On the postnatal development of the striate cortex (V1) in the tree shrew (Tupaia belangeri).

Histological serial sections, three-dimensional reconstructions and morphometry served to study the postnatal development of V1 in tree shrews. The main objectives were to evaluate the expansion of V1, the implications of its growth on the occipital cortex and, vice versa, the effects of the expanding neocortex on the topography of V1. The future V1 was identified on postnatal day 1 by its granular layer IV, covering the superior surface of the occipital cortices including the poles. A subdivision of layer IV, distinctive for the binocular part, was evident in the central region. V1 expanded c

Cerebellar cortical efferent fibers of the paraflocculus of tree shrew (Tupaia glis).

Efferent projections from the paraflocculus of the tree shrew (Tupaia glis) were studied utilizing the Fink and Heimer ('67) method. Cerebellar corticonuclear fibers of both dorsal (Dpf) and ventral (Vpf) divisions of the paraflocculus terminate in the lateral cerebellar nucelus (NL) and in the posterior interposed nucleus (NIP). These fibers are ipsilateral, topographically organized and arranged into at least two zones. Following injury to either the Dpf or Vpf, degenerated axons are found in lateral and caudal regions of the NIP respectively. Consequently, these two portions of the parafloc

Potassium oxonate induces acute hyperuricemia in the tree shrew (tupaia belangeri chinensis).

Potassium oxonate, a selectively competitive uricase inhibitor, produced hyperuricemia (HUA) in rodents in a previous study. In this study, we employed the tree shrew as an animal model to study potassium oxonate-induced HUA. The effect of allopurinol (ALLO), a uric acid reducer, was also examined in this model. Potassium oxonate at doses of 5, 20, 40, 60, 80, 100, and 1,000 mg/kg was given intraperitoneally to tree shrews. The results showed that potassium oxonate can effectively increase the levels of uric acid in tree shrews at doses ranging from 40 to 100 mg/kg. Semiquantitative RT-PCR sho

Characterization of hepatitis E virus infection in tree shrew (Tupaia belangeri chinensis).

BACKGROUND: Hepatitis E virus (HEV) is a major cause of hepatitis in developing countries and poses a threat to public health worldwide. Tree shrew (Tupaia belangeri chinensis) is a useful animal model in studies on hepatitis viruses, such as hepatitis B and C viruses. However, the use of this animal model for HEV research is yet to be developed. METHODS: Tree shrews were intravenously (IV) injected with swine genotype 4 HEV or infected by contact-exposure to IV infected tree shrews. RT-nPCR was performed to detect HEV RNA in the feces, tissues, and blood. HEV capsid protein in the different

[Morphological characteristics and cryodamage of Chinese tree shrew (Tupaia belangeri chinensis) sperm].

The tree shrew (Tupaia belangeri chinensis) is a small non-rodent mammal, which is a relatively new experimental animal in medicine due to its close evolutionary relationship to primates and its rapid propagation. Sperm characteristics and cryopreservation in the tree shrew were the main contents of our spermatological research. Epididymal sperm were surgically harvested from male tree shrews captured from the Kunming area. The rate of testis weight to body weight was (1.05±0.07)%, volume of both testis was (1.12 ± 0.10) mL, total sperm from epididymis and vas deferens were 2.2-8.8×10(7), and

Phyllodes tumor in the mammary gland of a northern tree shrew (Tupaia belangeri).

A rapidly growing mass was observed in the left third mammary gland of a 5-and-a-half-year-old intact female northern tree shrew (Tupaia belangeri). Histopathologically, the mass was composed of numerous irregularly dilated mammary ducts with a pericanalicular or intracanalicular pattern. These mammary ducts showed a leaf-like structure due to the proliferations of surrounding spindle-shaped neoplastic cells with moderate to severe nuclear atypia. We diagnosed this case as a phyllodes tumor in a mammary gland of a northern tree shrew (Tupaia belangeri), which closely resembled those occurring

A systematic study of Tupaia as a model for human acute hepatitis B infection.

The molecular features of hepatitis B virus (HBV) infection, eradication, and pathogenesis are poorly understood, partly due to the lack of an adequate animal model that faithfully reproduces the course of infection. Although Tupaia belangeri were previously recognized as HBV-susceptible animals, the course of infection in adult tupaias remains obscure. Herein, we performed a longitudinal study and demonstrated that adult tupaias were efficiently infected (90% infection rate) with 108 copies of the HBV genome. HBV replicated vigorously, produced high levels of covalently closed circular DNA (c

The corticosterone receptive system in the brain of Tupaia belangeri visualized by in vivo autoradiography.

The present investigation deals with in vivo binding of 3H-corticosterone in the brains of tree shrews as visualized by autoradiography. Tree shrews were injected with 3H-corticosterone and brain sections were mounted on slides which were subsequently exposed on tritium sensitive film. The relative labeling of 20 different brain structures was determined densitometrically. The indusium griseum, which demonstrated the highest binding for corticosterone of all brain regions in the autoradiograms, was taken as reference and defined as 100% relative labeling (RL). As in other species, the hippocam

A Golgi study of the medial geniculate body in the tree shrew (Tupaia glis).

The subdivisions of the medial geniculate body in Tupaia recognized in previous connectional and cytoarchitectural studies are identified in Golgi-impregnated material. They may be distinguished by the organization of the neuropil, especially the dendrites, and, in many cases, by differences in the neurons. The ventral nucleus contains tufted cells with disc-shaped dendritic fields which are aligned to form laminae. The caudomarginal and deep dorsal nuclei have less tufted, less precisely arranged cells with longer, thin appendages. Neurons in the suprageniculate and dorsal nuclei are similar

Anterior Segment Anatomy and Conventional Outflow Physiology of the Tree Shrew (Tupaia belangeri).

PURPOSE: Rodent and primate models are commonly used in glaucoma research; however, both have their limitations. The tree shrew (Tupaia belangeri) is an emerging animal model for glaucoma research owing in part to having a human-like optic nerve head anatomy, specifically a collagenous load-bearing lamina. However, the anterior segment anatomy and function have not been extensively studied in the tree shrew. Thus, the purpose of this study was to provide the first detailed examination of the anterior segment anatomy and aqueous outflow facility in the tree shrew. METHODS: Aqueous outflow dyna

An analysis of neurovascular disease markers in the hippocampus of Tupaia chinensis at different growth stages.

INTRODUCTION: It is considered that Tupaia chinensis can replace laboratory primates in the study of nervous system diseases. To date, however, protein expression in the brain of Tupaia chinensis has not been fully understood. METHOD: Three age groups of T. chinensis-15 days, 3 months and 1.5 years-were selected to study their hippocampal protein expression profiles. RESULTS: A significant difference was observed between the 15-day group and the other two age groups, where as there were no significant differences between the 3-month and 1.5-year age groups. Characteristics of olfactory ensheathing cells and microarray analysis in Tupaia belangeri (Wagner, 1841).

Tree shrews are most closely related to the primates and so possess a number of advantages in experimental studies; they have been used as an animal model in bacterial and virus infection, cancer, endocrine system disease, and certain nervous system diseases. Their olfactory ensheathing cells (OECs) are able to release several cytokines to promote neuronal survival, regeneration and remyelination. The present study used western blot analysis to identify antibody specificity in protein extracts from whole tree shrew brains to identify the specificity of p75 nerve growth factor receptor (NGFR) d

Susceptibility and initial immune response of Tupaia belangeri cells to dengue virus infection.

Dengue is an emerging disease of great public health significance worldwide. The lack of a suitable infection model has hampered dengue virus (DENV) pathogenesis study, and developing a suitable small animal model has been a long-standing challenge. The aim of this study was to develop a feasible experimental model of DENV infection using Tupaia belangeri. The susceptibility of tupaia to DENV infection and characteristics of its innate immune response were examined in vitro. We found that tupaia fibroblast cells support replication of DENV serotypes 1-4 with a linear increase in viral load 24-

Oxidative Stress and Immune Responses During Hepatitis C Virus Infection in Tupaia belangeri.

Hepatitis C virus (HCV) is a leading cause of chronic liver disease, cirrhosis, and hepatocellular carcinoma. To address the molecular basis of HCV pathogenesis using tupaias (Tupaia belangeri), we characterized host responses upon HCV infection. Adult tupaias were infected with HCV genotypes 1a, 1b, 2a, or 4a. Viral RNA, alanine aminotransferase, anti-HCV core and anti-nonstructural protein NS3 antibody titres, reactive oxygen species (ROS), and anti-3β-hydroxysterol-Δ24reductase (DHCR24) antibody levels were measured at 2-week intervals from 0 to 41 weeks postinfection. All HCV genotypes est

Scanning electron microscopic study on pineal vascularization of the common tree shrew (Tupaia glis).

The detailed blood supply including microvascularization of pineal gland in the common tree shrew (Tupaia glis) was investigated using a vascular corrosion cast technique in conjunction with scanning electron microscopy. Adult common tree shrews of both sexes, divided into 3 groups, were injected with red latex, blue vinyl resin, and Batson's No. 17 plastic mixture for the studies of arterial supply, venous drainage, and microvasculature of the pineal gland, respectively. It was found that the pineal gland is a highly vascularized organ. It receives two to four branches from the medial posteri

Molecular cloning and characterization of NPC1L1 in the Chinese tree shrew (Tupaia belangeri chinensis).

BACKGROUND: The Niemann-Pick C1-Like 1 protein, a multi-transmembrane domain molecule, is critical for intestinal cholesterol absorption, and is the entry factor for hepatitis C virus (HCV). The Chinese tree shrew (Tupaia belangeri chinensis) is closer to primates in terms of genetic evolution than rodents. Previous studies indicated that the tree shrew was suitable for HCV research; however, little is known about tree shrew NPC1L1. METHODS AND RESULTS: TsNPC1L1 cDNA was amplified by rapid amplification of cDNA ends (RACE) technology. The cDNA sequence, its encoded protein structure, and expr

Calretinin immunoreactivity in the prenatally developing olfactory systems of the tree shrew Tupaia belangeri.

The prenatal patterns of calretinin immunoreactivity were studied in the olfactory systems of Tupaia belangeri. We investigated the peripheral and primary central parts of the vomeronasal system and of the main olfactory system from the 19th to the 43rd (last) day of gestation and compared the findings with the known calretinin immunoreactivity patterns in adult T. belangeri and the published data on other mammals. The onset of calretinin immunoreactivity was noted in the main olfactory system on the 23rd day of gestation and, in the vomeronasal system, on the 25th day of gestation: single pre

The patterns of cell death and of macrophages in the developing forebrain of the tree shrew Tupaia belangeri.

The patterns of cell death and of macrophages were investigated in the forebrain and eyes of the tree shrew Tupaia belangeri during five phases of optic cup formation. Seventeen embryos were studied. Three- dimensional reconstructions were made from one embryo of each phase. In phase 1 (V-shaped optic evagination) a midline band of cell death passes through the closing anterior neuroporus. From phases 2 (optic vesicle) to 5 (far-advanced invagination) the midline band of cell death extends in the dorsal wall of the forebrain to its rostral pole and, further, into its ventral wall. Cardiac musculature of the cranial vena cava in the common tree shrew (Tupaia glis).

Cardiac musculature of the cranial vena cava in the common tree shrew (Tupaia glis) was examined by light and transmission electron microscopy. The common tree shrew has well developed cardiac myocyte layers in the tunica media of the cranial vena cava, extending from the right atrium to the root of the subclavian vein. Because the common tree shrew belongs to a primitive group of mammals, the occurrence of cardiac musculature in the cranial vena cava may be a common feature in lower mammals. The development of this musculature indicates that active contraction of the cranial vena cava wall oc
