Rhizosphere phosphatases can exhibit hormetic effects in response to cadmium (Cd) ion stimulation. However, understanding the mechanisms underlying hormesis effects on soil ecosystems is challenging as studies on hormesis are usually specific to an organism, cell, or organ. To comprehensively investigate the mechanism of phosphatase hormesis, this study utilized in situ zymography and metabolomics to analyze the rhizosphere of Trifolium repens L. (white clover). Zymograms showed that rhizosphere phosphatase displayed a hormetic effect in 10 mg kg-1 Cd contaminated soil, with a hotspot area 1.8

A method based on matrix solid-phase dispersion (MSPD) has been developed for the determination of 12 isoflavones in Trifolium pratense L. Dried leaf samples were blended with C(18), placed in small columns and isoflavones extracted with dichloromethane-methanol. Analyses were performed by high performance liquid chromatography with diode array detection (HPLC-DAD) with 2-methoxyflavone as internal standard. Several dispersants, eluents and clean-up steps were tested during the optimization of the process in order to obtain the best selectivity and yields. Mean recoveries ranged from 70% to 11

Landfilling is common in developing countries since it is the easiest and cheapest way of waste disposal, however, it leads to serious environmental problems such as soil, water, and air pollution. A landfill has a life span of fifteen years after which it is closed leaving the site unusable, as a result, effective methods are needed for restoring and reclaiming the closed landfill site for future use. Phytoremediation has emerged as a viable and environmentally friendly method, which uses green plants to remove pollutants from soil, air, and water. In this study, Medicago sativa (alfalfa) and

Biorefining of grasses and legume forages is carried out commercially to recover feed proteins for monogastric animals. The biorefining produces a fiber-rich pulp byproduct with additional valorization potential. Here, we explored whether pulp products from several biorefined plants could exhibit bioactivity against animal parasites. Among the tested plants, extracts derived from red clover possessed exceptionally high bioactivity against Ascaris suum larvae. GC-MS analysis revealed the antiparasitic compounds to be phytoestrogens, especially biochanin A and to a lesser extent formononetin. Hi

Industrialization-driven surface ozone (O3) pollution significantly impairs plant growth. This study evaluates the effectiveness of exogenous protectants [3 mg L⁻1 abscisic acid (ABA), 400 mg L⁻1 ethylenediurea (EDU), and 80 mg L⁻1 spermidine (Spd)] on Trifolium repens subjected to O3 stress in open-top chambers, focusing on plant growth and dynamics of culturable endophytic fungal communities. Results indicate that O3 exposure adversely affects photosynthesis, reducing root biomass and altering root structure, which further impacts the ability of plant to absorb essential nutrients such as po

Species that repeatedly evolve phenotypic clines across environmental gradients have been highlighted as ideal systems for characterizing the genomic basis of local environmental adaptation. However, few studies have assessed the importance of observed phenotypic clines for local adaptation: conspicuous traits that vary clinally may not necessarily be the most critical in determining local fitness. The present study was designed to fill this gap, using a plant species characterized by repeatedly evolved adaptive phenotypic clines. White clover is naturally polymorphic for its chemical defence

PREMISE OF THE STUDY: The maintenance of adaptive polymorphisms within species requires fitness trade-offs reflecting selection for each morph. Cyanogenesis, the ability to produce hydrogen cyanide (HCN) after tissue damage, occurs in >3000 plant species and exists as a discrete polymorphism in white clover. This polymorphism is spatially distributed in recurrent clines, with higher frequencies of cyanogenic plants in warmer climates. The HCN autotoxicity hypothesis proposes that cyanogenic plants are selected against where frosts are common, as freezing liberates HCN and could impair cellular

White clover (Trifolium repens) is naturally polymorphic for cyanogenesis (hydrogen cyanide release following tissue damage). The ecological factors favouring cyanogenic and acyanogenic plants have been examined in numerous studies over the last half century, making this one of the best-documented examples of an adaptive polymorphism in plants. White clover cyanogenesis is controlled by two, independently segregating Mendelian genes: Ac/ac controls the presence/absence of cyanogenic glucosides; and Li/li controls the presence/absence of their hydrolysing enzyme, linamarase. In this study, we e

PREMISE: β-Cyanoalanine synthase (β-CAS) and alternative oxidase (AOX) play important roles in the ability of plants to detoxify and tolerate hydrogen cyanide (HCN). These functions are critical for all plants because HCN is produced at low levels during basic metabolic processes, and especially for cyanogenic species, which release high levels of HCN following tissue damage. However, expression of β-CAS and Aox genes has not been examined in cyanogenic species, nor compared between cyanogenic and acyanogenic genotypes within a species.
METHODS: We used a natural polymorphism for cyanogenesis

Mercury is a global pollutant in the modern world. There is a large number of areas in the world where mercury is present in soils in significant quantities. Remediation methods which have traditionally been proposed may pose a risk of secondary mercury contamination and/or adverse health effects for cleaners. Phytoextraction of heavy metals from the soil environment is currently considered one of the promising non-invasive methods of remediation. But this approach has limited effectiveness. Chemically induced phytoextraction can increase the efficiency of this process both by converting less

Glycyrrhetinic acid (GA) is a pharmacologically important oleanane-type triterpenoid best known from licorice, but its sustainable production is limited by inefficient enzymes. Here, we report the complete GA biosynthetic pathway in Trifolium repens, a legume not previously known to produce GA. Targeted metabolomics confirmed GA accumulation in multiple tissues, highest in roots. Transcriptomic analyses identified key biosynthetic genes, and heterologous reconstruction in yeast verified their catalytic functions. The pathway comprises three β-amyrin synthases, two β-amyrin 11-oxidases (TrCYP88

Understanding the molecular evolution of genes that underlie intraspecific polymorphisms can provide insights into the process of adaptive evolution. For adaptive polymorphisms characterized by gene presence/absence (P/A) variation, underlying loci commonly show signatures of long-term balancing selection, with gene-presence and gene-absence alleles maintained as two divergent lineages. We examined the molecular evolution of two unlinked P/A polymorphisms that underlie a well-documented adaptive polymorphism for cyanogenesis (hydrogen cyanide release with tissue damage) in white clover. Both c

Variation in cyanogenesis (hydrogen cyanide release following tissue damage) was first noted in populations of white clover more than a century ago, and subsequent decades of research have established this system as a classic example of an adaptive chemical defence polymorphism. Here, we document polymorphisms for cyanogenic components in several relatives of white clover, and we determine the molecular basis of this trans-specific adaptive variation. One hundred and thirty-nine plants, representing 13 of the 14 species within Trifolium section Trifoliastrum, plus additional species across the

Biorefining of grassland biomass (e.g., red clover) has attracted attention as a source of high-value protein that can serve as an alternative, more sustainable feed for livestock. In addition, extracts from red clover pulp (the byproduct of this process) have shown potent antiparasitic activity, suggesting an added valorization of the biorefining process through development of a dietary supplement that can potentially be used to control parasite infection. Here, we demonstrate that the biorefinery process disrupts plant cell walls, leading to an accumulation of the isoflavone biochanin A in t
