The effect of treating cattle with deltamethrin to control tsetse flies and ticks was investigated on 2 ranches 8 km apart in central Uganda where there is a high risk of trypanosomosis. This area had a moderate challenge of Glossina pallidipes. The cattle had very low challenge of Rhipicephalus appendiculatus, Amblyomma variegatum, Boophilus decolaratus and Rhipicephalus evertsi evertsi, due to regular treatment with dioxathion. On one ranch a dip was charged with deltamethrin to treat cattle regularly for 3 months. The other ranch was used as a control for the tsetse population, but the anim

Survival in and colonization of the tsetse fly midgut are essential steps in the transmission of many species of African trypanosomes. In the fly, bloodstream trypanosomes transform into the procyclic stage within the gut lumen and later migrate to the ectoperitrophic space, where they multiply, establishing an infection. Progression of the parasite infection in the fly depends on factors inherent to the biology of trypanosomes, tsetse, and the bloodmeal. Flies usually eradicate infection early on with both pre-existing and inducible factors. Parasites, in contrast, respond to these stimuli by

Tsetse flies (Glossina spp.), the vector for African trypanosomiasis, are highly attracted by blue and black surfaces. This phototactic behaviour has long been exploited to trap tsetse flies as one measure in the control of African trypanosomiasis. However, why blue and black are so attractive for tsetse flies is still unknown. We propose that the combination of blue and black is attractive for many Glossina species because when searching for a shady resting place to pass the day, the flies are probably guided by the blueness and darkness of daytime shadows. In contrast to people's experience

The tsetse fly Glossina pallidipes, the major vector of the parasite that causes animal African trypanosomiasis in Kenya, has been subject to intense control measures with only limited success. The G. pallidipes population dynamics and dispersal patterns that underlie limited success in vector control campaigns remain unresolved, and knowledge on genetic connectivity can provide insights, and thereby improve control and monitoring efforts. We therefore investigated the population structure and estimated migration and demographic parameters in G. pallidipes using genotypic data from 11 microsat

Human African Trypanosomiasis (HAT) transmitted by the tsetse fly continues to be a public health issue, despite more than a century of research. There are two types of the disease, the chronic gambiense and the acute rhodesiense-HAT. Fly abundance and distribution have been affected by changes in land-use patterns and climate. However, disease transmission still continues. Here, we review some aspects of HAT ecoepidemiology in the context of altered infestation patterns and maintenance of the transmission cycle as well as emerging options in disease and vector control.

Trypanosome infections identified by polymerase chain reaction on field-caught tsetse flies from various locations were analyzed with respect to factors intrinsic and extrinsic to the trypanosome-tsetse association. These factors were then simultaneously analyzed using artificial neural networks (ANNs) and the important factors were identified to predict and explain the presence of trypanosomes in tsetse. Among 4 trypanosome subgroups (Trypanosoma brucei s.l., T. congolense of the 'savannah' and of the 'riverine-forest' types, and T. simiae), the presence of the 2 types of T. congolense was pr

BACKGROUND: With the largest cattle population in Africa and vast swathes of fertile lands infested by tsetse flies, trypanosomosis is a major challenge for Ethiopian farmers. Managing the problem strategically and rationally requires comprehensive and detailed information on disease and vector distribution at the national level. To this end, the National Institute for Control and Eradication of Tsetse and Trypanosomosis (NICETT) developed a national atlas of tsetse and African animal trypanosomosis (AAT) for Ethiopia.
METHODS: This first edition of the atlas focused on the tsetse-infested ar

Tsetse flies have a highly regulated and defined microbial fauna made of 3 bacterial symbionts (obligate Wigglesworthia glossinidia, commensal Sodalis glossinidius and parasitic Wolbachia pipientis) in addition to a DNA virus (Glossina pallidipes Salivary gland Hypertrophy Virus, GpSGHV). It has been possible to rear flies in the absence of either Wigglesworthia or in totally aposymbiotic state by dietary supplementation of tsetse's bloodmeal. In the absence of Wigglesworthia, tsetse females are sterile, and adult progeny are immune compromised. The functional contributions for Sodalist are le

Tsetse flies (Diptera: Glossinidae: Glossina spp.) are vectors for African trypanosomiasis, a devastating disease that kills both people and animals in sub-Saharan Africa. Trypanosomes ingested with an infected bloodmeal reside within the gut of tsetse and eventually move to the salivary glands where they become transmissible during blood feeding. Although tsetses are efficient vectors for disease transmission, infection prevalence in the field is surprisingly low, a trait referred to as refractoriness. Refractoriness is relatively more pronounced in palpalis subgroup flies, although certain s

Salivarian trypanosomes pose a substantial threat to livestock, but their full diversity is not known. To survey trypanosomes carried by tsetse in Tanzania, DNA samples from infected proboscides of Glossina pallidipes and G. swynnertoni were identified using fluorescent fragment length barcoding (FFLB), which discriminates species by size polymorphisms in multiple regions of the ribosomal RNA locus. FFLB identified the trypanosomes in 65 of 105 (61.9%) infected proboscides, revealing 9 mixed infections. Of 7 different FFLB profiles, 2 were similar but not identical to reference West African Tr

The prevalence of trypanosome infections in tsetse flies in the Chiawa area of Lower Zambezi in Zambia, with endemic trypanosomosis, was determined by a polymerase chain reaction (PCR) method that allowed the detection of trypanosome DNA and determination of the type of animal host fed on by the tsetse fly Glossina pallidipes, using tsetse-derived DNA extracts as templates. Ninety G. pallidipes (82 females and 8 males; 18.3%) of the 492 flies captured by baited biconical traps tested positive for the presence of Trypanosoma brucei species genomic DNA. Of the 90 T. brucei-positive flies, 47 (52

The sterile insect technique (SIT) is an environment friendly and sustainable method to manage insect pests of economic importance through successive releases of sterile irradiated males of the targeted species to a defined area. A mating of a sterile male with a virgin wild female will result in no offspring, and ultimately lead to the suppression or eradication of the targeted population. Tsetse flies, vectors of African Trypanosoma, have a highly regulated and defined microbial fauna composed of three bacterial symbionts that may have a role to play in the establishment of Trypanosoma infec

Tsetse flies are vectors of parasites that cause diseases responsible for significant economic losses and health issues in sub-Saharan Africa, including sleeping sickness in humans and nagana in domestic animals. Efficient vector-control campaigns require good knowledge of the demographic parameters of the targeted populations. In the last decade, population genetics emerged as a convenient way to measure population densities and dispersal in tsetse flies. Here, by revealing a strong negative density-dependent dispersal in two dimensions, we suggest that control campaigns might unleash dispers

BACKGROUND: Tsetse flies occur in much of sub-Saharan Africa where they transmit the trypanosomes that cause the diseases of sleeping sickness in humans and nagana in livestock. One of the most economical and effective methods of tsetse control is the use of insecticide-treated screens, called targets, that simulate hosts. Targets have been ~1 m2, but recently it was shown that those tsetse that occupy riverine situations, and which are the main vectors of sleeping sickness, respond well to targets only ~0.06 m2. The cheapness of these tiny targets suggests the need to reconsider what intensit

BACKGROUND: Here we set out to standardize long-lasting, visually-attractive devices for Glossina swynnertoni, a vector of both human and animal trypanosomiasis in open savannah in Tanzania and Kenya, and in neighbouring conservation areas used by pastoralists. The goal was to determine the most practical device/material that would induce the strongest landing response in G. swynnertoni for use in area-wide population suppression of this fly with insecticide-impregnated devices.
METHODS AND FINDINGS: Trials were conducted in wet and dry seasons in the Serengeti and Maasai Mara to measure the

An outbreak of human trypanosomiasis due to Trypanosoma brucei rhodesiense has been affecting the Busoga district of Uganda since 1976. More than 40,000 cases have been recorded up to 1990. Since two years the epidemic area has been extending to the Tororo district. The vector is Glossina fuscipes. In order to stop the disease transmission a vector control project was launched in 1988 in Busoga area. It is based on tsetse fly trapping, using pyramidal optic traps impregnated with deltamethrin (10 traps per square kilometer). The results were excellent. Everywhere fly populations were reduced b

Spatio-temporally heterogeneous environments may lead to unexpected population dynamics. Knowledge is needed on local properties favouring population resilience at large scale. For pathogen vectors, such as tsetse flies transmitting human and animal African trypanosomosis, this is crucial to target management strategies. We developed a mechanistic spatio-temporal model of the age-structured population dynamics of tsetse flies, parametrized with field and laboratory data. It accounts for density- and temperature-dependence. The studied environment is heterogeneous, fragmented and dispersal is s

Tsetse flies are the vectors of African trypanosomiasis affecting 36 sub-Saharan countries. Both wild and domestic animals play a crucial role in maintaining the disease-causing parasites (trypanosomes). Thus, the identification of animal reservoirs of trypanosomes is vital for the effective control of African trypanosomiasis. Additionally, the biotic and abiotic factors that drive gut microbiome diversity in tsetse flies are primarily unresolved, especially under natural, field conditions. In this study, we present a comprehensive DNA metabarcoding approach for individual tsetse fly analysis
