Trypanosomes are protozoan parasites that cause infectious diseases, including African trypanosomiasis (sleeping sickness) in humans and nagana in economically important livestock1,2. An effective vaccine against trypanosomes would be an important control tool, but the parasite has evolved sophisticated immunoprotective mechanisms-including antigenic variation3-that present an apparently insurmountable barrier to vaccination. Here we show, using a systematic genome-led vaccinology approach and a mouse model of Trypanosoma vivax infection4, that protective invariant subunit vaccine antigens can

In this study, we reported the first outbreak of the infection by Trypanosoma vivax in horses in southern Brazil, a non-endemic region where bovines have only recently been found infected by this trypanosome species. We evaluated 12 horses from a farm in southern Brazil, where four horses displayed pale mucous membranes, fever, weight loss, and swelling of abdomen, prepuce, or vulva. The diagnosis of T. vivax was confirmed in four horses by morphological parameters of trypomastigotes in blood smears and species-specific PCR. All T. vivax-infected animals showed anemia, and most showed increase

About 3,000 European and Zebu cattle sera from various rearing areas of French Guiana were examined by enzyme-linked immunosorbent assay (ELISA) for detection of specific circulating antigens of Trypanosoma vivax and for detection of antibodies to Trypanosoma species with reagents provided by ILRAD. For the whole country, the seroprevalence rate is 29%. Infection was detected everywhere in French Guiana, but seroprevalence rates were different from place to place, depending on the epidemiological situation, the abundance of vectors (stable-flies and horse-flies) and the management. We could ge

Trypanosoma vivax isolated from dairy cattle undergoing a haemorrhagic disease was inoculated into Ayrshire steers. Five of six infected animals experienced brief periods of diarrhoea and sublingual and gastro-intestinal haemorrhage. Gastro-intestinal bleeding coincided with markedly reduced numbers of thrombocytes and a high level of parasitaemia in the peripheral blood. Prothrombin times were extended and fibrinogen levels were elevated in infected animals. Plasma paracoagulation tests were positive for the presence of fibrin monomers and/or clottable early fibrin degradation products during

African animal trypanosomosis (AAT) is caused by the tsetse fly-transmitted protozoans Trypanosoma congolense and T. vivax and leads to huge agricultural losses throughout sub-Saharan Africa. Three drugs are available to treat nagana in cattle (diminazene diaceturate, homidium chloride, and isometamidium chloride). With increasing reports of drug-resistant populations, new molecules should be investigated as potential candidates to combat nagana. Dicationic compounds have been demonstrated to have excellent efficacy against different kinetoplastid parasites. This study therefore evaluated the

BACKGROUND: This study was conducted in response to recurring reports from eastern Sudan of camel trypanosomosis that can no longer be treated by currently available trypanocidal drugs. One hundred and eighty-nine blood samples were obtained from camels in different herds and local markets in the western part of Sudan, and a cross-sectional study was carried out between December 2015 and February 2016 to identify the causative agents and possible circulating genotypes.
RESULTS: The prevalence of trypanosomes detected using the conventional parasitological techniques of Giemsa-stained blood sm

This paper reports the first occurrence of bovine trypanosomosis due to Trypanosoma vivax in Bolivia. T. vivax was identified in thin blood smears of 159 cattle from the Provinces of Velaco (57), Nuflo de Chavez (20), Guayaros (30) and Chiquitos (52), and in 86.20% of 29 cattle from Laguna Concepción examined by microhematocrit test. The clinical signs observed were fever, anemia, abortion, progressive weakness, loss of appetite, lethargy, substantial weight loss in a relatively short time, and progressive emaciation.

The haematocrit centrifuge technique (HCT), or Woo test, is the technique most commonly used for diagnosis of animal trypanosomosis, but its sensitivity is not well defined. Depending on the authors, the sensitivity of the Murray technique (dark' ground buffy coat method, or DG/BCM) is higher, equal or lower than that of the Woo test for the detection of Trypanosoma vivax. Most authors have compared particular techniques relatively to other techniques, rather than attempting to measure the sensitivity of the tests in relation to a fixed reference. The relative sensitivity of particular tests a

The course of experimental Trypanosoma vivax infection in eight Uda rams was studied. All the infected animals became parasitaemic 2 days post-inoculation and remained so throughout the study period. A three-phase disease pattern was recognized, i.e. acute, subacute and chronic stages lasting 17-85 days. The disease was characterized by fever and a terminal decrease in rectal temperature despite an increase in parasitaemia with time for rams with acute and subacute infections. Mean weight loss was most marked in subacute followed by chronic cases. Gross pathological changes observed in some in

African trypanosomes (Trypanosoma) are vector-borne haemoparasites that survive in the vertebrate bloodstream through antigenic variation of their Variant Surface Glycoprotein (VSG). Recombination, or rather segmented gene conversion, is fundamental in Trypanosoma brucei for both VSG gene switching and for generating antigenic diversity during infections. Trypanosoma vivax is a related, livestock pathogen whose VSG lack structures that facilitate gene conversion in T. brucei and mechanisms underlying its antigenic diversity are poorly understood. Here we show that species-wide VSG repertoire i

It is widely held that cyclical development of Trypanosoma vivax in Glossina is confined to the proboscis. This view has been re-examined in a series of experiments. Teneral G. morsitans centralis were fed on a goat infected with T. vivax IL 1392 and dissected 1-2 h after feeding. The infection rates in the labrum and hypopharynx were 40% and 0%, in contrast to 82% and 58%, respectively, observed in a control group dissected on day 25. This suggested that in a significant number of tsetse, cyclical development of T. vivax was initiated at sites other than the proboscis. Subsequent experiments

Trypanosoma vivax produced a progressive macrocytic normochromic anaemia in sheep during the acute phase of infection. Reticulocytes were absent from the blood of healthy sheep and of sheep with T. vivax-induced anaemia. However, anaemia induced artificially (AHA) in sheep by in vitro heat treatment of red cells, which was comparable in classification and degree to the anaemia of T. vivax infection, produced a reticulocytosis of 1.5 +/- 1.0 per cent. When plasma from the anaemic blood of sheep infected with T. vivax for two and four weeks was inoculated subcutaneously into mice, the reticulocy

Trypomastigotes attached to the wall of the hypopharynx in tsetse flies infected with Trypanosoma vivax are believed to represent the true metacyclic stage of this trypanosome. Electron microscopy demonstrates that attachment is mediated by hemidesmosome-like junctions along the flagellar membrane and that none of the trypomastigotes, either attached or free in the hypopharynx lumen, possesses a surface coat comparable with that on the metacyclics of T. brucei and T. congolense and on the bloodstream stages of all salivarian trypanosomes. As the variable antigen of bloodstream and metacyclic T

Sera from goats and cattle that were infected with two Trypanosoma vivax clones (ILDat 1.2 and ILDat 2.1) derived from different stocks were analysed for antibody activity against the variable surface glycoproteins (VSGs) of the infecting clones by enzyme-linked immune assays (ELISA) and immune lysis. To obtain purified VSG, lysed trypanosomes were separated on dodecyl sulphate-polyacrylamide gels. The gels were copper stained and the VSG protein band was excised from the gel. After destaining, the proteins were electroeluted from the gel slices and used as antigens in ELISA. High titres of Ig

Infection of Ayrshire cattle with a stock of Trypanosoma vivax from the Galana Ranch, Kenya, resulted in an acute disease characterised by profound anaemia and haemorrhage, which reached maximum severity between 3 and 5 weeks after infection. Bleeding from the ears, nose and rectum occurred. At necropsy, petechial and ecchymotic haemorrhages were widespread, but were particularly severe in the gastrointestinal tract. In confirmation of the gross findings, congestion, haemorrhage and degenerative changes in most tissues and organs were found histologically. Thrombi were seen in the lymphatic ve
