MYC at the tumor-immune interface: mechanisms of immune escape and immunotherapy resistance.

Immunotherapies have transformed cancer treatment by harnessing the immune system to recognize and eliminate malignant cells, offering durable clinical benefit across diverse tumor types. Despite successes with immune checkpoint inhibitors (ICIs) and other strategies like cytokines, oncolytic viruses, cancer vaccines, bispecific antibodies, and adoptive cell therapies, substantial fractions of patients still fail to respond or develop resistance. The oncogene MYC, deregulated in ~70% of human cancers, has emerged as a central driver of immune evasion and a key contributor to immunotherapy fail

Adenosine signaling in tumor immune escape: metabolic checkpoints, myeloid suppression, and combination immunotherapy.

Adenosine is a central metabolic regulator of tumor immune escape, shaping the tumor microenvironment (TME) through suppression of effector lymphocytes and reprogramming of myeloid populations. Extracellular adenosine is generated primarily via the ectonucleotidases CD39 and CD73 and signals through A2A (A2AR) and A2B (A2BR) receptors on T cells, NK cells, tumor-associated macrophages (TAMs), myeloid-derived suppressor cells (MDSCs), and dendritic cells. This pathway promotes T-cell exhaustion, inhibits cytotoxicity, and enhances myeloid-driven immunosuppression, creating metabolic and spatial

Harnessing exosomes: From tumor immune escape to therapeutic innovation in gastric cancer immunotherapy.

Gastric cancer ranks fifth among the most prevalent cancers globally, with a dismal prognosis. In recent years, immunotherapy, particularly immune checkpoint inhibitors, has emerged as a glimmer of hope for advanced gastric cancer patients. However, not all patients can benefit from this treatment modality, as the tumor microenvironment significantly influences treatment efficacy. Exosomes, pivotal mediators of intercellular communication, exert intricate and diverse effects in shaping and regulating the tumor microenvironment. This review provides a comprehensive overview of the functional me

UTX is an escape from X-inactivation tumor-suppressor in B cell lymphoma.

To explain the excess cancer rate in males, several candidates for "escape from X-inactivation tumor-suppressor" (EXITS) were recently identified. In this report we provide direct experimental evidence supporting UTX's role as an EXITS gene. Using a mouse lymphoma model, we show clear dosage effect of UTX copy number during tumorigenesis, which strongly supports the EXITS theory. Importantly, UTX deletion not only accelerates lymphomagenesis, it also strongly promotes tumor progression. UTX-knockout tumors are more aggressive, showing enhanced brain dissemination and formation of blood vessels

Tumor-microenvironment-mediated immune escape drives trastuzumab resistance in HER2-positive gastric cancer.

The combination of trastuzumab and chemotherapy, with or without pembrolizumab, is the current first-line standard of care for patients with human epidermal growth factor receptor 2 (HER2)-positive advanced gastric cancer and gastroesophageal junction cancer. However, upon disease progression, subsequent therapies often yield limited clinical benefit. The mechanisms of drug resistance to trastuzumab remain unresolved. Current studies on its resistance mechanisms have largely focused on alterations in the HER2 pathway, including HER2 heterogeneity, reduced or absent HER2 expression, variations

The Upregulation of Molecules Related to Tumor Immune Escape in Human Pituitary Adenomas.

Human pituitary adenomas are one of the most common intracranial neoplasms. Although most of these tumors are benign and can be treated medically or by transsphenoidal surgery, a subset of these tumors are fast-growing, aggressive, recur, and remain a therapeutic dilemma. Because antibodies against immune checkpoint receptors PD-1 and CLTA-4 are now routinely used for cancer treatment, we quantified the expression of mRNA coding for PD-1, CLTA-4, and their ligands, PD-L1, PD-L2, CD80, and CD86 in human pituitary adenomas and normal pituitary glands, with the ultimate goal of exploiting immune

Exosomal PD-L1: New Insights Into Tumor Immune Escape Mechanisms and Therapeutic Strategies.

As a classical immune checkpoint molecule, PD-L1 on the surface of tumor cells plays a pivotal role in tumor immunosuppression, primarily by inhibiting the antitumor activities of T cells by binding to its receptor PD-1. PD-1/PD-L1 inhibitors have demonstrated unprecedented promise in treating various human cancers with impressive efficacy. However, a significant portion of cancer patients remains less responsive. Therefore, a better understanding of PD-L1-mediated immune escape is imperative. PD-L1 can be expressed on the surface of tumor cells, but it is also found to exist in extracellular

The soluble form of CD160 acts as a tumor mediator of immune escape in melanoma.

Melanoma is responsible for 90% of skin cancer-related deaths. Major therapeutic advances have led to a considerable improvement in the prognosis of patients, with the development of targeted therapies (BRAF or MEK inhibitors) and immunotherapy (anti-CTLA-4 or -PD-1 antibodies). However, the tumor constitutes an immunosuppressive microenvironment that prevents the therapeutic efficacy and/or promotes the development of secondary resistances. CD160 is an activating NK-cell receptor initially described as delineating the NK and CD8+ T-cell cytotoxic populations. Three forms of CD160 have been de

Mechanism of tumor cells escaping from immune surveillance of NK cells.

Natural killer (NK) cells play an important role in anti-tumor and anti-infection, and perform their immune surveillance function in various ways. However, no matter what kind of cancer, the functional activity of NK cells in the tumor microenvironment (TME) is suppressed. Understanding the relationship between tumor cells and NK cells is very critical for tumor immunotherapy. This review discusses the mechanism of tumor cells escaping the immune surveillance of NK cells. These include a variety of factors that inhibit the activity of NK cells, an imbalance of activating receptors and inhibiti

Involvement of HLA class I molecules in the immune escape of urologic tumors.

CONTEXT AND OBJECTIVE: To analyze the influence of different alterations in human leukocyte antigen class I molecules (HLA I) in renal cell carcinoma, as well as in bladder and prostate cancer. We also study the correlation between HLA I expression and the progression of the disease and the response after immunotherapy protocols. EVIDENCES ACQUISITION: It has been shown, experimentally, that the immune system can recognize and kill neoplastic cells. By analyzing the expression of HLA I molecules on the surface of cancer cells, we were able to study the tumor escape mechanisms against the immu

Tumor-infiltrating T lymphocytes: A promising immunotherapeutic target for preventing immune escape in cholangiocarcinoma.

Cholangiocarcinoma (CCA) is becoming more common and deadly worldwide. Tumor-infiltrating T cell subtypes make distinct contributions to the immune system; collectively, they constitute a significant portion of the tumor microenvironment (TME) in CCA. By secreting cytokines and other chemicals, regulatory T cells (Tregs) decrease activated T cell responses, acting as immunosuppressors. Reduced CD8+ T cell activation results in stimulating programmed death-1 (PD-1), which undermines the immunological homeostasis of T lymphocytes. On the other hand, cancer cells are eliminated by activated cytot

A lysosomal escape-enabled endoplasmic reticulum-targeting BODIPY photothermal agent for enhanced tumor ablation.

Photothermal therapy (PTT) has emerged as a promising minimally invasive strategy for tumor treatment due to its high selectivity, minimal drug resistance, and precise controllability. However, the efficacy of PTT is often limited by the inability of photothermal agents (PTAs) to effectively target subcellular organelles after cellular uptake. In this study, we designed and synthesized an endoplasmic reticulum (ER)-targeting BODIPY-based photothermal agent (ER-BDP) by functionalizing a trifluoromethyl-substituted BODIPY scaffold with a p-toluenesulfonyl group. The resulting ER-BDP nanoparticle

TCF3 safeguards tumor-intrinsic interferon competence to restrain prostate cancer progression and immune escape.

Prostate cancer commonly remains refractory to checkpoint blockade, consistent with an immune-cold microenvironment and tumor-intrinsic defects in interferon signaling. We report that loss of the basic helix-loop-helix transcription factor TCF3 defines a tumor-intrinsic mechanism that simultaneously drives aggressive progression and immune evasion. TCF3 expression is reduced in advanced and metastatic prostate cancer across multiple cohorts and in a Pten-driven model. In androgen-independent prostate cancer cells and organoids, TCF3 depletion enhances proliferation, clonogenicity, migration, a

Amino acid metabolic regulatory network in tumor immune escape: mechanistic exploration and intervention directions.

Tumor immune escape is not determined solely by immune checkpoints, suppressive cytokines, or changes in immune-cell composition; it is also organized by the metabolic architecture of the tumor microenvironment. Reprogrammed amino acid metabolism contributes to this process by redistributing nutrients, generating immunoregulatory metabolites, and reshaping immune-cell states. Rather than viewing individual amino acid pathways as independent mechanisms, this review proposes a network framework in which nutrient competition, metabolite-mediated communication, and immune-state stabilization are m

Bibliometric and visualized analysis of global distribution and research frontiers in tumor immune escape.

BACKGROUND: Tumor cells employ various mechanisms to evade detection and attack by the immune system, a phenomenon known as tumor immune escape, which represents a significant target for immunotherapy. Both primary and secondary immune escape mechanisms pose substantial challenges that hinder the efficacy of immunotherapy. This study aims to systematically examine the knowledge structure, hotspot frontiers, emerging trends, and future directions in the field of tumor immune escape through the application of bibliometric methods and knowledge mapping analysis. METHODS: A comprehensive search o

Lung cancer tumor immune microenvironment: analyzing immune escape mechanisms and exploring emerging therapeutic targets.

Lung cancer is the most common malignant tumor in the world. Presently, there are still problems, including a high recurrence rate, resistance, and serious toxic side effects, even if conventional treatments like chemotherapy, radiotherapy, and targeted therapy have somewhat improved patient survival. Even though immune checkpoint inhibitors that target programmed cell death-1/programmed cell death ligand 1 have fundamentally altered the therapeutic paradigm, the core mechanism is strongly linked to tumor immune escape, and some patients continue to have poor response rates or treatment resist

Enhancement of Ferroptosis in Escape Variant Tumor Cells by IFNγ Derived from Antigen-Specific T Cells Controls Tumor with Heterogeneity.

Tumor masses often exhibit heterogeneity, including escape variant clones that lack antigen-presenting machinery and/or tumor antigens, which poses a major challenge to immunotherapy. Ferroptosis, a form of regulated cell death driven by iron-dependent lipid peroxidation, has been shown to effectively induce cell death in various tumor cells. Recent studies have reported that IFNγ suppresses the expression of system Xc-, thereby enhancing the induction of ferroptosis. Based on this, we hypothesized that combining immunotherapy with ferroptosis inducers could enhance antitumor effects against b

Targeting Intercellular Communication in the Bone Microenvironment to Prevent Disseminated Tumor Cell Escape from Dormancy and Bone Metastatic Tumor Growth.

Metastasis to the bone is a common feature of many cancers including those of the breast, prostate, lung, thyroid and kidney. Once tumors metastasize to the bone, they are essentially incurable. Bone metastasis is a complex process involving not only intravasation of tumor cells from the primary tumor into circulation, but extravasation from circulation into the bone where they meet an environment that is generally suppressive of their growth. The bone microenvironment can inhibit the growth of disseminated tumor cells (DTC) by inducing dormancy of the DTC directly and later on following forma
