This paper examines UK farmers' levels of confidence in vaccinating badgers against bovine tuberculosis (bTB) and their trust in the Government's ability to deal with bTB. In 2010, a badger vaccine based on the BCG vaccine was licensed following field trials and used as part of the UK Government's Badger Vaccination Deployment Project. A stratified random sample of cattle farmers in five different locations of England was surveyed using a telephone survey to elicit their views of badger vaccination. The survey provided a total of 341 responses with a response rate of 80 per cent. Results sugge

The development of vaccines and effective diagnostic methods for bovine tuberculosis requires an understanding of the immune response against its causative agent, Mycobacterium bovis. Although this disease is primarily investigated and diagnosed through the assessment of cell-mediated immunity, the role of B cells and antibodies in bovine tuberculosis has been relatively undervalued and understudied. Current evidence indicates that circulating M. bovis-specific antibodies are not effective in controlling the disease. However, local humoral immune responses may contribute to either defence or p

Despite the compulsory test and slaughter campaigns in cattle, bovine tuberculosis (bTB) is still present in Spain, and the role of wildlife reservoirs is increasingly recognized. We provide an update on recent progress made in bTB control in Spanish wildlife, including aspects of epidemiology, surveillance, host-pathogen interaction and wildlife vaccination. At the high densities and in the particular circumstances of Mediterranean environments, wild ungulates, mainly Eurasian wild boar and red deer, are able to maintain Mycobacterium bovis circulation even in absence of domestic livestock. To evaluate the potential of Mycobacterium tuberculosis Rv0394c protein for use in the diagnosis of bovine tuberculosis, the Rv0394c protein was expressed and purified via a prokaryotic expression system. Its immunogenicity was assessed in mouse and guinea pig models, and its stimulatory effect in peripheral blood IFN-γ release assays was analyzed. The gene fragment of Rv0394c was introduced into the prokaryotic expression vector pColdI via homologous recombination to construct the recombinant plasmid pColdI-Rv0394c, which was transformed into Escherichia coli BL21(DE3) for induced expression.

Tuberculosis (TB), caused by members of the Mycobacterium tuberculosis complex (MTBC), remains a significant global health concern. While zoonotic transmission of M. bovis from cattle to humans is well documented, reverse zoonotic transmission of M. tuberculosis from humans to cattle has received far less attention. This review provides the first comprehensive synthesis of M. tuberculosis infections in cattle, drawing on evidence from farms, households, and slaughterhouses where human-animal contact is most intense. Available data indicate that such spillover events are uncommon compared with

The use of a bacillus Calmette-Guerin (BCG)-based vaccine could represent a viable strategy for controlling bovine tuberculosis (TB), principally in those cases where a wildlife disease vector exists. This article focuses on recent progress in animal TB vaccinology, outlining that oral-route vaccination represents the most feasible means of distributing a vaccine to control disease in wildlife. Drawing on historical successes of previous wildlife vaccination programs, the article suggests how, and in what form, an oral-delivery BCG-based vaccine might become operational, considering the wide d

Coinfections can alter the adaptive immune responses to bovine tuberculosis (bTB) and hinder accurate diagnosis; however, their effect on host immunity to Mycobacterium bovis and disease progression is not well understood. In this study, we assessed the impact of natural coinfection with bovine leukemia virus (BLV) on immune responses to M. bovis. Among 108 cattle examined, those coinfected with BLV showed a significantly higher intragranulomatous mycobacterial burden and more severe pathological lesions compared to animals infected with M. bovis alone. Additionally, coinfected animals demonst

BACKGROUND: Bovine tuberculosis (bTB) is the most important animal health concern in Wales. Annual testing across all cattle herds in Wales commenced in 2010. In 2017, a new geographic division of Wales was conceived, with bespoke cattle controls and eradication milestones reflecting the geographical heterogeneity of bTB distribution.
METHODS: This observational study uses descriptive analysis and Poisson regression modelling to analyse bTB surveillance data from all herds in Wales.
RESULTS: Since 2010, there has been a significant decrease (p < 0.0001) in bTB incidence (8.6%‒6.3%) and plate

The emergence of wildlife reservoirs of Mycobacterium bovis infection in cattle as well as increased inter-regional trade with associated spread of M. bovis has led to renewed interest in the use of vaccines for the control of bovine tuberculosis (TB). Field efficacy trials performed in the early 20th century demonstrated the partial effectiveness of bacilli Calmette-Guerin (BCG) for the control of bovine TB. Recent experimental trials with cattle have demonstrated that: (1) subunit vaccines may boost immunity elicited by BCG in cattle, (2) T cell central memory immune responses evoked by prot

Bovine tuberculosis, caused by Mycobacterium bovis, is rapidly increasing in cattle herds in developed countries such as the UK, New Zealand and the USA. In addition, persistence of M. bovis in other parts of the world may account for up to 10% of cases of human tuberculosis. Thus, a rise in the number of M. bovis infections poses an increased human health risk and is also a major economic problem. In the UK, the incidence of bovine tuberculosis continues to rise despite the use of a skin test and slaughter control policy, highlighting the need for an effective vaccination strategy to control

Understanding the gross lesion distribution associated with bovine tuberculosis (bTB) and the relationship between antemortem test results is important for optimizing surveillance activities. Antemortem diagnostic test results and postmortem examination results from animals infected with bTB in the United States are routinely collected during surveillance and outbreak response. From 2017 to 2022, data were compiled and analyzed from 403 infected animals, representing both beef and dairy cattle from a variety of herds with different levels of disease prevalence. Overall, 95.3% of cattle infecte

The aim of this study was to obtain a contemporary data set of pathology in tuberculin reactor and in-contact cattle in England and Wales. Four hundred animals (200 reactors and 200 in-contacts) from 242 farms located in 14 counties in Western England and Wales were examined. The mean number of lymph nodes (LNs) with tuberculosis (TB)-like lesions per TB-confirmed animal was 1.7 in reactors and 1.5 in in-contact animals. Tuberculous lesions in both reactor and in-contact animals were most commonly observed in the LNs of the thorax, followed by the head and abdomen, particularly the mediastinal

Bovine tuberculosis (TB) is a disease characterised by progressive development of specific granulomatous lesions or tubercles in lung tissue, lymph nodes or other organs. Mycobacterium bovis is the causative agent of the disease. Bovine species, including bison and buffaloes, are susceptible to the disease, but nearly all warm-blooded animals can be affected. All species are not equally susceptible to the disease; some are spill-over (end) hosts and others maintenance hosts. In Africa, bovine TB primarily affects cattle; however, infection in other farm and domestic animals, such as sheep, goa

We consider a model for the spread of bovine tuberculosis in herds comprising three species (bovids, caprids and equids) in Chad. The epidemiological model is built on top of a classic Lotka-Volterra competition model, which is exploited in a regime where stable coexistence of the three species holds. The epidemiological model itself is an SLI model, because of the absence of treatment for herds in the area. After studying some mathematical properties of the model, we perform a short computational analysis, investigating sensitivity of the model and comparing solutions with and without competi

Bovine tuberculosis (bTb) is endemic in Bangladesh but the true prevalence has not yet been reported. Our objectives for this study were to determine the true prevalence and identify risk factors for bTb at the animal- and herd-level in Bangladesh. A total of 510 cows were randomly selected during January 2018 to December 2018. Caudal fold (CFT) and comparative cervical tuberculin tests (CCT) were serially interpreted. Animal- and herd-level risk factor data were collected using a pre-tested questionnaire. The hierarchical true prevalence of bTb was estimated within a Bayesian framework. The h

Bovine tuberculosis (bTB) is a major zoonotic disease of cattle that is endemic in much of the world, limiting livestock productivity and representing a global public health threat. Because the standard tuberculin skin test precludes implementation of Bacille Calmette-Guérin (BCG) vaccine-based control programs, we here developed and evaluated a novel peptide-based defined antigen skin test (DST) to diagnose bTB and to differentiate infected from vaccinated animals (DIVA). The results, in laboratory assays and in experimentally or naturally infected animals, demonstrate that the peptide-based

Mycobacterium bovis, the causative agent of bovine tuberculosis (bTB), poses an economic risk to the United States (U.S.) cattle industry. Although management has reduced herd-level prevalence, repeated spillback from wildlife reservoirs, such as white-tailed deer (Odocoileus virginianus), remains a major barrier to eradication. Here, we developed a stochastic simulation that incorporates seasonal changes in deer abundance (birth pulses and hunter harvest) and heterogeneity in contact behavior to predict bTB dynamics in wildlife. We then used this framework to identify mechanisms that shape bT
