Study on the Non-Target Metabolomics Effects of Tylosin on Pasteurella multocida.

Pasteurella multocida (P. multocida) is not only the core pathogen of bovine respiratory disease (BRDC) but also a significant zoonotic agent, posing a dual threat to global animal husbandry and public health. This study utilized untargeted metabolomics to systematically dissect the metabolic regulatory network of P. multocida in response to tylosin within a One Health framework. The results revealed significant "defense-growth" metabolic reprogramming: activation of amino sugar and nucleotide sugar pathways (e.g., CDP-glucose) indicated cell wall remodeling, while directional shifts in the ph

Bibliometric analysis of pharmacokinetic research on the combination of norfloxacin and tylosin in poultry and reptiles.

BACKGROUND: Bibliometric analysis is a method for analyzing article data, such as those from books and journals. Pharmacokinetic studies are one way to determine the course of drugs in the body, and the irrational use of drugs, especially antibiotics, will cause antibiotic residues and antimicrobial resistance. AIM: To assess the pharmacokinetic research trends of norfloxacin and tylosin in poultry and reptiles, bibliometric analyses focusing on pharmacokinetics and antimicrobial resistance are necessary, in as much as improper dosage can lead to increased antimicrobial resistance, especially

Development and assessment of an immobilized bacterial alliance that efficiently degrades tylosin in wastewater.

Microbial degradation of tylosin (TYL) is a safe and environmentally friendly technology for remediating environmental pollution. Kurthia gibsonii (TYL-A1) and Klebsiella pneumonia (TYL-B2) were isolated from wastewater; degradation efficiency of the two strains combined was significantly greater than either alone and resulted in degradation products that were less toxic than TYL. With Polyvinyl alcohol (PVA)-sodium alginate (SA)-activated carbon (AC) used to form a bacterial immobilization carrier, the immobilized bacterial alliance reached 95.9% degradation efficiency in 1 d and could be reu

Acquired resistance to the 16-membered macrolides tylosin and tilmicosin by Mycoplasma bovis.

The molecular mechanism of acquired resistance to the 16-membered macrolides tylosin (Ty) and tilmicosin (Tm) was investigated in Mycoplasma bovis field isolates. Sequence analysis of domains II and V of the two 23S rRNA alleles and ribosomal proteins L4 and L22 was performed on 54 M. bovis isolates showing different minimal inhibitory concentrations (MIC). The presence of any one of the point mutations G748A, C752T, A2058G, A2059G or A2059C (Escherichia coli numbering) in one or both alleles of the 23S rRNAs was correlated with decreased susceptibility to Ty (8-1024 μg/ml) and to Tm (32 to >2

Structural Insight into Interaction between C20 Phenylalanyl Derivative of Tylosin and Ribosomal Tunnel.

Macrolides are clinically important antibiotics that inhibit protein biosynthesis on ribosomes by binding to ribosomal tunnel. Tylosin belongs to the group of 16-membered macrolides. It is a potent inhibitor of translation whose activity is largely due to reversible covalent binding of its aldehyde group with the base of A2062 in 23S ribosomal RNA. It is known that the conversion of the aldehyde group of tylosin to methyl or carbinol groups dramatically reduces its inhibitory activity. However, earlier we obtained several derivatives of tylosin having comparable activity in spite of the fact t

[Analytical Method for Tylosin in Livestock Products and Honey Using LC-MS/MS].

This study proposes a method to determine tylosin A in livestock products and tylosin A and B in honey using LC-MS/MS. The chilled samples were homogenized with a chilled mixture of ethanol and 20% (v/v) acetic acid (1 : 1, v/v),which were further cooled with ice until extraction was initiated. Acetone was used to extract analytes from the samples. These extracts were defatted using n-hexane (this step was omitted for honey) and purified with an octadecyl silylated silica gel cartridge column (Bond Elut C18).HPLC was performed on an Inertsil ODS-4 column using a gradient formed from 0.1% (v/v)

Concurrent biodegradation of tylosin and tilmicosin by a novel Paracoccus versutus 4-B-2.

The co-contamination of macrolide antibiotics tylosin (TYL) and tilmicosin (TLM) poses a persistent environmental threat, exacerbating the spread of antimicrobial resistance. Effective bioremediation has been hampered by the lack of microorganisms capable of degrading both compounds simultaneously. Here, we report the isolation of Paracoccus versutus 4-B-2, a novel bacterial strain that achieves complete co-degradation of TYL and TLM within 144 h, with first-order degradation half-lives of 1.28 days for TLM and 1.47 days for TYL, representing a 1.6-fold enhancement over previously reported str

Identification of novel tylosin analogues generated by a wblA disruption mutant of Streptomyces ansochromogenes.

BACKGROUND: Streptomyces, as the main source of antibiotics, has been intensively exploited for discovering new drug candidates to combat the evolving pathogens. Disruption of wblA, an actinobacteria-specific gene controlling major developmental transition, can cause the alteration of phenotype and morphology in many species of Streptomyces. One wblA homologue was found in Streptomyces ansochromogenes 7100 by using the Basic Local Alignment Search Tool. It is interesting to identify whether novel secondary metabolites could be produced by the wblA disruption mutant as evidenced in other Strept

Impact of subtherapeutic administration of tylosin and chlortetracycline on antimicrobial resistance in farrow-to-finish swine.

The use of antimicrobial agents in swine production at subtherapeutic concentrations for the purpose of growth promotion remains controversial due to the potential impact on public health. Beginning at weaning (3 weeks), pigs received either nonmedicated feed or feed supplemented with subtherapeutic levels of either tylosin (11-44 ppm) or chlortetracycline (5.5 ppm). After only 3 weeks, pigs given feed supplemented with tylosin had significantly higher levels of tylosin-resistant anaerobes (P < 0.0001) compared with the control group, increasing from 11.8% to 89.6%, a level which was stable fo

Efficacy of tylosin tablets for the treatment of pyoderma due to Staphylococcus intermedius infection in dogs.

Tylosin tablets (20 mg/kg, q12h) were administered orally to 21 dogs with superficial or deep staphylococcal pyodermas. Response to therapy was excellent in 90.5% of the dogs, and in vitro susceptibility testing correlated perfectly with therapeutic response. Duration of therapy varied from 17 to 91 days, with an average of 33 days. Relapses occurred in 28.6% of the dogs within a three-month period. No side effects were reported. Under the conditions of the study, tylosin was an effective and safe antibiotic for the treatment of staphylococcal pyoderma in dogs.

Discovery of a highly efficient TylF methyltransferase via random mutagenesis for improving tylosin production.

Macrolides are currently a class of extensively used antibiotics in human and animal medicine. Tylosin is not only one of the most important veterinary macrolides but also an indispensable material for the bio- and chemo-synthesis of new generations of macrolide antibiotics. Thus, improving its production yield is of great value. As the key rate-limiting enzyme catalyzing the terminal step of tylosin biosynthesis in Streptomyces fradiae (S. fradiae), TylF methyltransferase's catalytic activity directly affects tylosin yield. In this study, a tylF mutant library of S. fradiae SF-3 was construct

A high prevalence of tylosin resistance among Staphylococcus aureus strains isolated from bovine mastitis.

The macrolides appear to have considerable effects for treatment of bovine mastitis because of excellent diffusion into the mammary gland, long half-life, low protein binding, high intracellular concentration and lipid solubility. Acquired resistance to macrolides in Staphylococcus aureus is primarily related to target-site modification through acquisition of an erm gene. In the present study the prevalence of both phenotypic and genotypic tylosin resistance in S. aureus isolates (n = 103) from subclinical mastitis in nine dairy farms belonging to three different province of Iran were investig

Collaborative sorption behavior of tylosin and enrofloxacin in loess soil: implications for veterinary antibiotic fate.

Multiple veterinary antibiotics (VAs) coexist in soils, but their interaction and their effects on sorption in soils have not been extensively studied. This study focused on investigating the sorption properties of two widely used VAs, tylosin (TYL) and enrofloxacin (ENR), in loess soil. Through a comprehensive batch sorption method, we systematically evaluated single and binary sorption behaviors using varying ions (Ca2+, Mg2+, NH4+, and K+), ionic strengths (from 0.01 to 0.1 M) and pH (4-10). The results showed that batch experiments fitted well with pseudo-second-order kinetics (equilibrium

An intricate regulation of WblA controlling production of silent tylosin analogues and abolishment of expressible nikkomycin.

Genome sequencing has revealed that actinomycetes possess the potential to produce many more secondary metabolites than previously thought. The existing challenge is to devise efficient methods to activate these silent biosynthetic gene clusters (BGCs). In Streptomyces ansochromogenes, disruption of wblA, a pleiotropic regulatory gene, activated the expression of cryptic tylosin analogues and abolished nikkomycin production simultaneously. Overexpressing pathway-specific regulatory genes tylR1 and tylR2 can also trigger the biosynthesis of silent tylosin analogues, in which TylR1 exerted its f
