Targeting the Ubiquitin-conjugating Enzyme for Oral Squamous Cell Carcinoma Therapy: Discovering Natural Inhibitors.

BACKGROUND: Oral Squamous Cell Carcinoma (OSCC) is a multiple-phase carcinogenic disease that concurrently involves malignant lesions, invasion, and metastasis. It has been reported that Ubiquitin-conjugating enzymes play a significant role in the progression of OSCC and other fatal cancers through the process of ubiquitination. Among them, UBE2D1 represents a promising target for therapeutic intervention. Strategies aimed at inhibiting UBE2D1 could restore the function of tumor suppressors, such as p53, and potentially enhance the effectiveness of existing cancer therapies. OBJECTIVE: This s

Carabrone alleviates neuroinflammation by covalently targeting ubiquitin conjugating enzyme UBE2D3 in experimental autoimmune encephalomyelitis.

Multiple sclerosis (MS) is an incurable and progressive inflammatory disease of the central nervous system (CNS) characterized by inflammatory demyelination, chronic axonal damage, and neurodegeneration. Carabrone (CA), a sesquiterpene lactone from the traditional Chinese herb Carpesium abrotanoides L., demonstrates diverse pharmacological properties. However, its therapeutic effects on MS and the underlying mechanisms remain unexplored. In the present study, we reported the beneficial effects of CA on experimental autoimmune encephalomyelitis (EAE), a well-established mouse model of MS, for t

UBE2O, a host ubiquitin-conjugating enzyme, is a key regulator of hepatitis B virus maturation and egress.

A critical step in Hepatitis B virus (HBV) maturation and egress is the ubiquitination of the capsid/core protein (HBc), which enables its recognition by the endosomal sorting complex required for transport (ESCRT) machinery and recruitment to multivesicular bodies (MVBs). This study investigates the role of UBE2O, an atypical E2 ubiquitin-conjugating enzyme with intrinsic E3 ligase activity, in nucleocapsid assembly and virion egress. Loss of UBE2O in HBV-infected primary human hepatocytes (PHH) and HepG2-NTCP cells led to a reduction in viral replication, as evidenced by decreased levels of

Ubiquitin-conjugating enzyme E2-25K increases aggregate formation and cell death in polyglutamine diseases.

Polyglutamine diseases are characterized by neuronal intranuclear inclusions of expanded polyglutamine proteins, which are also ubiquitinated, indicating impairment of the ubiquitin-proteasome system. E2-25K (Hip2), an ubiquitin-conjugating enzyme, interacts directly with huntingtin and may mediate ubiquitination of the neuronal intranuclear inclusions in Huntington disease. E2-25K could thus modulate aggregation and toxicity of expanded huntingtin. Here we show that E2-25K is involved in aggregate formation of expanded polyglutamine proteins and polyglutamine-induced cell death. Both a trunca

A human ubiquitin conjugating enzyme, L-UBC, maps in the Alzheimer's disease locus on chromosome 14q24.3.

We have identified a novel ubiquitin conjugating enzyme gene, L-UBC, which maps to human Chromosome (Chr) 14q24.3. This is also the location of the major early onset familial Alzheimer's disease gene (FAD3). L-UBC encodes a protein that demonstrates homology to the yeast ubiquitin conjugating enzyme, UBC-4, and human UbcH5. Their functions are to ubiquitinate specific proteins targeted for degradation. The protein also exhibits very strong homology to a rabbit protein, E2-F1, which mediates p53 degradation driven by papilloma virus E6 protein in vitro. The accumulation of specific proteins tha

UBC13 is an RNF213-associated E2 ubiquitin-conjugating enzyme, and Lysine 63-linked ubiquitination by the RNF213-UBC13 axis is responsible for angiogenic activity.

Moyamoya disease (MMD) is a cryptogenic vascular disorder in the intracranial arteries. RING protein 213 (RNF213) is the susceptibility gene for MMD, and encodes a RING domain and a Walker motif. Herein, we identified UBC13 (UBE2N) as an E2 ubiquitin-conjugating enzyme for RNF213 E3 ubiquitin ligase by yeast two-hybrid screening with a fragment containing RNF213 RING domain as bait, and the immunocomplex of RNF213-UBC13 was detected in vivo. Analysis of the ubiquitin chain on RNF213 by monitoring autoubiquitination showed that RNF213 was autoubiquitinated in a K63 chain fashion, but not in a K

Upregulation of ubiquitin-conjugating enzyme E2T (UBE2T) predicts poor prognosis and promotes hepatocellular carcinoma progression.

Reportedly, ubiquitin-conjugating enzyme E2T (UBE2T) is closely related to the progression of several malignancies. This work is aimed to probe the role of UBE2T in the progression of hepatocellular carcinoma (HCC) patients. The microarray analysis was executed to screen the differentially expressed genes (DEGs) in HCC tissues. The Cancer Genome Atlas (TCGA) and Gene Expression Profiling Interactive Analysis (GEPIA2) databases, PCR and immunohistochemistry were utilized to validate the dysregulation of UBE2T in HCC. Kaplan-Meier analysis was employed to determine the relationship between UBE2T

The ubiquitin-conjugating enzyme CDC34 is essential for cytokinesis in contrast to putative subunits of a SCF complex in Trypanosoma brucei.

The ubiquitin-proteasome system is a post-translational regulatory pathway for controlling protein stability and activity that underlies many fundamental cellular processes, including cell cycle progression. Target proteins are tagged with ubiquitin molecules through the action of an enzymatic cascade composed of E1 ubiquitin activating enzymes, E2 ubiquitin conjugating enzymes, and E3 ubiquitin ligases. One of the E3 ligases known to be responsible for the ubiquitination of cell cycle regulators in eukaryotes is the SKP1-CUL1-F-box complex (SCFC). In this work, we identified and studied the f

Aged monkey brains reveal the role of ubiquitin-conjugating enzyme UBE2N in the synaptosomal accumulation of mutant huntingtin.

Although misfolded proteins are ubiquitinated and cleared by the proteasome, they can accumulate in synapses in aged neurons to promote synaptic dysfunction in a variety of neurodegenerative diseases, including Huntington's disease (HD), which is caused by polyglutamine expansion in huntingtin. The mechanism behind this aging-related phenomenon is unknown and has been difficult to investigate using animals with short life spans. With brain tissues from longer-lived rhesus monkeys of different ages, we found that aging reduces ubiquitin-proteasomal activity and also increases the level of ubiqu

Burn injuries in rats upregulate the gene expression of the ubiquitin-conjugating enzyme E2(14k) in skeletal muscle.

Burn injuries are associated with muscle cachexia, which mainly reflects protein breakdown in the ubiquitin-proteasome pathway. Ubiquitination of proteins degraded by this mechanism is regulated by multiple enzymes, including the 14-kd ubiquitin-conjugating enzyme, E2(14k). In this study, burn injuries in rats resulted in increased levels of the 1.2 kilobase E2(14k) transcript in the white, fast-twitch extensor digitorum longus muscle with no changes or only minor changes in the red, slow-twitch soleus muscle, liver, and kidney. The results provide the first evidence that burn injuries upregul

Rice ubiquitin-conjugating enzyme OsUbc13 negatively regulates immunity against pathogens by enhancing the activity of OsSnRK1a.

Ubc13 is required for Lys63-linked polyubiquitination and innate immune responses in mammals, but its functions in plant immunity still remain largely unknown. Here, we used molecular biological, pathological, biochemical, and genetic approaches to evaluate the roles of rice OsUbc13 in response to pathogens. The OsUbc13-RNA interference (RNAi) lines with lesion mimic phenotypes displayed a significant increase in the accumulation of flg22- and chitin-induced reactive oxygen species, and in defence-related genes expression or hormones as well as resistance to Magnaporthe oryzae and Xanthomonas

Role of Ubiquitin-conjugating enzyme E2 (UBE2) in two immune-mediated inflammatory skin diseases: a mendelian randomization analysis.

Psoriasis vulgaris (PV) and Atopic dermatitis (AD) are the two major types of immune-mediated inflammatory skin disease (IMISD). Limited studies reported the association between Ubiquitin-conjugating enzyme E2 (UBE2) and IMISD. We employed a two-sample Mendelian randomization (MR) study to assess the causality between UBE2 and PV & AD. UBE2 association genome-wide association study (GWAS) data were collected. The inverse variance weighted (IVW) method was utilized as the principal method in our Mendelian randomization (MR) study, with additional using the MR-Egger, weighted median, simple mode

Analysis of Ubiquitin-Conjugating Enzyme E2T (UBE2T) Protein Levels in the Bone Marrow Biopsy Specimens of Patients With Multiple Myeloma.

Introduction Several studies have reported that the ubiquitin-conjugating enzyme E2 T (UBE2T) is overexpressed in multiple myeloma (MM). This has been assessed via bioinformatic analysis, which represents mRNA expression. However, biological experimental evidence is lacking. Moreover, the protein levels of UBE2T in tissue obtained via bone marrow biopsy are not clear. To identify newer potential therapeutic targets for MM, this study aimed to assess the expression of the UBE2T protein by immunohistochemical (IHC) staining of bone marrow biopsy specimens and also collected clinical data for a c

Immunoprotective effect of recombinant ubiquitin-conjugating enzyme (UCE) and elongation factor 1-alpha (EF1α) in rabbits against Eimeria media.

Eimeria media, one of the primary rabbit coccidia, causes rabbits diarrhea, dehydration, and hindered growth, leading to economic losses. Anticoccidial drugs are the current primary ways of preventing and controlling rabbit coccidiosis. However, increasing drug resistance has become a serious concern, and vaccine research and application for E. media remain limited, with current efforts mainly focused on precocious strains. To screen candidate antigens for a recombinant subunit vaccine against E. media, this study expressed ubiquitin-conjugating enzyme (UCE) and elongation factor 1-alpha (EF1α

Ubiquitin-conjugating Enzyme E2M Destabilizes Heat Shock Protein 70 and Mediates Sleep Deprivation-Induced Cognitive Impairment in Mice.

Sleep deprivation (SD) contributes to cognitive impairment. Heat shock protein 70 (HSP70) protects against SD-induced neuroinflammation and cognitive decline; however, how endogenous HSP70 is regulated under SD conditions, and in particular whether it is controlled by post-translational modifications such as ubiquitination, remains unclear. The present study examined whether the ubiquitin-conjugating enzyme E2M (UBE2M) governs HSP70 protein stability and thereby contributes to SD-related inflammatory and cognitive outcomes. A mouse model of SD was established using the modified multiple-platfo

African swine fever virus ubiquitin-conjugating enzyme pI215L inhibits IFN-I signaling pathway through STAT2 degradation.

African swine fever virus (ASFV) is the causative agent of one of the most lethal diseases affecting domestic pig and wild boar, which is endangering the swine industry due to its rapid expansion. ASFV has developed different mechanisms to evade the host immune response, including inhibition of type I IFN (IFN-I) production and signaling, since IFN-I is a key element in the cellular antiviral response. Here, we report a novel mechanism of evasion of the IFN-I signaling pathway carried out by the ASFV ubiquitin-conjugating enzyme pI215L. Our data showed that pI215L inhibited IFN-stimulated resp

Mind the Metal: A Fragment Library-Derived Zinc Impurity Binds the E2 Ubiquitin-Conjugating Enzyme Ube2T and Induces Structural Rearrangements.

Efforts to develop inhibitors, activators, and effectors of biological reactions using small molecule libraries are often hampered by interference compounds, artifacts, and false positives that permeate the pool of initial hits. Here, we report the discovery of a promising initial hit compound targeting the Fanconi anemia ubiquitin-conjugating enzyme Ube2T and describe its biophysical and biochemical characterization. Analysis of the co-crystal structure led to the identification of a contaminating zinc ion as solely responsible for the observed effects. Zinc binding to the active site cystein
