2,4,6-trinitrotoluene induces neurotoxicity by affecting the G protein pathways in Caenorhabditis elegans.

2,4,6-trinitrotoluene (TNT) is a chemical widely used to make explosives, and its use of residues can lead to potential threats to both ecosystems and human health. A thorough understanding of the various toxic effects of TNT is essential for developing effective environmental protection and health safety measures. Thus, we employed Caenorhabditis elegans (C. elegans), a typical model organism, to explore the neurotoxic effects of TNT and the signaling pathways involved. The results showed that neurotoxicity induced by 10-100 ng/mL TNT was manifested in reduced behavioral capacity (head thrash

Phototoxicology. 2. Near-ultraviolet light enhancement of Microtox assays of trinitrotoluene and aminodinitrotoluenes.

Coexposure of 2,4,6-trinitrotoluene (TNT), 2-amino-4,6-dinitrotoluene (2A), or 4-amino-2,6-dinitrotoluene (4A) to near-ultraviolet (nuv) light (lambda max-354 nm) significantly enhanced their toxicity toward Photobacterium phosphoreum (Microtox bioassay) during 30 min but not 15 min. Based on the slopes of the dose-response lines, the nuv coexposure and dark toxic mechanisms of action for TNT, 2A, and 4A appeared to be similar. nuv coexposure of binary mixtures significantly enhanced (supraadditivity) the toxicity of these compounds to P. phosphoreum. Under normal laboratory lighting, the toxi

Nanocomposite of europium organic framework and quantum dots for highly sensitive chemosensing of trinitrotoluene.

Luminescent metal-organic frameworks (MOFs) are considered as next-generation sensor materials for small molecules and explosives. In the present work, a nanocomposite of luminescent europium organic framework (EuOF) and CdSe quantum dots (QDs) has been first time investigated for photoluminescence (PL) based highly sensitive detection of trinitrotoluene (TNT). The nanocomposite EuOF/QD has been synthesized by initiating the growth of EuOF in the presence of QDs. The successful synthesis of the product has been verified with the help of electron microscopy, X-ray diffraction analysis, and surf

Alkaline hydrolysis/polymerization of 2,4,6-trinitrotoluene: characterization of products by 13C and 15N NMR.

Alkaline hydrolysis has been investigated as a nonbiological procedure for the destruction of 2,4,6-trinitrotoluene (TNT) in explosives contaminated soils and munitions scrap. Nucleophilic substitutions of the nitro and methyl groups of TNT by hydroxide ion are the initial steps in the alkaline degradation of TNT. Potential applications of the technique include both in situ surface liming and ex situ alkaline treatment of contaminated soils. A number of laboratory studies have reported the formation of an uncharacterized polymeric material upon prolonged treatment of TNT in base. As part of an

Highly-sensitive voltammetric detection of trinitrotoluene on reduced graphene oxide/carbon nanotube nanocomposite sensor.

This work presents the highly-sensitive detection of 2,4,6-trinitrotoluene (TNT) on reduced graphene oxide/multi-walled carbon nanotube (rGO/MWCNT) nanocomposite sensor. The formation of a thin film of this nanocomposite occurred at the cyclohexane/water immiscible interface of a mixture of MWCNT and rGO in the biphasic solution. The film was transferred to a boron-doped diamond (BDD) electrode for the square-wave voltammetric detection of TNT, which presented improved analytical characteristics in comparison with bare BDD and after modification with precursors. L-cysteine-capped CdTe QD-based sensor for simple and selective detection of trinitrotoluene.

Trinitrotoluene, usually known as TNT, is a kind of chemical explosive with hazardous and toxic effects on the environment and human health. National and societal security concerns have dictated an increasing need for the analytical detection of TNT with rapidity, high sensitivity and low cost. This work demonstrates a novel method using L-cysteine-capped CdTe quantum dots (QDs) to assay TNT, based on the formation of a Meisenheimer complex between TNT and cysteine. The fluorescence (FL) of quantum dots quench because electrons of the QDs transfer to the TNT molecules via the formation of a Me

A Simple Determination of Trinitrotoluene (TNT) Based on Fluorescence Quenching of Rhodamine 110 with FRET Mechanism.

Sensitive and selective detection of nitroaromatic explosives is an important issue in regard to human health, environment, public security and military issues. In this study, a simple and sensitive fluorescence quenching - based assay utilizing Rhodamine 110 as fluorophore probe was developed for the determination of trinitrotoluene (TNT). This sensitive fluorometric method could measure the decrease in fluorescence of Rhodamine 110 (λex = 490 nm, λem = 521 nm) owing to the primary amine groups of Rhodamine 110 (different from other rhodamines) capable of donor-acceptor interaction with TNT.

Fast Detection of 2,4,6-Trinitrotoluene (TNT) at ppt Level by a Laser-Induced Immunofluorometric Biosensor.

The illegal use of explosives by terrorists and other criminals is an increasing issue in public spaces, such as airports, railway stations, highways, sports venues, theaters, and other large buildings. Security in these environments can be achieved by different means, including the installation of scanners and other analytical devices to detect ultra-small traces of explosives in a very short time-frame to be able to take action as early as possible to prevent the detonation of such devices. Unfortunately, an ideal explosive detection system still does not exist, which means that a compromise

A theoretical insight into the reaction mechanisms of a 2,4,6-trinitrotoluene nitroso metabolite with thiols for toxic effects.

2,4,6-Trinitrotoluene (TNT) is a class C carcinogen as rated by the Environmental Protection Agency. One of the toxicity mechanisms of TNT is the covalent binding of its metabolites to critical proteins. However, knowledge about their molecular reaction mechanisms is scarce. Herein, we have provided density functional theory (DFT) simulation evidences for the reaction mechanisms of the nitroso metabolite of TNT with the sulfhydryl group of model thiols for the first time. The results show that the solvent-mediated proton-transfer mechanism plays a significant role in the entire process. For th

A study on the determination of the metabolites of 2,4,6-Trinitrotoluene using a dual-drift tube ion mobility spectrometer.

2,4,6-Trinitrotoluene (TNT) and its four metabolites, namely 2-ADNT, 4-ADNT, 2,4-DANT, and 2,6-DANT, are highly toxic substances. These metabolites also serve as biomarkers for assessing the health of individuals exposed to TNT. In this study, a homemade DDT-IMS apparatus was utilized to detect these metabolites. Under negative detection mode, the drift times of 2-ADNT and 4-ADNT showed subtle shifts within a drift tube temperature range of 100 °C-120 °C, aiding in their differentiation. In positive detection mode for 2,4-DANT and 2,6-DANT, significant variations were observed in both the numb

Acute toxicity of 2,4,6-trinitrotoluene, 2,4-dinitrotoluene, and 2,6-dinitrotoluene in the adult bullfrog (Lithobates catesbeiana).

2,4,6-Trinitrotoluene (TNT) is one of the most prevalent high explosives in the environment. 2,4-Dinitrotoluene (2,4-DNT) and 2,6-dinitrotoluene (2,6-DNT) are the most common isoforms of dinitrotoluene. The goal of this study was to determine the acute toxic effects of TNT, 2,4-DNT, and 2,6-DNT in adult male bullfrogs. The LD(50) for TNT was 1,060 mg/kg BW while the LD(50 )for 2,4-DNT and 2,6-DNT was 1,098 mg/kg BW. All three compounds elicited similar symptoms of toxicity including changes of skin color, body weight, development of seizures, liver and kidney necrosis, and lung cyanosis. Relat

Cytotoxicity and mutagenicity of a 2,4,6-trinitrotoluene (TNT) and hexogen contaminated soil in S. typhimurium and mammalian cells.

The toxicity and mutagenicity of aqueous and organic extracts of soil contaminated with TNT, TNT metabolites and hexogen was determined in mammalian cell lines and in prokaryotic cells. The prokaryotic toxicity was determined via the colony forming ability of Salmonella typhimurium (strains TA 98 and TA 100). The same strains were used to test mutagenicity in the Ames test. The mammalian toxicity was analyzed in human fibroblasts by the inhibition of cell growth and cell viability (MTT assay). The mammalian mutagenicity was tested with the HPRT test in V79 cells (hamster lung). The aqueous soi

Phototoxicology. 3. Comparative toxicity of trinitrotoluene and aminodinitrotoluenes to Daphnia magna, Dugesia dorotocephala, and sheep erythrocytes.

2,4,6-Trinitrotoluene (TNT) and compounds associated with its production are toxic and phototoxic to a wide range of biota. The planarian Dugesia dorotocephala, but not Daphnia magna, metabolized TNT (1 mg/liter) to 4-amino-2,6-dinitrotoluene (4A; 0.4 mg/liter) and 2-amino-4,6-dinitrotoluene (2A; 0.2 mg/liter). Coexposure to near-ultraviolet (nuv) light enhanced the toxicity of 2A more than that of TNT and 4A. The toxicities of TNT, 4A, and 2A to Du. dorotocephala were all decreased by glutathione (GSH) conjugation. This suggests that all had mechanisms of toxic action involving formation of q

Ultra-sensitive gas phase detection of 2,4,6-trinitrotoluene by non-covalently functionalized graphene field effect transistors.

The high energy density (4.2 MJ kg-1) and low vapour pressure (7.2 × 10-9 atm) of chemical explosives such as TNT (2,4,6-trinitrotoluene) pose a grave security risk demanding immediate attention. Detection of such hazardous and highly challenging chemicals demands specific, ultra-sensitive and rapid detection platforms that can concomitantly transduce the signal as an electrical readout. Although chemo-sensitive strategies have been investigated, the majority of them are restricted to detecting TNT from solutions and are therefore not implementable in real-time, on-field situations. Ultrasensitive SERS detection of trinitrotoluene through capillarity-constructed reversible hot spots based on ZnO-Ag nanorod hybrids.

A simple and efficient self-approach strategy was used to apply ultrasensitivity and self-revive ZnO-Ag hybrid surface-enhanced Raman scattering (SERS) sensors for the highly sensitive and selective detection of explosive TNT in both solution and vapour conditions. The good ultrasensitive sensing performance is a result of the abundant Raman hot spots, which were spontaneously formed in a reversible way by the self-approaching of flexible ZnO-Ag hybrid nanorods driven by the capillary force of solvent evaporation. Moreover, the enhancement effect was repeatedly renewed by the reconstruction of

Mechanisms and kinetics of alkaline hydrolysis of the energetic nitroaromatic compounds 2,4,6-trinitrotoluene (TNT) and 2,4-dinitroanisole (DNAN).

The environmental impacts of energetic compounds can be minimized through the design and selection of new energetic materials with favorable fate properties. Building predictive models to inform this process, however, is difficult because of uncertainties and complexities in some major fate-determining transformation reactions such as the alkaline hydrolysis of energetic nitroaromatic compounds (NACs). Prior work on the mechanisms of the reaction between NACs and OH(-) has yielded inconsistent results. In this study, the alkaline hydrolysis of 2,4,6-trinitrotoluene (TNT) and 2,4-dinitroanisole

Extremely sensitive and selective antibodies against the explosive 2,4,6-trinitrotoluene by rational design of a structurally optimized hapten.

Antibodies are a promising tool for the fast and selective trace detection of explosives. Unfortunately, the production of high-quality antibodies is not trivial and often expensive. Therefore, excellent antibodies are a rare and limiting resource in fields such as biosensing, environmental analysis, diagnostics, cancer therapy, and proteomics. Here, we report the synthesis, bioconjugation, and application of the structurally optimized hapten 6-(2,4,6-trinitro)-phenylhexanoic acid to improve the selectivity and sensitivity of antibodies for the detection of one of the most important explosives
