Tumor Microenvironment: Extracellular Matrix Alterations Influence Tumor Progression.

The tumor microenvironment (TME) is composed of various cell types embedded in an altered extracellular matrix (ECM). ECM not only serves as a support for tumor cell but also regulates cell-cell or cell-matrix cross-talks. Alterations in ECM may be induced by hypoxia and acidosis, by oxygen free radicals generated by infiltrating inflammatory cells or by tumor- or stromal cell-secreted proteases. A poorer diagnosis for patients is often associated with ECM alterations. Tumor ECM proteome, also named cancer matrisome, is strongly altered, and different ECM protein signatures may be defined to s

Microenvironment of Solid Tumors.

The tumor microenvironment (TME) plays a fundamental role in tumor progression. Cancer cells interact with their surroundings to establish a supportive niche through structural changes and paracrine signaling. Cells around transformed tumor cells contribute to cancer development, while infiltrating immune cells in this aggressive TME often become exhausted. Solid tumors, especially the most invasive types such as pancreatic ductal adenocarcinoma, are notably stiff mechanically, with cross-linking enzymes significantly affecting the survival of cancer cells in both primary tumors and metastatic

Natural medicines target tumor vascular microenvironment to inhibit tumor.

The abnormally active tumor vasculature provides a good blood supply for the rapid proliferation of tumors. The tumor microenvironment of tumor cells is associated with the secretion of a lot of angiogenic factors to promote the formation of blood vessels. However, the blood vessels are often irregular and immature. Additionally, the tumor tissue, in the process of its rapid proliferation, oppresses tumor blood vessels, causing hypoperfusion and leading to high interstitial pressure and hypoxia, which also results in changes to the fluid mechanics in the tumor microenvironment. Different fluid

Stroma-oriented tumor microenvironment facilitates intraosseous ameloblastoma tumor growth.

Ameloblastoma (AM) is a locally aggressive odontogenic epithelial tumor with poorly understood intraosseous pathogenesis. Here, we established an orthotopic intraosseous mouse model using AM epithelial cells in combination with AM-derived mesenchymal stromal cells (AMMSCs) or their secretome. AMMSC-derived extracellular vesicle-enriched secretome enhances stemness, epithelial-mesenchymal transition (EMT) phenotypes, and tumorigenic ability of AM epithelial cells. A unique signature of microRNAs in AMMSC-derived secretome is functionally associated with the upregulation of EMT-regulatory transc

Multifunctional Liposomes Remodeling Tumor Immune Microenvironment for Tumor Chemoimmunotherapy.

In the treatment of solid tumors, the complex barriers composed of cancer-associated fibroblasts (CAFs) prevent drug delivery and T cells infiltration into tumor tissues. Although nanocarriers hold great prospects in drug delivery, fibrosis causes the biological barrier and immunosuppressive tumor microenvironment (ITM) that impairs the anti-tumor efficacy of nanocarriers. Here, a small dendritic macromolecule loaded with doxorubicin (PAMAM-ss-DOX) (DP) is synthesized and encapsulated into pH-responsive nanoliposome, together with adjuvant toll-like receptor 7/8 (TLR7/8) agonist resiquimod (R8

Studying Ferroptosis in the Tumor Microenvironment.

Despite strong preclinical evidence, ferroptosis-based strategies have not yet translated into the clinic, in part due to the disconnect between conventional in vitro models and the complex tumor microenvironment. In this commentary, we consider ferroptosis as a microenvironmentally regulated process and call for multidisciplinary approaches that bridge foundational ferroptosis biology with clinically actionable interventions grounded in the context of the tumor microenvironment.

Oncolytic virotherapy and tumor microenvironment modulation.

Oncolytic viruses (OVs) have emerged as a transformative approach in cancer therapy, offering tumor-specific lysis while sparing normal tissues. In addition to their direct cytolytic effects, OVs actively reshape the tumor microenvironment (TME) by enhancing immune infiltration, disrupting immunosuppressive signals, and promoting tumor antigen presentation. However, the complexity of the TME poses challenges, often necessitating combination therapies to improve OV efficacy and overcome tumor resistance. This review explores the evolution of oncolytic virotherapy, from the early use of naturall

Mimicking and analyzing the tumor microenvironment.

The tumor microenvironment (TME) is increasingly appreciated to play a decisive role in cancer development and response to therapy in all solid tumors. Hypoxia, acidosis, high interstitial pressure, nutrient-poor conditions, and high cellular heterogeneity of the TME arise from interactions between cancer cells and their environment. These properties, in turn, play key roles in the aggressiveness and therapy resistance of the disease, through complex reciprocal interactions between the cancer cell genotype and phenotype, and the physicochemical and cellular environment. Understanding this comp

Vascular Microenvironment, Tumor Immunity and Immunotherapy.

Immunotherapy holds great promise for treating cancer. Nonetheless, T cell-based immunotherapy of solid tumors has remained challenging, largely due to the lack of universal tumor-specific antigens and an immunosuppressive tumor microenvironment (TME) that inhibits lymphocyte infiltration and activation. Aberrant vascularity characterizes malignant solid tumors, which fuels the formation of an immune-hostile microenvironment and induces tumor resistance to immunotherapy, emerging as a crucial target for adjuvant treatment in cancer immunotherapy. In this review, we discuss the molecular and ce

Androgen Signaling in the Tumor Microenvironment.

The key function of mesenchymal/stromal androgen receptor (AR) signaling for prostate development has been well documented by tissue recombination experiments. Some studies have addressed the expression and function of AR in stromal cells in prostate cancer, yet our understanding of the role of stromal AR in other tissues beyond prostate is still insufficient.Genomic analysis has revealed that cellular responses to androgens differ between epithelial and stromal cells. AR in stromal cells seems not to act via classical AR transcription factors such as FOXA1 but rather depends on the JUN/AP1 co

The Tumor Microenvironment in Cholangiocarcinoma Progression.

Cholangiocarcinoma (CCA) is an aggressive and heterogeneous malignancy of the biliary tree. A typical hallmark of CCA is that cancer cells are embedded into a dense stroma containing fibrogenic cells, lymphatics and a variety of immune cells. Functional roles of the reactive tumor stroma are not fully elucidated; however, recent studies suggest that the tumor microenvironment plays a key role in the progression and invasiveness of CCA. CCA cells exchange autocrine/paracrine signals with other cancer cells and the infiltrating cell types that populate the microenvironment. This crosstalk is und

Role of prostaglandins in tumor microenvironment.

Tumor tissue is composed of tumor cells and surrounding non-tumor endothelial and immune cells, collectively known as the tumor microenvironment. Tumor cells manipulate tumor microenvironment to obtain sufficient oxygen and nutrient supply, and evade anti-tumor immunosurveillance. Various types of signaling molecules, including cytokines, chemokines, growth factors, and lipid mediators, are secreted, which co-operate to make up the complex tumor microenvironment. Prostaglandins, cyclooxygenase metabolites of arachidonic acid, are abundantly produced in tumor tissues. Ever since treatment with

The tumor microenvironment in esophageal cancer.

Esophageal cancer is a deadly disease, ranking sixth among all cancers in mortality. Despite incremental advances in diagnostics and therapeutics, esophageal cancer still carries a poor prognosis, and thus, there remains a need to elucidate the molecular mechanisms underlying this disease. There is accumulating evidence that a comprehensive understanding of the molecular composition of esophageal cancer requires attention to not only tumor cells but also the tumor microenvironment (TME), which contains diverse cell populations, signaling factors and structural molecules that interact with tumo

Bone marrow microenvironment and tumor progression.

The bone marrow constitutes an unique microenvironment for cancer cells in three specific aspects. First, the bone marrow actively recruits circulating tumor cells where they find a sanctuary rich in growth factors and cytokines that promote their proliferation and survival. When in the bone marrow, tumor cells profoundly affect the homeostasis of the bone and the balance between osteogenesis and osteolysis. As a consequence, growth and survival factors normally sequestered into the bone matrix are released, further fueling cancer progression. Second, tumor cells actively recruit bone marrow-d

Chemokine Landscapes of the Tumor Microenvironment.

Chemokines are well-recognized for orchestrating immune cell traffic between tissues via the blood and lymph, yet how they guide the formation of cellular neighborhoods and niches within inflamed tissues remains largely unknown. Here, we use spatial transcriptomics to comprehensively map the chemokine landscape in the chronic inflammatory environment of solid tumors. In murine models representing melanoma, sarcoma, and carcinoma, we identify conserved and tumor type-specific patterns for individual chemokines, including exclusive or preferential expression in tumor core versus stroma and disti

Phase separation and the tumor microenvironment.

Liquid-Liquid Phase Separation (LLPS) is a physicochemical process which involves biomolecules spontaneously assembling into membrane-less biomolecular condensates through multivalent weak interactions. It emerges as a key factor regulating cellular homeostasis, and is widely involved in the spatiotemporal regulation of core cellular activities including gene expression and signal transduction. Notably, the dynamic process of LLPS is highly sensitive to extracellular microenvironmental cues, especially in the context of cancer. The Tumor Microenvironment (TME) is a dynamically heterogeneous en

The immunosuppressive tumor microenvironment in glioblastoma.

Glioblastoma (GBM) remains one of the most lethal primary brain tumors, with limited therapeutic improvement despite maximal surgical resection, radiotherapy, and temozolomide. A major barrier to durable treatment response is the profoundly immunosuppressive tumor microenvironment, which is characterized by immune exclusion, defective antigen presentation, myeloid dominance, and severe T-cell dysfunction. Tumor-associated macrophages, resident microglia, myeloid-derived suppressor cells, neutrophils, regulatory T cells, and glioma-derived extracellular vesicles collectively establish a suppres

Which dominates recurrence: tumor or microenvironment?

Lung squamous cell carcinoma (LUSC) is a major challenge in oncology due to its high recurrence rate, highlighting the need to identify recurrence drivers. Based on the data provided by Zhang Zemin’s team in their research, we selected survival metadata from 119 patients with lung squamous cell carcinoma and data on the proportions of 51 immune cell types. We then combined machine learning methods to analyze the intrinsic tumor factors and tumor microenvironment (TME) in the recurrence of lung squamous cell carcinoma (LUSC). Chi-square tests identified immune cell subsets enriched in recurrent

Targeting tumor microenvironment with photodynamic nanomedicine.

Photodynamic therapy (PDT) is approved for the treatment of certain cancers and precancer lesions. While early Photosensitizers (PS) have found their way to the clinic, research in the last two decades has led to the development of third-generation PS, including photodynamic nanomedicine for improved tumor delivery and minimal systemic or phototoxicity. In terms of nanoparticle design for PDT, we are witnessing a shift from passive to active delivery for improved outcomes with reduced PS dosage. Tumor microenvironment (TME) comprises of a complex and dynamic landscape with myriad potential tar

Studying the Tumor Microenvironment in Zebrafish.

The tumor microenvironment significantly contributes to tumor initiation, progression, neo-angiogenesis, and metastasis, and a better understanding of the role of the different cellular players would facilitate the development of novel therapeutic strategies for cancer treatment. Towards this goal, intravital imaging is a powerful method to unravel interaction partners of tumor cells. Among vertebrate model organisms, zebrafish is uniquely suited for in vivo imaging studies. In recent years zebrafish has also become a valuable model in cancer research. In this chapter, we will summarize, how z
