Nematicidal activity of allyl bromide and dibromo(nitro)methane under laboratory conditions.

BACKGROUND: Restrictions on soil fumigants are prompting the development of new compounds for controlling nematodes, other soilborne pathogens and weeds. We evaluated the nematicidal activity of five bromine compounds against Meloidogyne javanica in vitro, and tested the two most effective ones against Pratylenchus penetrans and Xiphinema index in vitro and in soil. RESULTS: Only allyl bromide and dibromo(nitro)methane showed nematicidal activity against M. javanica juveniles in vitro at <320 mg L(-1) . Allyl bromide killed M. javanica and P. penetrans at 20 mg L(-1), and X. index at 10 mg L(

MgCBP1 Synergistically Enhances MgENG2-Mediated Cellulose Degradation to Promote Parasitism in Meloidogyne graminicola.

Plant-parasitic nematodes secrete effector proteins, including cell wall-degrading enzymes (CWDEs) and cell wall-modifying proteins (CWMPs), to facilitate infection. However, whether cellulose-binding proteins (CBPs), which are a class of CWMPs, can synergize with cellulases (a subgroup of CWDEs) to promote parasitism remains unclear. Here, we identified and characterized a CBP (MgCBP1) and a β-1,4-endoglucanase (MgENG2) from the rice root-knot nematode Meloidogyne graminicola. MgCBP1 contains a conserved cellulose-binding module (CBM) and is specifically expressed in the subventral oesophagea

Integrated omics reveal phenylpropanoid biosynthesis as a key defense pathway against Meloidogyne incognita in Cucumis metuliferus.

The southern root-knot nematode (Meloidogyne incognita, M. incognita) poses a severe threat to cucurbit production, yet no resistant germplasm exists among cultivated species. Although Cucumis metuliferus (C. metuliferus), a wild Cucurbitaceae species, exhibits resistance to M. incognita and prior transcriptomic studies have identified key responsive genes and signaling pathways, an integrated understanding of the dynamic transcriptional and metabolic reprogramming is still lacking. Here, an integrated transcriptomic and metabolomic analysis was performed on resistant (CM064) and susceptible (

First report of nematicidal activity of Planococcus plakortidis against the citrus nematode Tylenchulussemipenetrans.

This study reports, for the first time, the nematicidal potential of the cell-free culture filtrate of Planococcus plakortidis against the citrus nematode Tylenchulus semipenetrans. In vitro bioassays revealed a strong nematicidal activity, reaching 83.06% mortality after five days of incubation. The highest nematicidal effect coincided with the maximum bacterial population density (5.6 × 106 cells ml-1), suggesting a close association between the exponential growth phase of the bacterium and the production of bioactive metabolites. Under greenhouse conditions, application of the culture filtr

Application of array tomography to elucidate nuclear clustering architecture in giant-feeding cells induced by root-knot nematodes.

Plant-parasitic nematodes like root-knot nematodes (RKN; Meloidogyne spp.) cause great losses in agriculture by inducing root swellings, named galls, in host roots disturbing plant growth and development. Previous two-dimensional studies using different microscopy techniques revealed the presence of numerous nuclear clusters in nematode-induced giant cells within galls. Here, we show in three dimensions (3D) that nuclear clustering occurring in giant cells is revealed to be much more complex, illustrating subclusters built of multiple nuclear lobes. These nuclear subclusters are unveiled to be

3-Furoic Acid from Sea-Derived Aspergillus luchuensis Hy-6 as a Valuable Lead Compound against Plant-Parasitic Nematodes in Cucumber.

Root-knot nematodes pose a significant threat to crop growth and yield, thereby impacting global food security. Here, the sea-derived Aspergillus luchuensis hy-6 was identified as a producer of 3-furoic acid by liquid chromatography-mass spectrometry. Low doses of 3-furoic acid, an acidic compound with strong nematicidal activity, can reduce the motility, lifespan, and egg hatchability of nematodes in a dose-dependent manner. Additionally, 3-furoic acid was found to affect the chemotaxis of Meloidogyne incognita toward cucumber by decreasing the root's attractive activity to the nematodes. Met

Parasitic Nematodes of Sugarcane: A Major Productivity Impediment and Grand Challenges in Management.

Sugarcane is an important cash crop grown in 137 countries, accounting for 80% of global sugar production. It supports the livelihood of more than 100 million people and up to 25% of the rural population in some countries. Plant-parasitic nematodes are one significant constraint in sugarcane production and can lead to a loss of up to 30% in productivity. More than 300 species of parasitic nematodes have been discovered in sugarcane soil. Owing to limited data, the potential damage to sugarcane crops caused by parasitic nematodes is often underestimated. The main nematodes present in sugarcane

SlWRKY16 and SlWRKY31 of tomato, negative regulators of plant defense, involved in susceptibility activation following root-knot nematode Meloidogyne javanica infection.

The involvement of WRKY transcription factors in plant-nematode interactions, and in particular, how these WRKYs participate in regulating the complex morphological and physiological changes occurring after nematode infection, are the topic of active research. We characterized the functional role of the unstudied tomato WRKY genes SlWRKY16 and SlWRKY31 in regulating tomato roots' response to infection by the root-knot nematode Meloidogyne javanica. Using promoter-GUS reporter gene fusions and qRT-PCR, we show that both SlWRKYs are predominantly expressed during the first half of the parasitic

The impact of fluazaindolizine on free-living nematodes and the nematode community structure in a root-knot nematode infested vegetable production system.

BACKGROUND: Fluazaindolizine is a novel sulfonamide nematicide that is currently under commercialization in various countries. Four trials (two in tomato, two in zucchini) were carried out over two growing seasons in a root-knot nematode (RKN; Meloidogyne incognita)-infested plastic house to test the effects of a single application of fluazaindolizine at 1000 and 2000 g ha-1 , on RKN, the resident nematode community and to estimate any side effects on the soil food web and soil ecological functions. The composition of the nematode community was evaluated at three sampling times, pre-treatment

Mechanosensation of rice root border cells activates serotonin-mediated pre-invasion behaviors in Meloidogyne graminicola via DEG/ENaC channels.

BACKGROUND: Plant-parasitic nematodes (PPNs) locate host roots primarily through chemosensation, yet the sensory events that convert root contact into the stylet thrusts required for host penetration remain obscure. Root border cells (RBCs), terminally differentiated cells shed from the root cap, constitute the first point of physical contact between the root and invading nematodes, but whether and how RBCs trigger pre-invasion behaviors is unknown. RESULTS: Here we showed that rice (Oryza sativa) RBCs induce preparasitic second-stage juveniles (pre-J2) of the rice root-knot nematode Meloidog

Volatile Organic Compounds of Bacillus velezensis GJ-7 against Meloidogyne hapla through Multiple Prevention and Control Modes.

The Bacillus velezensis GJ-7 strain isolated from the rhizosphere soil of Panax notoginseng showed high nematicidal activity and therefore has been considered a biological control agent that could act against the root-knot nematode Meloidogyne hapla. However, little was known about whether the GJ-7 strain could produce volatile organic compounds (VOCs) that were effective in biocontrol against M. hapla. In this study, we evaluated the nematicidal activity of VOCs produced by the fermentation of GJ-7 in three-compartment Petri dishes. The results revealed that the mortality rates of M. hapla J2

Toxicity of fungal-derived volatile organic compounds against root-knot nematodes.

BACKGROUND: Root-knot nematodes (RKNs), including Meloidogyne species, are among the most destructive plant-parasites worldwide. Recent evidence suggests that entomopathogenic fungi (EPF) can antagonize RKNs. Such antagonistic effects are likely mediated by toxic metabolites, including volatile organic compounds (VOCs), produced by the fungi. However, how widespread these effects are across EPF species, and which VOCs mediate negative interactions between EPF and RKNs needs to be further elucidated. RESULTS: First, we evaluated the nematicidal effect of VOCs emitted by 46 EPF isolates against

Transcriptome reveals the toxicity difference of dimethyl disulfide by contact and fumigation on Meloidogyne incognita through calcium channel-mediated oxidative phosphorylation.

The prevention and control of root-knot nematode disease has been posing a severe challenge worldwide. Fumigant dimethyl disulfide (DMDS) has excellent biological activity against nematodes. However, DMDS displays significant differences in contact and fumigation toxicity on nematodes. The specific regulatory mechanisms of DMDS on nematodes were investigated by characterizing the ultrastructure of nematodes, examining the physiological and biochemical indicators, and conducting transcriptome high-throughput sequencing. As indicated by the results, DMDS fumigation exhibited the biological activ
