrigeration (4 °C), and incubation (27 °C). Fecal parasite concentration was assessed at 24 h and at 2, 5, 7, 10, 15, 20, 25, and 30 days p

Attempts to improve xenodiagnosis: comparative test of sensibility using Rhodnius neglectus, Panstrongylus megistus, Triatoma vitticeps and Triatoma infestans in endemic areas of Brazil.

From June 1984 to July 1992, 392 xenodiagnostic tests were applied on 264 patients with chronic Chagas disease from Brazilian endemic areas of Virgem da Lapa and Coronel Murta, situated in the Jequitinhonha Valley, in the State of Minas Gerais. The susceptibilities of Rhodnius neglectus, Panstrongylus megistus, Triatoma vitticeps and Triatoma infestans were compared. Most of the time 20 nymphs (fourth instar) of each species were applied to 161 women and 103 men aged between 5 and 83 years of age. The tests were prepared to compare the susceptibilities of two species at a time, using the same

Dynamics of feeding and defecation behavior of Triatoma infestans hybrids.

Triatoma platensis is occasionally found coexisting with Triatoma infestans in chicken coops in Argentina. Some authors have reported the presence of hybrid specimens of both species in chicken coops and other peridomestic habitats. Given the coexistence of T. infestans with T. platensis and the possibility of generating fertile hybrids, it is important to evaluate the vectorial competence of these hybrids. The objective of this study was to record the dynamics of feeding-defecation behavior in fifth-stage nymphs and adults of hybrids between both species and to compare it with T. platensis an

[About an insecticidal paint for controlling Triatoma infestans, in Bolivia].

Preliminary evaluations of an insecticidal paint based on diazinon, chlorpyrifos and pyriproxyfen in a micro-encapsulated formulation (Inesfly 5A IGR) have shown that it has effective and persistent activity against Triatoma infestans inside homes and in areas surrounding homes, in a highly infested region of the Bolivian Chaco. Furthermore, the evaluations have highlighted that the product presents good handling characteristics and gives a good appearance to houses and outhouses that have been treated, and that its acceptance among the population and the local sanitary authorities is excellen

The use of aggregation signals by Triatoma brasiliensis (Heteroptera: Reduviidae).

Triatoma brasiliensis is considered the main vector of Chagas disease in the semiarid zones of North-eastern Brazil. In this work, we describe the spatial pattern of defecation in T. brasiliensis and the spatial relationship of faeces with the shelters of these bugs. Afterwards, we analyse the attraction response of T. brasiliensis larvae to potential odour cues present in their dry faeces, to substances present in their cuticule (footprints) and to the dry faeces of Triatoma pseudomaculata. We show that faecal spots are deposited in a larger density inside the shelter than in the remaining ar

Salivary apyrases of Triatoma infestans are assembled into homo-oligomers.

Apyrase activity is present in the saliva of haematophagous arthropods. It is related to blood-feeding because of the apyrase ability to hydrolyse ADP, a key component of platelet aggregation. Five apyrases with apparent molecular masses of 88, 82, 79, 68 and 67 kDa were identified in the saliva of the vector of Chagas disease, Triatoma infestans. The large size observed during purification of these enzymes suggested oligomerization. In the present study, we confirmed, using gel-filtration and analytical ultracentrifugation, the presence of apyrase oligomers with molecular masses of 200 kDa in

Copulation and mate guarding behavior in Triatoma brasiliensis (Hemiptera: Reduviidae).

The hematophagous bug Triatoma brasiliensis (Neiva) is an important vector of Chagas disease in Brazil. We describe the mating behavior of this species in a comparative manner and analyze the effect of time elapsed since the first imaginal bloodmeal on motivation of males to mate, on female mate-rejections, and on overall mating frequency. We also evaluate whether females of this species copulate with several males in short time intervals, and we examine whether the duration of copulation may vary in the presence of other males compared with what is observed with isolated pairs. Age affected t

Impacts of residual insecticide spraying on the abundance and habitat occupancy of Triatoma sordida and co-occurrence with Triatoma infestans: A three-year follow-up in northeastern Argentina.

Triatoma infestans, the main vector in the Gran Chaco region, may competitively displace other sympatric species such as Triatoma sordida. We conducted a three-year longitudinal study of site- and house-level infestation and abundance of triatomine bugs before and after an area-wide insecticide spraying campaign followed by sustained vector surveillance in a well-defined rural section of the Argentine Chaco encompassing 368-411 houses. Here, we tested whether insecticide applications targeting and virtually suppressing T. infestans reduced the abundance of T. sordida and modified its habitat o

Morphological diversity of Triatoma infestans across biogeographic zones of Chile.

Triatoma infestans, the primary domiciliary vector of Chagas disease in Chile, has a broad geographic distribution and is characterized by high synanthropy and strong colonization capacity. Landscape characteristics are key factors in understanding population distribution and the processes that shape morphological variation. This study aimed to characterize the structures associated with dispersal and feeding using geometric morphometrics in domiciliary adults of T. infestans (n = 353 total specimens analyzed) along a latitudinal gradient (∼1,255 km). Furthermore, the specimens' localities wer

Biological parameters of Triatoma longipennis recently introduced into Central Mexico.

There are many invasive alien insect species, several of these species can affect human health by causing disease. These include vectors of vector-borne diseases, such as the triatomines that transmit Chagas disease. Triatoma longipennis Usinger (Hemiptera: Reduviidae) is one of the most epidemiologically important vector species of Trypanosoma cruzi Chagas (Trypanosomatida: Trypanosomatidae) in Mexico. This study reports the third collection of T. longipennis from the state of Hidalgo, Mexico, where the prevalence of T. cruzi infection in humans has historically exceeded 3.38%. This study eva

Biogeographical factors determining Triatoma recurva distribution in Chihuahua, México, 2014.

INTRODUCTION: Triatoma recurva is a Trypanosoma cruzi vector whose distribution and biological development are determined by factors that may influence the transmission of trypanosomiasis to humans. OBJECTIVE: To identify the potential spatial distribution of Triatoma recurve, as well as social factors determining its presence. MATERIALS AND METHODS: We used the MaxEnt software to construct ecological niche models while bioclimatic variables (WorldClim) were derived from the monthly values of temperature and precipitation to generate biologically significant variables. Complete Mitochondrial Genome of the Chagas Disease Vector, Triatoma rubrofasciata.

Triatoma rubrofasciata is a wide-spread vector of Chagas disease in Americas. In this study, we completed the mitochondrial genome sequencing of T. rubrofasciata. The total length of T. rubrofasciata mitochondrial genome was 17,150 bp with the base composition of 40.4% A, 11.6% G, 29.4% T and 18.6% C. It included 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and one control region. We constructed a phylogenetic tree on the 13 protein-coding genes of T. rubrofasciata and other 13 closely related species to show their phylogenic relationship. The determination of T. rubrofasciata mitogeno
