Non-functional ubiquitin C-terminal hydrolase L1 drives podocyte injury through impairing proteasomes in autoimmune glomerulonephritis.

Little is known about the mechanistic significance of the ubiquitin proteasome system (UPS) in a kidney autoimmune environment. In membranous nephropathy (MN), autoantibodies target podocytes of the glomerular filter resulting in proteinuria. Converging biochemical, structural, mouse pathomechanistic, and clinical information we report that the deubiquitinase Ubiquitin C-terminal hydrolase L1 (UCH-L1) is induced by oxidative stress in podocytes and is directly involved in proteasome substrate accumulation. Mechanistically, this toxic gain-of-function is mediated by non-functional UCH-L1, which

A Unique Case of Pediatric Orbital Nodular Fasciitis with a Confirmed USP6 Fusion Mutation.

Nodular fasciitis is a benign, pseudoneoplastic condition often misdiagnosed as sarcoma due to its rapid growth, high cellularity, and mitotic activity. Herein is reported a case of a 7-year-old patient with a painless, enlarging nodule in the left superolateral orbit. Ultrasound revealed a 7.5 mm cystic mass, which was excised via an upper eyelid crease anterior orbitotomy. Histopathologic studies showed benign spindle cells, and molecular testing identified a UBC::USP6 fusion, confirming the diagnosis of nodular fasciitis. Nodular fasciitis is most common in the limbs, trunk, and head or nec

UCHL3-mediated proteasomal degradation of GOPC drives microcystin-LR-induced acrosomal dysgenesis.

Growing evidence links environmental exposure to microcystin-leucine-arginine (MC-LR) with declining male fertility, yet its molecular pathogenesis remains poorly understood. In this study, we observed that MC-LR disrupted acrosome biogenesis, a critical process for sperm function. Mechanistically, MC-LR induced proteasomal degradation of Golgi-associated PDZ- and coiled-coil motif-containing protein (GOPC), a key acrosome assembly protein, by disrupting its ubiquitin homeostasis. This degradation resulted from dual inhibition of MC-LR on the deubiquitinase ubiquitin carboxyl terminal hydrolas

Antibody-free approach for ubiquitination profiling by selectively clicking the ubiquitination sites.

Ubiquitination is a reversible post-translational modification that plays a pivotal role in numerous biological processes. Antibody-based approaches, as the most used methods for identifying ubiquitination sites, exist sequence recognition bias, high cost, and ubiquitin-like protein modification interference, limiting their widespread application. Here, we proposed an Antibody-Free approach for Ubiquitination Profiling, termed AFUP, by selectively clicking the ubiquitinated lysine to enrich and profile endogenous ubiquitinated peptides using mass spectrometry. Briefly, protein amines were bloc

Deubiquitination and the regulation of stress granule assembly.

Stress granules (SGs) are evolutionarily conserved ribonucleoprotein (RNP) structures that form in response to a variety of environmental and cellular cues. The presence of these RNP granules has been linked to a number of human diseases, including neurodegenerative disorders like amyotrophic lateral sclerosis (ALS) and spinocerebellar ataxia type 2 (Li et al., J Cell Biol 201:361-372, 2013; Nonhoff et al., Mol Biol Cell 18:1385-1396, 2007). Understanding how the assembly of these granules is controlled could, therefore, suggest possible routes of therapy for patients afflicted with these cond

Targeting USP7 in sarcomas: opportunities and challenges.

Sarcomas are a heterogeneous group of aggressive mesenchymal tumors with limited therapeutic options in the advanced setting. Novel actionable targets and biology-driven therapies are needed to improve patients' outcome. The identification of common pathways engaged by different disease drivers may suggest new therapeutic strategies exploitable across sarcomas. Deregulation of the ubiquitin-proteasome system (UPS) has provided attractive targets for therapeutic intervention also in these malignancies. As sarcoma cells hijack UPS to stabilize aberrant oncogenic signaling and/or inactivate tumor

Spotlights on ubiquitin-specific protease 12 (USP12) in diseases: from multifaceted roles to pathophysiological mechanisms.

Ubiquitination is one of the most significant post-translational modifications that regulate almost all physiological processes like cell proliferation, autophagy, apoptosis, and cell cycle progression. Contrary to ubiquitination, deubiquitination removes ubiquitin from targeted protein to maintain its stability and thus regulate cellular homeostasis. Ubiquitin-Specific Protease 12 (USP12) belongs to the biggest family of deubiquitinases named ubiquitin-specific proteases and has been reported to be correlated with various pathophysiological processes. In this review, we initially introduce th

Clinical and genetic analysis of familial neuromyelitis optica spectrum disorder in Chinese: associated with ubiquitin-specific peptidase USP18 gene variants.

BACKGROUND: Familial clustering of neuromyelitis optica spectrum disorder (NMOSD) was present in Chinese. This study was to investigate the clinical characteristics and genetic background of familial NMOSD. METHODS: Through questionnaires in four medical centres in 2016-2020, we identified 10 families with NMOSD aggregation. The statistical differences of clinical characteristics between familial and sporadic NMOSD (22 cases and 459 cases) were summarised. The whole-exome sequencing (WES) for seven families (13 cases and 13 controls) was analysed, compared with our previous WES data for spora

[The role of the ubiquitin-proteasome system in the development of acute myocardial injury. Case report].

BACKGROUND: Acute myocardial injury (AMI) is a universal pathological process that complicates both cardiac and non-cardiac pathologies. However, understanding of the molecular mechanisms underlying the various types of AMI remains insufficient. One of the current concepts for the development of AMI is the regulation of the ubiquitin-proteasome system (UPS), which mediates apoptosis and the adaptive response to injury. However, the patterns of its activation in the myocardium in various clinical situations remain poorly understood. AIM: To evaluate and compare the expression of ubiquitin and

USP18 regulates skeletal muscle regeneration independently of its deISGylation activity.

OBJECTIVES: USP18 is a multifunctional protein that attenuates type I interferon (IFN-I) signalling and regulates ISG15-mediated ISGylation. Although USP18 has been suggested to suppress myogenic differentiation in vitro, its role in muscle regeneration and inflammatory muscle disease remains unclear. This study aimed to define the spatial localization and functional contribution of USP18 and ISG15 in inflamed and regenerating muscle. METHODS: USP18 function in myogenesis was examined using inducible knockout and catalytic-inactive mutant muscle cell models. Spatial expression of USP18 and IS

USP10 in Neurological Disorders: Mechanistic Insights and Emerging Therapeutic Strategies.

Various cellular processes, such as DNA repair and signal transduction, are regulated through ubiquitination and deubiquitination. Dysregulation of ubiquitination cascade enzymes and deubiquitinating enzymes leads to various diseases. Among them, deubiquitinating enzymes have been shown to be closely associated with cancer, cardiovascular disease, and metabolic diseases. Recent studies have found that deubiquitinating enzymes play an important role in controlling neuronal fate, synaptic development, and maintaining normal nervous system function. USP10, a member of the deubiquitinating enzyme

Molecular basis of ubiquitin-specific protease 8 autoinhibition by the WW-like domain.

Ubiquitin-specific protease 8 (USP8) is a deubiquitinating enzyme involved in multiple membrane trafficking pathways. The enzyme activity is inhibited by binding to 14-3-3 proteins. Mutations in the 14-3-3-binding motif in USP8 are related to Cushing's disease. However, the molecular basis of USP8 activity regulation remains unclear. This study identified amino acids 645-684 of USP8 as an autoinhibitory region, which might interact with the catalytic USP domain, as per the results of pull-down and single-molecule FRET assays performed in this study. In silico modelling indicated that the regio

Role of UCH-L1/ubiquitin in acute testicular ischemia-reperfusion injury.

The most significant complication of testicular torsion is loss of the testis, which may lead to impaired fertility. Molecular mechanisms how spermatogenesis impairs owing to testicular torsion remain unknown. This investigation, by using mouse model of testicular torsion, was undertaken to gain insight into the cellular and molecular mechanism underlying torsion-induced germ cell loss. Male mice were subjected to 2h ischemia-inducing torsion, and testes were examined at 24, 48, and 72h after the repair of torsion (reperfusion). Ischemia-reperfusion (IR) of the testes resulted in germ cell, mo

An overview of BAP1 biological functions and current therapeutics.

BRCA1-associated protein 1 (BAP1) is a tumor suppressor gene that was first identified in 1998. Germline loss-of-function variants in BAP1 are associated with a tumor predisposition syndrome with at least four cancers: uveal melanoma (UM), malignant mesothelioma (MMe), renal cell carcinoma (RCC), and cutaneous melanoma (CM). Furthermore, somatic BAP1 mutations are important drivers for several cancers most notably UM, MMe, RCC, intrahepatic cholangiocarcinoma (ICC) and hepatocellular carcinoma (HCC). Emerging evidence substantiates the fundamental role of BAP1 in suppressing cancer initiation

USP25 inhibition ameliorates Parkinson's disease by restoring mitophagy.

Parkinson's disease (PD) is a progressive neurodegenerative disease that casts a significant shadow over global health and the identification of therapeutic targets for PD will empower more effective clinical treatment. The gene encoding the deubiquitinating enzyme USP25 has been identified as a susceptible locus for PD, but the role of USP25 in PD remains unknown. In this study, we found that USP25 exacerbated dopaminergic neuronal loss and motor deficits in murine models of PD by sabotaging the mitophagy machinery. USP25 physically interacted with the autophagy receptor optineurin and disrup
