Observations on the fine structure, enzyme histochemistry, and innervation of parathyroid gland and ultimobranchial body of Chthonerpeton indistinctum (Gymnophiona, Amphibia).

Fine structural and enzyme histochemical observations on ultimobranchial body and parathyroid gland of the caecilian Chthonerpeton are presented. The cell clusters and follicles of the ultimobranchial body consist mainly of granulated cells which are termed C-cells and obviously belong to the APUD cell series. In the larger follicles additional possibly exhausted degranulated cells and replacement cells occur. A rich supply of nerve fibres has been found in this gland. Frequently nerve terminals were observed to come into synaptic contact with the C-cells. Two categories of nerve fibres occur:

FRS2alpha is required for the separation, migration, and survival of pharyngeal-endoderm derived organs including thyroid, ultimobranchial body, parathyroid, and thymus.

The docking protein FRS2alpha plays an important role in fibroblast growth factor (FGF)-induced intracellular signal transduction by linking FGF receptors (FGFRs) to a variety of intracellular signaling pathways. In FRS2alpha(2F/2F) mutant mice at embryonic day (E)18.5, in which the Shp2-binding sites of FRS2alpha were disrupted, the thyroid glands were aplastic or hypoplastic. C cells were absent or present in low numbers and rarely formed a compact mass of cells. Parathyroid glands were mostly connected to thymus tissues. At E10.5, the formations of pharyngeal pouches and thyroid primordium

Mash1 regulates the development of C cells in mouse thyroid glands.

In mammals, the ultimobranchial body derived from the fourth pharyngeal pouch gives rise to thyroid C cells. The C cells of newborn mice are immunoreactive for calcitonin, calcitonin gene-related peptide (CGRP), protein gene product (PGP) 9.5 and NeuroD, and transiently exhibit the neuronal markers TuJ1 and somatostatin during fetal development. The basic helix-loop-helix (bHLH) transcription factor Mash1 plays a role in the differentiation of autonomic neurons. We show that in wild-type mouse embryos, Mash1 is expressed in the ultimobranchial body at embryonic day (E) 12.5, when the body is l

Follicular cell lineage in persistent ultimobranchial remnants of mammals.

It has been a subject of much debate whether thyroid follicular cells originate from the ultimobranchial body, in addition to median thyroid primordium. Ultimobranchial remnants are detected in normal dogs, rats, mice, cattle, bison and humans and also in mutant mice such as Eya1 homozygotes, Hox3 paralogs homozygotes, Nkx2.1 heterozygotes and FRS2α2F/2F. Besides C cells, follicular cell lineages immunoreactive for thyroglobulin are located within these ultimobranchial remnants. In dogs, the C cell complexes, i.e., large cell clusters consisting of C cells and undifferentiated cells, are prese

Innervation of the C cells of chicken ultimobranchial glands studied by immunohistochemistry, fluorescence microscopy, and electron microscopy.

Innervation of the ultimobranchial glands in the chicken was investigated by immunohistochemistry, fluorescence microscopy and electron microscopy. The nerve fibers distributed in ultimobranchial glands were clearly visualized by immunoperoxidase staining with antiserum to neurofilament triplet proteins (200K-, 150K- and 68K-dalton) extracted from chicken peripheral nerves. The ultimobranchial glands received numerous nerve fibers originating from both the recurrent laryngeal nerves and direct vagal branches. The left and right sides of the ultimobranchial region were asymmetrical. The left ul

Spectrum of lesions derived from branchial arches occurring in the thyroid: from solid cell nests to tumors.

There is a group of lesions in the head and neck region derived from branchial arches and related structures which, when inflamed, are characterized by the formation of cysts lined by squamous or glandular epithelium and surrounded by a heavy inflammatory infiltrate rich in germinal centers. In the thyroid, the main source of various structures which may cause diagnostic dilemma is the ultimobranchial body. To investigate the spectrum of such thyroid lesions, the consultation files were reviewed for thyroid samples containing pathological structures regarded to arise from the ultimobranchial b

The development of pharyngeal endocrine organs in mouse and chick embryos.

Some interesting differences exist between development of the pharyngeal endocrine organs in mouse and chicken. In both, the thyroid forms as an evagination of the pharyngeal floor in the midline at the level of pharyngeal arch II and moves caudally to the base of the neck. In the chicken, the thyroid divides to form paired organs whereas in the mouse it forms a moustache-shaped bar that later fuses with the parathyroid and ultimobranchial bodies. The thymi, parathyroids and ultimobranchial bodies form as lateral evaginations of the pharyngeal pouches. The chick forms two pairs of thymi and pa

The carotid body of the pheasant (Phasianus colchicus L.). An electron microscopy study.

In the pheasant, the carotid body lies laterally to the common carotid artery, is partly surrounded by parathyroid tissue and is immediately adjacent to the ultimobranchial body. In some cases these tissues intermingle. The cellular component of the carotid body consists primarily of granular endocrine cells, which are arranged in islands and lie in immediate proximity to wide capillaries and nerve fibres. A quantitative evaluation of granule size frequencies in these main cells, which we divided into three types, showed, inter alia, that the types represented three stages of cell differentiat

Cellular and molecular events on the development of mammalian thyroid C cells.

Thyroid C cells synthesize and secrete calcitonin, a serum calcium-lowering hormone. This review provides our current understanding of mammalian thyroid C cells from the molecular and morphological perspectives. Several transcription factors and signaling molecules involved in the development of C cells have been identified, and genes expressed in the pharyngeal pouch endoderm, neural crest-derived mesenchyme in the pharyngeal arches, and ultimobranchial body play critical roles for the development of C cells. It has been generally accepted, without much-supporting evidence, that mammalian C c

DiGeorge's or the III-IV pharyngeal pouch syndrome: pathology and a theory of pathogenesis.

Six new cases of the III-IV pharyngeal pouch syndrome were encountered at autopsy among 897 consecutive pediatric autoposies. All occurred in patients with conotruncal cardiac anomalies. The anatomic characteristics of the heart defect suggest a possible mechanism for the pathogenesis of this developmental anomaly. It is postulated that premature involution of the thyroidea ima artery, which is the principal embryonic blood source to the 3rd and 4th pharyngeal pouches and the ultimobranchial body, may be the critical event in the embryogenesis of the syndrome. Hemodynamic alterations in the co

C-cell differentiation in the wall of an aberrant ultimobranchial sinus in the thyroid gland of an old rat.

BACKGROUND: In mammals, the thyroid gland possesses two types of endocrine cells, follicular cells and C cells, which have different functions but share a similar endodermal origin (although from different regions of the primitive pharynx). Specifically, follicular cells derive from the ventral pharyngeal floor, while C cells derive from the fourth pair of pharyngeal pouches through the ultimobranchial bodies (UBBs). Disruptions to human midline thyroid morphogenesis are relatively frequent and known as thyroid dysgenesis, which is the leading cause of congenital hypothyroidism. In contrast, f

Expression of the epithelial marker E-cadherin by thyroid C cells and their precursors during murine development.

Studies of chick-quail chimeras have reported that avian ultimobranchial C cells originate from the neural crest. It has consequently been assumed, without much supporting evidence, that mammalian thyroid C cells also originate from the neural crest. To test this notion, we employed both Connexin43-lacZ and Wnt1-Cre/R26R transgenic mice, because their neural crest cells can be marked. We also examined the immunohistochemical expression of a number of markers that identify migratory or postmigratory neural crest cells, namely, TuJ1, neurofilament 160, nestin, P75NTR, and Sox10. Moreover, we exa

Proneural gene Mash1 (Ascl1) is expressed in multiple lineages and regulates their differentiation and specification.

Ascl1 (Mash1), a bHLH transcription factor, is widely expressed by neuronal progenitors. The gene plays a key role in the differentiation of the autonomic nervous system, i.e., sympathetic, parasympathetic, and enteric ganglia; all of which are derived from neural crest cells. In Ascl1-null mutants, defective development of these ganglia is induced. The differentiation of neuroendocrine cells, including the carotid body, ultimobranchial body and thyroid C cells, the neuroepithelial body in the lung, and the adrenal medulla, is also controlled by Ascl1. Although the carotid body glomus cells an

The ultimobranchial gland and congenital thyroid abnormalities in man.

This study was carried out to investigate the fate of the ultimobranchial contribution to the thyroid in man when the thyroid lobe is not fully descended. Thyroid abnormalities were recorded in 41 of 29,000 autopsies at the London Hospital--a prevalence of 1.41/1000. The 18 major abnormalities included four cases of lingual thyroid, in two of which cystic structures were noted in the neck in the region of the upper parathyroid. Three further cases of lingual thyroid with upper parathyroid cysts were collected from other sources. The multilobulated cystic structures were up to 1.5 cm in diamete

Fluorescence microscopy of the avian (Gallus domesticus) carotid body.

Sterba's technique to detect neurosecretory substances was applied to carotid body tissue, surrounding branchial derivatives, ganglion nodosum and the carotid arterial wall. Mainly a yellowish-green, a yellowish-red and a bright yellow fluorescence was observed as well as greenish-yellow fibres with varicosities and terminal knobs. The metachromatic reaction of the technique was briefly discussed.
